Objective: The present study employed 5-aza-2'-deoxycytidine (5-Aza-CdR) to treat non-small cell lung cancer (NSCLC) cell line A549 to investigate the effects on proliferation and expression of the TFPI-2 gene. Methods: Proliferation was assessed by MTT assay after A549 cells were treated with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR, a specific demethylating agent, for 24, 48 and 72h. At the last time point cells were also analyzed by flow cytometry (FCM) to identify any change in their cell cycle profiles. Methylation-specific polymerase chain reaction (MSPCR), real time polymerase chain reaction(real-time PCR) and western blotting were carried out to determine TFPI-2 gene methylation status, mRNA expression and protein expression. Results: MTT assay showed that the growth of A549 cells which were treated with 5-Aza-CdR was significantly suppressed as compared with the control group (0 ${\mu}mol/L$ 5-Aza-CdR). After treatment with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h, FCM showed their proportion in G0/G1 was $69.7{\pm}0.99%$, $76.1{\pm}0.83%$, $83.8{\pm}0.35%$, $95.5{\pm}0.55%$ respectively (P<0.05), and the proportion in S was $29.8{\pm}0.43%$, $23.7{\pm}0.96%$, $15.7{\pm}0.75%$, $1.73{\pm}0.45%$, respectively (P<0.05), suggesting 5-Aza-CdR treatment induced G0/G1 phase arrest. MSPCR showed that hypermethylation in the promoter region of TFPI-2 gene was detected in control group (0 ${\mu}mol/L$ 5-Aza-CdR), and demethylation appeared after treatment with 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h. Real-time PCR showed that the expression levels of TFPI-2 gene mRNA were $1{\pm}0$, $1.49{\pm}0.14$, $1.86{\pm}0.09$ and $5.80{\pm}0.15$ (P<0.05) respectively. Western blotting analysis showed the relative expression levels of TFPI-2 protein were $0.12{\pm}0.01$, $0.23{\pm}0.02$, $0.31{\pm}0.02$, $0.62{\pm}0.03$ (P<0.05). TFPI-2 protein expression in A549 cells was gradually increased significantly with increase in the 5-Aza-CdR concentration. Conclusions: TFPI-2 gene promoter methylation results in the loss of TFPI-2 mRNA and protein expression in the non-small cell lung cancer cell line A549, and 5-Aza-CdR treatment could induce the demethylation of TFPI-2 gene promoter and restore TFPI-2 gene expression. These findings provide theoretic evidence for clinical treatment of advanced non-small cell lung cancer with the demethylation agent 5-Aza-CdR. TFPI-2 may be one molecular marker for effective treatment of advanced non-small cell lung cancer with 5-Aza-CdR.

시진핑정권 출범 후, 중국의 반부패운동은 18기 공산당중앙회를 거쳐 더 강화되고 있다. 많은 연구자들은 중국의 반부패운동이 일시적일 수도 있으며, 정권을 공고화하기 위한 수단으로 보여주기 정책이라고 하였다. 하지만 본 논문은 중국의 반부패운동이 한번의 일시적인 이벤트에 거치는 것이 아니라 시진핑정권 전반에 걸쳐 장기적인 통치수단으로 작용할 것으로 보고 있다. 본 논문에서 구체적으로 18기 중국공산당회의 이후 “호랑이”와 “파리”를 모두 잡는 반부패운동의 특징을 보여 주려고 구체적인 사건과 반부패의 ‘전형적 인물’들을 중심으로 반부패의 특징과 성격을 밝혀내고자 하였으며, 반부패운동이 지속될 것이라는 추이를 내놓았다. Since the 18th CPC National Congress, Chinese Communist Party Central Committee has been improving the Party’s style of work and strengthening the building of a clean government while a campaign in order to combat corruption is being carried out firmly, strongly and boldly to severely punish corruption , fighting both “tigers” and “flies” perseveringly, which reveals the confidence and determination of CPC to combat corruption and build a clean government. In this article, based on how to view the corruption in China under the new situation, the current corruption is analyzed objectively, furthermore, the current moves, achievements and features in combating corruption are taken apart profoundly, the coming difficulties of China in combating corruption are faced straightly, in the end, the routing of combating corruption and building a clean government in the future of China is thought rationally and penetratingly.
Orthogonal 방법을 통한 Poly(AM-DMDAAC)/MMT 고흡수성나노복합체 제조 연구

A novel poly(AM-DMDAAC)/MMT superabsorbent nanocomposites are prepared by radical polymerizationusing ammonium persulfate (APS) and anhydrous sodium sulfite as a free radical initiator and N,N-methylene bisacrylamide(MBA) as a crosslinker. In this paper, an optimization study on the synthesis of superabsorbent nanocompositesis carried out. Orthogonal array experiment indicates that the optimized conditions is acrylamide (AM) content 23 wt%,diallyl dimethyl ammonium chloride (DMDAAAC) content 6 wt%, montmorillonite (MMT) content 4 wt%, initiatorcontent 0.2 wt% and crosslinker content 0.02 wt%. Under the optimization syntheses conditions concluded, the maximumwater absorbency in distilled water is 659.53 g·g-1 and in 2 wt% sodium chloride solution is 116.25 g·g-1. Compared withthe range values of different factors (Rj), the order of significance factors in distilled water is C (MMT) > B (DMDAAC)> A (AM) > D (crosslinker) > E (initiator). MMT is intercalated during polymerization reaction and a nanocompositestructure is formed as shown by TEM analysis and XRD analysis.

Al-Cu-Mg aluminum alloy sheet is widely used for aircraft skin. Multi-stage forming combined with Solution Heat Treatment (SHT) is necessary for some geometrically complex productions in aircraft skin components. However, the effects of pre-strain and natural ageing time on the formability of Al-Cu-Mg aluminum alloy sheet are not fully understood. In this study, the deformation responses of an Al-Cu-Mg aluminum alloy sheet, over a range of pre-strain levels (0% to 6%) and natural ageing time levels (0 h to 2 h) were investigated. A modified Hollomon constitutive model coupling with pre-strain and natural ageing time was proposed to characterize the deformation responses of the Al-Cu-Mg aluminum alloy sheet. The hemispherical dome tests were performed at natural ageing time levels ranging from 0.25 h to 1.5 h. Marciniak and Kuczynski (M-K) theory was used to obtain the forming limit prediction model which takes pre-strain and natural ageing time into account. The theoretical Forming Limit Curves (FLCs) were compared with the experimental results.
Galectin-9 Acts as a Prognostic Factor with Antimetastatic Potential in Hepatocellular Carcinoma

Considerable research has been conducted concerning galectin-9 and carcinomas, but little information is available about any relation with the hepatocellular carcinoma. In this study, we employed a small interfering RNA (siRNA) targeting galectin-9 to down-regulate the expression in HepG2 cells. As a result, after galectin-9 expression was reduced, cell aggregation was suppressed, while other behaviour such as the proliferation, adhesion and invasion to ECM, cell-endothelial adhesion and transendothelial invasion of the cells were markedly enhanced. When tumors of 200 patients with hepatocellular carcinoma were tested for galectin-9 expression by immunohistochemistry, binding levels demonstrated intimate correlations with the histopathologic grade, lymph node metastasis, vascular invasion and intrahepatic metastasis (P<0.05). Moreover, survival analysis indicated that patients with galectin-9 expression had much longer survival time than those with negative lesions, and the Log-rank test indicated that this difference was statistical significant (P<0.0001). The Cox proportional hazards model suggested that negative galectin-9 expression in hepatocellular carcinoma represented a significant risk factor for patient survival. We propose that galectin-9 might be a new prognostic factor with antimetastatic potential in patients with hepatocellular carcinoma.
Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.
