        Phosphodiesterase inhibitor improves renal tubulointerstitial Hypoxia of the Diabetic rat Kidney

        Background/Aims: Renal hypoxia is involved in the pathogenesis of diabetic nephropathy. Pentoxifyllin (PTX), a nonselective phosphodiesterase inhibitor, is used to attenuate peripheral vascular diseases. To determine whether PTX can improve renal hypoxia, we investigated its effect in the streptozocin (STZ)-induced diabetic kidney. Methods: PTX (40 mg/kg, PO) was administered to STZ-induced diabetic rats for 8 weeks. To determine tissue hypoxia, we examined hypoxic inducible factor-1α (HIF-1α), heme oxygenase-1 (HO-1), vascular endothelial growth factor (VEGF), and glucose transporter-1 (GLUT-1) levels. We also tested the effect of PTX on HIF-1α in renal tubule cells. Results: PTX reduced the increased protein creatinine ratio in diabetic rats at 8 weeks. HIF-1α, VEGF, and GLUT-1 mRNA expression increased significantly, and the expression of HO-1 also tended to increase in diabetic rats. PTX significantly decreased mRNA expression of HIF-1α and VEGF at 4 and 8 weeks, and decreased HO-1 and GLUT-1 at 4 weeks. The expression of HIF-1α protein was significantly increased at 4 and 8 weeks in tubules in the diabetic rat kidney. PTX tended to decrease HIF-1α protein expression at 8 weeks. To examine whether PTX had a direct effect on renal tubules, normal rat kidney cells were stimulated with CoCl2 (100 μM), which enhanced HIF-1α mRNA and protein levels under low glucose conditions (5.5 mM). Their expressions were similar even after high glucose (30 mM) treatment. PTX had no effect on HIF-1α expression. Conclusions: PTX attenuates tubular hypoxia in the diabetic kidney.

      • 분리된 뇌조직의 수초화세포에 관한 연구

        중추신경계통의 신경세포들은 긴 축삭돌기로서 다른 신경세포와 연접을 이루기 때문에 뇌손상시 나타나는 반응도 신경로의 방향과 관계가 깊다. 대부분의 긴 축삭은 수초집으로 둘러싸여 보호를 받고 있고 또 수초는 축삭의 대사활동에 중요한 기능을 발휘한다. 뇌가 손상받아 축삭들이 절단될 때 퇴행되는 축삭을 둘러싼 수초집의 다양한 변화는 손상부위의 괴사과정, 치유과정 및 부분적 재생기능에 중요한 요소가 된다. 본 실험에서는 뇌속에 주위의 다른 신경세포와 의 연결이 단절된 뇌조직의 기둥을 만들고 그속에서 일어나는 형태학적 반응 가운데서 특히 수초구조물의 특수한 반응을 연구하였다. 3Kg 정도의 고양이를 nembutal로 마취시키고 뇌정위고정대에 고정시킨 다음 전두골의 일부를 절개한 후 직경 5mm 크기의 원통형칼날을 내려 눌러서 뇌조직의 기둥이 주변으로 부터 분리되도록 했다. 이때 부위선택은 뇌지도를 이용하여 중간줄무늬체동맥이 아래로 부터 올라오는 것이 손상되지 않도록 해서, 분리된 뇌조직이 계속하여 혈액공급을 받을 수 있도록 하였다. 3~4주동안 생존 시킨 후 1% glutaraldehyde 와 1% paraformaldehyde 용액으로 뇌를 고정시켰고 일반적인 술식으로 표본을 작성하여 전자현미경으로 관찰하였다. 관찰 결과 손상된 줄무늬체기저핵 안에서는 많은 수초구주물들이 불규칙한 모습을 지녔으며, 특히 수초집이 팽되되고 심하게 쭈그러드는 것들이 많았으나 이때도 수초의 기본구조는 흐트러지지 않고 정상에 가까운 것이 많았다. 이는 비록 절단된 축삭이 괴사하여 소멸되더라도, 이를 둘러싼 수초집을 만들고 유지해 온 희돌기아교세포가 건재하기 때문에 아직도 기본구조를 유지한 것으로 생각되었다. 특히 전체적인 모양 심하게 변하는 것은 희돌기아교세포가 오히려 활성화되어 새로운 변형을 시도하는 것으로 생각되며, 그 대표적인 예로서 여유분의 수초집이 신경종말을 비롯한 다른 구조물을 흡인하거나 둘러싸는 현상을 보였다. 가장 특이한 경우로는 거대화된 수초집 속에 상당수의 세포들이 들어 있는 수초화세포의 출현이었다. 이들 세포의 성분을 확정하기는 어려우나 형태학적 특징과 세포의 이동특성, 포식형태등으로 볼때 이 세포들이 별아교세포 성분인 것으로 판단하였다. 이와 같은 별아교세포의 수초화는 이 세포들이 손상받은 뇌조직이 치유과정에서 수초집 안팎의 섬유화를 유도하고, 새로이 재생되려는 축삭의 자라나갈 자리를 차지하므로써 손상된 축삭의 재생을 막는 원인이 될 수 있다고 결론지었다. Since the neurones in the central nervous system make contact with other neurones through their long axons, the tissue reactions following brain damage are complicate, and the reactions appear largely along the course of transected axons. Myelin sheaths around transected axons have to show curious reactions. because they belong to oligodendroglia instead of belong to component. In this experiment, a tissue column of fundus striati nucleus was made by pressing a tube-knife of 5 mm diameter down toward the basal forebrain area, where the middle striatal arteries penetrate the brain. Three to four weeks after the stereotaxic isolation, tissue blocks are punched out from fundus striati nucleus. Ultrathin sections are obtained through the observations are discussed and the following results are obtained. Many axons and their myelin sheaths showed various transformations, including the redundant myelin sheaths and myelinated cells. Many redundant myelin sheaths are wrapped around, or sucked in the axon terminals or other structures. Redundant myelin sheath should be very active structures, even after the degeneration of axon within it, since the myelin sheath is a component of oligodendroglia instead of axonal component. Myelinated cells, the cells dwelled within some giant-swelled myelin sheaths, are observed. They contained relatively light cytoplasm with scanty cell other cell components or axon terminals, within the giant myelin capsule. The structural characteristics and the moldability into the myelin sheath, of these myelinated cells suggested that they are activated astroglial cells. Astroglial activities within or out of the transected myelin may prohibit the new axonal sprouts growing into the myelin sheath during regeneration course of nerve fiber in the central nervous system.

      • 유기질비료 시비량에 따른 오미자 묘목의 초기 생육특성에 관한 연구

        This study was carried out to have basic and applied informations to find the optimum nitrogen fertilization level for organic cultivation and to investigate early growth characteristics of Schizandra chinensis Baillon. Different application levels of the organic fertilizer were based on the recommended rate of N fertilizer. Five different levels of the organic fertilizer were applied to S. chinensis and early growth responses such as the length of vine, the diameter of vine, the number of leaves, and the number of branches were investigated. Lengths of main and total vines were shown longer at the level of 1.0 time organic fertilizer than those at others. The vine length at early growth was the highest at the level of 1.0 time organic fertilizer among the treatments. Stem diameter was shown thicker level of 1.0 time organic fertilizer than others on August and September, whereas on October it was the thickest in the conventional cultivation. Number of leaf was increased higher at the level of 1.0 and 2.0 times organic fertilizer than that in the compared to conventional. The number of branches was appeared the lowest at the level of 1.0 time organic fertilizer at early growth, however they were increased through progressing their growth stages. They were not significantly difference among the treatments except for the level of 2.0 times organic fertilizer. Based on the research results, the growth of S. chinensis was commonly higher at the level of 1.0 time organic fertilizer compared to other treatments.

      • 비젼 제어시스템에 사용된 카메라의 최적개수에 대한 실험적 연구

        The vision system model used for this study involves the six parameters that permits a kinds of adaptability in the relationship between the camera space location of manipulable visual cues and the vector of robot joint coordinates is estimated in real time. Also this vision control method requires the number of cameras to transform 2-D camera plane from 3-D physical space, and be used irrespective of location of cameras, if visual cues are displayed in the same camera plane. Thus, this study is to investigate the optimal number of cameras used for the developed vision control system according to the change of the number of cameras. This study is processed in the two ways : a) effectiveness of vision system model b) optimal number of cameras c) These results show the evidence of the adaptability of the developed vision control method using the optimal number of cameras.

      • 단클론성 IgG 침착을 동반한 증식성 사구체 신염 1예

        Proliferative glomerulonephritis with monoclonal IgG deposits is a novel phenotype of glomerular injury with the diagnostic criteria of (1) the presence of glomerular monoclonal IgG deposits restricted to a single IgG subclass and a single light chain isotype, associated with endocapillary proliferative, membranoproliferative, or membranous features; (2) the presence of granular("immune complex type") deposits by electron microscopy; and (3) the absence of clinical and laboratory evidence of cryoglobulinemia. We report a case of 65-year-old man with generalized edema who was diagnosed as proliferative glomerulonephritis with monoclonal IgG deposits by renal biopsy.

      • Propylthiouracil에 의한 중증 급성간염 1예

        PTU에 의한 간염의 발생은 매우 드물게 발생하는 것으로 알려져 있으나 저자들은 Graves씨 병으로 진단 받고 propylthiouracil 투여를 받고있던 환자에서 중증의 급성 간염이 발생한 1예를 경험하였기에 보고하는 바이다. Propylthiouracil is widely used to treat patients with hyperthyroidism. This drug has been associated with severe hepatotoxicity rarely. We presented the case of jaundice and profound liver dysfunction from a 23-year old woman treated with propylthiouracil for hyperthyroidism. Viral, metabolic, and autoimmune liver disease could be excluded and liver biopsy revealed a pattern of acute hepatitis. After discontinuing the drug, there was a progressive resolution of hepatic symptoms and decrease in biochemical data of the liver. Despite propylthiouracil induced hepatitis in rare case, patients receiving propylthiouracil are exposed to develope severe hepatotoxicity. Therefore it might be advisable to monitor level of the transaminase on regular base from patients receiving propylthiouracil.
