        Coenzyme Q10: a progress towards the treatment of neurodegenerative disease

        Coenzyme Q10 (CoQ10, or ubiquinone) is an electron carrier of the mitochondrial respiratory chain (electron transport chain) with antioxidant properties. In view of the involvement of CoQ10 in oxidative phosphorylation and cellular antioxidant protection a deficiency in this quinone would be expected to contribute to disease pathophysiology by causing a failure in energy metabolism and antioxidant status. Indeed, a deficit in CoQ10 status has been determined in a number of neuromuscular and neurodegenerative disorders. Primary disorders of CoQ10 biosynthesis are potentially treatable conditions and therefore a high degree of clinical awareness about this condition is essential. A secondary loss of CoQ10 status following HMG-CoA reductase inhibitor (statins) treatment has been implicated in the pathophysiology of the myotoxicity associated with this pharmacotherapy. CoQ10 and its analogue, idebenone, have been widely used in the treatment of neurodegenerative and neuromuscular disorders. These compounds could potentially play a role in the treatment of mitochondrial disorders, Parkinson’s disease, Huntington’s disease, amyotrophic lateral sclerosis, Friedreich’s ataxia, and other conditions which have been linked to mitochondrial dysfunction. This article reviews the physiological roles of CoQ10, as well as the rationale and the role in clinical practice of CoQ10 supplementation in different neurological diseases, from primary CoQ10 deficiency to neurodegenerative disorders. These will help in future for treatment of patients suffering from neurodegenerative disease.

        Development of male sterile transgenic lines in rice by tapetum specific expression of barnase gene

        The key to development of barnase-barstar transgene based hybrid seed technology is the availability of tightly regulated tapetum specific promoter, as any leaky expression of the barnase gene leads to several unintended effects. In the present study, we used two different tapetum specific promoters i.e. promoter of the RTS gene isolated from rice cultivar IR64 and the OsG6b promoter from japonica rice cultivar Hayayuki to express the barnase gene in rice transgenic lines. While viable male sterile transgenic lines could not be obtained with RTS promoter we could develop single copy male sterile lines when the barnase gene was expressed under the OsG6b promoter.

        A prospective study evaluating impact on renal function following percutaneous nephrolithotomy using Tc99m ethylenedicysteine renal scan: Does multiplicity of access tracts play a role?

        Purpose: A prospective study evaluating impact of percutaneous nephrolithotomy (PCNL) on renal function following PCNL using ethylenedicysteine (EC) renal scan. Does multiplicity of access tracts play a role?Materials and Methods: A prospective observational study was done and patients undergoing PCNL for renal calculi at our urology department were selected. Renal Tc99m EC scan was done pre-surgery and at 3 months follow-up. An assessment was done on decline in glomerular filtration rate (GFR) in postoperative period based on number of access tracts required. Results: A total of 110 patients were enrolled in the study. The total number of punctures was 170 with 141 being supra-costal puncture and 29 infra-costal. The total number of single punctures were 60 and classified as Group I whereas patients with double puncture and triple puncture (40 and 10 patients, respectively) were classified as Group II. Mean postoperative split renal function (in Tc99m EC scans) of patients of Group I (40.93±19.62%) was found to be higher than that of Group II (32.82±16.98%). Mean change (decline) in GFR for single, double and triple tracts were 2.68 mL/min, 3.80 mL/min, and 4.2 mL/min, respectively. Conclusions: PCNL used for stone removal can improve renal function by eradicating obstruction; however, this procedure may itself negatively impact the functions of the targeted kidney. Our study showed post PCNL decrease in GFR which worsens with an increasing number of accesses.

        Remittances and Household Expenditure Patterns of Selected Families in Kathmandu Valley

        This paper made an attempt to study the household expenditure patterns of remittance receiving families in Kathmandu. A field survey was conducted in order to see the contribution of remittance for improving the livelihood of the respondents` family. The factors incorporated in the study were; basic consumption, education, health, entertainment, saving pattern, building a new house and repayment of loan expenses. Non-probability convenient sampling technique was used and a structured questionnaire was distributed to collect the information from the respondents in Kathmandu. The study depicted that the highest portion of the remittance was used on purchasing food expenses and for the education expenses of the children. Other portion of it was used for health expenses, saving; repayment of the loan and etc. The analysis predicts that remittance-recipient households experienced a significant effect of remittance on the household`s consumption, education and health expenses as well as saving which leads to improve the wellbeing of the family.

        Major Challenges of Public Governance and Public Affairs in India

        The challenges for public governance and public affairs in India are multitudinous. The challenge is that of reincarnating governance from a process structure to the result structure. Multiple stakeholders have sneaked into policy formulation. This article deals with several challenging issues including employment, food, education, the Direct Benefit Transfer, electoral reforms, poverty, corruption, decentralization and empowerment, service delivery, and institutional developments. The bureaucracy, which is cracking under the heavy burden of delivery on the one hand and the lack of resources on the other hand, has led to a stagnation of delivery of services or a decline in the quality standard of service. Accordingly, this paper explores the importance and challenges of public administration from a governance perspective and highlights how governance structures and administrative culture have been altered to deliver better services to citizens in India and also highlights what needs to be done to overcome the problems in the process of governance whereby efficiency and effectiveness of both policy formulation and policy execution can be made more in sync with the changing times.

        Techniques for Formulation of Nanoemulsion Drug Delivery System: A Review

        Nanoemulsion drug delivery systems are advanced modes for delivering and improving the bioavailability of hydrophobic drugs and the drug which have high first pass metabolism. The nanoemulsion can be prepared by both high energy and low energy methods. High energy method includes high-pressure homogenization, microfluidization, and ultrasonication whereas low energy methods include the phase inversion emulsification method and the self-nanoemulsification method. Low energy methods should be preferred over high energy methods as these methods require less energy, so are more efficient and do not require any sophisticated instruments. However high energy methods are more favorable for food grade emulsion as they require lower quantities of surfactant than low energy methods. Techniques for formulation of nanoemulsion drug delivery system are overlapping in nature, especially in the case of low energy methods. In this review, we have classified different methods for formulation of nanoemulsion systems based on energy requirements, nature of phase inversion, and self-emulsification.

        Vibration upshot of operating mechanical sewing machine

        Background: Most of the people associated with tailoring occupation in Nepal are still using mechanical sewing machine as an alternative of new technology for tailoring. Common peroneal nerves of both right and left legs are exposed to strenuous and chronic stress exerted by vibration and paddling of mechanical sewing machine. Methods: The study included 30 healthy male tailors and 30 healthy male individuals. Anthropometric variables as well as cardio respiratory variables were determined for each subject. Standard Nerve Conduction Techniques using constant measured distances were applied to evaluate common peroneal nerve (motor) in both legs of each individual. Data were analyzed and compared between study and control groups using Man Whitney U test setting the significance level p ≤ 0.05. Results: Anthropometric and cardio respiratory variables were not significantly altered between the study and control groups. The Compound muscle action potential (CMAP) latency of common peroneal nerves of both right [(11.29 ± 1.25 vs. 10.03 ± 1.37), P < 0.001] and left [(11.28 ± 1.38 vs. 10.05 ± 1.37), P < 0.01] legs was found to be significantly prolonged in study group as compared to control group. The Amp-CMAP of common peroneal nerves of both right [(4.57 ± 1.21 vs. 6.22 ± 1.72), P < 0.001] and left [(4.31 ± 1.55 vs. 6.25 ± 1.70), P < 0.001] legs was found significantly reduced in study group as compared to control group. Similarly, the motor nerve conduction velocity (MNCV) of common peroneal nerves of both right [(43.72 ± 3.25 vs. 47.49 ± 4.17), P < 0.001] and left [(42.51 ± 3.82 vs. 46.76 ± 4.51), P < 0.001] legs was also found to be significantly reduced in study group in comparison to control group. Conclusion: Operating mechanical sewing machine by paddling chronically and arduously could have attributed to abnormal nerve conduction study parameters due to vibration effect of the machine on right and left common peroneal nerves. The results of present study follow the trend towards presymptomatic or asymptomatic neuropathy similar to subclinical neuropathy.

        Photo Physical Studies of PVP Arrested ZnS Quantum Dots

        Monodispersed polyvinylpyrrolidone (PVP) arrested ZnS quantumdots (QDs) having diameter in range ~2-5 nm are synthesized by acolloidal precipitation method using PVP as the stabilizing agent. X-ray diffraction (XRD), transmission electron microscopy (TEM),high resolution transmission electron microscopy (HRTEM),selective area electron diffraction (SAED) and Fourier transforminfrared (FT-IR) spectroscopy are probed to investigate thestructural information. The optical properties are studied usingdiffuse UV-visible reflectance and photoluminescence (PL)spectroscopy techniques. TEM images as well as XRD reflectionpeak broadening indicate the nanometer size particles formationwith cubic (sphalerite) phase within the polymer matrix. Opticalabsorbance studies reveal an excitonic peak at around ~310 nmdictates the effect of quantum confinement effect in the ZnS QDs. PL emission spectra for ZnS QDs in PVP exhibit four emissionpeaks at ~382 nm, ~414 nm, ~480 nm and ~527 nm are observed. These excitonic emissions from ZnS QDs are caused by theinterstitial sulfur/Zn vacancies and surface states.
