      • Brine Shrimp Bioassay를 이용한 해양생물의 세포독성검색

        As a part of chemical study on the bioactive metabolites from marine organisms, we have investigated cytotoxicity using brine shrimp bioassay for each solvent fractions of the marine algae(12 species), marine sponges(3 species), coelenterates(2 species), echinoderms(4 species), marine molluscs(17 species), and ascidians(2 species), respectively. As the results, chloroform extract of Stichopus japonicus (LC_50 : 274 ㎍/ml), ethyl acetate extract of Anthocidaris crassispina(LC_50 : 121 ㎍/ml), n-butanol extract of Undaria pinnatifida (LC_50 : 178 ㎍/ml), and water extract of Thais clavigera (LC_50 : 61 ㎍/ml), displayed the most significant cytotoxic activity against brine shrimp. Among the marine organisms tested, echinoderms and marine molluscs were thought to be the most active Phylums on screening of new bioactive compounds.

      • 소아에서 성장호르몬 결핍증 진단을 위한 혈중 IGF-1과 IGFBP-3의 절단값

        Background : It is essential to have reference criteria for the diagnosis of growth hormone deficiency in children. The objective of study was to decide the cut-off value for Insulin-like Growth Factor-Ⅰ(IGF-Ⅰ) and Insulin-like Growth Factor Binding Protein-3(IGFBP-3) for diagnosis of growth hormone deficiency. Methods : We studied the serum level of IGF-Ⅰ and IGFBP-3 in 231 subjects(131 boys and 100 girls) with idiopathic short stature(ISS) and 37 subjects(23 boys and 14 girls) with complete or partial growth hormone deficiency(GHD). IGF-Ⅰ were measured by radioimmunoassay(RIA) method and IGFBP-3 were measured by immnoradiometric assay(IRMA) method. We conducted chi-square test, Fisher's exact test, student t-test, receiver operating characteristic(ROC) curve and discriminant anaylsis using MedCalc and SPSS Program. Results : The ROC curves showed that the best IGF-Ⅰ and IGFBP-3 cut-off value. Cut-off value of serum IGF-Ⅰ concentration was 346ng/mL(sensitivity 86.49%, specificity 85.71%) and of serum IGFBP-3 concentration was 3196ng/mL(sensitivity 81.82%, specificity 81.66%). If discriminant value (Z=0.442+0.661×IGF-Ⅰ+0.469×IGFBP-3) was below -1.47, the subject is discriminated GHD respectively. Conclusion : Our study results support that the measurement of serum IGF-Ⅰ and IGFBP-3 concentration might be an essential supplementary tools in the diagnostic evaluation of children with GHD.

        Microorganisms producing bioemulsifier were isolated from the sea water in Pusan costal aera. The isolated strain which had the highest emulsification activity and stability was identified as the genus Acinetobacter from the results of morphological, cultural and biochemical tests and named Acinetobacter sp. EL-C6 for convinience. The compositions of optimum medium for emulsification of crude oil by Acinetobacter sp. EL-C6 were crude oil 2%, NH_4NO_3 0.2%, K_2HPO_4 0.01%, MgSO_4·7H_2O 1%, CaCl_2·2H_2O 0.1%, NaCl 3% and the initial pH 7.5. The cultivation for emulsification of crude oil was carried out in 500m1 shaking flask containing 100m1 of the optimum medium at 30℃. The highest emulsification was observed after 5 days. The utilization on the various hydrocarbon of the Acinetobacter sp. EL-C6 was showed that the utilization of n-alkane compounds were better than that of aromatic compounds. Among the petroleum compounds, by Acinetobacter sp. EL-06.

        Pseudomonas sp. EL-G527에 의한 환경친화성 생물계면활성제의 생산최적조건

        A biosurfactant-producing microorganism was isolated from activated sludge by enrichment culture when grown on a minimal salt medium containing n-hexadecane as a sole carbon source. This microorganism was identified as Pseudomonas sp. and it was named Pseudomonas sp. EL-G527. It's optimal culture condition is 2% n-hexadecane, 0.2% NH_4NO_3, 0.3% KH_2PO_4, 0.3% K_2HPO_4, 0.02% MgSO_4·7H_2O, 0.0025% CaCl_2·6H_2O, 0.0015% FeSO_4·7H_2O in 1ℓ distilled water and initial pH 7.0. Cultivation was initiated with a 2% inoculum obtained from starter cultures grown in 30 ㎖ of the same medium in 250 ㎖ flask. They were cultivated at 30 ℃ in reciprocal shaking incubator and the highest biosurfactant production was observed after 4 days.

        The rates of reaction of 4-substituted-2, 6-dinitrochlorobenzenes with para-substituted anilines in methanol-acetonitrile mixtures were measured by conductometry. It was observed that the rate constant increase in the order of Y=α-CH3>β-CH3>Py-H>α-CH3, where Y is a substituent in the pyridine ring. The electrophilic catalysis by methanol may be ascribed to the formation of hydrogen bonds between alcoholic hydrogen and nitrogen of amines in the ground state.

        The quantitative analysis of heavy metals(As, Cd, Cr, Hg, Pb, Cu) in the sea water around Kaduck island was conducted during the period of March, 1998 to September, 1998. The sea water in the four regions was sampled four times and the heavy metals were analyzed. The Heavy metal concentration of the sea water around Kaduck island was As 127.7㎍/ℓ , Cd 49.28㎍/ℓ , Cr 0.462㎍/ℓ, Hg 0.111㎍/ℓ, Pb 1.713㎍/ℓ, Cu 2.716㎍/ℓ in the average value, respectively.

        간호사의 이직의도 구조모형

        Purpose: The purpose of this study was to construct and test a structural equation model of nurses' turnover intention. Method: The participants were 716 nurses at 4 university hospitals. Data were collected by a self-report questionnaire in 2008 and analyzed using SPSS 12.0 and AMOS 4.0 Win program. Results: The fit indices of modified model were x²=285.928(p<.001),x²/df=1.958GFI=0.964, RMSEA=0.037, AGFI=0.944, NFI=0.947, NNFI=0.961, CFI=0.973, PNFI=0.658 and showed high compatibility with the empirical data. In the final model, job stress and satisfaction were found to have significant direct effects on nurses' turnover intention, while organizational culture and emotional labor were very important factors to have indirect effects on turnover intention via job stress and satisfaction. Nurses' turnover intention was accounted for 41.2% of covariance by these factors. Conclusions: Comprehensive nursing interventions for decreasing nurses' turnover intention should be focused on reducing job stress and increasing job satisfaction. Also, Nursing strategy for enhancing positive organizational culture and decreasing emotional labor would be helpful for decreasing nurses' turnover intention, too. Further studies are needed to prospectively conduct to verify these causal relationships with larger sample.

        우리나라 일부 석면사업장의 석면폐 유병률

        Asbestos industry has been in operation over 60 years in Korea. However, the prevalence of asbestosis has not been yet reported. With rapid turn-over of workers, previous cross-sectional studies of current workers on the job could not find cases with exposures long enough for the development of asbestosis. This study was done to evaluate asbestosis prevalence of those worksites with operation history of more than 20 years. In total, 139 workers from 5 worksites were examined. Asbestos industries covered in the study include 2 asbestos textile, 1 brake lining, and 2 ship repairing worksites. Chest x-ray was taken from all workers and read by two experts familiar with pneumoconiosis classification according to 1980 ILO guidelines. Those with findings compatible with asbestosis were further ckecked with high resolution computerized tomography (HRCT). Pulmonary function tests were done according to ATS guidelines, and occupational and previous medical history was taken through a standardized interview. Air-borne asbestos was measured according to NIOSH method 7400. The air-borne asbestos concentrations ranged from 0.2-1.3 f/cc for asbestos textile, from 0.7-1.0 f/cc for brake lining, and from 6.3-7.8 f/cc during asbestos removal at ship repairing worksite. Of the 139 workers 25 had abnormal chest radiographic findings, and 10 of them had findings compatible with pneumoconiosis. When work history and current asbestos measurements were accounted, 9 workers who had more than 10 years of asbestos exposure history showed chest radiographic findings of pneumoconiosis with Finally, 4 workers showed finding of pulmonary fibrosis and/or pleural thickening at HRCT, and 2 of them had restrictive lung function changes. The study results showed that, among 139 subjects, there were 4(3%) definite asbestosis cases confirmed with HRCT. The prevalence of probable asbestosis was 7% for 10-14 years of exposure, 13% for 15-19 years of exposure, and 23% for 20 or more years of exposure. The prevalence of compensable asbestosis with abnormal lung function was 4-6% for those with 15 or more years of exposure.
