      • Vibrio cholerae균 증식에 있어서 Ferric Citrate가 미치는 영향

        This study was performed to find a method of more high isolation rate of Vibrio cholerae from contaminated foods and the carrier of cholera. Ferric citrate, an ingredient of TCBS agar that has been used most freguently as a selective media for identification of Vibrio spp., was known to inhibit growth of enteric bacteria than Vibrio species. So, We investigated the effect of the ferric citrate on the growth of Vibrio cholerae 1. The growth of Vibrio cholerae was not inhibited in agar plate medium including low concentration of ferric citrate but Vibrio cholerae was inhibited in medium containing high concentration of ferric citrate. 2. TCBS agar medium was not suitable for isolation of Vibrio cholerae.

      • 강유전체박막/반도체 구조의 형성 및 특성

        Use of a rapid thermal annealing(RTA) technique is shown to improve the properties of metal-ferroelectric BaMgF_(4)-silicon structures The fluoride film was deposited in an ultra-high vacuum system at a substrate temperature of 300℃. A post-deposition annealing was conducted for 10 seconds at 600℃ in a vacuum of 0.1 Torr, using a home-made RTA apparatus. The results were found out to increase the resistivity of the ferroelectric BaMgF_(4) film from a typical value of 1-2×10^(11) Ω · cm before the annealing to about 5×10^(13) Ω · cm and reduce the interface state density of the BaMgF_(4)/Si interface to about 8×10^(10) /cm^(2) ·eV. Ferroelectric hysteresis measurements using a Sawyer-Tower circuit yielded remanent polarization and coercive field values of about 0.5 μC/cm^(2) and 80㎸/cm, respectively. The typical remanent polarization of the BaMgF_(4) films on the (100) amd (111) orientated silicon wafers were 0.5-0.6 μC/cm^(2) and that of the films on the (110) wafers was 1.2 μC/cm^(2).

        페이스트 오펙 제조에 관한 연구 : Ⅱ. 페이스트 오펙 포세린의 금속과의 결합강도 비교 Ⅱ. Comparison on the Bond Strength of Paste Opaque Porcelain to Metal

        Metal-ceramic system has been widely using for aesthetic dental prosthesis. Opaque porcelain is used fore the masking of the metallic color of coping and the enhancing of the bond strength between metal and porcelain for PFM system. Paste opaque is a new type of opaque porcelain to easily apply onto metal coping and to get uniform thickness and good bonding characteristics to metal. The paste opaque was prepared by mixing of powder opaque with organic substances for this study. Three kinds of paste opaque which are in market were selected to compare the particle size distribution and bond strength with the paste opaque prepared in this study. The particle size distributions of all paste opaques were measured by particle size analyser, and the bond strengths were examined by using universal testing machine. This study shows that the maximum size of all samples is under 50㎛ and Ceramco has the finest mean particle size. The bond strength of the prepared paste opaque is about 35 MPa, and it is satisfied with ISO standard(25MPa).

      • Helicobacter pylori 편모 유전자의 클로닝 및 염기서열 분석

        A λgt11 expression libary of H. pylori DNA in E. coli Y1090 was screened with flagellin-specific rabbit antiserum for molecular cloning of the flagellin gene of H. pylori. A positive clone, λHPF4, was obtained and the recombinant antigen expressed from λHPF4 was a fusion protein with the molecular weight of 168kd. Sequence analysis of antigen-encoding DNA showed that an open reading frame composed of 1,536 nucleotides encodes a polypepride with a oredicted molecular size of 54kd. This open reading frame did not show the homology with flaA gene encoding 56kd protein of H. pylori and was confirmed as a unique sequence through homoligy searching. Therefore, the cloned antigen is supposed to be the carboxy-terminal region of the other flagellin protein of H. pylori, flaB, with the molecular weight of 58kd.

        허혈 조건부여가 심근기능의 회복에 미치는 영향 : 적출 심장모델을 이용한 실험연구 Isolated heart experimental study

        Background: Brief episode of coronary artery occlusion(i.e.,ischemic preconditioning) makes the heart more resistant to injury from a subsequent ischemic insult. Although a great deal of effort has been made in studying ischemic preconditioning, the underlying mechanism of ischemic preconditioning and its effect on hypothermic insult has not been elucidated. This study was performed to see whether ischemic preconditioning protects against the depression of cardiac contractility induced by hypothermic cardioplegic arrest/reperfusion. And recently, adenosine was known to have some correlation with the mechanism of preconditioning. If so, does this effect remain after the blockade of adenosine receptor by 8-phenyl theophyline? Method: Twenty-four Sprague-Dawley rat weighed 250-350g were used and divided into three groups. Rat hearts were removed rapidly, and each isolated heart paced with a rate of 180/min was perfused by modified Krebs-Hensleit buffer(KHB) solution on a Langendorff apparatus for an hour. After obtaining baseline data including left ventricular pressure(LVP), dp/dt, and coronary flow, cardiac arrest was induced by perfusion of 0℃ crystalloid cardioplegic(St Thomas) solution. After that, all hearts were stored in the same St Thomas solution at same temperature for 2 hours. In group Ⅰ(control group), the heart was reperfused by KHB solution. In group Ⅱ(preconditioning group), the heart was subjected to two 2-minute episode of global ischemia followed by 5 minute reperfusion with KHB solution(preconditioning) before cardiac arrest. In group Ⅲ(phenyl theophylline group), the heart was subjected to preconditioning procedure and 8-phenyl theophylline at 10μM in concentration was added to KHB solution at time of reperfusion. Observing parameter was obtained in each group at 10, 20, 40 and 60 minutes after starting reperfusion and compared statistically by use of one way ANOVA test(STASTICA, release 4.5). p-value less than 0.05 was considered significant. Results: Although depressed LVP, dp/dt, and coronary flow were seen in all groups during the reperfusion period, the preconditioned group showed more effective recovery of LVP than that of the control group, especially at 10, 20 and 40 minutes(p<.05). We failed to demonstrate the difference between the phenyl theophylline group and the control group(p=NS). Conclusion: These results suggest that ischemic preconditioning has protective effect on recovery state of hypothemic cardioplegic arrest/reperfusion. Its protective effect was limited during early reperfusion stage and was blocked by adenosine blocker.

        The purposes of this study are to examine the current status of local educational governance system, to diagnose the problems of local educational governance system, to inspect the validity of enforcement of local educational governance system at primary administrative units, and to develop the enforcement model for local educational governance system at primary units. To accomplish these purpo`ses, survey research method by questionnaire for 860 subjects of study as residents in Pohang City is applied to this study. Though local educational governance system at a wide area administrative units has been enforced for the past six years since 1991, the degree of residents interest in local educational governance system at a wide area administrative units is very low, and the degree of residents' satisfaction and participation in local educational governance system at a wide area administrative units is also very low. Consequently, it is very important to enforce local educational governance system at primary administrative units for developing local educational governance system as soon as possible. Based on the findings of residents opinion for the enforcement of local educational governance system at primary administrative units, basic principles and theoretical background on local educational governance system, the enforcement model for local educational governance system at primary administrative units was developed in this study.

        현재 업체에서 제공하고 있는 반도체 정보의 수집 및 가공이 모두 수동으로 이루어져 있어 많은 시간의 노력과 인건비가 들어가고 있다.현재 개발되어 있는 에이전트는 범용적이고 일반적인 형태의 에이전트로 특화된 반도체 정보의 수집 및 가공에는 부적합하다. 따라서 본 연구를 통해 반도체 정보 시스템에 특화된 웹 에이전트를 개발하여 현재 수동으로 이루어지고 있는 반도체 정보 시스템의 정보처리 과정을 자동으로 수행하는 웹 에이전트를 개발하여 인건비의 절감과 정보 시스템의 신뢰성 향상 및 정보처리 과정의 자동화, 체계화를 이루고자 한다.

      • CFRP 적층판의 초음파 특성평가

        Ultrasonic NDE of composite laminates is usually analyzed on the backwall echoes of the specimen. In the inspection of actual composite structures, the back surface of the part was curved or contained complex geometry and did not provide a usable backwall echo. In this work, a one-sided pitch-catch setup was used in the detection and evaluation of flaws and material anomalies in solid composite laminates. Two miniature potted Rayleigh wave transducers (for generating surface waves in contact mode on steel) were joined head-to-head and used in the pitch-catch mode on the surface of composites. The pitch-catch signal was found to be more sensitive than normal incidence backwall echo of longitudinal wave to fiber orientation of the CFRP composites, including low level porosity, ply waviness, and cracks.

        우리나라 재난의학적 처치에 대한 연구 : 1993년 3월 29일 구포 열차 전복사고를 중심으로 한 연구 ANALYSIS OF RAILROAD COLLAPSED ACCIDENT OF GUPO

        Disaster in an unexpected, chaotic, horrendous catastrophes and it may be either natural or human origin. Now a days, disasters are occurring at on increasing rate due to population growth and technological advancement. Medical disaster in an incident which produces such a number of causalities that makes the routine methods for patient care ar4e inadequate. The public no linger accept these consequences as "act of God" and they demand that disaster risks to be minimized and that any impact to be management in an orderly fashion. So emergency medical personnel must strive to achieve themselves to achieve the greatest good for the greatest number of potential survivors at the disaster scene. On 28 March 1993, a train carrying 600 passengers rolled over due to railroad collapse. About 170 passengers dies(25 before extrication) and 22 severe wounded survived. Problems in the organization at the scene, triage, and the communication contributed to the confusion in the management of victims. As analysis of this accident allows several lessons to be learn which might prevent or reduce the fatalities from future disaster. Although Korean community has relied on a model of disaster response deprived from the experience of military and civil defense planners for years. A better approach to organizing medical personnel for the incident with multiple causalities, which is based on the medical response of the emergency medical services organization, will be needed in the near future.

      • Helicobacter pylori와 대장균의 Shuttle Vector 개발

        In this study, a vehicle vector using cryptic plasmids was constructed for gene transfer in Helicobacter pylori. pHP51(3.9 kb) and pHP489(1.2 kb) were selected for constructing vectors from cryptic plasmid of H. pylori isolates in Korea. The HindⅢ-digestedDNA fragment(1.2kb) of pHP489 and 1.6kb DNA fragment of pHP51 were ligated with a kanamycin resistance gene(aph3'-Ⅲ) from C. jejuni to produce the recombinant plasmids pHP489K and pHP51K, respectively. Transformation frequency of pHP51K by electroporation was low. But pHP489K could be effectively transformed into various H. pylori strains. In order to design an intermdiate vehicle vector for gene transfer into H. pylori, pBlueHP489K was prepared by recloning pHP489K DNA into pBluescript and pTZ19R vector. This vector permitted the DNA fragment containing pHP489 sequence, aph3'-Ⅲ, and cloned DNA to be cut and self-ligated in the SacⅠ site after cloning. ureA and ureB gene were inserted into pBlueHP489K, resulting in pBlueHP489K/AB. The DNA fragment containing pHP489, kanamycin resistance gene(aph3'-Ⅲ), and urease structural gene was cut away from pBlueHP489K/AB and self-ligated to generate pBlueHP489K/AB. pBlueHP489K/AB made urease-negative H. pylori strains restore their urease activity. By this experiment, pBlueHP489K was confirmed to be the vehicle system for transferring H. pylori genes.
