The eukaryotic genome is packed into chromatin, which is important for the genomic integrity and gene regulation. Chromatin structures are maintained through assembly and disassembly of nucleosomes catalyzed by histone chaperones. Asf1 (anti-silencing function 1) is a highly conserved histone chaperone that mediates histone transfer on/off DNA and promotes histone H3 lysine 56 acetylation at globular core domain of histone H3. To elucidate the role of Asf1 in the modulation of chromatin structure, we screened and identified small molecules that inhibit Asf1 and H3K56 acetylation without affecting other histone modifications. These pyrimidine- 2,4,6-trione derivative molecules inhibited the nucleosome assembly mediated by Asf1 in vitro, and reduced the H3K56 acetylation in HeLa cells. Furthermore, production of HSV viral particles was reduced by these compounds. As Asf1 is implicated in genome integrity, cell proliferation, and cancer, current Asf1 inhibitor molecules may offer an opportunity for the therapeutic development for treatment of diseases. Heterotopic Gastric Mucosa causing leading to Recurrent jejunal Intussusceptions

Heterotopic gastric mucosa (HGM) is a rare anomaly in the small bowel and supposed to be of vitellointestinal tract origin. In addition, HGM may cause intussusception as a lead point in the jejunum. A 5-year-old girl presented with chief complaints of vomiting and abdominal pains for 2 weeks. Her past medical and family histories were unremarkable. There was history of constipation. Medical therapy aimed at vomiting was unsuccessful. The abdominal computed tomography (CT) scan revealed the intussusception at proximal jejunal loops. Three times air reduction and once saline reduction was performed. At exploratory laparotomy, the intussusception could not be found. The patient continued to be symptomatic and referred for endoscopic evaluation. Enteroscopy withgastroscope showed some variable size polypoid mucosal lesions with erosions on proximal jejunum. We performed endoscopic mucosal resection to remove the jejunal lesion. Histopathologic evaluation showed hyperplastic polyps arising from heterotopic gastric mucosa. In immunohistochemical staining, these polyps expressed MUC4, MUC5AC, MUC6, MUC2, and c-erbB2, but negative for MUC1. Despite its rarity, HGM should be considered as a cause of recurrent intussusceptions and diagnostic studies should be performed

Background/Aims: The immediate and long-term results of percutaneous transhepatic cholangioscopic lithotomy for the intrahepatic stones were analyzed according to degree of biliary stricture, severity of cirrhosis, and Tsunoda classification. Methods: We studied retrospectively medical records of 92 patients with hepatolithiasis who received percutaneous transhepatic cholangioscopy (PTCS) treatment from September 1992 to May 1997. Sixty-eight patients were followed up for 24 to 60 months (median 42 months). Results: Complete clearance of the intrahepatic stones was achieved in 74 patients (80%). In group A (Tsunoda class I and II), no recurrence was observed upto 28 months of follow-up. However, in the patients of group B (Tsunoda class Ⅲ and Ⅳ), recurrence rates were increased continuously, and 50% of them had eventually recurrence of stones or cholangitis. Conclusions: PTCS is an effective and safe procedure for management of hepatolithiasis in patients who are not candidates for surgical resection. In cases of severe andultiple strictures, adequate dilatation of bile duct strictures and close follow-up are necessary.

<P>The mammalian genome encodes multiple variants of histone H3 including H3.1/H3.2 and H3.3. In contrast to H3.1/H3.2, H3.3 is enriched in the actively transcribed euchromatin and the telomeric heterochromatins. However, the mechanism for H3.3 to incorporate into the different domains of chromatin is not known. Here, taking the advantage of well-defined transcription analysis system of yeast, we attempted to understand the molecular mechanism of selective deposition of human H3.3 into actively transcribed genes. We show that there are systemic H3 substrate-selection mechanisms operating even in yeasts, which encode a single type of H3. Yeast HIR complex mediated H3-specific recognition specificity for deposition of H3.3 in the transcribed genes. A critical component of this process was the H3 A-IG code composed of amino acids 87, 89 and 90. The preference toward H3.3 was completely lost when HIR subunits were absent and partially suppressed by human HIRA. Asf1 allows the influx of H3, regardless of H3 type. We propose that H3.3 is introduced into the active euchromatin by targeting the recycling pathway that is mediated by HIRA (or HIR), and this H3-selection mechanism is highly conserved through the evolution. These results also uncover an unexpected role of RI chaperones in evolution of variant H3s.</P>

Background/Aims: Anti-tumor necrosis factor (TNF) therapy for active ulcerative colitis (UC) and Crohn`s disease (CD) is associated with increased risks of tuberculosis (TB) infection. We analyzed the incidence and clinical features of Korean patients with inflammatory bowel disease (IBD) who developed active TB during anti-TNF therapy. Methods: Ten cases of active TB developed in patients treated with infliximab (n=592) or adalimumab (n=229) for UC (n=160) or CD (n=661) were reviewed. We analyzed demographics, interval between start of anti-TNF therapy and active TB development, tests for latent TB infection (LTBI), concomitant medications, and the details of diagnosis and treatments for TB. Results: The incidence of active TB was 1.2% (10/821): 1.5% (9/592) and 0.4% (1/229) in patients receiving infliximab and adalimumab, respectively. The median time to the development of active TB after initiation of anti-TNF therapy was three months (range: 2-36). Three patients had past histories of treatment for TB. Positive findings in a TB skin test (TST) and/or interferon gamma releasing assay (IGRA) were observed in three patients, and two of them received anti-TB prophylaxis. Two patients were negative by both TST and IGRA. The most common site of active TB was the lungs, and the active TB was cured in all patients. Conclusions: Active TB can develop during anti-TNF therapy in IBD patients without LTBI, and even in those with histories of TB treatment or LTBI prophylaxis. Physicians should be aware of the potential for TB development during anti-TNF therapy, especially in countries with a high prevalence of TB. (Intest Res 2016;14:146-151)
A Remote Sensed Data Combined Method for Sea Fog Detection

Steam and advection fogs are frequently observed in the Yellow Sea from March to July except for May. This study uses remote sensing (RS) data for the monitoring of sea fog. Meteorological data obtained from the Ieodo Ocean Research Station provided a valuable information for the occurrence of steam arid advection fogs as a ground truth. The RS data used in this study were GOES-9, MTSAT-1R images and QuikSCAT wind data. A dual channel difference (DCD) approach using IR and shortwave IR channel of GOES-9 and MTSAT-1R satellites was applied to detect sea fog. The results showed that DCD, texture-related measurement and the weak wind condition are required to separate the sea fog from the low cloud. The QuikSCAT wind data was used to provide the wind speed criteria for a fog event. The laplacian computation was designed for a measurement of the homogeneity. A new combined method, which includes DCD, QuikSCAT wind speed and laplacian computation, was applied to the twelve cases with GOES-9 and MTSAT-1R. The threshold values for DCD, QuikSCAT wind speed and laplacian are -2.0 K, 8 m s-1 and 0.1, respectively. The validation results showed that the new combined method slightly improves the detection of sea fog compared to DCD method: improvements of the new combined method are 5~6% increases in the Heidke skill score, 10% decreases in the probability of false detection, and 30~40% increases in the odd ratio.
