        Introduction of heterogeneous NADH reoxidation pathways into Torulopsis glabrata significantly increases pyruvate production efficiency

        The aim of this study was to increase pyruvate production efficiency in a multi-vitamin auxotrophic yeast,Torulopsis glabrata. This was achieved by decreasing intracellular NADH content through the introduction of two different NADH reoxidation pathways: one in the cytoplasm and the other in mitochondria. A nox (encoding a cytoplasmic H_2O-forming NADH oxidase) and an AOX1 (encoding a mitochondrial alternative oxidase) were successfully expressed heterologously in T. glabrata, resulting in a decrease in the NADH and ATP contents of 55% and 26%, (in T. glabrata NOX) and 45% and 47% (in T. glabrata AOX), respectively. The decreases in nucleotide concentrations led to increases in the glucose consumption rate, the pyruvate yield and pyruvate productivity of 27%, 15% and 22% (in T. glabrata NOX) and 38%, 21%, and 29% (in T. glabrata AOX), respectively, compared with the corresponding values of the control. We conclude that this method provides an alternative way to enhance the production efficiency of NADHrelated metabolites.

        Predictive Value of Exercise Blood Pressure Changes for Orthostatic Hypotension in Patients With Parkinson’s Disease

        Background and Purpose Orthostatic hypotension (OH) is common in patients with Parkinson’s disease (PD). Early recognition OH is required with sensitive assessments. The purpose of this study was to determine whether blood pressure (BP) changes during exercise can predict the occurrence of OH in PD. Methods This prospective cohort study included 80 consecutive patients with PD. All patients agreed to participate in a baseline evaluation and cardiopulmonary exercise test (CPET). According to the initial active standing test (AST), those without OH (PD-nonOH) at baseline had their AST results followed up for 6 months. The main outcome was defined as whether patients without OH at baseline would develop OH after 6 months. Logistic regression analysis was applied to identify the relevant variables. A nomogram was constructed based on clinical features and identified variables. The concordance index (C-index) and area under the receiver operating characteristic curve (AUC) were used to evaluate the accuracy and predictive ability of the nomogram, respectively. Results CPET results indicated that peak load, peak heart rate, heart rate recovery at 1 min, and systolic BP change (ΔSBP) were lower in those with OH than in the PD-nonOH group (p<0.05) at baseline. Logistic regression analysis indicated that peak load and ΔSBP during CPET had significant effects on OH (p<0.05). Age, sex, peak load, and ΔSBP were used to construct the nomogram model (C-index=0.761). The prediction model had an AUC of 0.782 (95% confidence interval=0.649–0.889) and a specificity and sensitivity of 70.0% and 81.8%, respectively. Conclusions This study has identified predictive factors for OH development in patients with PD. CPET could be used as a complementary examination to identify patients at a high risk of OH.

        GPH1 is involved in glycerol accumulation in the three-dimensional networks of the nematode-trapping fungus Arthrobotrys oligospora

        Turgor is very important for the invasive growth of fungal pathogens. Glycerol, a highly osmotic solvent, is considered to play an important role in turgor generation. The nematophagous fungus Arthrobotrys oligospora mainly lives as a saprophyte. In the presence of nematodes, A. oligospora enters the parasitic stage by forming three-dimensional networks (traps) to capture nematodes. In A. oligospora, we found that glycerol accumulated during nematode-induced trap formation. We demonstrated that deleting gph1, which encodes glycogen phosphorylase, decreased the glycerol content, compared with that of a wild-type strain. Although the number of traps induced by nematodes was not affected in the Δgph1 mutant, the capture rate was lower. Meanwhile, deleting gph1 also affected the growth rate and conidiation capacity of the fungus. These results indicate that glycerol derived from GPH1 is essential for the full virulence of A. oligospora against nematodes.

      • Construction of a Transposon-mediated Baculovirus Vector Hanpvid and a New Cell Line for Expressing Barnase

        In this study we developed the transposon-mediated shuttle vector 'Hanpvid', which composed of HaNPV (Heliothis armigera nuclear polyhedrosis virus) genomic DNA and a transposon cassette from Bacmid of Bac-to-Bac system. Hanpvid replicates in E. coli in the same way as Bacmid and retains infective function in cotton bollworm cells (Hz-AM1). Using Hanpvid we constructed a recombinant virus, which could infect Hz-AM1 cells and generate recombinant HaNPV (rHa-Bar) containing the barnase gene, a ribonuclease gene from Bacillus amyloliquefaciens. Since the expression vector carrying barnase gene cannot replicate in the absence of barstar, a specific inhibitor of barnase, we constructed a new cotton bollworm cell line (AM1-NB) using the marker rescue method. In AM1-NB barstar was integrated into the cellular chromosome to sustain the replication of rHa-Bar. To screen out recombinant HaNPV for potential use as biopesticide, Hz-AM1 and AM1-NB cell lines were infected with rHa-Bar, respectively. The results obtained indicate that Viral progenies in AM1-NB were 23 and 160 times greater than those in Hz-AM1 48 h and 72 h after infection, respectively. With additional insertion of the polyhedron gene from AcNPV (Autographa californica nuclear polyhedrosis virus) into the Hanpvid genome, rHa-Bar regained the polyhedron phenotype and its pest-killing rate greatly improved. Toxic analysis showed that the lethal dosages ($LD_{50}$) and the lethal time(s) ($LT_{50}$) of rHa-Bar were reduced by 20% and 30%, respectively, compared to wt-HaNPV in the third instar larvae of cotton bollworm. This study shows that in AM1-NB barnase can be effectively produced and used as pest-killing agent for the biological control of cotton pests.

      • Exogenous Morphine Inhibits Human Gastric Cancer MGC-803 Cell Growth by Cell Cycle Arrest and Apoptosis Induction

        Morphine is not only an analgesic treating pain for patients with cancer but also a potential anticancer drug inhibiting tumor growth and proliferation. To gain better insight into the involvement of morphine in the biological characteristics of gastric cancer, we investigated effects on progression of gastric carcinoma cells and the expression of some apoptosis-related genes including caspase-9, caspase-3, survivin and NF-${\kappa}B$ using the MGC-803 human gastric cancer cell line. The viability of cells was assessed by MTT assay, proliferation by colony formation assay, cell cycle progression and apoptosis by flow cytometry and ultrastructural alteration by transmission electron microscopy. The influences of morphine on caspase-9, caspase-3, survivin and NF-${\kappa}B$ were evaluated by semi-quantitative RT-PCR and Western blot. Our data showed that morphine could significantly inhibit cell growth and proliferation and cause cell cycle arrest in the G2/M phase. MGC-803 cells which were incubated with morphine also had a higher apoptotic rate than control cells. Morphine also led to morphological changes of gastric cancer cells. The mechanism of morphine inhibiting gastric cancer progression in vitro might be associated with activation of caspase-9 and caspase-3 and inhibition of survivin and NF-${\kappa}B$.

        Field Emission Properties of -C:N Films Deposited on Diamond Substrates

        Amorphous carbon-nitride films were grown on the nitridated diamond substrates by pulsed discharge of nitrogen gas by using graphite rods as the electrodes. The deposition parameters were optimized by monitoring the discharge plasma by optical-emission spectroscopy. It is demonstrated that the films were mainly a mixture of sp2 C-N, sp3 C-N and graphite nanocrystallites. Preliminary results show that deposited films have a cold-cathode-emission property. The threshold field for field emission is about 4.0 V/μm. The linear Fowler-Nordheim characteristic reveals that the fieldemission process is based on the tunneling mechanism.

        중국 상장기업의 자본구조 결정요인

        상해증권거래소는 2011년 3월 말 기준 상장 회사 905개, 상장증권 1,537개, 상장주식 949개, 상장주식 총수 2조 2,000억 주, 시가총액 19조 3,000억 위안 규모로 신흥시장의 최대 증권거래소로 자리매김을 하였으며, 앞으로 더욱 많은 발전을 할 것으로 예측되고 있다. 다른 국가의 증권거래소와는 달리 중국의 증권거래소는 상장기업 중 국유기업의 비중이 높은 특징을 지니고 있기 때문에 증권거래소에 상장된 기업들의 자본구조가 어떠한지, 더 나아가 자본구조를 결정하는 요인들이 무엇인지를 파악하는 것은 의미가 있다. 따라서 본 연구의 목적은 중국의 증권거래소 중 신흥시장의 최대 증권거래소로 부상한 중국의 대표적인 증권거래소인 상해증권거래소에 상장된 기업들 중 60%를 차지하고 있는 제조업에 속한 기업들을 대상으로 다중회귀분석을 이용하여 자본구조를 결정하는 요인들이 무엇인지를 파악하는데 있다. 본 연구결과를 요약하면 다음과 같다. 첫째, 기업규모는 부채비율에 1% 유의수준에서 유의적인 정(+)의 영향을 미치는 것으로 나타났다. 둘째, 수익성은 부채비율에 1% 유의수준에서 유의적인 부(-)의 영향을 미치는 것으로 나타났다. 셋째, 성장성은 부채비율에 1% 유의수준에서 유의적인 정(+)의 영향을 미치는 것으로 나타났다. 넷째, 현금흐름, 최대주주 지분율, 유통주 비율은 부채비율에 부(-)의 영향을 미치는 것으로 나타났으나 통계적으로 유의하지 않았다. 본 연구결과는 투자자에 대한 정보제공 및 재무구조 개선의 기초자료로 활용될 수 있다는 점에서 의의가 있다. Shanghai Stock Exchange is the largest stock exchange of emerging markets that there were listed firms 905, listed securities 1,537, listed stocks 949, total number of listed stocks 2 trillion 2000 billion shares. There is more development that is expected to occur in the future . The purpose of this study is to find determinants of capital structure to listed manufacturing firms in Shanghai Stock Exchange using multiple regression. Conclusions of this study are summarized as follows. First, firm size is positively related to debt ratio significantly at 1% significance level.. Second, the profitability is negatively related to debt ratio significantly at 1% significance level. Third, the growth ability is positively related to debt ratio significantly at 1% significance level. fourth, cash flow, the largest shares ownership, negotiable shares ratio are negatively related to debt ratio but they are not significant statistically. The result of this study provides information for investors and can be utilized to improvement of financial structure.

      • Prostate Cancer Risk in Relation to a Single Nucleotide Polymorphism in the Insulin-like Growth Factor-binding Protein-3 (IGFBP3) Gene: a Meta-analysis

        Insulin-like growth factor-binding protein-3 (IGFBP3) has been identified as a putative tumor suppressor with multifunctional roles in the IGF axis. Recently, there have been a growing body of studies investigating the relation between the IGFBP3 A-202C polymorphism, circulating IGFBP3 and prostate cancer risk, but their outcomes varied leading to controversy. Hence, it is necessary to perform a meta-analysis covering all eligible studies to shed a light on the association of IGFBP3 A-202C and cancer risk. Finally, we included a total of 11 relevant articles between 2003 and 2010 covering 14 case-control studies including 9,238 cases and 8,741 controls for our analysis. Our results showed that A-202C was a marginal risk factor of prostate cancer (allele contrast: OR=1.08, 95% CI :1.01-1.16; dominant model: OR=1.11, 95% CI :1.01-1.22; heterozygote codominant model: OR=1.11, 95% CI :1.03-1.18; homozygote contrast: OR=1.19, 95% CI :1.03-1.37). Stratification analysis revealed that sample size and control source were two major heterogeneous meta-factors especially in the recessive model (source: Population-based control group :p=0.30,I2=16.7%, Hospital-based control group: p=0.20, I2=30.3%; sample size: Small: p=0.22,I2= 32.8%, Medium: p=0.09,I2=48%, Large p=0.60,I2=0.0%); However, contrary to previous findings, no significance was found in racial subgroups. No significant publication bias was found in our analysis. Considering the robustness of the results and the discrepancy among some studies, there might be some unsolved confounding factors, and further more critical large studies are needed for confirmation.
