      • Personalized Context-aware Recommendation Approach for Web Services

        With the increasing number of Web services, the goal of consumers becomes to discover and use services that lead to their experiencing the highest quality. Quality of Service (QoS) is important to evaluate the QoS performance of services to differentiate the qualities of service candidates. QoS is highly related to context information since service consumers are typically distributed in different geographical locations. Their experience is usually different. Invoking a huge number of Web services for consumers to predict the quality is time-consuming, resource- consuming, and sometimes even impractical. To address the challenge, this paper proposes a personalized context-aware recommendation approach for predicting the QoS of Web services and designs a prediction framework. This algorithm is a hybrid of the model-based and memory-based collaborative filtering algorithms. In our experiment, we collect QoS information from geographically distributed service consumers through the framework. Based on the QoS and context information, we predict the quality of services. As a result, we can obtain a list of recommended services for selection. Finally, the experiment shows that the algorithm using context information achieves better prediction.

        Quantitative Investigation into the Relation between Force Chains and Stress Transmission During High-Velocity Compaction of Powder

        High-velocity compaction (HVC), an innovative approach to obtain green compacts with high and uniform density, is widely used in the powder metallurgy industry. In this study, meso force chains, macro stress transmission, and their relation were investigated using the discrete element method. The simulation details of HVC and the quantitative characterization of force chains and stress transmission were shown. Then, the relation between force chains and stress was investigated. The evolution of force chains showed the same change tendency as the stress distribution. They evolved from top to bottom and then reflected backwards in HVC while they did not show this trend in conventional compaction. The strength of the force chains maintained good consistency with the stress magnitude. Meanwhile, the length of the force chains presented a negative correlation with the stress magnitude, and high stress may cause new force chains to shorten. The average collimation coefficient was affected by the transmission of stress, and the short force chains had better straightness. Furthermore, force chains parallel to the direction of gravity were observed in the region with no stress concentration. The directional coefficient of force chains also had the same fluctuation trend as the variation in the principal stress angle.

        piRNA-1742 promotes renal cell carcinoma malignancy by regulating USP8 stability through binding to hnRNPU and thereby inhibiting MUC12 ubiquitination

        Accumulating studies have confirmed that PIWI-interacting RNAs (piRNAs) are considered epigenetic effectors in cancer. We performed piRNA microarray expression analysis on renal cell carcinoma (RCC) tumor tissues and paired normal tissues and performed a series of in vivo and in vitro experiments to explore piRNAs associated with RCC progression and investigate their functional mechanisms. We found that piR-1742 was highly expressed in RCC tumors and that patients with high piR-1742 expression had a poor prognosis. Inhibition of piR-1742 significantly reduced tumor growth in RCC xenograft and organoid models. Mechanistically, piRNA-1742 regulates the stability of USP8 mRNA by binding directly to hnRNPU, which acts as a deubiquitinating enzyme that inhibits the ubiquitination of MUC12 and promotes the development of malignant RCC. Subsequently, nanotherapeutic systems loaded with piRNA-1742 inhibitors were found to effectively inhibit the metastasis and growth of RCC in vivo. Therefore, this study highlights the functional importance of piRNA-related ubiquitination in RCC and demonstrates the development of a related nanotherapeutic system, possibly contributing to the development of therapeutic approaches for RCC.

        Chemical Constituents from the Aerial Parts of Isodon coetsa and their Cytotoxicity

        Three new compounds (1-3), including a neolignan, a triterpenoid, and a diterpenoid, together with twenty known compounds (4-23), were isolated from the aerial parts of Isodon coetsa. Their structures and relative configurations were elucidated on the basis of spectroscopic data. Compounds 1, 3, 5-9, 11-13, 16-17, and 19-23 were evaluated for their cytotoxicity against HT-29, BEL-7402, and SK-OV-3 human tumor cell lines. Compound 7 showed significant inhibitory effects on all three types of cells, with IC50 values of 2.52, 3.06, 2.14 μM, respectively.

        Genome-wide identification and analysis of expression pattern of REVEILLE transcription factors in castor (Ricinus communis L.)

        REVEILLE (RVEs) transcription factors play an important role in regulating circadian clock, plant growth and development, and stress responses. While RVEs have been identifi ed in various plant species, comprehensive genome exploration and analysis of RVEs in castor ( Ricinus communis L.) have not been previously reported. In this study, we identifi ed RcRVEs in the castor genome through sequence alignment and conducted an analysis encompassing gene structures, protein motifs, physicochemical properties, transcription patterns under cold stress, diff erent tissues, and diurnal rhythm. A total of fi ve RcRVEs were pinpointed in the castor genome. Evolutionary tree analysis categorized these RcRVEs into three distinct groups, implying that diff erent RcRVEs may fulfi ll distinct biological functions. Furthermore, several abiotic stresses and hormone-responsive cis -acting elements were found in the promoters of the RcRVEs . RNA-sequencing (RNA-seq) analysis revealed that RcRVE1 and RcRVE7 were induced by cold stress and exhibited upregulation over time, indicating their involvement in regulating cold adaptation in castor plants. Five RcRVEs have notably higher expression levels in leaves compared to other tissues. The expression of all RcRVEs displayed diurnal rhythms under light/dark cycles. This study provides valuable insights into the potential functions of RcRVEs in regulating castor’s growth and development, and responses to cold stress.

        Analysis the role of arabidopsis CKRC6/ASA1 in auxin and cytokinin biosynthesis

        The crosstalk between auxin and cytokinin (CK) is important for plant growth and development, although the underlying molecular mechanisms remain unclear. Here, we describe the isolation and characterization of a mutant of Arabidopsis Cytokinin-induced Root Curling 6 (CKRC6), an allele of ANTHRANILATE SYNTHASE ALPHA SUBUNIT 1 (ASA1) that encodes the á-subunit of AS in tryptophan (Trp) biosynthesis. The ckrc6 mutant exhibits root gravitropic defects and insensitivity to both CK and the ethylene precursor 1-aminocyclopropane-1-carboxylicacid (ACC) in primary root growth. These defects can be rescued by exogenous indole-3-acetic acid (IAA) or tryptophan (Trp) supplementation. Furthermore, our results suggest that the ckrc6 mutant has decreased IAA content, differential expression patterns of auxin biosynthesis genes and CK biosynthesis isopentenyl transferase (IPT) genes in comparison to wild type. Collectively, our study shows that auxin controls CK biosynthesis based on that CK sensitivity is altered in most auxin-resistant mutants and that CKs promote auxin biosynthesis but inhibit auxin transport and response. Our results also suggest that CKRC6/ASA1 may be located at an intersection of auxin, CK and ethylene metabolism and/or signaling.

        Intranasal Oxytocin Increases Perceptual Salience of Faces in the Absence of Awareness

        Objective The neuropeptide oxytocin has been found to improve human social cognition and promote prosocial behavior. However, it is still unclear about the mechanisms underlying these effects of oxytocin on neural processes, such as visual perception and awareness. Especially, it is still unclear whether oxytocin influences perceptual salience of social stimuli in the absence of awareness. Methods In a randomized double-blind, placebo-controlled trial we applied an interocular suppression paradigm and eye tracking methods to investigate the influence of intranasally administered oxytocin on perceptual salience of social stimuli. Suppression times and pupillometric data were measured during subjects being presented with gradually introduced pictures of social stimuli (neutral expression faces) or nonsocial stimuli (grayscale watch pictures) that were suppressed and invisible in 10 men who were administered 24 IU oxytocin and 10 men who were administered a placebo. Results The results demonstrated that the oxytocin group perceived social stimuli more quickly accompanied by subsequent larger increasing pupil diameter than nonsocial stimuli, indicating an increased unconscious salience of social stimuli. Conclusion These findings provided new insights into oxytocin’s modulatory role to social information processing, suggesting that oxytocin might enhance attentional bias to social stimuli even after removal of awareness.

      • Disruption of endothelial barrier function is linked with hyposecretion and lymphocytic infiltration in salivary glands of Sjögren's syndrome

        <P><B>Abstract</B></P> <P>Sjögren's syndrome (SS) is an inflammatory autoimmune disease that causes hyposecretion in salivary glands. Endothelial tight junctions (TJs) play crucial roles in salivation and barrier function of blood vessels. However, whether the alteration of endothelial TJs were involved in pathogenesis of SS was still unknown. Here, the ultrastructure and function of endothelial TJs in submandibular glands (SMGs) were detected by transmission electron microscopy and in vivo paracellular permeability assay in different aged NOD mouse model for SS. CFSE-labeled lymphocytes were injected into tail vein to trace the infiltration, while claudin-5 expression and distribution were detected by immunofluorescence, qRT-PCR, and western blot. Results showed that the stimulated salivary flow rate was gradually decreased and lymphocytic infiltration was found as age increased in 12- and 21-week-old NOD mice, but not 7-week-old NOD mice. Blood vessels were dilated, while endothelial TJ width and paracellular tracer transport were increased in 12-week-old NOD mice. Moreover, the injected CFSE-labeled lymphocytes were observed in SMGs of 12-week-old NOD mice. Claudin-5 level was increased and relocalized from the apical portion of neighboring endothelial cells to lateral membranes and cytoplasm in 12-week-old NOD mice. Additionally, the alteration of claudin-5 expression and distribution was further confirmed in labial salivary glands and bilateral parotid glands from SS patients. In cultured human microvessel endothelial cell line (HMEC-1), IFN-γ stimulation significantly increased claudin-5 expression.         Successful salvage treatment of acute graft-versus-host disease after liver transplantation by withdrawal of immunosuppression

        Acute graft-versus-host disease (GVHD) following liver transplantation is a rare but fatal complication. The correct diagnosis and management of GVHD after liver transplantation are still major challenges. Herein, we reported successful salvage treatment of acute GVHD by withdrawal of immunosuppression in a patient who presented with fever, skin rashes, and decreased blood cell counts after liver transplantation. This case highlights the need for awareness of drug-induced liver injury if liver function tests are elevated during treatment, especially in patients taking multiple potentially hepatotoxic drugs, such as broad-spectrum antibiotics. When occurs, an artificial liver support system is a useful tool to provide temporary support of liver function for the patient in the event of drug-induced liver injury.
