Osmotic stress‐induced phosphorylation by NLK at Ser128 activates YAP

<P>YAP is the major downstream effector of the Hippo pathway, which controls cell growth, tissue homeostasis, and organ size. Aberrant YAP activation, resulting from dysregulation of the Hippo pathway, is frequently observed in human cancers. YAP is a transcription co-activator, and the key mechanism of YAP regulation is its nuclear and cytoplasmic translocation. The Hippo pathway component, LATS, inhibits YAP by phosphorylating YAP at Ser127, leading to 14-3-3 binding and cytoplasmic retention of YAP. Here, we report that osmotic stress stimulates transient YAP nuclear localization and increases YAP activity even when YAP Ser127 is phosphorylated. Osmotic stress acts via the NLK kinase to induce YAP Ser128 phosphorylation. Phosphorylation of YAP at Ser128 interferes with its ability to bind to 14-3-3, resulting in YAP nuclear accumulation and induction of downstream target gene expression. This osmotic stress-induced YAP activation enhances cellular stress adaptation. Our findings reveal a critical role for NLK-mediated Ser128 phosphorylation in YAP regulation and a crosstalk between osmotic stress and the Hippo pathway.</P>

A 2,656 bp fragment of chicken ghrelin gene was cloned and SNPs were detected by PCR-RFLP and Allele Specific PCR (ASP) in 12 Chinese indigenous chicken breeds and a commercial chicken population. The results showed that there were 23 base variations and an amino acid change (Gln->Arg) in cloned chicken ghrelin gene. Three SNPs were confirmed in 13 populations and associations between this gene and growth traits of Tibetan chicken (TC) and Recessive White chicken (RW) were investigated. The results of haplotype analysis revealed that 26 haplotype genotypes were composed of eight haplotypes. The results of x2 tests indicated that there were significant differences between genotypes or haplotype genotype frequencies in some of the breeds or sexes at 0.05 or 0.01 levels. The results of ANOVA revealed that there were significant differences between genotypes or haplotype genotypes on some growth traits of TC and RW chicken breeds at 0.05 or 0.01 levels. Multiple comparisons showed that there were significant associations between genotype CT at site 71 and some growth traits of two chicken breeds and between genotype AG at site 1,215 and body weight at 16 wk of two chicken breeds, and there was a significant association between haplotype genotype CAA/CAG and body weight and shank girth at 16 wk of two chicken breeds.

R&D Status of High-current Accelerators at IFP

High-current accelerators have many important applications in Z-pinches, high-power microwaves, and free electron lasers, imploding liners and radiography and so on. Research activities on Zpinches, imploding liners, radiography at the Institute of Fluid Physics (IFP) are introduced. Several main high-current accelerators developed and being developed at IFP are described, such as the Linear Induction Accelerator X-Ray Facility Upgrade (LIAXFU, 12 MeV, 2.5 kA, 90 ns), the Dragon-I linear induction accelerator (20 MeV, 2.5 kA, 60 ns), and the Primary Test Stand for Z-pinch (PTS, 10 MA, 120 ns). The design of Dragon-II linear induction accelerator (20 MeV, 2.5 kA, 3 × 60 ns) to be built will be presented briefly.

<P>The device performance improvement of ridge waveguide (RWG) laser diodes (LDs), operating at a wavelength (lambda) of similar to 960 nm, with a metal-dielectric high-reflection (HR) mirror consisting of Au/Ti-SiO2 layers on the back facet was demonstrated. To determine the optimum thickness of each layer, optical reflection calculations were performed using a rigorous coupled-wave analysis method, which leads to the resultant Au (80 nm)/Ti (5 nm)-SiO2 (164 nm) layers. The layers exhibited a broad high reflection band of >91% over a wavelength range of 920-1000 nm, indicating the reflectivity of similar to 91.2% at lambda similar to 960 nm. For 2 mm-cavity RWG LDs with the Au/Ti-SiO2 HR mirror, an enhanced maximum output power (P-max) of 499.3 mW at an injection current of 3000 mA and a decreased threshold current (Ith) of 516 mA (i.e., P-max = 259.4 mW and Ith = 650 mA for the uncoated LDs) were obtained, showing an increased slope efficiency percentage of 82%. The external differential quantum efficiency was also increased from similar to 17.1 to similar to 31.1%. Also, the full widths at half maximum values of beam divergences of the device were 38 degrees (vertical direction) and 4 degrees (horizontal direction). (C) 2015 Elsevier B.V. We investigated the device characteristics of GaN-based blue vertical light-emitting diodes (VLEDs) with two different package structures (i.e., lead frame with metal/plastic body (MPLF package) and lead frame with metal body (MLF package)) under various measurement conditions. In comparison with the MPLF packaged VLEDs, the MLF packaged VLEDs exhibited relatively lower junction temperature and thermal resistance values due to the better heat dissipation capability, leading to further improved optical, spectral, and thermal device characteristics. Thermal simulations of the VLEDs with two different packages were performed using three-dimensional steady-state device models to theoretically calculate their thermal and mechanical behaviours. The maximum temperatures, internal temperature distributions, and thermomechanical stresses were analysed by a finite element method.

Located at the important tumor suppressor locus, 3p22, PLCD1 encodes an enzyme that mediates regulatory signaling of energy metabolism, calcium homeostasis and intracellular movements. We identified PLCD1 as a downregulated gene in aerodigestive carcinomas through expression profiling and epigenetic characterization. We found that PLCD1 was expressed in all normal adult tissues but low or silenced in 84% (16/19) gastric cancer cell lines, well correlated with its CpG island (CGI) methylation status. Methylation was further detected in 62% (61/98) gastric primary tumors, but none of normal gastric mucosa tissues. PLCD1 methylation was significantly correlated with tumor high stage. Detailed methylation analysis of 37 CpG sites at the PLCD1 CGI by bisulfite genomic sequencing confirmed its methylation. PLCD1 silencing could be reversed by pharmacological demethylation with 5-aza-2′-deoxycytidine, indicating a direct epigenetic silencing. Ectopic expression of PLCD1 in silenced gastric tumor cells dramatically inhibited their clonogenicity and migration, possibly through downregulating MMP7 expression and hampering the reorganization of cytoskeleton through cofilin inactivation by phosphorylation. Thus, epigenetic inactivation of PLCD1 is common and tumor-specific in gastric cancer, and PLCD1 acts as a functional tumor suppressor involved in gastric carcinogenesis.Oncogene (2009) 28, 2466–2475; doi:10.1038/onc.2009.92; published online 18 May 2009
Decode-and-Forward Two-Path Half-Duplex Relaying: Diversity-Multiplexing Tradeoff Analysis

<P>Two-path or successive relaying has recently emerged as a promising cooperative communication protocol to improve spectral efficiency in half-duplex relaying networks. In this paper, we consider decode-and-forward (DF) version of the two-path relaying protocol. We analyze the fundamental performance of this protocol in terms of the diversity-multiplexing tradeoff (DMT). We first derive the DMT for this protocol, where perfect decoding at the relays is assumed, and show that it approaches the 3 x 1 multiple-input single-output (MISO) DMT. We then remove the assumption of perfect decoding at the relays and derive the closed-form expression of the achievable DMT based on the relative distances between nodes. Specifically, we found that for sufficiently long transmission length, if the average source-relay SNR is at least 2.5 times (measured in dB) of other links, the 3 x 1 MISO DMT is achieved. Successive interference cancellation at the relays is also proposed to further improve the performance of the DMT.</P>

In order to improve the availability of phytase and probiotics together, a phytase gene from Aspergillus ficuum has been expressed in Lactobacillus. In this study, the transformed Lactobacillus with phytase gene was fed to pigs to determine its effect on pig production, feed conversion and gut microbes. Forty eight, 60-day-old, castrated pigs (Duroc${\times}$Landrace${\times}$Pietrain) were assigned to 6 groups, 8 pigs for each group. Group 1 was the control, group 2 was added with chlortetracycline (500 mg/kg), group 3 was added with the transformed Lactobacillus (500 mg/kg) with 20% (w/w) of calcium monohydrogen phosphate (CMP, $CaHPO_{4}$) removed, group 4 was added with the natural Lactobacillus (500 mg/kg) with 20% (w/w) of CMP removed, group 5 was added with the transformed Lactobacillus (500 mg/kg) with 40% (w/w) of CMP removed, group 6 was added with phytase (500 mg/kg) with 40% (w/w) of CMP removed. The results showed: i) the average daily gain (ADG) was improved in groups 2, 3 and 4 (p<0.05); ii) the diarrhea rates in the groups added with Lactobacillus were lower than in the other groups (p<0.05), in which the transformed Lactobacillus had more effect on reducing digestive disease; iii) the transformed Lactobacillus was most effective in improving the digestibilities of crude protein (CP), calcium (Ca), phosphorus (P), compared with the other groups (p<0.05); iv) Lactobacillus could increase lactic acid bacterium number and ammonia concentrations, and decrease pH values and E. coli number in pig feces (p<0.05); v) the phytase activity in the feces of pigs fed with the transformed Lactobacillus was 133.32 U/g, which was higher than in group 4 (9.58 U/g, p<0.05), and was almost the same as group 6 (135.94 U/g); vi) the transformed Lactobacillus could increase serum concentrations of IgA, triglyceride, and glutamic oxaloacetic transaminase activity (p<0.05), and had no significant effect on other serum indexes (p>0.05).
