        Infinite time interval RBSDEs with non-Lipschitz coefficients

        Semi-Rational Screening of Probiotics from the Fecal Flora of Healthy Adults against DSS-Induced Colitis Mice by Enhancing Anti-Inflammatory Activity and Modulating the Gut Microbiota

        Ulcerative colitis (UC), a chronic inflammatory bowel disease, substantially impacts patients’ health-related quality of life. In this study, an effective strategy for discovering high-efficiency probiotics has been developed. First, in order to survive in the conditions of the stomach and intestine, high bile salt-resistant and strong acid-resistant strains were screened out from the fecal flora of healthy adults. Next, the probiotic candidates were rescreened by examining the induction ability of IL-10 (anti-inflammatory factor) production in dextran sodium sulfate (DSS)-induced colitis mice, and Lactobacillus sakei 07 (L07) was identified and selected as probiotic P. In the end, fourteen bifidobacterium strains isolated from stools of healthy males were examined for their antimicrobial activity. Bifidobacterium bifidum B10 (73.75% inhibition rate) was selected as probiotic B. Moreover, the colonic IL-6 and TNF-α expression of the DSS-induced colitis mice treated with L. sakei 07 (L07) - B. bifidum B10 combination (PB) significantly decreased and the IL-10 expression was up-regulated by PB compared to the DSS group. Furthermore, Bacteroidetes and Actinobacteria decreased and Firmicutes increased in the DSS group mice, significantly. More interestingly, the intestinal flora biodiversity of DSS colitis mice was increased by PB. Of those, the level of B. bifidum increased significantly. The Bacteriodetes/Firmicutes (B/F) ratio increased, and the concentration of homocysteine and LPS in plasma was down-regulated by PB in the DSS-induced colitis mice. Upon administration of PB, the intestinal permeability of the the DSS-induced colitis mice was decreased by approximately 2.01-fold. This method is expected to be used in high-throughput screening of the probiotics against colitis. In addition, the L. sakei 07 - B. bifidum B10 combination holds potential in UC remission by immunomodulatory and gut microbiota modulation.

        Exogenous 5-aminolevulinic acid promotes plant growth and salinity tolerance of grape rootstocks in coastal areas

        Salinity stress is a key factor aff ecting grape production in coastal areas. Two grape rootstock cultivars, ‘Beta’ (sensitive to salt stress) and ‘3309 C’ (resistant to salt stress), were used to investigate the physiological role of 5-aminolevlinic acid (5-ALA) in salinity tolerance. Functional capacities in the rhizosphere, leaf phenotype, photosynthetic parameters, antioxidant enzymes activities and certain physiological indexes were determined after treatment with seawater or seawa- ter followed by 100 mg/L 5-ALA application. The results showed that the functional capacities of the rhizosphere were distinct between the two management regimes. Microbial biomass and soil enzyme activities were reduced by salinity but promoted by 5-ALA. Salinity also suppressed the overall plant growth, leaf chlorophyll content, photosynthetic rate, maxi- mal photochemical effi ciency (Fv/Fm), photochemical quenching (qP), electron transport rate (ETR), root activity, ATP content, ATPase activity, and activities of the antioxidant enzymes ascorbate peroxidase (APX), peroxidase (POD), cata- lase (CAT), and superoxide dismutase (SOD) in both cultivars. Although plant growth, chlorophyll content, and activities of antioxidant enzymes in Beta were decreased to a greater extent than those in 3309 C under salinity stress, exogenous application of 5-ALA signifi cantly improved plant growth along with enhancement of all the above photosynthetic param- eters and activities of the four antioxidant enzymes in both cultivars. Seawater treatment alone signifi cantly increased non- photochemical quenching (NPQ), relative electrical conductivity (REC), and malondialdehyde (MDA) contents in leaves, especially for Beta. However, 5-ALA treatment decreased the levels of NPQ, REC, and MDA, but increased ATP content and ATPase activity in roots and leaves of both cultivars. Thus, application of 5-ALA would be benefi cial to improve the salt tolerance of grape rootstocks grown in coastal areas.

        Combinatorial nanococktails via selfassembling lipid prodrugs for synergistically overcoming drug resistance and effective cancer therapy

        Background: Circular RNAs (circRNAs) have important functions in many fields of cancer biology. In particular, we previously reported that the oncogenic circRNA, circPRMT5, has a major role in bladder cancer progression. Therapy based on circRNAs have good prospects as anticancer strategies. While anti-circRNAs are emerging as therapeutics, the specific in vivo delivery of anti-circRNAs into cancer cells has not been reported and remains challenging. Methods: Synthesized chrysotile nanotubes (SCNTs) with a relatively uniform length (~ 200 nm) have been designed to deliver an siRNA against the oncogenic circPRMT5 (si-circPRMT5) inhibit circPRMT5. In addition, the antitumor effects and safety evaluation of SCNTs/si-circPRMT5 was assessed with a series of in vitro and in vivo assays. Results: The results showed that SCNTs/si-circPRMT5 nanomaterials prolong si-circPRMT5’s half-life in circulation, enhance its specific uptake by tumor cells, and maximize the silencing efficiency of circPRMT5. In vitro, SCNTs encapsulating si-circPRMT5 could inhibit bladder cancer cell growth and progression. In vivo, SCNTs/si-circPRMT5 inhibited growth and metastasis in three bladder tumor models (a subcutaneous model, a tail vein injection lung metastatic model, and an in situ model) without obvious toxicities. Mechanistic study showed that SCNTs/sicircPRMT5 regulated the miR-30c/SNAIL1/E-cadherin axis, inhibiting bladder cancer growth and progression. Conclusion: The results highlight the potential therapeutic utility of SCNTs/si-circPRMT5 to deliver si-circPRMT5 to treat bladder cancer.         Structural evolution of the thermal conversion products of modified coal tar pitch

        Coal tar pitch is a product with high carbon content and aromatic compounds. Modified coal tar pitch is a high quality raw material for the preparation of intermediate phase pitch, needle coke, carbon microspheres, et al. In this paper, modified coal tar pitch was used as raw material, nitrogen was used as protective gas, and thermal conversion was carried out at constant temperatures (370, 390, 410, 420 °C). Polarized light microscopy, SEM, elemental analysis, FTIR spectroscopy, Raman spectroscopy and XRD diffraction combined with split-peak fitting were used to characterize the microstructures of the thermal transformation products. The results showed that the Iar and CH3/ CH2 contents of the products increased with the gradual increase of the thermal conversion temperature, and the aromatic content increased. And the higher the temperature at the same heating rate, the more the ideal graphite microcrystal content, and the defective graphite microcrystals are converted into ideal graphite microcrystals during the thermal conversion process. When the reaction temperature exceeds 390 °C, the microstructure of the thermal transformation products is anisotropic spheres, and the small spheres fuse with each other and tend to be basin-like and mosaic structure as the temperature increases.

        Application of the French Codes to the Pressurized Thermal Shocks Assessment

        The integrity of a reactor pressure vessel (RPV) related to pressurized thermal shocks (PTSs)has been extensively studied. This paper introduces an integrity assessment of an RPVsubjected to a PTS transient based on the French codes. In the USA, the “screening criterion”for maximum allowable embrittlement of RPV material is developed based on theprobabilistic fracture mechanics. However, in the French RCC-M and RSE-M codes, whichare developed based on the deterministic fracture mechanics, there is no “screening criterion”. In this paper, the methodology in the RCC-M and RSE-M codes, which are used forPTS analysis, are firstly discussed. The bases of the French codes are compared with ASMEand FAVOR codes. A case study is also presented. The results show that the method in theRCC-M code that accounts for the influence of cladding on the stress intensity factor (SIF)may be nonconservative. The SIF almost doubles if the weld residual stress is considered. The approaches included in the codes differ in many aspects, which may result in significantdifferences in the assessment results. Therefore, homogenization of the codes inthe long time operation of nuclear power plants is needed.

        Back-Calculation Method of Rock Mass Pressure in a Shallow-Buried Super Large-Span Tunnel Using Upper-Bench CD Method

        Rock mass pressure has always been a research hotspot in the field of tunnel engineering, especially in the super large-span tunnel, which is characterized by flat section, large excavation span, and complex stress field. Based on the Letuan Tunnel (a bi-directional tunnel with eight traffic lanes) of Binlai expressway expansion project in Shandong Province, China, this paper focused on the calculation method of rock mass pressure and the evolution law of load release in the construction process of the super large-span tunnel excavated by upper-bench central diaphragm (CD) method. Based on field measured data of Letuan Tunnel, the deformation behavior of primary lining and the distribution state of rock mass pressure during the tunnel construction were analyzed. According to the bearing mode of supporting structure, the mechanical models of different construction stages were established. Then, the rock mass pressures in different construction stages were back-calculated using mechanical models and compared with the measured values, and the evolution law of load release during the tunnel construction was discussed. The study results show that the tunnel deformation and rock mass pressure were significantly affected by the construction process and support form, and the excavation span was the key factor affecting the stability of rock mass. For the shallow-buried super large-span tunnel constructed by upper-bench CD method, the primary support of upper bench was under eccentric pressure. The comparison between the back-calculated value and the field measured value indicated that they were similar, and the average relative error was 17.23%. According to the concept of load release coefficient proposed in this paper, the load release coefficient after the pilot tunnel ahead (Part I) excavation reached 63%, and the load release coefficient after the pilot tunnel behind excavation (Part II) was 37%, which means that the rock mass pressure of Part I is increased about 59% due to the excavation of Part II.

        Curcumin protects against the intestinal ischemia-reperfusion injury: involvement of the tight junction protein ZO-1 and TNF-α related mechanism

        Present study aimed to investigate the effect of curcumin-pretreatment on intestinal I/R injury and on intestinal mucosa barrier. Thirty Wistar rats were randomly divided into: sham, I/R, and curcumin groups (n=10). Animals in curcumin group were pretreated with curcumin by gastric gavage (200 mg/kg) for 2 days before I/R. Small intestine tissues were prepared for Haematoxylin & Eosin (H&E) staining. Serum diamine oxidase (DAO) and tumor necrosis factor (TNF)-α levels were measured. Expression of intestinal TNF-α and tight junction protein (ZO-1) proteins was detected by Western blot and/or immunohistochemistry. Serum DAO level and serum and intestinal TNF-α leves were significantly increased after I/R, and the values were markedly reduced by curcumin pretreatment although still higher than that of sham group (p<0.05 or p<0.001). H&E staining showed the significant injury to intestinal mucosa following I/R, and curcumin pretreatment significantly improved the histological structure of intestinal mucosa. I/R insult also induced significantly down-regulated expression of ZO-1, and the effect was dramatically attenuated by curcumin-pretreatment. Curcumin may protect the intestine from I/R injury through restoration of the epithelial structure, promotion of the recovery of intestinal permeability, as well as enhancement of ZO-1 protein expression, and this effect may be partly attributed to the TNF-α related pathway.
