        PTEN mutation predicts unfavorable fertility preserving treatment outcome in the young patients with endometrioid endometrial cancer and atypical hyperplasia

        Objective: This study aimed to investigate the impact of molecular classification and PTEN, KRAS and PIK3CA gene mutation on the outcome of fertility-preserving treatment in the patients with endometrioid endometrial cancer (EEC) and endometrial atypical hyperplasia (EAH). Methods: This is a single-center retrospective study. A total of 135 patients with EEC and EAH receiving fertility-preserving treatment and molecular classification were reviewed. The distribution of the four types of molecular classification was described. The impact of non-specific molecular profile (NSMP), mismatch repair-deficiency (MMRd), and PTEN, KRAS and PIK3CA gene mutation on the outcome of fertility-preserving treatment was analyzed. Results: Of the patients analyzed, 86.7% (117/136) were classified as having NSMP; 14 (10.4%), MMRd; 1 (0.7%), POLEmut EAH; and 3 (2.2%), p53abn EEC. The patients having NSMP and MMRd achieved similar 16-, 32-, and 48-week complete response rates. The patients harboring tier I and tier II PTEN mutations (PTENmut-Clin) achieved lower cumulative 32-week CR rates than those with PTEN-others (without PTENmut-Clin) (22/47, 46.8% vs. 50/74, 67.6%; p=0.023; odds ratio=0.422; 95% confidence interval [CI]=0.199–0.896). Insulin-resistance (hazard ratio [HR]=0.435; 95% CI=0.269–0.702; p=0.001) and PTENmut-Clin (HR=0.535; 95% CI=0.324–0.885; p=0.015) were independent negative predictors for lower 32-week CR rates. Conclusion: PTENmut-Clin is an independent risk factor for unfavorable fertility-preserving treatment outcomes in the patients with EEC and EAH. The patients with MMRd receiving fertility-preserving treatment achieved outcomes similar to those of the patients with NSMP. The molecular profiles might guide fertility-preserving treatment in the prognosis and clinical decisions.

        Genistein Alleviates β-Amyloid-Induced Inflammatory Damage Through Regulating Toll-Like Receptor 4/Nuclear Factor κB

        Genistein (GEN), a major soybean isoflavone (SIF), might possess neuroprotective properties through its antiinflammatory activity. We hypothesized that GEN could prevent the inflammatory damage detected in C6 cells induced by β-amyloid peptides 25-35 (Aβ25-35). Accordingly, we evaluated the inflammatory damage induced by Aβ25-35 and the protective effect of GEN against Aβ25-35 in C6 cells. In our study, the C6 glial cells (rats glioma cell lines) were preincubated with or without GEN for 2 h following incubation with Aβ25-35 for another 24 h. Then, methylthiazolyl tetrazolium (MTT) assay was used to assess the cell viability. Immunofluorescence staining was used to identify the C6 cells. Inflammatory factors tumor necrosis factor (TNF)-α and interleukin (IL)-1β were analyzed by using enzyme-linked immunosorbent assay (ELISA). Western blot analysis and reverse transcription-polymerase chain reaction analysis were performed to assess the expression of Toll-like receptors 4 (TLR4), inhibitor of kappaB-alpha (IκB-α). The current results showed that GEN could alleviate Aβ25-35-induced cell apoptosis and prevent Aβ25-35-induced TNF-α and IL-1β release from C6 cells. In addition, GEN prevented Aβ25-35-induced upregulation of the gene and protein expression of TLR4, and GEN significantly upregulated the expression of IκB-α in C6 cells damaged by Aβ25-35. These results suggest that GEN can alleviate the inflammatory stress caused by Aβ25-35 treatment, which might be associated with the neuroprotective effect of GEN regulating the TLR4/NFκB signaling pathway.

        Semi-Rational Screening of Probiotics from the Fecal Flora of Healthy Adults against DSS-Induced Colitis Mice by Enhancing Anti-Inflammatory Activity and Modulating the Gut Microbiota

        Ulcerative colitis (UC), a chronic inflammatory bowel disease, substantially impacts patients’ health-related quality of life. In this study, an effective strategy for discovering high-efficiency probiotics has been developed. First, in order to survive in the conditions of the stomach and intestine, high bile salt-resistant and strong acid-resistant strains were screened out from the fecal flora of healthy adults. Next, the probiotic candidates were rescreened by examining the induction ability of IL-10 (anti-inflammatory factor) production in dextran sodium sulfate (DSS)-induced colitis mice, and Lactobacillus sakei 07 (L07) was identified and selected as probiotic P. In the end, fourteen bifidobacterium strains isolated from stools of healthy males were examined for their antimicrobial activity. Bifidobacterium bifidum B10 (73.75% inhibition rate) was selected as probiotic B. Moreover, the colonic IL-6 and TNF-α expression of the DSS-induced colitis mice treated with L. sakei 07 (L07) - B. bifidum B10 combination (PB) significantly decreased and the IL-10 expression was up-regulated by PB compared to the DSS group. Furthermore, Bacteroidetes and Actinobacteria decreased and Firmicutes increased in the DSS group mice, significantly. More interestingly, the intestinal flora biodiversity of DSS colitis mice was increased by PB. Of those, the level of B. bifidum increased significantly. The Bacteriodetes/Firmicutes (B/F) ratio increased, and the concentration of homocysteine and LPS in plasma was down-regulated by PB in the DSS-induced colitis mice. Upon administration of PB, the intestinal permeability of the the DSS-induced colitis mice was decreased by approximately 2.01-fold. This method is expected to be used in high-throughput screening of the probiotics against colitis. In addition, the L. sakei 07 - B. bifidum B10 combination holds potential in UC remission by immunomodulatory and gut microbiota modulation.

        Bayesian Rules Based Optimal Defense Strategies for Clustered WSNs

        Considering the topology of hierarchical tree structure, each cluster in WSNs is faced with various attacks launched by malicious nodes, which include network eavesdropping, channel interference and data tampering. The existing intrusion detection algorithm does not take into consideration the resource constraints of cluster heads and sensor nodes. Due to application requirements, sensor nodes in WSNs are deployed with approximately uncorrelated security weights. In our study, a novel and versatile intrusion detection system (IDS) for the optimal defense strategy is primarily introduced. Given the flexibility that wireless communication provides, it is unreasonable to expect malicious nodes will demonstrate a fixed behavior over time. Instead, malicious nodes can dynamically update the attack strategy in response to the IDS in each game stage. Thus, a multi-stage intrusion detection game (MIDG) based on Bayesian rules is proposed. In order to formulate the solution of MIDG, an in-depth analysis on the Bayesian equilibrium is performed iteratively. Depending on the MIDG theoretical analysis, the optimal behaviors of rational attackers and defenders are derived and calculated accurately. The numerical experimental results validate the effectiveness and robustness of the proposed scheme.

        SSR analysis reveals molecular variation and genetic diversity in Malus sieversii with different stress resistance levels in China

        Malus sieversii, a valuable crop in Xinjiang, China, is an important natural resource for researching the genetic diversity and phylogenetic evolution of the genus Malus worldwide. Samples from 152 M. sieversii individuals with different symptom grades of deadwood were collected from Gongliu and Xinyuan and analysed with 21 pairs of simple sequence repeat (SSR) primers, revealing molecular genetic characteristics and phylogeny of different groups and individuals. The samples showed high genetic diversity at the species level (Na = 10.76, Ne = 4.37, H = 0.73, I = 1.65, Ho = 0.71, and He = 0.73). Variation was mainly found within groups with lower genetic differentiation (Fst = 0.092) and higher gene flow (Nm = 2.67). A total of 226 alleles were obtained, of which 25 exclusive alleles were from samples with < 40% deadwood rate, and 23 exclusive alleles were from samples with > 40% deadwood rate. Specific bands relating to individuals with deadwood rate < 10% (10 exclusive alleles) or > 60% (7 exclusive alleles) were amplified with 11 pairs of SSR primers. The number of exclusive alleles from M. sieversii plants collected in Gongliu was 52 and from those collected in Xinyuan was 24. Using unweighted pair-group method with arithmetic cluster analysis, the groups with different symptom grades from different sampling sites were shown to be clearly differentiated and formed several discrete subclades. Significantly, the six groups from Gongliu were further classified into two subclusters: Gongliu 1, including three groups with < 40% deadwood rate, and Gongliu 2, including three groups with > 40% deadwood rate.

        Density measurement and equal density temperature of CO2+brine from Dagang - formation from 313 to 363 K

        Densities of CO2+Dagang - formation brine solution were measured by a magnetic suspension balance(MSB) in the pressure range from (10 to 18) MPa, at the temperatures from (313.15 to 363.15) K and CO2 mass frac-tions at 0, 0.0101, 0.0198 and 0.0299. The experimental results revealed that the solution densities increased linearlywith the increasing pressure and CO2 concentration, while decreasing with the increasing temperatures in the experi-mental range. When the temperature increased from (313.15 to 363.15) K, the slopes of the densities versus (vs.) CO2mass fractions decreased from (0.193 to 0.106) g·cm−3. A correlation equation was developed based on thermody-namic theory and experimental data. The absolute average deviation between the correlation equation and the experi-mental data was 0.05%, and the maximum deviation was 0.37% for the density of CO2+water/brine solution in com-mon geological storage conditions. According to the density of CO2 - free brine and apparent molar volume of CO2 inbrine, the equal density temperature (Te) of CO2+Dagang brine solution was obtained at 464.67 K when pressure is10MPa, which means that the density of brine dissolved with CO2 will be less than that of CO2-free brine when thetemperature is higher than 464.67 K at 10MPa. In this work the formation temperature of the Dagang oilfield reser-voir is from 313.15 K to 363.15 K, which is lower than the equal density temperature. Therefore, the safety of CO2 stor-age in Dagang oilfield reservoir can be guaranteed.

      • A facile approach to enhance antigen response for personalized cancer vaccination

        <P>Existing strategies to enhance peptide immunogenicity for cancer vaccination generally require direct peptide alteration, which, beyond practical issues, may impact peptide presentation and result in vaccine variability. Here, we report a simple adsorption approach using polyethyleneimine (PEI) in a mesoporous silica microrod (MSR) vaccine to enhance antigen immunogenicity. The MSR-PEI vaccine significantly enhanced host dendritic cell activation and T-cell response over the existing MSR vaccine and bolus vaccine formulations. Impressively, a single injection of the MSR-PEI vaccine using an E7 peptide completely eradicated large, established TC-1 tumours in about 80% of mice and generated immunological memory. When immunized with a pool of B16F10 or CT26 neoantigens, the MSR-PEI vaccine eradicated established lung metastases, controlled tumour growth and synergized with anti-CTLA4 therapy. Our findings from three independent tumour models suggest that the MSR-PEI vaccine approach may serve as a facile and powerful multi-antigen platform to enable robust personalized cancer vaccination.</P>
