        Potential of the Quantitative Trait Loci Mapping Using Crossbred Population

        In the process of crossbreeding, the linkage disequilibria between the quantitative trait loci (QTL) and their linked markers were reduced gradually with increasing generations. To study the potential of QTL mapping using the crossbred population, we presented a mixed effect model that treated the mean allelic value of the different founder populations as the fixed effect and the allelic deviation from the population mean as random effect. It was assumed that there were fifty QTLs having effect on the trait variation, the population mean and variance were divided to each QTL in founder generation in our model. Only the additive effect was considered in this model for simulation. Six schemes (S1-S6) of crossbreeding were studied. The selection index was used to evaluate the synthetic breeding value of two traits of the individual in the scheme of S2, S4 and S6, and the individuals with high selection index were chosen as the parents of the next generation. Random selection was used in the scheme of S1, S3 and S5. In this study, we premised a QTL explained 40% of the genetic variance was located in a region of 20 cM by the linkage analysis previously. The log likelihood ratio (log LR) was calculated to determine the presence of a QTL at the particular chromosomal position in each of the generations from the fourth to twentieth. The profiles of log LR and the number of the highest log LR located in the region of 5, 10 and 20 cM were compared between different generations and schemes. The profiles and the correct number reduced gradually with the generations increasing in the schemes of S2, S4 and S6, but both of them increased in the schemes of S1, S3 and S5. From the results, we concluded that the crossbreeding population undergoing random selection was suitable for improving the resolution of QTL mapping. Even experiencing index selection, there was still enough variation existing within the crossbred population before the fourteenth generation that could be used to refine the location of QTL in the chromosome region.

        Ultrasonic Vibration and Rheocasting for Refinement of Mg–Zn–Y Alloy Reinforced with LPSO Structure

        In this work, ultrasonic vibration (UV) and rheo-squeeze casting was first applied on the Mg alloy reinforced with long periodstacking ordered (LPSO) structure. The semisolid slurry of Mg–Zn–Y alloy was prepared by UV and processed by rheosqueezecasting in succession. The effects of UV, Zr addition and squeeze pressure on microstructure of semisolid Mg–Zn–Yalloy were studied. The results revealed that the synergic effect of UV and Zr addition generated a finer microstructure thaneither one alone when preparing the slurries. Rheo-squeeze casting could significantly refine the LPSO structure and α-Mgmatrix in Mg96.9Zn1Y2Zr0.1alloy without changing the phase compositions or the type of LPSO structure. When the squeezepressure increased from 0 to 400 MPa, the block LPSO structure was completely eliminated and the average thickness ofLPSO structure decreased from 9.8 to 4.3 μm. Under 400 MPa squeeze pressure, the tensile strength and elongation of therheocast Mg96.9Zn1Y2Zr0.1alloy reached the maximum values, which were 234 MPa and 17.6%, respectively, due to its fineα-Mg matrix (α1-Mg and α2-Mg grains) and LPSO structure.

        Effective Pre-rating Method Based on Users' Dichotomous Preferences and Average Ratings Fusion for Recommender Systems

        With an increase in the scale of recommender systems, users’ rating data tend to be extremely sparse. Some methods have been utilized to alleviate this problem; nevertheless, it has not been satisfactorily solved yet. Therefore, we propose an effective pre-rating method based on users’ dichotomous preferences and average ratings fusion. First, based on a user-item ratings matrix, a new user-item preference matrix was constructed to analyze and model user preferences. The items were then divided into two categories based on a parameterized dynamic threshold. The missing ratings for items that the user was not interested in were directly filled with the lowest user rating; otherwise, fusion ratings were utilized to fill the missing ratings. Further, an optimized parameter λ was introduced to adjust their weights. Finally, we verified our method on a standard dataset. The experimental results show that our method can effectively reduce the prediction error and improve the recommendation quality. As for its application, our method is effective, but not complicated.

        Expression Characterization, Polymorphism and Chromosomal Location of the Porcine Calsarcin-3 Gene

        Calcineurin is a calmodulin dependent protein that functions as a regulator of muscle cell growth and function. Agents capable of interacting with calcineurin could have important applications in muscle disease treatment as well as in the improvement of livestock production. Calsarcins comprise a family of muscle-specific calcineurin binding proteins which play an important role in modulating the function of calcineurin in muscle cells. Recently, we described the first two members of the calsarcin family (calsarcin-1 and calsarcin-2) in the pig. Here, we characterized the third member of the calsarcin family, calsarcin-3, which is also expressed specifically in skeletal muscle. However, unlike calsarcin-1 and calsarcin-2, the calsarcin-3 mRNA expression in skeletal muscle kept rising throughout the prenatal and postnatal development periods. In addition, radiation hybrid mapping indicated that porcine calsarcin-3 mapped to the distal end of the q arm of pig chromosome 2 (SSC2). A C/T single nucleotide polymorphism site in exon 5 was genotyped using the denaturing high performance liquid chromatography (DHPLC) method and the allele frequencies at this locus were significantly different among breeds.

        The role of roxadustat in chronic kidney disease patients complicated with anemia

        The incidence of chronic kidney disease (CKD) is increasing worldwide and the current prevalence rate is 13.4%. There are > 120 million CKD patients in China and this number is expected to increase. One of the main abnormalities in patients with CKD and kidney impairment is decreased synthesis of erythropoietin (EPO), which causes anemia and affects iron metabolism. The probability of developing is higher in anemia patients with CKD than in the general population, and the incidence increases as kidney function decreases. Deficient EPO production by the kidney is the most important cause of renal anemia. Notably, anemia in patients with CKD has multiple causes, such as bleeding caused by platelet dysfunction, iron deficiency due to digestive and absorption disorders of the gastrointestinal tract, and shorter red blood cell life. Anemia is also a leading cause of hospitalization in patients with CKD. A new oral medication to treat renal anemia, the hypoxia-inducible factor prolyl hydroxylase inhibitor called roxadustat (FG-4592), regulates iron metabolism and promotes erythropoiesis. This drug has a therapeutic effect on patients with CKD. Roxadustat showed advantages over EPO in clinical experiments. This review summarizes the mechanisms of action, clinical applications, effectiveness, and safety of roxadustat.

        In this paper, we consider the density-dependent magnetohydrodynamic equations, and provide a regularity criterion involving the velocity field in BMO space, which extends [J.S. Fan, F.C.Li, G. Nakamura, Z. Tan, Regularity criteria for the three-dimensional magnetohydrodynamic equations, J. Differential Equations, 256 (2014), 2858-2875]. Moreover, we applied our method of proof to the density-dependent Hall-MHD system.

        Transcriptome sequencing combined with bioinformatics predicts potential genes and pathways associated with bupivacaine-induced apoptosis

        This study aimed to explore the potential genes and pathways associated with bupivacaine-induced apoptosis. Human neuroblastoma cell line SH-SY5Y was used in this study. The effect of bupivacaine on cell viability of SH-SY5Y was detected by Cell Counting Kit-8. Transcriptome sequencing was performed for SH-SY5Y cells that were treated and untreated with bupivacaine based on the HiSeq 4000 sequencing platform. The sequencing results were analyzed using bioinformatics methods, including differentially expressed genes (DEGs) identification, functional enrichment analysis, protein–protein interaction (PPI) network analysis and module analysis. The cell viability of SH-SY5Y cells decreased significantly after bupivacaine treatment (p < .01). Based on the HiSeq 4000 sequencing platform, we obtained a global overview of the transcriptome of SH-SY5Y treated with/ without bupivacaine. Bioinformatics analysis identified 335 up-regulated and 294 down-regulated DEGs in bupivacaine group. They were significantly enriched in cell cycle-associated functions and pathways and cAMP signaling pathway. In the PPI network, proliferating cell nuclear antigen (PCNA), v-Akt murine thymoma viral oncogene homolog 3 (AKT3), cyclin-dependent kinase inhibitor 1A (CDKN1A) and cell division cycle 6 (CDC6) had high topology scores. Module analysis obtained two sub-network modules (cluster 1 and cluster 2). PCNA, CDC6, CDKN1A and AKT3 may play important roles in bupivacaine-induced apoptosis. Additionally, bupivacaine may also induce apoptosis via pathways of cell cycle and cAMP signaling pathway.

        A reduced time-varying model for a long beam on elastic foundation under moving loads

        Dynamics of a long beam on the elastic foundation subjected to moving loads is studied in the present paper. The sliding window technique is used to dynamically truncate the long beam and a reduced time-varying beam system is obtained. The Hamilton’s principle is employed to establish the equations of motion of the reduced system. The variable separation method is adopted to solve dynamical responses of the reduced system. Examples of a long simply supported Timoshenko beam on the nonlinear foundation subjected to a single moving load and multiple loads are included. Numerical results of the reduced model compared with the ones obtained from the moving element model adapted in literature are carried out to show the validity and the good efficiency of the method proposed in the present paper.

        Evaluation of the Genetic Relationship among Ten Chinese Indigenous Pig Breeds with Twenty-six Microsatellite Markers

        The genetic diversities and relationships of 10 Chinese indigenous pig breeds and three exotic pig breeds have been evaluated using 26 microsatellites recommended by the Food and Agriculture Organization & the International Society of Animal Genetics (FAO-ISAG). The allele frequencies, genetic heterozygosity (H) and polymorphism information content (PIC) have been calculated. The results showed that genetic diversity of Chinese indigenous pig breeds is higher than that of the introduced pig breeds. The clustering of 10 breeds is generally consistent with their geographical distribution.

        A retrospective study on the efficacy of Roxadustat in peritoneal dialysis patients with erythropoietin hyporesponsiveness

        Background/Aims: Roxadustat, an oral medication for treating renal anemia, is a hypoxia-inducible factor prolyl hydroxylase inhibitor used for regulating iron metabolism and promoting erythropoiesis. To investigate the efficacy and safety of roxadustat in patients undergoing peritoneal dialysis (PD) with erythropoietin hyporesponsiveness. Methods: Single-center, retrospective study, 81 PD patients (with erythropoietin hyporesponsiveness) were divided into the roxadustat group (n = 61) and erythropoiesis-stimulating agents (ESAs) group (n = 20). Hemoglobin (Hb), total cholesterol, intact parathyroid hormone (iPTH), brain natriuretic peptide (BNP), related indicators of cardiac function and high-sensitivity C-reactive protein (hs-CRP) were collected. Additionally, adverse events were also recorded. The follow-up period was 16 weeks. Results: The two groups exhibited similar baseline demographic and clinical characteristics. At baseline, the roxadustat group had a mean Hb level of 89.8 ± 18.9 g/L, while the ESAs group had a mean Hb level of 95.2 ± 16.0 g/L. By week 16, the Hb levels had increased to 118 ± 19.8 g/L (p < 0.05) in the roxadustat group and 101 ± 19.3 g/L (p > 0.05) in the ESAs group. The efficacy of roxadustat in improving anemia was not influenced by baseline levels of hs-CRP and iPTH. Cholesterol was decreased in the roxadustat group without statin use. An increase in left ventricular ejection fraction and stabilization of BNP were observed in the roxadustat group. Conclusions: For PD patients with erythropoietin hyporesponsiveness, roxadustat can significantly improve renal anemia. The efficacy of roxadustat in improving renal anemia was not affected by baseline levels of hs-CRP0 and iPTH.
