        Electroacupuncture-Modulated MiR106b-5p Expression Enhances Autophagy by Targeting Beclin-1 to Promote Motor Function Recovery After Spinal Cord Injury in Rats

        Objective: Electroacupuncture (EA) has a definite effect on the treatment of spinal cord injuries (SCIs), but its underlying molecular mechanism remains unclear. Meanwhile, MiR-106b-5p is an autophagy- and apoptosis-related microribonucleic acid, but whether it regulates the progression of autophagy and apoptosis in SCIs is yet undetermined. As such, this study aimed to elucidate the involvement of miR-106b-5p in the EA treatment of an SCI. Methods: The miR-106b-5p level was detected by quantitative real-time polymerase chain reaction. In vitro, SH-SY5Y cells were transfected with miR-106b-5p mimics or inhibitors to regulate the miR-106b-5p expression, while in vivo, SCI rats were treated with EA for 7 days at the bilateral Zusanli (ST36) and Jiaji (EX-B2) acupoints. The motor function was evaluated using the Basso-Beattie-Bresnahan (BBB) criteria. Further, autophagic vacuoles, pathological damage, and neuronal cell morphology were observed by transmission electron microscopy, as well as by hematoxylin and eosin and Nissl staining, respectively. Results: The miR-106b-5p level, which can interact directly with Beclin-1 by influencing its expression, as well as the expressions of P62, Caspase-3, and Bax, was upregulated after an SCI, but it decreased after EA. Moreover, the ratio of LC3-II to LC3-I was upregulated after EA. EA can enhance autophagy, reduce neuronal apoptosis, and minimize motor dysfunction and histopathological deficits after an SCI. More importantly, however, all the above effects induced by EA can be reversed after an injection of miR-106-5p agomir to produce an overexpression of miR-106b-5p. Conclusion: EA treatment could downregulate miR-106b-5p to alleviate SCI-mediated injuries by promoting autophagy and inhibiting apoptosis.

        Salp swarm algorithm with iterative mapping and local escaping for multi-level threshold image segmentation: a skin cancer dermoscopic case study

        If found and treated early, fast-growing skin cancers can dramatically prolong patients’ lives. Dermoscopy is a convenient and reliable tool during the fore-period detection stage of skin cancer, so the efficient processing of digital images of dermoscopy is particularly critical to improving the level of a skin cancer diagnosis. Notably, image segmentation is a part of image preprocessing and essential technical support in the process of image processing. In addition, multi-threshold image segmentation (MIS) technology is extensively used due to its straightforward and effective features. Many academics have coupled different meta-heuristic algorithms with MIS to raise image segmentation quality. Nonetheless, these meta-heuristic algorithms frequently enter local optima. Therefore, this paper suggests an improved salp swarm algorithm (ILSSA) method that combines iterative mapping and local escaping operator to address this drawback. Besides, this paper also proposes the ILSSA-based MIS approach, which is triumphantly utilized to segment dermoscopic images of skin cancer. This method uses two-dimensional (2D) Kapur’s entropy as the objective function and employs non-local means 2D histogram to represent the image information. Furthermore, an array of benchmark function test experiments demonstrated that ILSSA could alleviate the local optimal problem more effectively than other compared algorithms. Afterward, the skin cancer dermoscopy image segmentation experiment displayed that the proposed ILSSA-based MIS method obtained superior segmentation results than other MIS peers and was more adaptable at different thresholds.

      • The stiffness and buckling behavior of panels with stamping-induced imperfections

        With the decrease of sheet thickness, the stiffness and buckling behavior of automobile body panel are more and more concerned. In this paper, the stiffness and buckling behavior of panels with stamping-induced imperfection is studied. Imperfections caused in drawing and springback are obtained by finite element analysis and introduced to define the initial state of the panel before indenting. The indenting procedure is simulated in ABAQUS Standard. In order to solve the buckling problem, a viscous pressure that is proportional to the relative velocity between surfaces is added artificially. For comparison, a simplified model in which imperfections are not considered is also established. Comparing the simulation results with the reference experiment, we find that the stamping-induced imperfections lead to a more severe oil canning phenomena under a buckling load, although they increase panel’s initial stiffness a bit. Moreover, among all stamping-induced imperfections, the geometrical imperfection (surface deflection) is beneficial to increase panel’s buckling load, and the material imperfections (residual stress and uneven thickness) help to decrease panel’s buckling load and can make oil canning more severe during post-buckling. Influences of blank holder force, sheet thickness and material property on panel stiffness and buckling behavior are also studied. The work of this paper is beneficial to processing optimization, material selection, and reinforcement design for automotive exterior panels.

        Tannic Acid and Ferrous Sulfate Modified Kapok Fiber for Oil-water Separation

        Biodegradable oil absorbing materials have attracted extensive attention in dealing with oil spill pollution. Todevelop more excellent oleophilic oil sorbent, the combination of tannic acid and ferrous sulfate was employed to treat therenewable and biodegradable natural kapok fiber. The as-prepared kapok showed superhydrophobic characteristics with highwater contact for the formation of tannic acid and ferrous ion complex on kapok surface, which was confirmed by scanningelectron microscopy observation and infrared spectroscopy and X-ray diffraction analysis. The preparation process of themodified kapok fiber was optimized. When the sodium chlorite pretreated kapok fiber was firstly treated with 4.25 g/l tannicacid and subsequently immersed in 4.25 g/l tannic acid and 6.25 g/l ferrous sulfate mixture at 60 °C for 15 min, the aspreparedkapok fiber presented the best oil absorption capacity and hydrophobicity (water contact angle 150.7 °). The treatedkapok had good adsorption capacity for oils and organic solvents, 48.2, 41.6, 53.4, 52.8, 49.2, 41.2 g/g for vegetable oil,diesel oil, silicone oil, carbon tetrachloride, DMF and n-hexane, respectively. In addition, the tannic acid and ferrous sulfatetreated kapok was recycled for six times through simple mechanical squeezing, and showed good oil-water separationperformance oil, indicating its potential application in the removal of spilled oil on water.

        Experiences of East Asian International Counseling Students during the COVID-19 Pandemic

        International counseling students, especially those from East Asia, were more vulnerable during the coronavirus (COVID-19) pandemic than their domestic counterparts. In the current study, we explored the lived experiences of ten master’s level East Asian international counseling students (EAICS) during the COVID-19 pandemic using a phenomenological design. Four overarching themes emerged from the analysis: (a) transition in COVID-19 pandemic, (b) being culturally minoritized, (c) positive experiences of the counseling program, and (d) positive action. The critical role of the academic community in supporting this marginalized group against racism and xenophobia and practical ways of facilitating EAICS’s professional development are discussed.

        The evolution of microstructure and resistance in electroplated copper films by linear integrated laser scanning annealing

        Thermal treatment is an effective way to decrease the resistivity and internal stress by inducing grain growth accompaniedwith redistribution of embedded impurities. With the narrowing of Cu interconnects in IC packaging, the increased resistanceis becoming the main issue that hindering the electrical performance of IC. Herein, a laser annealing method by linerscanning (LALS) to anneal the Cu interconnects were reported which provide a gradient thermal field for the crystallographic/microstructure transition. The impacts of laser annealing on the sheet resistance of the electroplating Cu films wereinvestigated in aspects of microstructure and phase field simulation. Cu films treated by LALS owned larger average grainsize, better recrystallization fraction, and significantly higher average grain aspect ratio than conventional annealing, whichindicated the increased driving force for grain boundaries evolution by LALS method. This study exhibited the direct evidenceon the impacts of laser annealing process on the resistance of electroplated Cu films. The laser annealing process witha local temperature gradient caused a significant decline in Cu electrical resistance compared to the conventional annealingprocess, indicating its extraordinary potential in improving Cu wire conductivity. This work will provide a scientific basis forselecting the post-treatment process for electrodeposited Cu films to achieve ideal electrical properties and microstructurein electronics industry applications.

        Curcumin Ameliorates Reserpine-Induced Gastrointestinal Mucosal Lesions Through Inhibiting IκB-α/NF-κB Pathway and Regulating Expression of Vasoactive Intestinal Peptide and Gastrin in Rats

        The objective of our study was to investigate whether curcumin protects against reserpine-induced gastrointestinal mucosal lesions (GMLs) in rats and to explore the mechanism of curcumin’s action. Sprague-Dawley rats were randomly divided into four groups: control group, reserpine-treated group, reserpine treatment group with curcumin at high dose (200 mg/kg), and reserpine treatment group with curcumin at low dose (100 mg/kg). Rats in reserpine-treated group were induced by intraperitoneally administered reserpine (0.5 mg/kg) for 28 days. TUNEL staining and hematoxylin and eosin staining were used to evaluate the apoptotic cells and morphologic changes. In addition, to explore the mechanism of curcumin in protecting GMLs, we used serum of experimental rats to assess the level of vasoactive intestinal peptide (VIP), gastrin, interleukin-6, interleukin-10, tumor necrosis factor-α and interferon-γ by ELISA and radioimmunoassay. The protein levels of NF-κB, p-IκB-α, IκB-α, Bcl-2, Bax, and cleaved-caspase-3 were examined by western blot analysis. Data were analyzed with SPSS 19.0 software package. Curcumin treatment prevented tissue damage and cell death in the reserpinetreated rats and effectively decreased inflammatory response and balanced the expression of VIP and gastrin in the reserpinetreated rats. NF-κB, p-IκB-α, Bax, and cleaved-caspase-3 were increased in the reserpine group, but the curcumin high-dose group inhibited them. Curcumin can target the IκB-α/NF-κB pathway to inhibit inflammatory response and regulate the level of VIP and gastrin in reserpine-induced GML rats.

      • Preparation and Characterization of Paclitaxel-loaded PLGA Nanoparticles Coated with Cationic SM5-1 Single-chain Antibody

        The purpose of this study was to develop paclitaxel-loaded poly(lactide-co-glycolide) (PLGA) nanoparticles coated with cationic SM5-1 single-chain antibody (scFv) containing a polylysine (SMFv-polylys). SM5-1 scFv (SMFv) is derived from SM5-1 monoclonal antibody, which binds to a 230 kDa membrane protein specifically expressed on melanoma, hepatocellular carcinoma and breast cancer cells. SMFv-polylys was expressed in Escherichia coli and purified by cation-exchange chromatography. Purified SMFv-polylys was fixed to paclitaxel-loaded PLGA nanoparticles to form paclitaxel-loaded PLGA nanoparticles coated with SMFv-polylys (Ptx-NP-S). Ptx-NP-S was shown to retain the specific antigen-binding affinity of SMFv-polylys to SM5-1 binding protein-positive Ch-hep-3 cells. Finally, the cytotoxicity of Ptx-NP-S was evaluated by a non-radioactive cell proliferation assay. It was demonstrated that Ptx-NP-S had significantly enhanced in vitro cytotoxicity against Ch-hep-3 cells as compared with non-targeted paclitaxel-loaded PLGA nanoparticles. In conclusion, our results suggest that cationic SMFv-polylys has been successfully generated and may be used as targeted ligand for preparing cancer-targeted nanoparticles.

        The deubiquitinating enzyme STAMBP is a newly discovered driver of triple-negative breast cancer progression that maintains RAI14 protein stability

        Triple-negative breast cancer (TNBC) is a heterogeneous malignancy in women. It is associated with poor prognosis, aggressive malignant behavior, and limited treatment options. In the ubiquitin‒proteasome system (UPS), deubiquitinases (DUBs) are potential therapeutic targets for various tumors. In this study, by performing unbiased siRNA screening, we identified STAMBP, a JAMM metalloprotease in the DUB family, as a driver of human TNBC tumor growth. Functionally, the knockdown of STAMBP inhibited the proliferation, migration, and invasion of multiple TNBC cell lines. Immunoprecipitation–mass spectrometry combined with functional and morphological analysis verified the interaction between STAMBP and the actin-binding protein RAI14. Mechanistically, STAMBP stabilized the RAI14 protein by suppressing the K48-linked ubiquitination of RAI14 and thus prevented its proteasomal degradation. Therefore, knocking down STAMBP resulted in the reduction in RAI14 protein levels and suppression of tumor growth in vitro and in vivo. Importantly, high levels of STAMBP were correlated with poor prognosis in TNBC patients. In summary, we reveal a previously unrecognized DUB pathway that promotes TNBC progression and provides a rationale for potential therapeutic interventions for the treatment of TNBC.
