Areca palm yellow leaf (AYL) disease caused by the 16SrI group phytoplasma is a serious threat to the development of the Areca palm industry in China. The 16S rRNA gene sequence was utilized to establish a rapid and efficient detection system efficient for the 16SrI-B subgroup AYL phytoplasma in China by loopmediated isothermal amplification (LAMP). The results showed that two sets of LAMP detection primers, 16SrDNA-2 and 16SrDNA-3, were efficient for 16SrIB subgroup AYL phytoplasma in China, with positive results appearing under reaction conditions of 64oC for 40 min. The lowest detection limit for the two LAMP detection assays was the same at 200 ag/μl, namely approximately 53 copies/μl of the target fragments. Phytoplasma was detected in all AYL disease samples from Baoting, Tunchang, and Wanning counties in Hainan province using the two sets of LAMP primers 16SrDNA-2 and 16SrDNA-3, whereas no phytoplasma was detected in the negative control. The LAMP method established in this study with comparatively high sensitivity and stability, provides reliable results that could be visually detected, making it suitable for application and research in rapid diagnosis of AYL disease, detection of seedlings with the pathogen and breeding of diseaseresistant Areca palm varieties.

Aflatoxin contamination of food and grain poses a serious economic and health problem globally. Aflatoxin B1 (AFB1) is extremely mutagenic and toxic as well as a potent carcinogen to both humans and livestock. In this study, the degradation of AFB1 by extracellular extract of Cladosporium uredinicola was examined using high-performance liquid chromatography (HPLC), thin-layer chromatography (TLC), and liquid chromatography mass spectrometry. Within 24 h of incubation, AFB1 was efficiently eliminated by the culture supernatant of C. uredinicola (84.5±5.7%) at 37oC; the elimination was proven to be enzymatic, and the enzyme was thermostable. The biotransformation products of AFB1 detected by HPLC and TLC were proven to be the same compound. Analysis with LCMS showed that AFB1 was bio-transformed to a structurally different compound (m/z=365 [M+Na]+), which is first reported. The cytotoxicity study to HeLa cells indicated that culture supernatant-treated AFB1 is less toxic as compared with AFB1.

Lots of minor constituents in herbal medicines have shown great potential to devolve into leadcompounds, and their extraction is attracting more and more attention in pharmaceutical industry. Thisstudy provided an example that showed efficient and selective extraction of minor bioactive naturalproducts (prenylatedflavonoids with anti-cancer activities) from an herbal medicine (licorice) using pureionic liquids as green solvents. After investigation of a variety of hydrophobic ionic liquids withultrasonic-assisted extraction, we found that [C8MIM]BF4 (1-octyl-3-methylimidazolium tetrafluor-oborate) was the most effective one for extraction offive representative compounds (isoangustone A,glycycoumarin, licoisoflavanone, licoricidin and glabridin), and solvent to solid ratio, extraction time,extraction temperature and soaking time were then optimized through single-factor experiments andresponse surface methodology. As a result, the established method had significantly higher extractionselectivity than conventional organic solvents, which could be attributed to significant physical change oflicorice microstructures after extraction, as well as strong interactions (especially hydrogen bondinteractions) between [C8MIM]BF4 and prenylatedflavonoids by mechanism study. In addition, 78.92% ofthe extracted prenylatedflavonoids could be readily recovered from [C8MIM]BF4 by using reversed-phasesolid phase extraction, and the recyclability of [C8MIM]BF4 was also confirmed.

This paper proposes a current sharing method for interleaved LLC resonant converter with secondary partial energy phase shift regulation. An AC voltage source with phase shift regulation is constructed and in series with main transformer at secondary side to balance the output currents. With secondary regulation ability, the current imbalance caused by the resonant tank tolerance can be compensated. The major part of power is delivered to load by main transformer, and small portion power is converted by the auxiliary parts, therefore, the energy loss of the conversion is almost as high as conventional LLC resonant converter. The steady state operating principle of the proposed converter is analyzed, and a prototype with two phases current sharing ability is built-up based on the theoretical analysis.

Loess is mostly distributed in an unsaturated state in nature, and the complexity of the engineering properties of unsaturated soil is mainly due to the existence of matric suction. Therefore, matric suction must be considered in investigating the mechanical properties of unsaturated loess. However, soils are most often subjected to three principal stresses with different magnitudes in practical engineering. For the sake of examining and discussing the mechanical behaviour of unsaturated natural loess under a complex stress path, a suction-controlled true triaxial apparatus with a rigid-flexible boundary is used to test unsaturated natural loess under a complicated stress path. Four trials of isotropic consolidation tests are conducted on natural loess under suction-controlled conditions via the true triaxial apparatus. The consolidation yield characteristics of the natural loess under different matric suctions are investigated. Forty-eight trials of consolidated drained true triaxial tests under suction-controlled conditions are conducted on unsaturated natural loess to examine and discuss the influence of the matric suction and intermediate principal stress parameter (b-value). The consolidated drained trials are performed under a constant net mean stress and a constant matric suction with different intermediate principal stress parameters (b-values). Stress-strain curves and failure envelopes of the natural loess are also presented. The results indicate that the stress-strain-strength response of unsaturated natural loess depends on the matric suction and intermediate principal stress parameter under true triaxial conditions.

Some long noncoding RNAs (lncRNAs), which harbor microRNAs in their gene sequence and are also known as microRNA host gene derived lncRNAs (lnc-MIRHGs), play a dominant role alongside miRNAs, or both perform biological functions synergistically or independently. However, only a small number of lnc-MIRHGs have been identified. Here, multiple liver injury datasets were analyzed to screen and identify the target lncRNA Mir122hg. Mir122hg was mainly enriched in liver tissues with human-mouse homology. In both CCl4-induced acute liver injury and Dgal/LPS-induced fulminant liver failure in mice, Mir122hg was sharply downregulated at the early stage, while a subsequent significant increase was only found in the CCl4 group with liver recovery. Overexpression and silencing assays confirmed that Mir122hg played a protective role in acute injury by promoting hepatocyte proliferation in vivo and in vitro. Consistent with the results of gene enrichment analysis, Mir122hg binding to C/EBPα affected its transcriptional repression, promoted gene transcription of downstream chemokines, Cxcl2, Cxcl3, and Cxcl5, and exerted pro-proliferative effects on hepatocytes through activation of the AKT/GSK-3β/p27 signaling pathway by CXC/CXCR2 complexes. This study identifies a novel lncRNA with protective effects in acute liver injury and demonstrates that the binding of Mir122hg-C/EBPα promotes hepatocyte proliferation via upregulation of CXC chemokine and activation of AKT signaling.

Two metal compounds, $[Co(phen)_2(H_2O)_2]{\cdot}2H_2SIP{\cdot}2H_2O$ 1 and $[Ni(phen)_3]{\cdot}2H_2SIP{\cdot}3H_2O$ 2, have been obtained by incorporating 1,10-phenanthroline (phen) and 5-sulfoisophthalic acid monosodium salt ($NaH_2SIP$) ligands under hydrothermal conditions. Meanwhile, the two compounds were characterized by element analysis, IR, XRD, TG-DTA and single-crystal X-ray diffraction. Both 1 and 2 present 3D supramolecular structures via O-H${\cdots}$O hydrogen bond interactions. Luminescent properties for 1 and 2 were also studied. The compound 1 has two fluorescence emission peaks centered at 398 nm attributed to the intraligand emission from the SIP ligand and at 438 nm assigned to the combined interaction of intraligand ${\pi}^*-{\pi}$ transitions of the phen ligand and ligand-to-metal-charge-transfer (LMCT) transitions (${\lambda}_{ex}$ = 233 nm). The compound 2 shows one emission band centered at 423 nm with a shoulder peak at 434 nm which may be originated from the intraligand ${\pi}^*-{\pi}$ transitions of the phen ligand (${\lambda}_{ex}$ = 266 nm).
