      • Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells

        IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.

        A Lectin with Mycelia Differentiation and Antiphytovirus Activities from the Edible Mushroom Agrocybe aegerita

        A lectin named AAL has been purified from the fruiting bodies of the edible mushroom Agrocybe aegerita. AAL consisted of two identical subunits of 15.8 kDa, its pI was about 3.8 determined by isoelectric focusing, and no carbohydrate was discerned. Being treated by pyrogultamate aminopeptidase, the blocked N-terminus of AAL was sequenced as QGVNIYNI. AAL agglutinated human and animal erythrocytes regardless of blood type or animal species. Its hemagglutinating activity was unaffected by acid or alkali treatment and demetalization or addition of divalent metals Mg^(2+), Ca^(2+) Zn^(2+). ALL was toxic to mice: its LD50 was 15.85 mg per kilogram body weight by intraperitoneal injection. In this study, two novel activities of AAL were proved. It showed inhibition activity to infection of tobacco mosaic virus on Nicotiana glutinosa. The result of IEF suggested that AAL attached to TMV particles. Mycelia differentiation promotion was the other interesting activity. AAL promoted the differentiation of fruit body primordia from the mycelia of Agrocybe aegeritu and Auricularia polytricha. AAL antiserum was prepared and immunologically cross-reactived with several proteins from five other kinds of mushrooms. These results suggested that AAL probably was a representative of a large protein family, which plays important physiological roles in mushroom.

        Prevalence, Risk Factors, and Psychological Effects of Primary Nocturnal Enuresis in Chinese Young Adults

        Purpose: This study aimed to investigate the prevalence, risk factors, and effects of primary nocturnal enuresis (PNE) on physical and mental health in young adults in mainland China. Methods: An anonymous questionnaire was used to collect information including the sociodemographic characteristics, history of PNE, family history, daytime voiding symptoms, Pittsburgh Sleep Quality Index (PSQI) scores, Self-Esteem Scale (SES), and Self-Rating Depression Scale (SDS). A total of 22,500 university students from 23 provinces and 368 cities in mainland China were included. Results: In total, 21,082 questionnaires were collected, and 20,345 of them qualified for statistical analysis. The PNE prevalence was 1.17%, and the distribution of monosymptomatic nocturnal enuresis (MNE) and nonmonosymptomatic nocturnal enuresis (NMNE) was 66.1% and 33.9%, respectively. In total, 28% of respondents with PNE reported bedwetting daily, 31.6% between 1 and 7 times weekly, and 40.4% between 1 and 4 times monthly; 80% of PNE cases had no history of treatment. The prevalence of PNE in patients with a family history, frequency, urgency, urinary incontinence, and recurrent urinary tract infections was significantly higher than in those without these conditions (P<0.001). PNE was significantly correlated with the PSQI total score (sleep quality) (P=0.011). The SES score was lower and the SDS was higher (P<0.001) in the PNE group than in those without PNE. Conclusions: In mainland China, the PNE prevalence among young adults was found to be high, and PNE had significant effects on physical and mental health. Risk factors included a family history, daytime voiding symptoms, and lack of treatment.

      • GSTP1, ERCC1 and ERCC2 Polymorphisms, Expression and Clinical Outcome of Oxaliplatin-based Adjuvant Chemotherapy in Colorectal Cancer in Chinese Population

        Aim: Platinum agents have shown to be effective in the treatment of colorectal cancer. We assessed whether single nucleotide polymorphisms (SNPs) in GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln might predict the overall survival in patients receiving oxaliplatin-based chemotherapy in a Chinese population. Methods: SNPs of GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln in 335 colorectal cancer patients were assessed using TaqMan nuclease assays. Results: At the time of final analysis on Nov. 2011, the median follow-up period was 37.7 months (range from 1 to 60 months). A total of 229 patients died during follow-up. Our study showed GSTP1 Val/Val (HR=0.44, 95% CI=0.18-0.98), ERCC1 C/C (HR=0.20, 95% CI=0.10-0.79) and ERCC2 G/G (HR=0.48, 95% CI=0.19-0.97) to be significantly associated with better survival of colorectal cancer. GSTP1 Val/Val, ERCC1 C/C and ERCC2 G/G were also related to longer survival among patients with colon cancer, with HRs (95% CIs) of 0.41 (0.16-0.91), 0.16 (0.09-0.74) and 0.34 (0.16-0.91), respectively. Conclusion: GSTP1, GSTP1, ERCC1 Asn118Asn and ERCC2 Lys751Gln genotyping might facilitate tailored oxaliplatin-based chemotherapy for colorectal cancer patients.

        Virtual Queue Based QoS Layered Vertical Mapping in Wireless Networks

        Wireless communication is one of most active areas in modern communication researches, QoS (Quality of Service) assurance is very important for wireless communication systems design, especially for applications such as streaming video etc., which requires strict QoS assurance. The modern wireless networks multi-layer protocol stack structure results in QoS metrics layered and acting in cascade and QoS metrics vertical mapping between protocol layers. Based on virtual buffer between protocol layers and queuing technology, a unified layered QoS mapping framework is proposed in this paper, in which we first propose virtual queue concept, give a novelty united neighboring protocol layers QoS metric mapping framework, and analysis method based on dicerete-time Markov chain, and numerical results show that our proposed framework represents a significant improvement over previous model.

        Experimental Simulation on Open-Ended Pipe Pile Penetration Using Transparent Granule

        In order to attain the soil movement and explore the penetration mechanism of open-ended pipe piles in saturated sand, the transparent granule, which could substitute the natural sand, was used to simulate the pile jacking with different pile sections. The granule displacement vector field, the plug neutral surface and the plug height were obtained. Test results showed that when the pile inner radius is larger, the squeezing displacement and the plug height are larger. Normalization of the horizontal and vertical components of granule movement employing the pile outer radius showed that the 4d range of the pile end was the area where the inner shaft resistance exerted. In addition, it was found that the linear relationship between incremental filling ratio (IFR) and plug length ratio (PLR).

        Facile Preparation of ZnO Nanocatalysts for Ozonation of Phenol and Effects of Calcination Temperatures

        ZnO nanoparticles were synthesized through a facile route and were used as ozonation catalysts. With the increase of calcination temperature (150-300 oC), surface hydroxyl groups and catalytic efficiency of asobtained ZnO decreased remarkably, and the ZnO obtained at 150 oC showed the best catalytic activity. Compared with ozonation alone, the degradation efficiency of phenol increased above 50% due to the catalysis of ZnO-150. In the reaction temperatures range from 5 oC to 35 oC, ZnO nanocatalyst revealed remarkable catalytic properties, and the catalytic effect of ZnO was better at lower temperature. Through the effect of tertbutanol on degradation of phenol and the catalytic properties of ZnO on degradation of nitrobenzene, it was proposed that the degradation of phenol was ascribed to the direct oxidation by ozone molecules based on solidliquid interface reaction.

        Comparative proteomic analysis of proteins expression changes in the mammary tissue of cows infected with Escherichia coli mastitis

        Cows infected with Escherichia (E.) coli usually experience severe clinical symptoms, including damage to mammary tissues, reduced milkyield, and altered milk composition. In order to investigate the host response to E. coli infection and discover novel markers for mastitistreatment, mammary tissue samples were collected from healthy cows and bovines with naturally occurring severe E. coli mastitis. Changesof mammary tissue proteins were examined using two-dimensional gel electrophoresis and label-free proteomic approaches. A total of 95differentially expressed proteins were identified. Of these, 56 proteins were categorized according to molecular function, cellular component,and biological processes. The most frequent biological processes influenced by the proteins were response to stress, transport, andestablishment of localization. Furthermore, a network analysis of the proteins with altered expression in mammary tissues demonstrated thatthese factors are predominantly involved with binding and structural molecule activities. Vimentin and -enolase were central “functionalhubs” in the network. Based on results from the present study, disease-induced alterations of protein expression in mammary glands andpotential markers for the effective treatment of E. coli mastitis were identified. These data have also helped elucidate defense mechanismsthat protect the mammary glands and promote the pathogenesis of E. coli mastitis.
