        Adiponectin Attenuates the Inflammation in Atopic Dermatitis-Like Reconstructed Human Epidermis

        Background: Atopic dermatitis (AD) is a chronic disorder, with a vicious cycle of repetitive inflammation and deterioration of the epidermal barrier function. Adiponectin, an adipokine, has anti-inflammatory effects on various metabolic and inflammatory disorders. Recently, its level was found to be reduced in serum and tissue samples from AD patients. Objective: We aimed to investigate the effects of adiponectin on epidermal inflammation and barrier structures in AD skin. Methods: A three-dimensional in vitro epidermal equivalent model mimicking AD was obtained by adding an inflammatory substance cocktail to normal human epidermal equivalents (HEEs). The expression of epidermal differentiation markers, primary inflammatory mediators, and lipid biosynthetic enzymes was compared between adiponectintreated AD-HEEs, untreated control AD-HEEs, and normal HEEs. Results: Adiponectin co-treatment 1) inhibited the increase in mRNA expression of major inflammatory mediators (carbonic anhydrase II, neuron-specific NEL-like protein 2, thymic stromal lymphopoietin, interleukin-8, tumor necrosis factor-alpha, and human beta-defensin-2) from keratinocytes in AD-inflammatory HEEs, 2) enhanced the expression of lipid biosynthetic enzymes (fatty acid synthase, HMG CoA reductase, and serine-palmitoyl transferase), and 3) promoted the expression of differentiation factors, especially filaggrin. We also found that the expression of adiponectin receptor-1 and -2 decreased in the epidermis of chronic AD lesion. Conclusion: Activation of the adiponectin pathway is expected to enhance epidermal differentiation and barrier function as well as attenuate inflammatory response to AD as a therapeutic approach.         2016년 3월 31일부터 7월 31일까지 서울특별시 서초구 서초동에 위치한 서울고등학교 교정 일대를 서울고등학교 환경생물부 부원들이 채집 조사한 결과 20과 43속 53종의 거미가 조사 연구되어 이에 보고하는 바이다. From March 31st to July 31st in 2016, members of an environmental biology club at Seoul High School located in Seocho-gu, Seoul, Dae-Hee Kim, Dong-Woo Park, Jae-Woong Seo and Hyun-woo Lee, collected 20 families 43 genera 53 species of spiders and report the result of the study here.

        호두 기름의 성분조성과 항알레르기 효과

        This study was conducted to investigate physicochemical properties and anti-allergic properties of walnut oil, pH, acid value and iodine value of walnut oil were respectively 4.9, 0.8 and 117. Most of general composition of walnut oil was crude fat(99.9%). Vitamin A and E were 0.06 and 10.25㎎/100g and the major fatty acids of walnut oil was linoleic acid(62.8%). Total phenolics and antiallergic effects of walnut oil were estimated 27㎎% and 62.82% at the concentration of 0.5% ethanol walnut oil. These results suggest that the walnut oil can provide one of the valuable resource for the functional foods.

        식중독 세균 5속을 검출하기 위한 4개 주요 REP-PCR 반응성분의 최적화

        반복성 염기서열을 응용한 PCR법인 REP-PCR을 사용하여 식중독 미생물의 분리와 동정을 위한 연구에 적용하기 위하여 주요 식중독 유발세균 5속(genus), 6종의 균주를 사용하여 실험을 실시하였다. PCR 구성성분인 MgCl₂, dNTPs, REP sequence primer, 주형 DNA의 농도를 최적화하기 위한 연구 결과, MgCl₂의 농도가 2.0mN과 2.5mN일 때, 균주들의 fingerprinting pattern의 변화가 없거나 적은 것으로 관찰되어, 모든 속의 균주를 단일조건으로 분리?동정하기 위해서 MgCl₂의 농도는 2.5mN이 최적인 것으로 결론을 내렸다. dNTPs는 50μM의 농도를 적용하였을 때부터 전체 fingertprinting pattern범위의 주 단편들을 나타내는 균주들도 있었으나 총 단편의 수가 완벽하게 나타나지는 않았고, 200μM의 적용시점에서 6종의 균주 모두 단편의 수나 강도의 변화가 관찰되지 않았기 때문에 fingerprinting pattern의 파악을 위한 목적에는 200μM의 dNTPs 농도만으로도 충분하였다. REP primer는 적용한 농도가 증가함에 따라 단편의 수와 강도가 증가하였으며 Vibrio를 제외한 5종의 균주가 2.0μM의 primer를 적용했을 때 고유한 fingerprinting pattern을 나타냈다. 주형 DNA의 양을 변화시켜 적용하였을 때 DNA 양의 비율이 2배에서 5배까지 증가하여도 초기 미량 적용 시 생성되었던 fingerprinting pattern은 많은 차이를 보이지는 않았다. 그러나 적용되는 DNA 양에 따라 단편들의 강도와 수가 약간씩 점증하는 양상을 보였다. This experiment was conducted to optimize REP(repetitve extragenic palindromic sequence)-PCR reaction conditions in order to simultaneously differentiate the five different foodborne pathogenic bacterial genera of Escherichia, Salmonella, Shigella, Vibrio and Listeria. The four major PCR cocktail components of MgCl₂, dNTPs, primers and teplate DNA were considered and their optimum concentrations were determined using six reference strains. the optimized condentration of MgCl₂ was determined to be 2.5 mM in order to obtatin a consistent fingerprinting pattern. The similar fingerprinting pattern was obtatined when REP primers and dNTPs were added up to the concentration of 2μM and 200μM, respectively. As for template DNA, the numbers of PCR fragments were not mostly affected, but their intensities were slightly increased as the concentrations of the DNA were increased from 2 times to 5 times.

        교잡종 찰옥수수에 대한 연구 III : Ⅲ. 국내 옥수수 유전자원 수집종에 대한 주요 특성 분포

        2006년 국내 각 지역으로부터 수집된 재래종옥수수 유전자원을 관행방법에 의해 파종하고 주요특성별로 분포도를 살펴본 결과는 다음과 같다. 1. 수집된 유전자원에 대한 발아율은 대부분 80∼100%이상의 분포를 보였으나, 일부 수집계통에는 30% 미만의 극히 저조한 발아율을 보였다. 2. 수집자원에 대한 초기생육을 살펴보면 극히 생육이 저조한 것이 40% 정도를 보였고, 극히 우수한 자원은 3%미만으로 나타났다. 3. 간장은 대부분이 150∼199cm범위에 55%가 분포하고 있었으나, l00cm이하가 5%, 250cm이상이 8%로 나타나 수집종간에 큰 차이를 보였다. 4. 주당 분얼수는 1개 미만이 40%의 분포를 보였으나, 일부 수집종에서 2 내지 3개의 분얼수를 보이는 계통이 확인되었다. 5. 개화소요일수는 61∼64일의 범위에서 40%, 57∼60일의 범위에서 30%의 분포를 보였으며, 특히 일부 수집종의 경우 55일 이하의 극조생종도 10% 정도의 분포를 보였다. 6. 이삭길이는 대부분 11∼15cm범위에 60%가 포함되어 있으나, 일부계통에서 5cm미만이 2%, 16∼20cm의 이삭길이도 10%가 분포하였다. Germination ratio on the 300 collected genetic resources showed 80 to 100 percentage, but those of some accessions were very low as below 30 percentage. Early growth phase of most of these lines were very poor while some collected lines were very vigor. Range of stem height showed from 150 to 199cm, but that of some accessions showed below 100cm as five percentage and some of them was over 250cm as eight percentage. Number of tillers per plant appeared less one but some collected lines have two to three tillers per plant. In days to tasseling, some accessions showed more four per plant. Ear length of these accession showed above 60 percentage as 11 to 15cm, while that of some collected lines showed over 10 percentage as 16 to 20cm.

      • 오프셋을 감소시킨 고속 Dynamic Latch 비교기 설계

        In this paper, we propose a technique to minimize the input offset of a dynamic latch used as a voltage comparator. The offsets in a dynamic latch comparator can be divided into three causes. The fist is VOM that is caused by mismatch between MOSs, and the second is ΔQ/CZ that is caused by charge injection mismatch. The last is VOCF that is the effect of positive feedback during sampling phase. The VOCF was only considered in the previous works, however, we analyze not only charge injection mismatch but also VOCF and propose a effective circuit that can minimize these in this paper.

        병리검사 처방오류 개선 시스템 구축

        문제: 조직병리검사의 처방이 복잡하고 전문화됨에 따라 오류처방이 빈번하게 발생되고 이를 해결하기위한 프로세스가 복잡하여 부서간의 갈등과 업무의 가중, 또한 병원 수입 감소가 초래되었다. 목적: 발생된 오류처방의 유형과 발생율, 오류처방을 해결하기 위한 프로세스를 파악하여 이에 대한 문제점을 개선하여 업무의 효율성과 관련부서간의 부가업무 감소와 갈등해소, 더불어 병원수익을 향상시키는 것에 목표를 두고 활동하였다. 의료기관: 서울시에 소재한 종합병원 질 향상 활동: 조직병리검사 처방의 오류처방 발생 원인과 유형, 발생율을 조사하여 원인을 파악하고 개선을 위한 프로세스를 구축하여 질 향상을 도모하였다. 개선효과: 수술실에서 의뢰한 외과 입원환자의 일반조직병리검사 오류율은 개선 전 7.8%에서 개선 후 3.5%, 동결절편조직검사 오류율은 개선 전 28.6%에서 개선 후 0%로 이전보다 모두 개선되었다. 또한 수술실 간호사의 업무 만족도는 일반조직병리검사의 처방시 95%, 동결절편조직검사 처방시 90%로 높은 만족도를 보였다. 단 병리과의 경우 업무가 증가됨에 따라 초과근무시간이 증가하였다.

        근관치료용 전기모터의 회전 안정성 평가

        수종의 근관치료용 전기모터들의 반복사용과 시간에 따른 회전속도의 변화를 비교하여 근관치료용 전기모터의 회전 안정성을 평가하고자 하였다. 시간에 따라 변화하는 실제회전속도 및 전압과 전류를 측정하고, Student t-test를 통한 실제회전속도와, 반복측정분석을 통한 상관관계를 비교분석 하였다. 동일 종류의 전기모터들 사이의 모든 측정 결과들은 차이를 보이지 않았다. 모든 전기모터에서 반복사용과 시간의 흐름에 따른 회전속도의 변화는 차이를 보이지 않았다. 이상의 결과로, 본 실험에 사용된 모든 전기모터는 일정한 회전속도를 유지해야 하는 회전 안정성의 조건에 충족됨을 알 수 있으며, 반복사용과 시간에 따른 신뢰성 있는 회전속도와 안정적인 전압 및 전류의 유지가 가능하리라 생각된다. The purpose of this study was to evaluate a rotational stability of endodontic electronic motors by comparing the changes of rotational speed, depending on the number of usages and with/without static load. Twelve new endodontic electronic motors were used in this study. Non contact type digital tachometer was used for measuring the rotational speed of handpiece. True RMS Multimeter was used for measuring the voltages and the electric currents. All measurements were recorded every 10 seconds during 10 minutes and repeated 9 times. Five repetition was done per each electronic motor. To statistical analysis, student t-test, repeated measures and Scheffe's post-hoc tests were performed. In the same motor group, there was no significant difference in all measurements. In all groups, there was no significant difference in the amount of rotational speed changes depending on the number of usages and with/without static load. In the limitation of this study, the results showed that all kinds of endodontic electronic motors in this study had an established rotational stability. Therefore they could be safely used in root canal treatment with areliable maintenance of rotational speed, regardless of the number of usages and with/without load.

      • 성선자극호르몬유리호르몬 길항제인 Antide가 수컷 생쥐의 유해감각예민도에 미치는 영향

        To investigate testosterone effect on nociception, tail flick latency(TFL, sec, 50±1℃) was measured before and after administration of antide(a gonadotropin releasing hormone antagonist) in male mice(Institute of Cancer Research, age of 7-8 weeks, weight 30.5-37.5gm). Testosterone replacement effect on TFL was also examined in antide pre-treated male mice. Experiment 1:30 male mice were randomized into 3 groups(N=10 each). Antide 100㎍/kg, 300㎍/kg or same volume(5.7ml/kg) of 0.2% bovine serum albumin was administered intraperitoneally. TFL was measured before and at 3,6, 9, 12 and 24 hours after injection. Experiment 2 : 40 novice male mice were randomized into 2 groups(N=20 each) and both groups were pre-treated with antide 300㎍/kg i.p.6 hours after antide injection, testosterone 1mg/kg or same volume(5.7ml/kg) of sesame oil was administered i.p. TFL was measured before and 6 hours after antide pre-treatment and 30 min after testosterone/vehicle injection. 1) TFL was significantly shortened by antide in dose dependent manner, In control group, baseline TFL(mean±SD) and those measured at 3,6,9,12 and 24 hours after injection were 4.8±0.7, 5.6±0.7, 5.3±0.6, 5.4±0.6 and 4.7±0.6 sec, respectively. In antide 100㎍/kg group, TFLs were 5.2±0.9, 5.1 ±1.1, 4.2±0.9, 4.4±0.8, 5.1±0.8, and 4.7±0.8 sec, respectively. Compared with baseline, significant hyperalgesia appeared 6 and 9 hours after treatment. TFL measured at 6 hours after treatment was significantly shorter than control. In antide 300㎍/kg group, TFLs were 5.6±0.4, 4.5±0.9, 4.3±0.6, 3.9±1.0, 4.6±0.8 and 4.7±0.9 sec. Significant hyperalgesia appeared 3 hours after treatment and continued thereafter. TFLs measured at 3, 6 and 9 hours after treatment were significantly shorter than control, and TFL at 9 hours was significantly shorter than antide 100㎍/kg group. 2) Hyperalgesia induced by antide pre-treatment was recovered by testosterone replacement. In control group. TFL was shortened by antide pre-treartment(baseline : 6.1±1.0 sec ,6 hr after antide : 4.7±1.0 sec) and there was no significant change after vehicle injection(4.6±0.8 sec). In testosterone group, shortening of TFL induced by antide pre-treatment(baseline: 6.2±0.8 sec; 6 hr after antide: 4.6±0.9 sec) disappeared after testosteronen replacement(0.6±0.8sec). From these results, it is suggested that testosterone has a role of maintaining baseline antinociception and acute decrease in testosterone level results in hyperalgesia.

      • Sr·페라이트 플라스틱자석의 제조에 관한 연구

        In this paper, we mention he fabrication of Sr ferrite plastic magnet. In order to compose Sr ferrite, first, a little CaO, SiO₂, Na₂SiO₃, and Al₂O₃ are added to SrO5.9Fe₂O₃. Then, the composition is calcinated at 1225℃ under N₂ enviroment for 1 hourwith addition of PVA 5wt% solution. The carbon coating method increase grinding effect on powder fabrication. To fabricate anisotropic Sr ferrite plastic magnet, 80-89wt% of Sr ferrite and polyamide6 are kneaded and then 0-12kOe of magnetic field is applied. On magnet fabrication, an optimal condition of injection mold is obtained under the injection temperature 270℃, the injection pressure 120㎏/㎠, the magnetic field 8kOe and magnetic field applied time 8sec. As the result of experiments, we obtain (BH)max = 2.09MGOe property on anisotropic plastic magnet which is fabricated by using injection mold method in magnetic field.
