      • Polysaccharide from Polygonatum Inhibits the Proliferation of Prostate Cancer-Associated Fibroblasts Cells

        Han, Shu-Yu,Hu, Ming-Hua,Qi, Guan-Yun,Ma, Chao-Xiong,Wang, Yuan-Yuan,Ma, Fang-Li,Tao, Ning,Qin, Zhi-Hai Asian Pacific Journal of Cancer Prevention 2016 Asian Pacific journal of cancer prevention Vol.17 No.8

Inhibition of cancer-associated fibroblasts (CAFs) may improve the efficacy of cancer therapy. Polysaccharide extracted from polygonatum can selectively inhibit the growth of prostate-CAFs (p<0.001) without inhibiting the growth of normal fibroblasts (NAFs). Polysaccharides from polygonatum stimulate autophagy of prostate-CAFs. 3-methyl-adenine(3-MA) is an autophagy inhibitor. 3-MA was added to prostate-CAFs with polysaccharide from polygonatum to determine whether autophagy plays an important role in the restrained effect. Finally, polysaccharide from polygonatum treatment significantly increased the activation of Beclin-1 and LC3, key autophagy proteins. Polysaccharide from polygonatum stimulates autophagy of prostate-CAFs and inhibits prostate-CAF growth, indicating that a novel anti-cancer strategy involves inhibiting the growth of prostate-CAFs.

      • Prognostic Significance of Beclin-1 Expression in Colorectal Cancer: a Meta-analysis

        Han, Ye,Xue, Xiao-Feng,Shen, Hu-Gang,Guo, Xiao-Bo,Wang, Xu,Yuan, Bin,Guo, Xing-Po,Kuang, Yu-Ting,Zhi, Qiao-Ming,Zhao, Hong Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.11

Objective: Beclin-1 has recently been observed as an essential marker of autophagy in several cancers. However, the prognostic role of Beclin-1 in colorectal neoplasia remains controversial. Our study aimed to evaluate the potential association between Beclin-1 expression and the outcome of colorectal cancer patients. Materials and Methods: All related studies were systematically searched in Pubmed, Embase, Springer and Chinese National Knowledge Infrastructure databases (CNKI), and then a meta-analysis was performed to determine the association of Beclin-1 expression with clinical outcomes. Finally, a total of 6 articles were included in our analysis. Results: Our data showed that high Beclin-1 expression in patients with CRC was associated with poor prognosis in terms of tumor distant metastasis (OR=2.090, 95%CI=1.061-4.119, p=0.033) and overall survival (RR=1.422, 95%CI=1.032-1.959, p=0.031). However, we did not found any correlation between Beclin-1 over-expression and tumor differentiation (OR=1.711, 95%CI=0.920-3.183, p=0.090). In addition, there was no evidence of publication bias as suggested by Egger's tests for tumor distant metastasis (p=1.000), differentiation (p=1.000) and OS (p=0.308). Conclusions: Our present meta-analysis indicated that elevated Beclin-1 expression iss associated with tumor metastasis and a poor prognosis in patients with CRC. Beclin-1 might serve as an efficient prognostic indicator in CRC, and could be a new molecular target in CRC therapy.

Direct ethanol production from dextran industrial waste water by Zymomonas mobilis

        Ming-xiong He,Han Qin,Xiao-bo Yin,Zhi-yong Ruan,Fu-rong Tan,Bo Wu,Zong-xia Shui,Li-chun Dai,Qi-chun Hu 한국화학공학회 2014 Korean Journal of Chemical Engineering Vol.31 No.11

The direct production of ethanol from dextran industrial waste water was investigated by using Zymomonasmobilis via batch and semi-continuous fermentation mode. In batch fermentation, pretreated waste water (unsterilizedand sterilized), pH value (3.8 and 6.0), and Mg2+(with and without) was compared with OD600, sugar and ethanol con-centration. After 24 h fermentation, sugar in the dextran waste water was almost exhausted, and the amount of ethanolaccumulated reached 24.33-29.92 g/l, which is nearly 99% of the theoretical yield of ethanol. Kinetic parameters ofZ. mobilis in batch fermentation were also investigated. The raw dextran waste water was also used in semi-continuousfermentation. After 48 h fermentation, the production of ethanol was 28.65 g/l. These results indicated that dextranwaste water may be used as a candidate substrate and Z. mobilis could convert the raw material into ethanol directly.

Privet golden leaves adapt unexpectedly well to light changes

        Ming Yuan,Bo Huang,Li-Hua Dong,Qiao-Hong Han,Yong Yang,Chun-Bang Ding,Chao Hu,Yang-Er Chen,Zhong-Wei Zhang,Shu Yuan 한국원예학회 2020 Horticulture, Environment, and Biotechnology Vol.61 No.4

Golden-leaf privet ( Ligustrum × vicaryi ) is widely used as a horticultural shrub because of its upper golden leaves, butits lower leaves are green. However, the putative mechanisms of its upper golden leaves and the leaf color changes inresponse to light shifts have not been well studied so far. Here, chlorophylls (Chl), carotenoids, and Chl precursors from bothgolden and green leaves grown in full sunlight (approximately 1200 μmol photons m −2 s −1 at noon) or low-light conditions(180 μmol m −2 s −1 ) were determined spectrophotometrically. In addition, their gas exchange parameters and Chl fl uorescencewere measured in situ. Metabolic fl ux analysis of chlorophyll intermediates indicated that the conversion of prochlorophyllideto chlorophyllide was signifi cantly blocked in golden leaves when compared with green leaves. Green leaves showed higherphotosynthetic capacity in low light than golden leaves, but golden leaves presented unexpectedly stronger photosyntheticcapacity and lower reactive oxygen species accumulation under the high-light condition. Furthermore, golden leaves showeda higher level of nonphotochemical quenching (NPQ) after the light-to-dark shift and presented a stronger adaptive abilityto a broad range of light environments. Higher NPQ values and less oxidative damage in golden leaves may be correlatedwith their higher carotenoid levels. The results imply that lower chlorophyll levels and higher carotenoid levels in canopyleaves may help privet plants acclimate better to illumination changes. This study demonstrates the key role of irradiance ingenerating the two types of Ligustrum × vicaryi leaves and sheds a light on cultivation of other ornamental foliage plants.

Effect of Salinity on Strength Behavior of Cement-treated Dredged Clay at High Initial Water Contents

        Jie Yin,Wen-xia Han,Gui-zhong Xu,Ming-ming Hu,Yong-hong Miao 대한토목학회 2019 KSCE JOURNAL OF CIVIL ENGINEERING Vol.23 No.10

This paper presents the experimental investigation into the effect of salinity on the strength behavior of cement-stabilized dredged clays as fills at high initial water contents. One source of dredged clay was dredged from Baima Lake of China. Specimens were prepared having different porewater salinities by blending the air-dried clays with NaCl solution at different salt concentrations. The prepared saline dredged clay specimens were thereafter adjusted to different initial water contents and blended with cement with different amounts. Unconfined compression tests were performed on cement-stabilized dredged clay specimens after 28-day curing. All tested specimens show strain softening behavior where the axial strain corresponding to the peak stress varies with amount of cement. Stress-strain curve for the specimen with lower salinity lies above on that of higher one. Initial water content has a negative effect on the strength of cement-treated clays while cement has a positive effect on the development of strength for cement-treated dredged clays due to the pozzolanic reactions. The presence of salt has an adverse influence on the obtaining of strength for cement-treated dredged clays because salt can inhibit cement from generating C-S-H and C-A-H. Unconfined compressive strength almost linearly decreases with the increasing salinity. A quantitative expression was presented to calculate the unconfined compressive strength for cement-treated saline dredged clays concerning the effects for initial water content, porewater salinity and amount of cement.

Quantitative trait locus mapping and genomic selection of tobacco (Nicotiana tabacum L.) based on high-density genetic map

        Tong Zhijun,Xiu Zhihui,Ming Yao,Fang Dunhuang,Chen Xuejun,Hu Yafei,Zhou Juhong,He Weiming,Jiao Fangchan,Zhang Chi,Zhao Shancen,Jin Han,Jian Jianbo,Xiao Bingguang 한국식물생명공학회 2021 Plant biotechnology reports Vol.15 No.6

Tobacco ( Nicotiana tabacum L.) is an economic crop and a model organism for studies of plant biology and genetics. As an allotetraploid plant generated from interspecific hybridization, tobacco has a massive genome (4.5 Gb). Recently, a genetic map with 45,081 single nucleotide polymorphism (SNP) markers was constructed using whole-genome sequencing data for a tobacco population including 274 individuals. This provides a basis for quantitative trait locus (QTL) mapping and genomic selection, which have been widely applied to other crops but have not been feasible in tobacco. Based on this high-density genetic map, we identified QTLs associated with important agronomic traits, chemical compounds in dry leaves, and hazardous substances in processed cigarettes. The LOD values for major QTLs were highest for agronomic traits, followed by chemical compounds and hazardous substances. In addition to the identification of molecular markers, we evaluated genomic selection models and found that BayesB had the highest prediction accuracy for the recombinant inbred line population. Our results offer new insights into the genetic mechanism underlying important traits, such as agronomic traits and quality-related chemical compounds in tobacco, and will be able to support the application of molecular breeding to tobacco.

Electrodeposited Palladium Coating on Co Micro-nano Cones Array for Low-Temperature Solid-State Bonding

        Jiunan Xie,Hua Hu,Peixin Chen,Han Lei,Anmin Hu,Yunwen Wu,Ming Li 대한금속·재료학회 2024 ELECTRONIC MATERIALS LETTERS Vol.20 No.3

A low-temperature solid-state bonding technology using palladium-coated Co micro-nano cones array (MCA) and Sn-3.0Ag-0.5Cu (wt%) solder was investigated. The Pd modifi cation layer on the surface of Co MCA reduced the growth of oxide fi lm. Low-temperature solid-state bonding was achieved using Co/Pd MCA under the bonding condition of 750 gf, 175 °C and150 s with the shear strength of 49.55 MPa, and there was no void found along the bonding interface. Microscopic observationrevealed that Co/Pd MCA was fully embedded in the soft solder. The average shear strength of the bonding joint wasmeasured and demonstrate that Co/Pd MCA has higher reliability than Co MCA. This work highlights the advantages ofbonding based on Co/Pd MCA, which has great potential for extensive practical applications.

Association of the PPAR-γ Gene with Altered Glucose Levels and Psychosis Profile in Schizophrenia Patients Exposed to Antipsychotics

        Yun-Ru Liu,El-Wui Loh,Tsung-Ming Hu,Tsuo-Hung Lan,Hsien-Jane Chiu,Yung-Han Chang,Shuo-Fei Chen,Yen-Hsin Yu,Cheng-Chung Chen 대한신경정신의학회 2014 PSYCHIATRY INVESTIGATION Vol.11 No.2

Objective Metabolic abnormalities, e.g., diabetes, are common among schizophrenia patients. Peroxisome proliferator activated receptor-γ (PPAR-γ) regulates glucose/lipid metabolisms, and schizophrenia like syndrome may be induced by actions involving retinoid X receptor-α/PPAR-γ heterodimers. We examined a possible role of the PPAR-γ gene in metabolic traits and psychosis profile in schizophrenia patients exposed to antipsychotics. Methods Single nucleotide polymorphisms (SNPs) of the PPAR-γ gene and a serial of metabolic traits were determined in 394 schizophrenia patients, among which 372 were rated with Positive and Negative Syndrome Scale (PANSS). Results SNP-10, -12, -18, -19, -20 and -26 were associated with glycated hemoglobin (HbA1c) whereas SNP-18, -19, -20 and -26 were associated with fasting plasma glucose (FPG). While SNP-23 was associated with triglycerides, no associations were identified between the other SNPs and lipids. Further haplotype analysis demonstrated an association between the PPAR-γ gene and psychosis profile. Conclusion Our study suggests a role of the PPAR-γ gene in altered glucose levels and psychosis profile in schizophrenia patients exposed to antipsychotics. Although the Pro12Ala at exon B has been concerned an essential variant in the development of obesity, the lack of association of the variant with metabolic traits in this study should not be treated as impossibility or a proof of error because other factors, e.g., genes regulated by PPAR-γ, may have complicated the development of metabolic abnormalities. Whether the PPAR-γ gene modifies the risk of metabolic abnormalities or psychosis, or causes metabolic abnormalities that lead to psychosis, remains to be examined.
