        Synthesis, Crystal Structure and Fungicidal Activities of New Type Oxazolidinone-Based Strobilurin Analogues

        A series of oxazolidinone-based strobilurin analogues were efficiently synthesized by the reaction of 3-(2-bromomethylphenyl)oxazolidin-2-one with 1-substituted phenyl-2H-pyrazolin-3-one. Their structures were confirmed and characterized by 1H-NMR, 13C-NMR, elemental analysis, and mass spectroscopy. In addition, the crystal structure of the target compound 3-(2-((1-phenyl-2H-pyrazol-3-yloxy)methyl)phenyl) oxazolidin-2-one was determined by single crystal X-ray diffraction. The bioassay results of these compounds indicated that some of the oxazolidin-2-one derivatives containing N-substituted phenyl 2H-pyrazol ring exhibited potential in vivo fungicidal activities against M. grisea at the dosage of 1 g L‒1.

        Polymer composite microspheres loading 177Lu radionuclide for Interventional Radioembolization therapy and Real-time SPECT imaging of Hepatic Cancer

        Background Transarterial radioembolization (TARE) with 90Y-labeled glass and resin microspheres is one of the primary treatment strategies for advanced-stage primary and metastatic hepatocellular carcinoma (HCC). However, difficulties of real-time monitoring post administration and embolic hypoxia influence treatment prognosis. In this study, we developed a new biodegradable polymer microsphere that can simultaneously load 177Lu and MgO nanoparticle, and evaluated the TARE therapeutic efficacy and biosafety of 177Lu-PDA-CS-MgO microspheres for HCC treatment. Methods Chitosan microspheres were synthesized through emulsification crosslink reaction and then conducted surface modification with polydopamine (PDA). The 177Lu and nano MgO were conjugated to microspheres using active chemical groups of PDA. The characteristics of radionuclide loading efficiency, biodegradability, blood compatibility, and anti-tumor effectwere evaluated both in vitro and in vivo. SPECT/CT imaging was performed to monitor bio-distribution and bio-stability of 177Lu-PDA-CS-MgO after TARE treatment. The survival duration of each rat was monitored. HE analysis, TUNEL analysis, immunohistochemical analysis, and western blot analysis were conducted to explore the anti-tumor effect and mechanism of composited microspheres. Body weight, liver function, blood routine examination were monitored at different time points to evaluate the bio-safety of microspheres. Results The composite 177Lu-PDA-CS-MgO microsphere indicated satisfactory degradability, biocompatibility, radionuclide loading efficiency and radiochemical stability in vitro. Cellular evaluation showed that 177Lu-PDA-CS-MgO had significant anti-tumor effect and blocked tumor cell cycles in S phase. Surgical TARE treatment with 177Lu-PDACS- MgO significantly prolonged the medial survival time from 49 d to 105 d, and effectively inhibited primary tumor growth and small metastases spreading. Moreover, these microspheres indicated ideal in vivo stability and allowed real-time SPECT/CT monitoring for up to 8 weeks. Immunostaining and immunoblotting results also confirmed that 177Lu-PDA-CS-MgO had potential in suppressing tumor invasion and angiogenesis, and improved embolic hypoxia in HCC tissues. Further evaluations of body weight, blood test, and pathological analysis indicated good biosafety of 177Lu-PDA-CS-MgO microspheres in vivo. Conclusion Our study demonstrated that 177Lu-PDA-CS-MgO microsphere hold great potential as interventional brachytherapy candidate for HCC therapy.       • Structure learning of exponential family graphical model with false discovery rate control

        Probabilistic graphical models enjoy great popularity in a wide range of domains due to their ability to model the conditional dependency relationships among random variables. This paper explores the structure learning for the exponential family graphical model with false discovery rate (FDR) control. Most existing FDR-con-trolled structure learning procedures have been designed for the Gaussian graphical model (GGM). A systematic approach for more general exponential family graphical models is still lacking. In this paper, we introduce a unified procedure to learn the structure of the exponential family graphical model with FDR control utilizing the symmetrized data aggregation (SDA) technique via sample splitting, data screening, and information pooling. We show that our method controls FDR asymptotically under some mild conditions. Extensive simulation results and two real-data examples validate the effectiveness of our method.

        Adaptive robust control of active magnetic bearings rigid rotor systems

        Aiming at the problems of diffi cult decoupling, complex controller models, and poor disturbance rejection capability caused by the inherent nonlinear and strong coupling characteristics of the active magnetic bearing (AMB), an adaptive H-infinity robust control (AHRC) method is proposed in the AMBs-rigid rotor system. The proposed method combines the active disturbance rejection control (ADRC) with the robust control to achieve stable operation of the AMBs-rigid rotor system under high-speed conditions. First, a four-degree-of-freedom (4-DOF) AMBs-rigid rotor model was established and transformed into a one-degree-of-freedom (1-DOF) model for the design of the robust controller, which weakens the coupling effect between each of the degrees of freedom (DOF). Then the multi-objective non-dominated sorting genetic algorithm II (NSGA-II) was used to fi nd the relative optimal weighting function value of the H-infinite robust control (HRC), which solved the difficulties in the design of the robust controller for known controlled system models. In addition, by combining the optimized H-infi nity robust controller with the original error feedback control rate in the ADRC, the proposed AHRC retains the strong robustness of the HRC and the decoupling capability of the ADRC, which improves the tracking performance and anti-disturbance performance of the controlled system. Simulation and experimental results verify the effectiveness of the proposed method.

        Mechanical behavior of composite gel periodic structures with the pattern transformation

        When the periodic cellular structure is loaded or swelling beyond the critical value, the structure may undergo a pattern transformation owing to the local elastic instabilities, thus leading to structural collapse and the structure changing to a new configuration. Based on this deformation-triggered pattern, we have proposed the novel composite gel materials. This designed material is a type of architectural material possessing special mechanical properties. In this study, the mechanical behavior of the composite gel periodic structure with various gel inclusions is studied further through numerical simulations. When pattern transformation occurs, it results in a different elastic relationship compared with the material at untransformed state. Based on the obtained nominal stress versus nominal strain behavior, the Poisson‟s ratio and corresponding deformed structure patterns, we investigate the performance of designed composite materials and the effects of the uniformly distributed gel inclusions on composite materials. A better understanding of the characteristics of these composite gel materials is a key to develop its potential applications on new soft machines.

        Identification of a Novel Linear B-Cell Epitope in the M Protein of Avian Infectious Bronchitis Coronaviruses

        This report describes the identification of a novel linear B-cell epitope at the C-terminus of the membrane (M) protein of avian infectious bronchitis virus (IBV). A monoclonal antibody (MAb) (designated as 15E2) against the IBV M protein was prepared and a series of 14 partially-overlapping fragments of the IBV M gene were expressed with a GST tag. These peptides were subjected to enzyme-linked immunosorbent assay (ELISA) and western blotting analysis using MAb 15E2 to identify the epitope. A linear motif, 199FATFVYAK206, which was located at the C-terminus of the M protein, was identified by MAb 15E2. ELISA and western blotting also showed that this epitope could be recognized by IBV-positive serum from chicken. Given that 15E2 showed reactivity with the 199FATFVYAK206 motif, expressed as a GST fusion protein, in both western blotting and in an ELISA, we proposed that this motif represented a linear B-cell epitope of the M protein. The 199FATFVYAK206 motif was the minimal requirement for reactivity as demonstrated by analysis of the reactivity of 15E2 with several truncated peptides that were derived from the motif. Alignment and comparison of the 15E2-defined epitope sequence with the sequences of other coronaviruses indicated that the epitope is well conserved among chicken and turkey coronaviruses. The identified epitope should be useful in clinical applications and as a tool for the further study of the structure and function of the M protein of IBV.

        Multicriteria Optimization via Parameter Space Investigation for Ultralow-Altitude Airdrop L1 Adaptive Controller

        This paper presents preliminary results of the application of the parameter space investigation method for the design of the L1 flight control system implemented on the ultralow-altitude airdrop process. In particular, the study has established the task performance level of airdrop mission which is chosen as the optimization objective in this paper. According to the characteristics of ultralow-altitude airdrop process, 12 optimization criteria are designated as the constraints during the multicriteria optimization. After two iterations, the task performance level of the airdrop is improved from “Moderate” to “Expected”. The results in the paper confirms the suitability of the parameter space investigation method for the multicriteria design optimization of an airdrop flight control system subject to desired control specifications. In addition, the robustness and dynamic response of the flight control system are significantly improved after optimization. Moreover, the analysis of optimization results has contributed to a deeper understanding of the relationship between L1 controller parameters and airdrop task performance.
