Background: Genome-wide association studies suggest that plasma triacylglyceride (TAG) in humans was associated with variation in the PLA2G6 locus, a gene that encodes calcium-independent phospholipase A2 (iPLA2Ղ). The objective of the present study is to understand the impact of genetic inactivation of iPLA2Ղ on hepatic TAG metabolism in C57BL/6 mice.Methods: Male iPLA2Ղ+/− mice were backcrossed with female iPLA2Ղ−/− mice for up to 10 generations prior to experiments. Lipid and lipoprotein metabolism from plasma, hepatocytes, thigh subcutaneous adipose and thigh skeletal muscle tissues of the mice were determined under various experimental conditions.Results: The iPLA2Ղ−/− mice, either male or female as compared with iPLA2Ղ+/+ littermates, showed no change in fasted or postprandial plasma TAG or total cholesterol at young (12-15 weeks) or old (40-44 weeks) ages under chow diet or high-fat diet (HFD) conditions. However, fractionation of plasma lipoproteins showed that under HFD conditions, there was a significant increase (by 40%) in apoB-100 association with VLDL1 fractions in iPLA2Ղ−/− mice as compared with iPLA2Ղ+/+ littermates. There was no significant difference in triglyceride or cholesterol contents in the liver, muscle, or adipose tissue between iPLA2Ղ−/− and iPLA2Ղ+/+ littermates. Metabolic labeling experiments with cultured primary hepatocytes isolated from iPLA2Ղ−/− mice also showed 2-fold increase in the secretion of [35S]methionine-labeled apoB-100 in VLDL1 fractions as compared with that from iPLA2Ղ+/+hepatocytes. Likewise, secretion of [3H]palmitate-labeled TAG from the iPLA2Ղ−/− hepatocytes was increased by 2-fold.Conclusions: Although iPLA2Ղ may play a role in TAG-rich VLDL1 production from cultured hepatocytes, there is no evidence that inactivation of iPLA2Ղ would lead to dyslipidemia in mice in vivo.

Purpose Drug resistance is one of the main causes of chemotherapy failure in patients with small cell lung cancer (SCLC), and extensive biological studies into chemotherapy drug resistance are required. Materials and Methods In this study, we performed lncRNA microarray, in vitro functional assays, in vivo models and cDNA microarray to evaluate the impact of lncRNA in SCLC chemoresistance. Results The results showed that KCNQ1OT1 expression was upregulated in SCLC tissues and was a poor prognostic factor for patients with SCLC. Knockdown of KCNQ1OT1 inhibited cell proliferation, migration, chemoresistance and promoted apoptosis of SCLC cells. Mechanistic investigation showed that KCNQ1OT1 can activate transforming growth factor-β1 mediated epithelial-to-mesenchymal transition in SCLC cells. Conclusion Taken together, our study revealed the role of KCNQ1OT1 in the progression and chemoresistance of SCLC, and suggested KCNQ1OT1 as a potential diagnostic and prognostic biomarker in SCLC clinical management.
Setosphapyrone C and D accelerate macrophages cholesterol effl ux by promoting LXRa/ABCA1 pathway

LXRα agonists have attracted signifi cant attentiondue to their potential biological activities on promotingcholesterol effl ux. This study was designed to investigatewhether setosphapyrone C and D have potential lipid-loweringcapacity and the underlying mechanisms in vitro. Ourdata showed that setosphapyrone C and D had weak cytotoxicitycompared to the liver X receptor α (LXRα) agonistT0901317. In RAW 264.7 macrophages, setosphapyroneC and D signifi cantly enhanced [ 3 H]-cholesterol effl ux by~ 21.3% and 32.4%, respectively; furthermore, setosphapyroneC and D enhanced the protein levels of ATP-bindingcassette transporter (ABC) A1 and LXRα by 58% and 69%,and 60% and 70% (8 μM), respectively; however, they had noeff ect on the protein levels of ABCG1 and scavenger receptorB type 1; additionally, they had minor eff ect on the mRNAexpression of lipogenic genes. Of note, setosphapyrone C and D signifi cantly enhanced LXRα/ABCA1pathway inmice primary macrophages. In BRL cells, setosphapyroneC and D signifi cantly improved the protein levels of ABCA1and ABCG1; setosphapyrone D signifi cantly enhanced theprotein expression of low-density lipoprotein. Collectively,setosphapyrone C and D with weak cytotoxicity exhibitedeff ective lipid-lowering eff ect via enhancing LXRα/ABCpathways. Setosphapyrones possess potential applicationfor the treatment of hyperlipidemic diseases.

To investigate the effects of AMP on letrozole-induced PCOS rats. All SD rats were randomly divided into normal, PCOS, metformin group (Met, 265 mg/kg) and AMP group (1 g/kg) and weighed every three days. After 21 days of continuous administration, oral glucose tolerance test was performed, and the blood glucose(BP) level was measured. After execution, calculate the relative weight of bilateral ovaries. Serum levels of FSH, LH, E2, T and FINS were detected by ELISA assay. Serum levels of TC, TG, HDL-C and LDL-C were detected by the kit and calculated the LH/FSH ratio and HOMA-IR level. Compared with the normal group, body weight and relative weight of ovaries, BP level and serum levels of T, LH, LH/FSH ratio, FINS, HOMA-IR, TC, TG, LDL-C, and E2, FSH, HDL-C in model group increased. Compared with the PCOS group, body weight in the Met group and AMP group was lower, BP level and serum levels of T, LH, FINS, HOMA-IR and LDL-C in the Met group and AMP group were significantly lower and the level of HDL-C was higher. Armillaria mellea polysaccharides can improve hormone disturbance, glucose metabolism and abnormal lipid level in letrozole-induced polycystic ovary syndrome model rats.

The GZO/Ag/GZO sandwich films were deposited on glass substrates by RF magnetron sputtering of Gadoped ZnO (GZO) and ion-beam sputtering of Ag at room temperature. The effect of GZO thickness and annealing temperature on the structural, electrical and optical properties of these sandwich films was investigated. The microstructures of the films were studied by X-ray diffraction (XRD). X-ray diffraction measurements indicate that the GZO layers in the sandwich films are polycrystalline with the ZnO hexagonal structure and have a preferred orientation with the c-axis perpendicular to the substrates. For the sandwich film with upper and under GZO thickness of 40 and 30 nm, respectively, it owns the maximum figure of merit of 5.3 × 10-2 Ω-1 with a resistivity of 5.6 × 10-5 Ω cm and an average transmittance of 90.7%. The electrical property of the sandwich films is improved by post annealing in vacuum. Comparing with the as-deposited sandwich film, the film annealed in vacuum has a remarkable 42.8% decrease in resistivity. The sandwich film annealed at the temperature of 350 ℃ in vacuum shows a sheet resistance of 5 Ω/sq and a transmittance of 92.7%, and the figure of merit achieved is 9.3 × 10-2 Ω-1.

A 0.5 wt% GNP/Al composite was prepared by powder metallurgy and hot pressing. The numerical simulations and experimentsof porthole die extrusion were performed at 440 °C, 460 °C, 480 °C and 500 °C. The effect of extrusion temperatureon the microstructure and mechanical properties of the composites was studied. The results showed that the composite nearthe welding zone underwent dynamic recrystallization due to the large strain to form fine grains, some of which grew tocoarse grains because of the local high temperature. The extrusion temperature influenced the properties of the 0.5 wt%GNP/Al composite along both the extrusion and transverse directions. This influence is combined with synergistic effectson the recrystallization of the grain, fluidity and plasticity of the matrix, determined by the delamination, distribution andstructure of graphene and welding behavior in the welding zone. Therefore, the comprehensive mechanical properties ofthe composite reached the highest values in the extrusion direction and the transverse direction at 460 °C; the TEM imagesshowed that the graphene and aluminum matrix were in contact with each other via a small wavy transition overlapping zoneor that interaction forces caused by welding and shearing allowed the formation of a direct interface with the Al matrix. Thedeformed graphene formed a good interface with the matrix, ensuring outstanding mechanical properties in the welding zone.

To observe the effect of SA on DSS-induced intestinal fibrosis in mice. The mice were randomly divided into normal, DSS and SA treated DSS mice group. Changes in body weight(BW), blood stool and diarrhea were observed. The serum levels of IL-1β, IL-6, IL-17A, IL-18 and TNF-α were determined by ELISA kits. The protein levels of NLRP3, caspase-1, ASC, IL-1 β, α-SMA, CollagenI, Beclin1, LC3II/I and p62 were detected by Western Blotting assay. Compared with the normal group, SA significantly inhibited the loss of BW and DAI score in colitis mice (P<0.05). H&E and Masson staining assay suggested the SA reduced epithelial cell damage, crypt structure loss, inflammatory cell infiltration, and the fibrogenesis in colon of colitis mice. The serum levels of IL-1β, IL-6, IL-17A, IL-18 and TNF-α were decreased by SA(P<0.05). SA was able to reduce the protein levels of NLRP3, Caspase-1, ASC, IL-1β, α-SMA, Collagen-I, p62, and increased the expressions of Beclin1 and LC3II/I in colitis mice. The study demonstrated that SA can modulate the activity of NLRP3 inflammasome and autophagy pathway to improve DSS induced intaestinal fibrosis in mice.
Preparation and properties of CNTs-Cu hybrids/epoxy superhydrophobic and anticorrosive coatings

In this work, hydrophobized copper powders combined siloxane-modified carbon nanotubes(SHMWCNTs) to obtain intertwined hybrids of carbon nanotubes and copper (CNTs-Cu hybrids), andsuperhydrophobic CNTs-Cu hybrids/epoxy coatings were prepared on the surface of AH36 carbon steelby drop coating. The surface morphology and wettability of coatings were changed by adjusting the ratiosof SHMWCNTs to hydrophobized copper powders. The obtained coating can show a water contact angleof 167.2, a water sliding angle of 2.9, a plant oil contact angle of 134.3, and self-cleaning properties. Thecoating maintains superhydrophobicity after 300 tape-peeling times, exhibiting excellent mechanicaldurability. The fouled area percentages of chlorella on the coating and a pure epoxy resin were 0.23(±0.07)% and 12.18(±1.22)%, respectively, displaying anti-biofouling resistances. The coating has excellentcorrosion resistance, and its corrosion current density reached 9.55 10-8 A/cm2.

To observe the effect of cyanidin-3,5-O-diglucoside(C3G) on intestinal fibrosis in a model of dextran sulphate sodium (DSS)-induced mice with colitis. Colitis was induced in C57BL/6 mice by addition of DSS to drinking water. Clinical (body weight, colon length and colon weight) and histologic changes were evaluated. In addition, colon levels of interleukin-1β, IL-6, IL-18, IL-17A, tumor necrosis factor(TNF)-α were detected by enzyme-linked immunosorbent assay. Relative to the DSS group, the mice in the C3G treatment group increased in body weight, colon length, and the ratio of colon weight to length was significantly lower (P<0.05). The DAI index of mice in the intervention group was lower than that in the colitis model group (P<0.05). Histological observation also found that the intervention of C3G is able to reduce the infiltration of inflammatory cells in colon tissue induced by DSS. The results of Masson staining suggest that the intervention of C3G also reduces the occurrence of intestinal fibrosis in mice with colitis. Our results suggested that C3G is an activity to improve inflammation and intestinal fibrosis in DSS colitis mice.
