        Differential Manifestation of Ocular Phenotypes in TALEN-mediated p19arf Knockout FVB/N and C57BL/6J mouse lines

        Background p19arf, primarily known as a tumor suppressor, has also been reported to play an essential role in normal development of mouse eyes. Consistently, lack of p19arf has been associated with ocular defects, but the mixed background of the knockout (KO) mouse strain used raised a concern on the accuracy of the phenotypes observed in association with the targeted gene due to genetic heterogeneity. Object We carried out a study to investigate into the efect of genetic background on the manifestation of p19arf KO associated phenotypes. Methods We characterized the phenotypes of novel p19arf KO mouse lines generated in FVB/N and C57BL/6J using a transcription activator-like efector nuclease (TALEN) system in comparison to the reported phenotypes of three other p19arf-defcient mouse lines generated using homologous recombination. Results Ninety-fve percent of FVB/N-p19arf KO mice showed ocular opacity from week 4 after birth which worsened rapidly until week 6, while such abnormality was absent in C57BL/6J-p19arf KO mice up to the age of 26 weeks. Histopathological analysis revealed retrolental masses and dysplasia in the retinal layer in FVB/N-p19arf KO mice from week 4. Besides these, both strains developed normally from birth to week 26 without increased tumorigenesis except for a subcutaneous tumor found in a C57BL/6J-p19arf KO mouse. Conclusion Our fndings demonstrated surprisingly variable manifestation of p19arf-linked phenotypes between FVB/N and C57BL/6J mice, and furthermore between our mouse lines and the established lines, indicating a critical impact of genetic background on functional study of genes using gene targeting strategies in mice.

      • Consumer Recognition Survey for Establishing the Reasonable Expiration Dates of Health Functional Foods

        This study investigated consumers` recognition on shelf life of health functional foods to set up their rational expiration date. For this study 107 male and female adults aged 20 to 59 years were randomly selected as major consumers of health functional foods. And their data were analyzed with SPSS 17.0. The questionnaire included questions asking a consumer`s recognition on the shelf life of health functional foods and how to treat health function food passing its shelf life. In this study, the consumers were found to pay attentions considerably to the shelf life. But most of them did not know an accurate meaning of the shelf life of health functional food and mistook it for expiry date. In addition, the consumers doubted safety of health functional foods passing the shelf life. Therefore, education for consumers about an accurate meaning of the shelf life was needed. The results of this study were considered to be helpful to set up a reasonable expiration date for health functional food.

        한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

        B 형 간염 에 있어서 IgM anti HBc 의 발현도 및 임상적 의의

        Testing for IgM antiHBc was useful to detecting the patients with negative and positive HBsAg viral hepatitis and may improve serodiagnostic accuracy when acute NANB and delta agent hepatitis occur in previousely unrecognized HBsAg carriers. Moreover, it may be a useful test in defining potentially high risk sources of exposure to hepatitis B virus. But it is not useful in distinguishing recent from remote infection because IgM antiHBc occasionally was found in chronic liver diseases, IgM antiHBc was found by ELISA in 32 patients with acute viral hepatitis B, 61 with chronic hepatitis B, 18 patients with liver cirrhosis, 36 healthy HBsAg carriers and l49 patients with positive antiHBc alone. We continuously evaluated HBsAg and liver function of the patients who had negative IgM antiHBc acute hepatitis. The results are summarized as follows: 1) IgM antiHBc was positive in 84.4% of acute hepatitis B, 4% of chronic hepatitis B, 5.5% of positive HBsAg liver cirrhosis, 2. 8% of HBsAg healthy carriers but was all negative in patients with positive antiHBc alone. 2) Three patients with negative HBsAg acute viral hepatitis was diagnosed by positive IgM antiHBc as acute viral hepatitis R. 3) During the follow-up of 5 patients with negative IgM antiHBc acute hepatitis, 2 patients were diagnosed as type B acute viral hepatitis because of disappearance of HBsAg and normalization of liver function. And another 3 patients were diagnosed as non-B acute viral hepatitis superimposed on HBsAg carrier or natural course of chronic hepatitis.

        일부지역의 암발생 특성에 관한 조사연구(2)

        Cerebrovascular disease have been replaced by neoplasm as first cause of death on 1980's. So, many epidemiological studies on cancer have been reported. However, most of them were performed on bests of biopy data or of histological examination. We had reported that valuable results could be obtained from surgical data for cancer patients, previously. In this paper, 5,103 surgically treated cancer patients in Daegu-Kyungbuk area during 1985-1989 were analyzed in terms of sex age and organ. Mean age on male cancer patients was increased, but in the case of female, the trend of age distribution is inverse to other reports. It is considered as regional specificity to cancer incidence and it is due to higher frequency of breast and uterine cervical cancer than other areas. Especially, in the case of breast cancer, the mean age of patients was 30's and be considered as most dangerous cancer to female.

        Intrusion arch wire activation시 상악 견치에 가해진 초기응력의 유한요소법을 통한 고찰

        본 연구는 유한요소법을 사용하여 상악견치에 intrusion arch wire를 강제로 engage시켰을 때 상악 견치와 치근막에서 발생되는 압축력과 인장력의 분포의 양을 알아보고 어느 부분에 집중되는가를 분석하였다. 또한 각 부분에서의 압축력과 인장력의 비율을 비교하기 위해서 시행하여 다음과 같은 결과를 얻었다. 1. 상악 견치의 FA point와 백악법랑경계부분에서는 압축력과 인장력의 비율이 거의 비슷하게 나타났다. 2. 치근첨 부분에서는 압축력이 인장력의 거의 4배정도 크게 나타났다. Intrusion force적용시 치근흡수 정도가 치근첨에서 발생되는 것을 정량화시켜서 보여준 결과이다. 3. 백악법랑경계부분은 즉 FA point를 제외하고는 압축력과 인장력이 가장 큰 값을 보이고 있다. 즉 치아에 교정력이 직접 적용된 부분을 제외하고는 치아와 치주인대가 처음 접촉되는 백악법랑경계부에서 응력이 집중되는 것을 유추할 수 있다. The purpose of this study was to find the distribution and measurement of compressive and tensile stress when intrusi-on arch wire is forced engage with upper canine and to analysis stress at each section through FEM. And we compare compressive and tensile ratio at each section. The results were as follows. 1. At FA point and cemento-enamel junction of upper canine, compressive and tensile force ratio is about the same. 2. At apex, compressive force is the four times as tensile force ; In intrusion, we show root resorption at apex. 3. At Cemento-enamel junction, the compressive and tensile force show the maximum value except FA point.

        Bubble detection is one of the most important measurements in the two-phase flows. For the purpose of the bubble detection, an optical fiber is used based on the fact that. An intensity of attenuated light varies according to the surrounding medium at the exposed core. Experiments are conducted to investigate. (1) the effect of length and diameter of the exposed core, (2) the effect of the shape of the exposed core (3) the effect of the size of the bubble, on the detection capability. The possibility of bubble detection with optical fibers is present in some geometrical conditions of optical fibers.

      • 食品工場廢水의 活性汚泥處理中 오존에 依한 酸化分解效果

        잔류염소를 함유한 發水를 活性汚泥에 의한 處理效率을 增加시키기 위하여 전압 100Volt, 유속 10ℓ-air/min, 0.8g-O₃/hr의 오존으로 처리했을때 COD의 除去效果및 투과율의 變化를 實驗한 結果를 要約하면 다음과 같다. 1. 잔류염소를 함유한 시료를 0.8g-O₃/hr의 오존으로 處理하였을때 pH의 變化 및 反應時間에 의한 COD의 除去率은 中性溶液(pH=7.2)에서 오존處理時間 50分 후 35%의 除去效果를 나타내어 酸性溶液(pH=3.2)의 20%, 알칼리性溶液(pH=12.0)의 10%의 除去效果보다 좋았다. 2. 잔류염소를 함유한 試料를 오존으로 50分間 處理하여 F/M比 0.1㎏·BOD/㎏·MLSS로 한 후 3. 6. 9 및 15時間 活性汚泥로 反應시켰을 때 反應時間 6時間 후 85.8%의 COD의 除去效果가 있어 티오황산나트륨(Na₂S₂O₃)으로 處理한 試料의 除去率 60.5%보다 좋았으며, 反應時間의 단축효과도 있었다. 3. Polyphenol色素 等 暗褐色을 띄는 試料를 酸性, 中性, 알칼리性 溶液으로 하여 오존으로 50分間 處理하였을 때 투과율이 30~40%에서 70% 以上 상승하였으며 알칼리성溶液에서가 투과율이 더 좋았다. For the increase of treatment efficiency when waste water which included residual chlorine was biologically treated with activated sludge, elimination efficiency of COD and change of transmittance by activated sludge method were examined after the treatment of residual chlorine with ozone of 100 volt and 10ℓ-air/min., 0.8g-O₃/hr. in velocity. The results were as follows. 1) When the sample which included residual chlorine was treated with ozone of 0.8g-O₃/hr., COD removal rate by the change of pH and ozonization time was more efficient than acidity solution of pH 3.2(20%) and alkaline solution of pH 12.0(10%). Since the COD removal rate become 35% after 50 minutes of ozonization time at neutral solution of pH 7.2. 2) After the residual chlorine included sample was treated with ozone for 50 minutes to make the F/M rate of 0.1㎏·BOD/㎏·MLSS, the sample was reacted with activated sludge for 3, 6, 9 and 15 hours. The 6 hours reaction gave 85.8% COD elimination which showed higher efficiency than 60.5% by Na₂S₂O₂ treatment togather with shorting efficiency of reaction time. 3) When such sample as polyphenol which gives dark brown color was treated with ozone for 50 minutes after being made into acidic, neutral and alkaline solution, the transmittance increased from 30-40% to 70% and gave better result in alkaline solution.

        구강 편평세포암종에서 P16ink⁴유전자의 Methylation에 대한 연구

        The p16 protein is a cyclin dependent kinase inhibitor that inhibits cell cycle progression from G1 phase to S phase in cell cycle. Many p16 gene mutations have been noted in many cancer-cell lines and in some primary cancers, and alterations of p16 gene function by DNA methylation have been noticed in various kinds of cancer tissues and cell-lines. There have been a large body of literature has accumulated indicating that abnormal patterns of DNA methylation (both hypomethylation and hypermethylation) occur in a wide variety of human neoplasma and that these aberrations of DNA methylation may play an important epigenetic role in the development and progression of neoplasia. DNA methylation is a part of the inheritable epigenetic system that influences expression or silencing of genes necessary for normal differentiation and proliferation. Gene activity may be silenced by methylation of up steream regulatory regions. Reactivation is associated with demethylation. Although evidence or a high incidence of p16 alterations in a variety of cell lines and primary tumors has been reported, that has been contested by other investigators. The precise mechanisms by which abnormal methylation might contribute to carcinogenesis are still not fully elucidated, but conceivably could involve the modulation of oncogene and other important regulatory gene expression, in addition to creating areas of genetic instability, thus predisposing to mutational events causing neoplasia. There have been many variable results of studies of head and neck squamous cell carcinoma(HNSCC). This investigation was studied on 13 primary HNSCC for p16 gene status by protein expression in immunohistochemistry, and DNA genetic/epigenetic analyzed to determine the incidence, the mechanisms, and the potential biological significance of its Inactivation. As methylation detection method of p16 gene, the methylation specific PCR(MSP) is sensitive and specific for methylation of any block of CpG sites in a CpG islands using bisulfite-modified DNA. The genomic DNA is modified by treatment with sodium bisulfate, which converts all unmethylated cytosines to uracil(thymidine) . The primers designed for MSP were chosen for regions containing frequent cytosines (to distinguish unmodified from modified DNA), and CpG pairs near the 5' end of the primers (to provide maximal discrimination in the PCR between methylated and unmethylated DNA) The two strands of DNA are no longer complementary after bisulfite treatment, primers can be designed for either modified strand. In this study, 13 paraffin embedded block tissues were used, so the fragment of DNA to be amplified was intentionally small, to allow the assessment of methyla-tion pattern in a limited region and to facilitate the application of this technique to samlples. In this 13 primary HNSCC tissues, there was no methylation of p16 promoter gene (detected by MSP and automatic sequencing). The p16 protein-specific immunohistochemical staining was performed on 13 paraffin embedded primary HNSCC tissue samples. Twelve cases among the 13 showed altered expression of p16 proteins (negative expression). In this study, The author suggested that low expression of p16 protein may play an important role in human HNSCC, and this study suggested that many kinds of genetic mechanisms including DNA methylation may play the role in carcinogenesis.

      • 한국인 지원자에서 니모디핀의 생체이용율연구

        A reversed phase HPLC method was developed and validated for the determination of nimodipine in human plasma. Nitrendipine was used as an internal standard. Calibration curves were linear in the concentration range of 5~300 ng/ml. The coefficient of variation of the intra- and inter-day precision was below 15%. The coefficient of variation of the accuracy was below 15% in the concentration range investigated. A bioavailability study was performed using the validated HPLC method. Eight healthy male volunteers were orally administered 30 mg of nimodipine. The pharmacokinetic parameters were calculated using WinNonlin. The mean values of AUC_(24hr) was 64.0±31.3 ng·hr/ml, C_(max) was 46.5±31.2 ng/ml, T_(max) was 0.54±0.17 hr and t_(1/2) was 3.03±2.26 hr. The pharmacokinetic parameters and the HPLC method can be used for the design of bioequivalence study of nimodipine.
