Effect of Myostatin (MSTN) g+6223G>A on Production and Carcass Traits in New Zealand Romney Sheep

Myostatin, which is also known as growth and differentiation factor 8 (GDF8), has been reported to act as a negative regulator of skeletal muscle development. Variation in the myostatin gene (MSTN) has been associated with variation in muscularity in certain "meaty" sheep breeds. Polymerase Chain Reaction-Single Strand Conformational Polymorphism (PCR-SSCP) analysis was used to investigate allelic variation in the previously described g+6223G>A single-nucleotide polymorphism (SNP) in the 3' untranslated region (3' UTR) of MSTN. The sheep studied were 79 New Zealand (NZ) Romney lambs derived from a single sire heterozyous for g+6223G>A, which is in itself notable as this polymorphism has not been described previously in this breed. Allelic variation was observed to be associated with an abnormal gender ratio (p = 0.046) in the progeny. The presence of allele A was observed to have an effect (p<0.05) on birth weight, mean loin yield, proportion yield loin and total muscle yield. Allelic variation did not significantly affect mean shoulder yield, leg yield, proportion yield shoulder and proportion yield leg. This preliminary result suggests that while the A allele at MSTN g+6223 appears to improve some valuable traits in NZ Romney sheep, further research is required to understand if and how it may affect other traits.

ZNF313 encoding a zinc-binding protein is located at chromosome 20q13.13, which exhibits a frequent genomic amplification in multiple human cancers. However, the biological function of ZNF313 remains largely undefined. Here we report that ZNF313 is an ubiquitin E3 ligase that has a critical role in the regulation of cell cycle progression, differentiation and senescence. In this study, ZNF313 is initially identified as a XIAP-associated factor 1 (XAF1)-interacting protein, which upregulates the stability and proapoptotic effect of XAF1. Intriguingly, we found that ZNF313 activates cell cycle progression and suppresses cellular senescence through the RING domain-mediated degradation of p21<SUP>WAF1</SUP>. ZNF313 ubiquitinates p21<SUP>WAF1</SUP> and also destabilizes p27<SUP>KIP1</SUP> and p57<SUP>KIP2</SUP>, three members of the CDK-interacting protein (CIP)/kinase inhibitor protein (KIP) family of cyclin-dependent kinase inhibitors, whereas it does not affect the stability of the inhibitor of CDK (INK4) family members, such as p16<SUP>INK4A</SUP> and p15<SUP>INK4B</SUP>. ZNF313 expression is tightly controlled during the cell cycle and its elevation at the late G1 phase is crucial for the G1-to-S phase transition. ZNF313 is induced by mitogenic growth factors and its blockade profoundly delays cell cycle progression and accelerates p21<SUP>WAF1</SUP>-mediated senescence. Both replicative and stress-induced senescence are accompanied with ZNF313 reduction. ZNF313 is downregulated during cellular differentiation process in vitro and in vivo, while it is commonly upregulated in many types of cancer cells. ZNF313 shows both the nuclear and cytoplasmic localization in epithelial cells of normal tissues, but exhibits an intense cytoplasmic distribution in carcinoma cells of tumor tissues. Collectively, ZNF313 is a novel E3 ligase for p21<SUP>WAF1</SUP>, whose alteration might be implicated in the pathogenesis of several human diseases, including cancers.

<P>Several studies have showed an association of gene polymorphisms with the development of glomerulonephritis (GN). We investigated the effects of gene polymorphisms on the development of GN by analyzing polymorphisms in the interleukin (IL)-18, transforming growth factor (TGF)-β, and vascular endothelial growth factor (VEGF) genes in Korean patients with primary GN. The study included 146 normal subjects (controls) and 100 patients diagnosed with primary GN by kidney biopsy. The gene polymorphisms A-607C and G-137C in <I>IL-18</I>, C-509T and T869C in <I>TGF-</I>β<I>1</I>, and C-2578A and C405G in <I>VEGF</I> were investigated in DNA extracted from peripheral blood. Significant differences were observed between the GN and control groups in the genotype and allele frequencies of A-607C <I>IL-18</I> and C405G <I>VEGF</I>. The frequencies of the <I>IL-18</I>−607CC genotype [<I>P</I> = 0.001, odds ratio (OR) = 2.473] and the <I>VEGF</I> 405GG genotype (<I>P</I> = 0.001, OR = 2.473) were significantly increased in the GN group. The combination of <I>IL-18</I>−607CC+ and <I>VEGF</I> 405GG+ genotypes had a higher risk for developing GN in comparison with the combination of <I>IL-18</I>−607CC− and <I>VEGF</I> 405GG− genotypes (<I>P</I> < 0.001, OR = 8.642). In the haplotype analysis of the <I>IL-18</I> gene, the CG haplotype was significantly more frequent in the GN group than the control group (61.5% <I>vs</I> 46.9%, <I>P</I> = 0.002). These results show that the −607CC genotype of the <I>IL-18</I> gene and the 405GG genotype of the <I>VEGF</I> gene are associated with susceptibility to and the development of primary GN.</P>

<P>We report the discovery of a planetary system from observation of the high-magnification microlensing event OGLE-2012-BLG-0026. The lensing light curve exhibits a complex central perturbation with multiple features. We find that the perturbation was produced by two planets located near the Einstein ring of the planet host star. We identify four possible solutions resulting from the well-known close/wide degeneracy. By measuring both the lens parallax and the Einstein radius, we estimate the physical parameters of the planetary system. According to the best-fit model, the two planet masses are similar to 0.11 M-J and 0.68 M-J and they are orbiting a G-type main-sequence star with a mass similar to 0.82 M-circle dot. The projected separations of the individual planets are beyond the snow line in all four solutions, being similar to 3.8 AU and 4.6 AU in the best-fit solution. The deprojected separations are both individually larger and possibly reversed in order. This is the second multi-planet system with both planets beyond the snow line discovered by microlensing. This is the only such system (other than the solar system) with measured planet masses without sin i degeneracy. The planetary system is located at a distance 4.1 kpc from the Earth toward the Galactic center. It is very likely that extra light from stars other than the lensed star comes from the lens itself. If this is correct, it will be possible to obtain detailed information about the planet host star from follow-up observation.</P>

Insulinomas are tumors that, in more than 90% or cases, are both single and benign, so the potential for surgical cure is very high. But, their localization is somewhat difficult, as they are small in size and located deep in pancreatic parenchyma. Because success of operation is depentdent on the localization rather than the surgical techniques, the availability of good licalization technique is mandatory. Recently, endoscopic ultrasonography(EUS) has been reported to be a very sensitive and specific test for the localization of insulinomas. We report a case of insulinoma that was localized successfully by EUS, after failure in localization by abdominal ultrasonography, abdominal CT, selective angiography, and even by transhepatic portal vein sampling. Purpose: The purpose of this study was to figure out general characteristics, health perception, self-efficacy, and family support influencing on health promotion behavior of middle-aged male. Method: In this descriptive correlation design study, 320 middle-aged male were recruited through convenient sampling between October and December, 2015. Questionnaires used for this study were HPLP(Health Promotion Lifestyle Profile), HPS(Health perception Scale), Self-Efficacy Scale developed by Hong, and Family Support Scale developed by Park. Data were statistically analyzed using SPSS 22.0 program with descriptive statistics, independent t-test, one way ANOVA with Scheffe's test, and multiple linear regression. Result: It had significant influence to correlation among health promotion behavior, health perception, self-efficacy, and family support of middle-aged male. Also, the most influenced factor to health promotion behavior showed health perception. Conclusion: The result of this study suggested that health perception significantly influenced to health promotion behavior. Therefore, health perception educating program for health promotion behavior of middle-aged male. And further studies are required with more sample of various group and systematic data collecting methods to confirm these results.

Injection molding is the most widely used process for the industrial forming of plastic articles. During an injection molding process of composites, the fiber-matrix separation and fiber orientation are caused by the flow of molten polymer/fiber mixture. As a result, the product tends to be nonhomogeneous and anisotropic. Hence, it is very important to clarify the relations between separation·orientation and injection molding conditions. So far, there is no research on the measurement of fiber orientation using image processing. In this study, the effects of fiber content ratio and molding condition on the fiber orientation-angle distributions are studied experimentally. Using the image processing method, the fiber orientation distribution of welding parts in injection-molded products is assessed. And the effects of fiber content and injection mold shapes on the fiber orientation in case of fiber reinforced polymeric composites are studied experimentally.
