        Seismic performance and optimal design of framed underground structures with lead-rubber bearings

        Lead-rubber bearings (LRBs) have been used worldwide in seismic design of buildings and bridges owing to their stable mechanical properties and good isolation effect. We have investigated the effectiveness of LRBs in framed underground structures on controlling structural seismic responses. Nonlinear dynamic time history analyses were carried out on the well-documented Daikai Station, which collapsed during the 1995 Hyogoken-Nanbu earthquake. Influences of strength ratio (ratio of yield strength of LRBs to yield strength of central column) and shear modulus of rubber on structural seismic responses were studied. As a displacement-based passive energy dissipation device, LRBs reduce dynamic internal forces of framed underground structures and improve their seismic performance. An optimal range of strength ratios was proposed for the case presented. Within this range, LRBs can dissipate maximum input earthquake energy. The maximum shear and moment of the central column can achieve more than 50% reduction, whereas the maximum shear displacement of LRBs is acceptable.

        Nomogram for predicting overall survival in children with neuroblastoma based on SEER database

        Purpose: This study was performed to establish and validate a nomogram for predicting the overall survival in children with neuroblastoma. Methods: The latest clinical data of neuroblastoma in Surveillance, Epidemiology, and End Results (SEER) database was extracted from 2000 to 2016. The cases included were randomly divided into training and validation cohorts. The survival curves were drawn with a Kaplan-Meier estimator to investigate the influences of certain single factors on overall survival. Also, least absolute shrinkage and selection operator regression was applied to further select the prognostic variables for neuroblastoma. Additionally, receiver operating characteristic (ROC) curves and calibration curves were used to evaluate the accuracy of the nomogram. Results: In total, 1,262 patients were collected and 8 independent prognostic factors were achieved, including patients’ age, sex, race, tumor grade, radiotherapy, chemotherapy, tumor site, and tumor size. Then we constructed a nomogram by using the data of the training cohort with 886 cases. Subsequently, the nomogram was validated internally and externally with 886 and 376 cases, respectively. The internal validation revealed that the area under the curves (AUC) of ROC curves of 1-, 3-, and 5-year overall survival were 0.69, 0.78, and 0.81, respectively. Accordingly, the external validation also showed that the AUC of 1-, 3-, and 5-year overall survival were all ≥0.69. Both methods of validation demonstrated that the predictive calibration curves were consistent with standard curves. Conclusion: The nomogram possess the potential to be a new tool in predicting the survival rate of neuroblastoma patients.

        Risk Factors for Anxiety in Major Depressive Disorder Patients

        Objective: To analyze the sociodemographic and clinical factors related to anxiety in patients with major depressive disorder (MDD). Methods: This study involved a secondary analysis of data obtained from the Diagnostic Assessment Service for People with Bipolar Disorders in China (DASP), which was initiated by the Chinese Society of Psychiatry (CSP) and conducted from September 1, 2010 to February 28, 2011. Based on the presence or absence of anxiety-related characteristics, 1,178 MDD patients were classified as suffering from anxious depression (n=915) or non-anxious depression (n=263), respectively. Results: Compared with the non-anxious group, the anxious-depression group had an older age at onset (t=−4.39, p<0.001), were older (t=−4.69, p<0.001), reported more lifetime depressive episodes (z=−3.24, p=0.001), were more likely to experience seasonal depressive episodes (χ2=6.896, p=0.009) and depressive episodes following stressful life events (χ2=59.350, p <0.001), and were more likely to have a family history of psychiatric disorders (χ2=6.091, p=0.014). Their positive and total scores on the Mood Disorder Questionnaire (MDQ) and the 32-item Hypomania Checklist (HCL-32) (p<0.05) were also lower. The logistic regression analysis indicated that age (odds ratio [OR]=1.03, p<0.001), a lower total MDQ score (OR=0.94, p=0.011), depressive episodes following stressful life events (OR=3.04, p<0.001), and seasonal depressive episodes (OR=1.75, p=0.039) were significantly associated with anxious depression. Conclusion: These findings indicate that older age, fewer subclinical bipolar features, an increased number of depressive episodes following stressful life events, and seasonal depressive episodes may be risk factors for anxiety-related characteristics in patients with MDD.

      • RASAL1 Attenuates Gastric Carcinogenesis in Nude Mice by Blocking RAS/ERK Signaling

        Recent studies have suggested that the RAS protein activator like-1 (RASAL1) functions as a tumor suppressor in vitro and may play an important role in the development of gastric cancer. However, whether or not RASAL1 suppresses tumor growth in vivo remains to be determined. In the present study, we investigated the role of RASAL1 in gastric carcinogenesis using an in vivo xenograft model. A lentiviral RASAL1 expression vector was constructed and utilized to transfect the human poorly differentiated gastric adenocarcinoma cell line, BGC-823. RASAL1 expression levels were verified by quantitative real-time RT-PCR and Western blotting analysis. Then, we established the nude mice xenograft model using BGC-823 cells either over-expressing RASAL1 or normal. After three weeks, the results showed that the over-expression of RASAL1 led to a significant reduction in both tumor volume and weight compared with the other two control groups. Furthermore, in xenograft tissues the increased expression of RASAL1 in BGC-823 cells caused decreased expression of p-ERK1/2, a downstream moleculein the RAS/RAF/MEK/ERK signal pathway. These findings demonstrated that the over-expression of RASAL1 could inhibit the growth of gastric cancer by inactivation of the RAS/RAF/MEK/ERK pathway in vivo. This study indicates that RASAL1 may attenuate gastric carcinogenesis.

        Molecular cloning of two novel stearoyl-acyl desaturase genes from winterness wheat

        Using RT-PCR and rapid amplification of cDNA ends, two new full-length cDNAs of SAD (TaSAD1 and TaSAD2) were obtained from a hardiest winter wheat cultivar (Mironovskaya808). Sequence comparison analysis showed that the deduced amino acid sequences of TaSAD1 and TaSAD2 had high similarity to those of other reported SAD proteins. They were also different each other by some substitutions, insertions and/or deletions involving single amino acid residues or motifs. Based on evolution analysis, it was clear that all SAD genes from Poaceae were closer than those from other genus such as Arabidopsis,Glycine, Triadica, Brassica, Sesamum and Bassia. All SAD genes clustered into two major groups in Poaceae. Meanwhile,TaSAD1 and TaSAD2 were clustered into different groups. The tertiary structure prediction indicated that both TaSAD1 and TaSAD2 proteins were a compact globular protein and their model structures almost were the same.

      • Meta-analysis of Associations between the MDM2-T309G Polymorphism and Prostate Cancer Risk

        The mouse double minute 2 (MDM2) gene plays a key role in the p53 pathway, and the SNP 309T/G single-nucleotide polymorphism in the promoter region of MDM2 has been shown to be associated with increased risk of cancer. However, no consistent results were found concerning the relationships between the polymorphism and prostate cancer risk. This meta-analysis, covering 4 independent case-control studies, was conducted to better understand the association between MDM2-SNP T309G and prostate cancer risk focusing on overall and subgroup aspects. The analysis revealed, no matter what kind of genetic model was used, no significant association between MDM2-SNP T309G and prostate cancer risk in overall analysis (GT/TT: OR = 0.84, 95%CI = 0.60-1.19; GG/TT: OR = 0.69, 95%CI = 0.43-1.11; dominant model: OR = 0.81, 95%CI= 0.58-1.13; recessive model: OR = 1.23, 95%CI = 0.95-1.59). In subgroup analysis, the polymorphism seemed more likely to be a protective factor in Europeans (GG/TT: OR = 0.52, 95%CI = 0.31-0.87; recessive model: OR = 0.58, 95%CI = 0.36-0.95) than in Asian populations, and a protective effect of the polymorphism was also seen in hospital-based studies in all models (GT/TT: OR = 0.74, 95%CI = 0.57-0.97; GG/TT: OR = 0.55, 95%CI = 0.38-0.79; dominant model: OR = 0.69, 95%CI = 0.54-0.89; recessive model: OR = 0.70, 95%CI = 0.51-0.97). However, more primary studies with a larger number of samples are required to confirm our findings.

        Safety and antifatigue effect of Korean Red Ginseng capsule: A randomized, double-blind and placebo-controlled clinical trial

        Background: In physical activity or labor, the human body is in a state of high intensity stress, and allparts or physiological functions of the body respond positively to maintain or balance the need formovement. The human body has many physiological changes in the process of movement, and fatigue isthe external manifestation of various complex changes inside the human body. Fatigue is also a physiologicalmechanism of self-protection after the body reaches a certain level of activity, which can preventthe occurrence of life-threatening excessive functional failure. The generation of fatigue is a very complexprocess, and its mechanism has not been concluded yet. Therefore, it is an important work to search andscreen the effective components of natural plants that have anti-fatigue effect and to explore theirmechanism. Methods: This was a 8-week, randomized, double-blind, placebo-controlled clinical trial. A total of 110subjects who passed physical examination were included according to the scheme design, and randomlydivided into a test group which was given KRG and a placebo control group. The calculation is carried outaccording to the standard of sub-high-intensity exercise test. Results: There was no adverse effect on safety index of subjects after taking red ginseng capsule. AfterKRG treatment, subjective strength grade is significant lower than placebo treatment. Blood lactic acidcontent is significantly get lower after trial in KRG group, and significant lower than placebo group. Creatine phosphokinase(CK) content is significantly get lower after trial in KRG group, and significantlower than placebo group. Conclusion: According to the criterion in the test scheme, the result shows that KRG is helpful onrelieving physical fatigue.

        Experimental and Numerical Investigation on the Dynamic Responses of the Remaining Structure under Impact Loading with Column Being Removed

        The dynamic response and stability of building structures under accidental loads such as earthquake, explosion, and impact have been focused by experts. The significant influence will be induced once the damaged structure begins to collapse. In this study, a reinforced concrete frame structure model was designed and tested to study the dynamic response law of the structure under the condition of the failure of the middle column of the model structure, the remaining structure successively failed the load-bearing column and continued to be subjected to impact load and uniform load. The analytical method was used to calculate the ultimate uniform load subjecting to the structure under the failure of four load-bearing columns, because its value was less than the load in this test, the structure collapsed. The finite element calculation model was established, and compared the simulation results to the test results to verify its rationality. Then, the influence of the impact energy and the structural stores on the dynamic response of the structure were analyzed. The results showed that the peak value of the dynamic response of the damaged structure did not increase strictly with the increase of the number of failure columns, which may be related to the sequence of failure columns.

        Angelica sinensis Supercritical Fluid CO2 Extract Attenuates D-Galactose-Induced Liver and Kidney Impairment in Mice by Suppressing Oxidative Stress and Inflammation

        Angelica sinensis (AS, Danggui in Chinese) is an important herbal component of various traditional formulae for the management of asthenia and its tonic effects. Although AS has been shown to ameliorate cognitive damage and nerve toxicity in D-galactose (D-gal)-elicited senescent mice brain, its effects on liver and kidney injury have not yet been explored. In this work, mice were subjected to hypodermic injection with D-gal (200 mg/kg) and orally gavaged with AS (20, 40, or 80 mg/kg) once a day for 8 successive weeks. Results revealed that AS significantly improved liver and kidney function as assessed by organ index and functional parameters. In addition, AS pretreatment effectively ameliorated the histological deterioration. AS attenuated the MDA level and markedly enhanced the activities and gene expressions of antioxidative enzymes, namely Cu, Zn-SOD, CAT, and GPx. Furthermore, AS markedly inhibited the D-gal-mediated increment of expressions of inflammatory cytokines iNOS, COX-2, IκBα, p-IκBα, and p65 and promoted the IκBα expression level in both hepatic and renal tissues. In sum, AS pretreatment could effectively guard the liver and kidney of mice from D-gal-induced injury, and the underlying mechanism was deemed to be intimately related to attenuating oxidative response and inflammatory stress.
