        한국인 본태성 고혈압환자에서 Angiotensinogen M235T 유전자 다양성

        N/A Background: Potential involvement of the angio-tensinogen gene(M235T) in the pathogenesis of essen-tial hypertension has been suggested by some inves- tigators. However, an association between M235T gene polymorphism and essential hypertension has been reported by some, but not by others. Since genetic diversity exists among different ethnic population, we addressed the question of whether there is an association between M235T gene poly-morphism and essential hypertension in the Korean. Methods: 100 patients with essential hypertension and 100 control subjects were recruited from out- patients at the Department of Internal Medicine, Soon-chunhyang Hospital, Chunan City. The criteria for hypertension was defined as systolic blood pressure higher than 160mmHg and/or diastolic blood pressure higher than 95mmHg. For detection of the M235T polymorphism at the angiotensinogen locus, the primer sequences were: sense primer : 5'-TGAAGGAG- AAGGTGTCTGCGGGA-3' and antisense primer : 5'- AGGACGGTGCGGTGAGAGTG-3'. The PCR product mixture was exposed to restriction enzyme Tthlll I and then submitted to electrophoresis in polyacryl-amide gel. Differences between the molecular variants of the gene in hypertensives and normotensives were com- pared by using the X²test. p<0.05 was considered statistically significant. The odds ratio and 95% confidence interval were calculated using Woolf's method. Results: Compared with the control subjects, hypertensives had higher values of three established risk factors for hypertension : age, BMI cholesterol (total and LDL). X analysis showed no difference in the distribution of genotype or allele frequency between the hypertensives and normotensives(X²=1.14, p=0.29). The crude odds ratio was 0.73 for CC over CT(95% confidence interval 0.41-1.30). The adjusted odds ratio with age, sex, lipid profilcs and BMI was 0.68 for CC over CT(9596 confidence interval 037-1,23). Conclusion : The molecular varient M235T of the angiotensinogen gene is not associated with essential hypertension in Korean population.

      • Antioxidant and cytoprotective effects of morin against hydrogen peroxide-induced oxidative stress are associated with the induction of Nrf-2-mediated HO-1 expression in V79-4 Chinese hamster lung fibroblasts

        <P>Natural phytochemicals of plant origin, including flavonoids, have been found to be potent antioxidants providing beneficial effects against oxidative stress-related diseases. The present study was carried out to investigate the antioxidant properties of morin, a flavonoid originally isolated from the flowering plants of the Moraceae family. Superoxide dismutase (SOD)-like activity and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) radical scavenging activity were determined. We also investigated the cytoprotective effects of morin against hydrogen peroxide (H2O2)-induced DNA damage and apoptosis in V79-4 Chinese hamster lung fibroblasts. Our results demonstrated that morin had strong scavenging effects against ABTS' radicals with enhanced SOD activity, which varied in a dose-dependent manner. Morin was found to reduce H2O2-induced intracellular reactive oxygen species generation and nuclear DNA damage, and it recovered cell viability damaged by H2O2 via inhibition of mitochondrial dysfunction-mediated apoptosis. Notably, the treatment of V79-4 cells with morin markedly enhanced the expression of heme oxygenase-1 (HO-1) but not quinone oxidoreductase-1, which was associated with the increased expression and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the downregulation of Kelch-like ECH-associated protein 1 expression. Based on our findings, we conclude that morin effectively ameliorated oxidative stress-induced DNA damage through intrinsic free radical scavenging activity and activation of the Nrf2/HO-1 pathway.</P>

        Caco-2 세포 단층막 투과 실험시 교반이 약물의 투과계수에 미치는 영향

        The unstirred water layer (UWL), which has been known to exist in the boundary of the intestinal lumen and intestinal wall, often behaves as an absorption barrier especially for lipophilic drugs. The intestinal absorption of drugs is often characterized using Caco-2 cell monolayers grown on Transwell polycarbonate membranes. The permeability (Pare) of drugs across the cell monolayer might be influenced by the agitation of the donor compartment, since the width of UWL on the surface of the cell monolayer would be reduced by the agitation. In this study, the effect of agitation of the donor compartment with 60 rpm on the permeability was measured for 12 drugs with a wide range of lipophilicity and permeability. The of mannitol, tributylmethyl ammonium, cimetidine, ranitidine, hydrocortisone, benzylpenicillin and loxoprofen was not influenced by the agitation, while the P_(app) of theophylline, propranolol, YH439, phenylpropanolarnine and testosterone was increased by the agitation. There was a significant correlation between the increase of P_(app) by agitation and the lipophilicity for the compounds having P_(app) > 2 x 10^(-5) cm/sec. No correlation was observed for the difference in P_(app) by agitation and the molecular weight, or lipophilicity of the drugs. Therefore, the agitation rate of the donor compartment in the Caco-2 cell monolayer study should be carefully controlled in order to estimate Pap, reproducibly especially for iipophilic drugs.

        The purpose of this study is to examine the possibility of a systematic theory and methodology for developing an interdisciplinary approach to secretarial science. As a branch of the applied social sciences, secretarial science has had problems which have resulted from disagreements on the concept of "secretary". As Professor Maurice Duverger of the University of Paris has observed, there is a striking contrast between the effectiveness of the applied social sciences and the anarchic state of their theory. As in the case of all other social sciences, secretarial science suffers form a confusion on its history and philosophy. This has produced two results: firstly, a mixture of the analysis of objective facts with the affirmation of normative principles: secondly, the predominance of a priori "practically". Therefore, secretarial science, as a discipline, is seen both a science (theory) and an art (practicality). Historically, in response to practical needs, social sciences have been developing logic, methodology and standards of relevance for scientific inquiry. In order further to refine the concept of secretary, it seems useful to begin by defining the concept of administration in generic terms, as a common social process involving certain common secretarial activities. These activities, which usually include decision-making, programming, staffing without supervision, may be seen in most forms of business or public administration as well. Indeed, to some extent, the differences between administration and secretarial service come to rest largely in the respective environment of each. The office of the secretary may be defined as that aspect of administration occurring under the formal aegis of administrative organization at every level. While recognizing the validity of secretarial science as a subfield in the study of public and business administration, we do not believe that it involves the analysis of administration (management) in all its forms and in all human groups. We must define a "Secretarial service" differently than the way it is commonly defined. For some, secretarial service may be characterized by its ultimate practically. For others, it may be seen as a administrative or managerial science. However, secretarial service should be seen as neither of these. It should be defined as something failing between public and business administration, in other words, as staff service within the framework of organization. The study of secretarial science should start from this point. Because secretarial science is, as mentioned above, a generalized human activity concerned with ordering the men and materials required to achieve organization goals, it has drown widely from the various social science. However, it is impossible to gain any adequate perspective of the field without tracing its background in the areas of public and business administration. Traditionally, the analysis of the field has reflected certain dominant contemporary trends in public and business administration science which are usually recognized as the parent discipline of the field. Each of these tends to be superseded as analytical tools and fashions change. An interdisciplinary approach represents an amalgam of all such styles : as role structural approach, behavioral approach and social philosophical approach. A new secretarial science as one of the applied social science should be pursued at the university level by adopting this interdisciplinary approach.

        In this study, a precipitation method, one of the most effective liquid phase reaction, was adopted in order to prepare high-tech Al₂O₃/ZrO₂ composite ceramics. Al₂(SO₄)₃·18H₂O, ZrOCl₂·8H₂O and YCl₃·6H₂O were used as starting materials and NH₄OH as a precipitation agent. Fine powders were prepared at optimum calcination condition. Sinterability of each fine powder and the effects of ZrO₂ on the grain size and mechanical properties of Al₂O₃ were investigated. The composition of Al₂O₃/ZrO₂ composites was fixed as Al₂O₃-15v/o ZrO₂(+3m/o Y₂O₃). The effect of MgO on the grain size of Al₂O₃ZrO₂ ceramics was also investigated.

        To evaluate the validity of the use of infraorbitale as anterior reference point of articulator system, the vertical distances form FH plane to Nasion, and that from FH plane to incisal edge of maxillary central incisor were measured on lateral cephalograms taken from 76 normal subjects. Results of this study were obtained as follows : 1. The average distance from FH plane to nasion was 22.57±2.67㎜. 2. The average distance from FH plane to the incisal edge of maxillary central incisor was 60.61±4.07㎜.

        This study is to represent basic datas of biomechanical work of human body which has measured precisely and datas on pathology and abnormal gaits paterns. 5 6th grader boys in Taejon city were attended in this experiment. Each student wore 8 landmarks on their lower leg joints and walked for 4 Km/h, 5 Km/h, and 6 Km/h on the tread mill. Mechanical energy of lower extremity segment by lower joint(thigh and shank) movement were measured in stance phase and swing phase by using 2 dimentional image analyzer, and which were evaluated energy of segment The conclusions were as fllowing; 1. Rotational kinetic energy change slightly in lower extremity segment, translational kinetic energy change in the biggest, and value of energy creat greater in swing phase than in stance phase. 2. Energy change in gait cycle increas in swing phase and decrease in stance phase. 3. Perfect energy change of lower extremity segment is not do.

        KLEINE-LEVIN 증후군 2례

        The Kleine-Levin syndrome is composed of recurring episodes of sleepiness lasting days, associated with excessive appetite and often with abnormal behavior. The authors reported two cases of the Kleine-Levin syndrome. The results were as follows: 1. They were adolescent male patients. 2. The onsets were predisposed by psychological stresses of school examinations. 3. Their electroencephalograms revealed abnormal findings only during episodes of hypersomnolence. 4. One of them was improved by treatment with imipramine and lithium.

        The purpose of this study is to present the mathematical interpretation on joint torque while being normal gait which include the muscle tension and use equation of motion with a double rigid-pendulum model. then, the estimates of the stance and swing phase were obtained from photoghaphs. When the angel of thigh(θ₁) and calf (θ₂) of lower leg measured from vertical direction to counterclockwise are compared, the results as follow; 1. It is possible to interpret the mathematical model on the joint torque of lower extremity with double rigid pendulum model. 2. With the conition of double rigid pendulum model, the length of center of mass does not effect at the change of angle.

      • 당뇨병 환자의 혈청 Lipoprotein에 관한 연구

        Lipoprotein cholesterol and triglyceride levels have been determined innormal and diabetes. The abnormalities of lipoprotein were investigated by SDS-polyacrylamide gel electrophoresis in the serum and the 3 major lipoprotein classes for diabetes. HDL cholesterol levels were lower in the diabetes compared to the normals. VLDL triglyceride levels also elevated in the diabetes. There were significant shifts in the distribution of lipoprotein cholesterol, with an increase in LDL cholesterol and decrease in HDL cholesterol. The results of this study suggest that diabetes may be associated with changes in both lipoprotein triglyceride and cholesterol levels. The electrohoretic patterns of VLDL and HDL in diabetes showed the abnorm-al pattern in comparison to normal.
