        한국인 본태성 고혈압환자에서 Angiotensinogen M235T 유전자 다양성

        N/A Background: Potential involvement of the angio-tensinogen gene(M235T) in the pathogenesis of essen-tial hypertension has been suggested by some inves- tigators. However, an association between M235T gene polymorphism and essential hypertension has been reported by some, but not by others. Since genetic diversity exists among different ethnic population, we addressed the question of whether there is an association between M235T gene poly-morphism and essential hypertension in the Korean. Methods: 100 patients with essential hypertension and 100 control subjects were recruited from out- patients at the Department of Internal Medicine, Soon-chunhyang Hospital, Chunan City. The criteria for hypertension was defined as systolic blood pressure higher than 160mmHg and/or diastolic blood pressure higher than 95mmHg. For detection of the M235T polymorphism at the angiotensinogen locus, the primer sequences were: sense primer : 5'-TGAAGGAG- AAGGTGTCTGCGGGA-3' and antisense primer : 5'- AGGACGGTGCGGTGAGAGTG-3'. The PCR product mixture was exposed to restriction enzyme Tthlll I and then submitted to electrophoresis in polyacryl-amide gel. Differences between the molecular variants of the gene in hypertensives and normotensives were com- pared by using the X²test. p<0.05 was considered statistically significant. The odds ratio and 95% confidence interval were calculated using Woolf's method. Results: Compared with the control subjects, hypertensives had higher values of three established risk factors for hypertension : age, BMI cholesterol (total and LDL). X analysis showed no difference in the distribution of genotype or allele frequency between the hypertensives and normotensives(X²=1.14, p=0.29). The crude odds ratio was 0.73 for CC over CT(95% confidence interval 0.41-1.30). The adjusted odds ratio with age, sex, lipid profilcs and BMI was 0.68 for CC over CT(9596 confidence interval 037-1,23). Conclusion : The molecular varient M235T of the angiotensinogen gene is not associated with essential hypertension in Korean population.

        생쥐의 적출관류간장을 이용한 Quinoneimine 화합물의 대사와 독성에 대한 기작

        Quinone compounds are widely distributed in nature and used in therapeutic medicines. Quinones, however, are known to cause severe hepatic toxicity. Generally, quinones undergo two distinctly reductive metabolic processes; one-electron reduction catalyzed by the microsomal NADPH: cytochrome P-450 reductase, producing the toxic semiquinone which may be detoxified by glutathione (GSH) conjugation, and two-electron reduction catalyzed by the cytoplasmic NAD(P)H: quinone reductase (QR), producing the nontoxic hydroquinone which is easily conjugated and excreted. To evaluate and to compare the protective role of GSH conjugation and QR reduction abilities from the quinone inducible hepatotoxicity, we have pretreated mice either with diethylmaleate (DEM), which causes the depletion of GSH, or with dicoumarol (DC), which causes the inhibition of QR. Livers were isolated from the mouse and perfused with 2, 6-dichlorophenone indophenone (DC1P), quinoneimine comound used as a model quinone. The rates of formations of two-electron reduced DCIP and its consecutive glucuronide conjugate (measure of QR-dependent protection), as well as the release of cytoplasmic lactate dehydrogenase (LDH), a marker enzyme of hepatotoxicity (measure of GSH-dependent protection) have been determined. Results obtained by infusing 100μM DCIP have shown that the formation rates of reduced DCIP and its consecutive glucuronide conjugate were decreased and the cytoplasmic LDH began to be released into perfusates much earlier, under the condition where the QR activity was severely inhibited with DC pretreatment. Also, when the hepatic content of GSH was depleted by the pretreatment with DEM and then the liver was infused with 100μM DCIP, the cytoplasmic LDH began to be released much earlier. In both of the control and DEM or DC pretreated mice livers, the releasing time of DCIP-glucuronide conjugates almost coincides with that of LDH after the prolonged infusion with DCIP. Therefore, this result indicates that the metabolic activation of quinoneimine compound leads to the hepatotoxicity and the protective involvement of GSH and/or QR does concurrently to the detoxification.

        2015 개정 교육과정을 반영한 고등학교 영어 I 교과서의 문화 내용 분석

        The present study aims at analyzing the cultural content in high school English I textbooks based on 2015 Revised Curriculum, using three categories: cultural subjects, cultural backgrounds, and cultural types. The findings of the study are 1) the textbooks dealt with the cultural subjects in a different rate ranged from 5 to 11; 2) in terms of cultural backgrounds, none of the textbooks include cultural comparison, and British/American culture was least dealt with in the textbooks; and 3) when it comes to cultural types, behavioral culture was included in the textbooks much less than mental culture and material culture. In general, it is required to be taken into consideration, when developing English textbooks, that various and balanced cultural content is need to be included in the textbook in terms of cultural subjects, backgrounds, and types in order to help EFL learners enhance their intercultural competence.

        유전 및 육종 : 버크셔종의 육질형질에 대한 유전모수 추정

        본 연구는 2004년~2008년에 미국에서 도입한 버크셔종의 육질형질에 대한 유전적 특성을 파악하고, 향후 선발에 이용한 유전적 그룹(genetic group)을 분리하여 선발 및 교배계획을 체계적으로 세워 효과를 극대화시키는 전략을 세우고자 수행하였다. 그 결과 사후 대사율의 척도인 근육의 사후 45분 pH, 24시간 pH에 대한 유전력은 각각 0.60과 0.61로 높게 조사되었다. 육색의 척도인 명도, 적색도 및 황색도의 유전력은 0.56, 0.58 및 0.62로 조사되었다. 근내지방도, 유리육즙량과 가열감량에 대한 유전력은 각각 0.57, 0.51 및 0.66으로 조사되었다. 본 연구 결과로 미루어 볼 때 육질형질의 개량에 있어 육종가를 기초로 하여 선발에 활용할 경우 유전적 개량량은 크게 나타날 것으로 사료된다. 또한 사후 대사율의 척도인 pH와 육질형질과의 상관도는 높은 연관성이 존재하므로, 종돈장에서 육질형질을 개량하고자 한다면 육종가를 기반으로 한 선발지수(selection index)를 활용하면 매우 효과적일 것으로 필자는 사료된다. 그러나 본 연구에 활용된 자료는 유전모수를 추정하는데 다소 부족한 것은 사실이지만 국내 최초로 돼지의 육질형질에 대한 유전모수를 추정하는데 있어 매우 의미가 있다고 할 수 있다. Swine industry in Korea plays an important role in providing the meat for domestic consumption, and the number of pigs in Korea was about 9.72 million heads as of June, 2010. Meat quality is used to describe any traits which impact the consumer acceptability of fresh meat products. Meat color, firmness, water holding capacity, ultimate muscle pH24h(measured 24 hours post-mortem), shear force, and intramuscular fat percentage(IMF) are generally accepted as important indicators of meat quality and ultimately, consumer acceptance of fresh pork. The objective of this study was to estimate genetic parameters for meat quality traits in Berkshire pigs. The heritability estimates for muscle pH24h, lightness(CIE L*), NPPC marbling were 0.61, 0.56 and 0.57, respectively, The heritability estimates for drip loss, cooking loss, shear force were 0.51, 0.66 and 0.56, respectively. The phenotypic correlations between pH24h and lightness(CIE L*), drip loss, cooking loss were negative, ranging from -.45~-.13. The genetic correlations between muscle pH24h and lightness(CIE L*), drip loss were negative, ranging from -.35~-.32. Genetic parameters obtained herein indicate that genetic improvement of muscle pH24h is not related to the NPPC marbling of meat, but rather to improved lightness(CIE L*) and drip loss. Genetic trends of meat quality traits showed increased muscle pH24h and decreased cooking loss and drip loss.

        Homocysteine 과다증과 관련된 반복 자연유산에서 Methylenetetrahydrofolate Reductase돌연변이에 대한 분석

        Objective: To analyze the methylenetetrahydrofolate reductase (MTHFR) mutation in recurrent spontaneous abortion associated with hyperhomocysteinemia. Material and Method: The blood sample of habitual aborter with high fasting homocysteine level was tested by PCR - RFLP method. Results: The patient was found to be a homozygosity for MTHFR gene mutation that was confirmed by the finding which is consistent with the mutation at the nucleotide 677 C to T, corresponding to Ala to Val. Conclusions: Hyperhomocysteinemia due to MTHFR mutation is a cause of recurrent spontaneous abortion. Therefore, the MTHFR mutation should be examined in the workup of recurrent spontaneous abortion showing hyperhomocysteinemia.

        생장 및 부상성이 향상된 남세균 돌연변이 균주 Arthrospira platensis M20CJK3

        A photosynthetic cyanobacterium Arthrospira platensis, well known for health food supplement, was studied as a target species for atmospheric $CO_2$ removal as well as biomass production. Although the biomass of A. platensis was massively produced in many countries, the recovery cost of its biomass is still high. The purpose of this study was to develop the A. platensis mutant strains which have enhanced growth rate and floatation activity to reduce the recovery cost. A. platensis KCTC AG20590 was treated with 0.24% ethyl methanesulfonate (EMS) for 20 min at room temperature. The mutant strain A. platensis M20CJK3 was finally selected by its morphological and physiological features. The morphology of the mutant A. platensis M20CJK3 was changed from loose-coiled form to tight-coiled form showing short pitch. The growth and $CO_2$ uptake rate of A. platensis M20CJK3 were improved about 15% and 17% compared with A. platensis KCTC AG20590, respectively. The floatation activity of A. platensis M20CJK3 was enhanced in 2-fold compared with that of A. platensis KCTC AG20590. Soluble proteins extracted from two strains were analyzed by two dimensional electrophoresis (2-DE) and MALDI-TOF MS/MS. Among 15 protein spots induced in 2-DE analysis, two spots were the proteins related to photosynthesis and electron transfer system of the other cyanobacteria. As a consequence, it seems that the tight-coiled mutant A. platensis M20CJK3 has an advantage of high growth rate and floatation activity which are beneficial for the mass cultivation and recovery. 건강보조식품 등으로 이용되는 Arthrospira platensis는 세계적으로 대량생산되고 있으나 생산공정 중 수확단계에서 많은 비용이 소요된다. 본 연구에서는 부상을 이용한 효과적인 수확을 위해 균주의 개량을 시도하였으며, 개량균주의 생리적 물리적 특성을 파악하고자 하였다. Ethyl methanesulfonate (EMS)를 모균주 A. platensis KCTC AG20590에 0.24%의 농도로 10, 20, 30분씩 처리하여, 형태 및 부상성이 우수한 균주 A. platensis M20CJK3 균주를 분리하였다. A. platensis M20CJK3은 느슨한 형태에서 촘촘한 형태로 세포사(trichome)의 길이 및 코일간 간격이 감소하였으며, 생장 및 $CO_2$ 고정능이 각각 15%, 17% 향상되었다. 또한, 개량균주의 부상성은 모균주에 비해 2배 이상 향상되었다. 이차원 전기영동 분석을 통해 모균주와 개량균주의 단백질 발현양상을 비교분석한 결과 광합성 관련 색소의 구조와 광전자전달계에 관련된 단백질의 발현 양상이 차이를 보였다. 본 연구에서 개발된 A. platensis M20CJK3은 고밀도 대량배양 및 수확에 유리하며, A. platensis 유전자 연구의 유용한 모델 균주로 활용될 수 있을 것으로 사료된다.
