      • Application of a wet oxidation method for the quantification of <sup>3</sup>H and <sup>14</sup>C in low-level radwastes

        Wet oxidation methods are utilized to separate and quantify <SUP>3</SUP>H and <SUP>14</SUP>C radionuclides in inorganic and organic radwastes generated at nuclear facilities. Because <SUP>3</SUP>H and <SUP>14</SUP>C are pure beta emitters with half-lives of 12.3 and 5,730 years, respectively, these radionuclides should be chemically separated from other radionuclides present in radwastes for accurate quantification. In particular, a collection technique for <SUP>14</SUP>C radionuclide in radwastes is needed because it is converted into <SUP>14</SUP>CO<SUB>2</SUB> gas by an oxidation reaction. To confirm the recoveries of <SUP>3</SUP>H and <SUP>14</SUP>C, various standard radioactive sources were used to verify the proposed method. Because the majority of <SUP>3</SUP>H radionuclides are distributed in tritiated water (HTO), only tritiated water was used as a standard for <SUP>3</SUP>H radionuclides. Additionally, <SUP>14</SUP>C-labeled methanol (<SUP>14</SUP>CH<SUB>3</SUB>OH), lauric acid (<SUP>14</SUP>CH<SUB>3</SUB>(CH<SUB>2</SUB>)<SUB>10</SUB>COOH), sodium bicarbonate (NaH<SUP>14</SUP>CO<SUB>3</SUB>), and toluene (C<SUB>6</SUB>H<SUB>5</SUB><SUP>14</SUP>CH<SUB>3</SUB>) were used as <SUP>14</SUP>C standards. The compounds were oxidized with chemical oxidants and then separated. The individual species were mixed with a scintillation cocktail and counted using a liquid scintillation counter. The recoveries of <SUP>14</SUP>C and <SUP>3</SUP>H were 82-97% and 98%, respectively. The wet oxidation method will be applied to RI wastes for clearance.

        Development of Naturally Degradable “Rice Polymer" For Organic Weed Management of Red Pepper and Rice

        Among the developed bío-degradable polymer fílms as compared wíth transparent fílm(Whíte), black polymer fílm was sígnífícantly more effectíve for controllíng weeds ín red pepper. Also, we found that whíte and black polymer mulchíng had 81.8% and 97.9% of managing weed controllíng effects in rice, respectively. Compared to nonmulched rice paddy with water supply, the non-mulched rice paddy wíthout any water supply has stopped its growth at 41 days after transplantíng, whíle polymer -mulched rice paddy without water supply had about 60% of normally growing rice plants. This shows the polymer treatment has a remarkable effect on water and power saving, solution of herbicidal resistance, avoidance of herbícidal influence to eco-system etc. When the naturally decomposíng polymer was used, a temperature was elevated as high as 4.7 ℃ on maximum and 2.6 ℃ on average. Also the naturally decomposing polymer accelerated rooting by 7 days and lowered a stress level from transplanting. The weed control effect mulched by polymer was remarkable as 98. 7%. The p이ymer now, after 294 days treated on the rice paddy, has been completely decomposed.

        Enhancement in carbon dioxide activity and stability on nanostructured silver electrode and the role of oxygen

        <P>Current energy production habits deplete fossil fuels and accumulate atmospheric CO2, which contribute to the global climate change. Electrochemical fuel production via CO2 reduction reaction is an idealistic yet an achievable process that mitigates CO2 emissions and simultaneously satisfies energy demands. Here, the enhancement of CO2 reduction activity and stability on size-controlled particulate Ag electrocatalysts derived from a simple, one-step cyclic voltammetry (CV) process by changing scan rates (1-200 mV/s) was demonstrated. Interestingly, larger nanoparticles prepared by slower scan rates (1-5 mV/s) have exhibited the most degree of enhancement for CO2 reduction to CO product. Compared to untreated Ag foil, nanostructured Ag electrode has shown an anodic shift of approximately 200 mV in the onset potential of CO partial current density (J(CO)), 160 mV reduction of overpotential j(CO) = 10 mA/cm(2), and increased Faradaic efficiency (F.E.) for CO production especially at lower biased potentials (-0.89 to -1.19V vs. RHE). Stability tests have demonstrated a drastic improvement in maintaining CO F.E. Xray photoelectron spectroscopy suggests that the enhancement is associated with stable oxygen species incorporated on the nanoparticle Ag surfaces during the CV fabrication process.         재생 불량성 빈혈 및 골수 이형성군 환자의 요중 Erythropoietin 활성도의 Bioassay

        Erythropoietin, a glycoprotein hormone, has an important and well-established role in regulating the developement of RBCs from committed erythropoietic precussor. Its activity can be measured by bioassay or RIA. Recently, we investigated urinary erythropoietin activity in A. A. patients and MDS patients by bioassay and RIA. In bioassay, we used as 3H-thymidine incorperation method to BACB-C mouse spleen cell. Thereafter we compared urinary Epo. acitivity in each disease group and like to find out prognostic significance to be influenced. The results were summarized as follows; 1. The urinary Epo. activities of the A.A. and MDS groups were higher than those of the normal control gorup. and the urinary Epo. activities of the A.A. group showed a marked increase over those of the MDS group (105.5±81.6 vs 1.7±1.5). But the after immune modulation therapyu, urinary Epo. acitivities of the A.A. group markedly decreased to the normal value (105.5±81.6 vs 1.8±1.7, p<0.001). 2. Age and sex differences were not related with urinary Epo. activities in each goup. 3. The hematocrits of the A.A. group did not correlate with any the urinary Epo. acitivities, but an inverse correlation was found in the MDS group. 4. The urinary Epo. activities measured by RIA were higher than those measured by bioassay, but there was a close corrlation between the two methods(R=0. 8, p< 0.001).

      • Gold catalyst reactivity for CO<sub>2</sub> electro-reduction: From nano particle to layer

        <P>CO2 electro-reduction is a promising method for sustainable production of carbon fuels as well as valuable chemicals. Herein, we investigated a direct electro-catalytic CO2 conversion to CO by nanostructured gold catalysts from the small nanoparticles to aggregated clusters to layered film. The selectivity of CO formation was found to increase as the gold amount increased and it reached a saturation point of 78% (-0.59 V vs. RHE) at the morphological transition from aggregated cluster to layered film. Furthermore, the similar to 4 nm gold nanoparticle exhibited a remarkable CO formation mass activity value at 166.1 A/g (-0.59 V vs. RHE). (C) 2015 Elsevier B.V. All rights reserved.</P>

        The genetic defects in human gametes and embryos can cause adverse effects on overall reproductive events. Biopsy of embryos for preimplantation genetic diagnosis (PGD) offers a new possibility of having children free of the genetic disease. In addition, advanced embryo culture method may enhance the effectiveness of embryo biopsy for the practical application of PGD. This experimental study was undertaken to evaluate the effects of coculture on the development in vitro of biopsied mouse embryos as a preclinical model for PGD of human embryos. Embryos were obtained after in vitro fertilization (IVF) from F1 hybrid mice (C57BLfemale/CBAmale). Using micromanipulation, 1, 2, 3 or 4 blastomeres of 8-cell stage embryos were aspirated through a hole made in the zona pellucida by zona drilling (ZD) with acidic Tyrode's solution (ATS). After biopsy of blastomeres, embryos were cultured in vitro for 110 hours in Ham's F-10 supplemented with 0.4% BSA or cocultured on the monolayer of Vero cells in the same medium. The frequence of blastocyst formation were recorded, and the embryos beyond blastocyst stage were stained with 10% Giemsa to count the total number of nuclei in each embryo. There was no significant difference in the blastocyst formation between the zona intact control group and the zona drilling (ZD) only, or biopsied groups. The hatching rate of all the treatment groups except 4/8 group was significantly higher than that of control group. In all the treatment groups, there was a significant reduction in the mean cell number of embryos beyond blastocyst stage ($50.2{\pm}14.0$ in control group vs. $41.2{\pm}7.9$ in ZD, $39.3{\pm}8.8$ in 7/8, $29.7{\pm}6.4$ in 6/8, $25.1{\pm}5.7$ in 5/8, and $22.1{\pm}4.3$ in 4/8 groups, p<0.05). When the same treatments were followed by coculture with Vero cells, a similar pattern was seen in the blastocyst formation and the hatching rate. However, in all the treatment groups, there was a significant increase in the mean cell number of embryos beyond blastocyst stage with coculture, compared with the parallel groups without coculture. In the cleavage rate of biopsied blastomeres cultured for 110 hours after IVF, there was no significant difference between coculture and non-coculture groups (87.2% vs. 78.7%). However, the mean cell number of embryos developed from the biopsied blastomeres was significantly higher in coculture group ($11.5{\pm}4.7\;vs.\;5.9{\pm}1.9$, p<0.05). In conclusion, biopsy of mouse embryos after ZD with ATS is a safe and highly efficient method for PGD, and coculture with Vero cells showed a positive effect on the development in vitro of biopsied mouse embryos and blastomeres as a preclinical model for PGD of human embryos.

        각종 갑상선질환에서의 항Thyroglobulin 항체 및 항Microsome 항체의 출현빈도에 관한 연구

        저자들은 각종 갑상선질환에서 갑상선자가항체의 출현빈도를 조사하여 이들 항체가 갖는 진단적 가치를 관찰하고저, 1978년 4월부터 1979년 4월사이에 서울대학교병원 내과에 내원하였던 각종 갑상선질환환자 108명과 정상인 15명을 대상으로 하여, 감작혈구응집반응을 이용하여 항 thyroglobulin 항체와 microsome 항체를 측정하여 다음과 같은 결론을 얻었다. 1) 정상인 15명에서의 항 thyroglobulin 항체는 모두 음성이었는데, 항 microsome 항체는 1명(6.7%)에서 양성이었다. 각종 갑상선질환환자 108명에서의 결과는 각각 34명(31.5%) 및 40명(37.0%)이 양성이었다. 2) Graves병 환자 25명에서는 항 thyroglobulin 항체는 7명(28.0%)이 양성이었으나 항 microsome 항체는 9명(36.0%)에서 양성이었고, 음성인 군과 양성군 사이에 임상소견 및 검사소견이 특별한 차이를 볼 수 없었다. 3) Hashimoto병 환자는 18명으로 이들은 모두 침생검소견으로 확진된 예들로서, 두가지 항체가 각각 16명(88.9%)과 17명(94.4%)에서 양성이었다. 4) Familial goiter 환자는 7명이었는데 각각 4명(57%) 및 6(85%)명에서 양성이었다. 5) 양성갑성선종 35명에서는 각각 2명(5.7%) 및 3명(8.6%)이 양성의 소견을 보였으며, 갑상선암종 16명에서는 각각 3명(18.8%)이 양성이었다 6) 2명의 아급성갑상선염환자는 두가지가 모두 음성이었던데 반하여, 원발성점액수종환자 1명은 수검검사에서 양성이었다. 7) 1:802 이상의 고력가를 보인 예는 항 thyroglobulin 항체의 경우는 16명(14.8%)이었고, 이중 10명(62.5%)이 Hashimoto병이었으며, 항 microsome항체의 경우는 20명중 13명(65.0%)이 Hashimoto병이었다. 8) Tg Ab와 Mc Ab의 상관계수는 0.76으로 상관관계가 비교적 높으며 Mc Ab의 양성빈도가 약간 높았다. 이상의 결과를 종합하면 감작혈구응집반응은 간편하고 예민하며 신뢰도 및 재현성이 높은 검사로 생각되며 이 방법으로 측정한 항 thyroglobulin 항체 및 항 microsome 항체는 Hashimoto병에서 그 검출빈도가 높고 그 역가도 높아 Hashimoto병의 진단에 유용하다고 사료된다. The authors investigated the incidence of antithyroglobulin antibodies and antibodies and antimicrosomal antibodies measured by tanned red cell hemagglutination method in subjects suffering from various thyroid disorders. 1) In 15 normal patients, neither suffering from any thyroid diseases nor from any other autoimmune disorders, the antithyroglobulin antibodies were all negative, but the antimicrosomal antibody was positive only in one patient(6.7%). 2) The antithyroglobulin antibodies were positive in 31.5%(34 patients) of 108 patients with various thyroid diseases, and the antimicrosomal antibodies were positive in 37.0%(40 patients). 3) of the 25 patients with Graves' diseases, 7 patients(28.0%) showed positive for the antithyroglobulin antibodies, and 9(36.0%) for the antimicrosomal antibodies. There was no definite differences in clinical and thyroid functions between the groups with positive and negative results. 4) Both antibodies were positive in 16(88.9%) and 17(94.4%) patients respectively among 18 patients with Hashimoto's thyroiditis, all of them were diagnosed histologically. 5) Three out of 33 patients with thyroid adenoma showed positive antibodies, and 3 of 16 patients with thyroid carcinoma revealed positive antibodies. 6) TRCH antibodies demonstrated negative results in 2 patients with subacute thyroiditis, but positive in one patient with idiopathic primary myxedema. 7) The number of patiencs with high titers($gt;l:802) was 16 for antithyroglobulin antibody, and 62.5%(10 patients) of which was Hashimoto's thyroiditis. Thirteen(65.0) of 20 patients with high titers($gt;l:802) for antimicrosomal antibody was Hashimoto's thyroiditis. TRCH test is a simple, sensitive method, and has high reliablity and reproducibility. The incidences and titers of antithyroglobulin antibody and antimicrosomal antibody are especially high in Hashimoto's thyroiditis.
