        Whole genome sequencing of Luxi Black Head sheep for screening selection signatures associated with important traits

        Objective: Luxi Black Head sheep (LBH) is the first crossbreed specialized for meat production and was developed by crossbreeding Black Head Dorper sheep (DP) and Small Tailed Han sheep (STH) in the farming areas of northern China. Research on the genomic variations and selection signatures of LBH caused by continuous artificial selection is of great significance for identifying the genetic mechanisms of important traits of sheep and for the continuous breeding of LBH.Methods: We explored the genetic relationships of LBH, DP, and several Mongolian sheep breeds by constructing phylogenetic tree, principal component analysis and linkage disequilibrium analysis. In addition, we analysed 29 whole genomes of sheep. The genomewide selection signatures have been scanned with four methods: heterozygosity (HP), fixation index (FST), cross-population extended haplotype homozygosity (XP-EHH) and the nucleotide diversity (θπ) ratio.Results: The genetic relationships analysis showed that LBH appeared to be an independent cluster closer to DP. The candidate signatures of positive selection in sheep genome revealed candidate genes for developmental process (HoxA gene cluster, BCL2L11, TSHR), immunity (CXCL6, CXCL1, SKAP2, PTK6, MST1R), growth (PDGFD, FGF18, SRF, SOCS2), and reproduction (BCAS3, TRIM24, ASTL, FNDC3A). Moreover, two signalling pathways closely related to reproduction, the thyroid hormone signalling pathway and the oxytocin signalling pathway, were detected.Conclusion: The selective sweep analysis of LBH genome revealed candidate genes and signalling pathways associated with developmental process, immunity, growth, and reproduction. Our findings provide a valuable resource for sheep breeding and insight into the mechanisms of artificial selection.

        LINC00662 Promotes Oral Squamous Cell Carcinoma Cell Growth and Metastasis through miR-144-3p/EZH2 Axis

        Purpose: Long non-coding RNA (lncRNA) is identified as an important regulator involved in oral squamous cell carcinoma(OSCC) tumorigenesis. This study aimed to investigate the functional role and underlying mechanism of LINC00662 in OSCC. Materials and Methods: The expression levels of LINC00662, miR-144-3p, and enhancer of zeste homolog 2 (EZH2) mRNA werequantified with quantitative real-time polymerase chain reaction in OSCC tissues and cell lines. Western blot analysis was used toassay the expression levels of E-cadherin, Vimentin, and EZH2. Cell proliferation, migration, and invasion were monitored by cellcounting kit-8 and Transwell assays. Dual-luciferase reporter and RNA immunoprecipitation assays were employed to verify theregulatory relationship between LINC00662 and miR-144-3p. Results: The expression of LINC00662, positively associated with the increased TNM stage and lymph node metastasis of the patients,was up-regulated in OSCC tissues and cells. The overexpression of LINC00662 facilitated the proliferation, migration, andinvasion of OSCC cells. MiR-144-3p could bind to LINC00662, and the promoting effect of LINC00662 overexpression was counteractedby miR-144-3p mimic. Moreover, EZH2 expression was negatively regulated by miR-144-3p and positively regulated byLINC00662. The silencing of EZH2 attenuated the promoting effects of overexpression of LINC00662 on cell proliferation, migration,invasion, and epithelial-mesenchymal transition. Conclusion: LINC00662, as an oncogenic lncRNA of OSCC, accelerates OSCC progression by repressing miR-144-3p expressionand increasing EZH2 expression.

        Ni Nanoparticle Anchored on MWCNT as a Novel Electrochemical Sensor for Detection of Phenol

        Increasing active sites and enhancing electric conductivity are critical factors to improve sensing performance toward phenol. Herein, Ni nanoparticle was successfully anchored on acidified multiwalled carbon nanotube (a-MWCNT) surface by electroless plating technique to avoid Ni nanoparticle agglomeration and guarantee high conductivity. The crystal structure, phase composition and surface morphology were characterized by XRD, SEM and TEM measurement. The as-prepared Ni/a-MWCNT nanohybrid was immobilized onto glassy carbon electrode (GCE) surface for constructing phenol sensor. The phenol sensing performance indicated that Ni/a-MWCNT/GCE exhibited an amazing detection performance with rapid response time of 4s, a relatively wide detection range from 0.01 mM to 0.48 mM, a detection limit of 7.07 μM and high sensitivity of 566.2 μAmM -1 cm -2. The superior selectivity, reproducibility, stability and applicability in real sample of Ni/a-MWCNT/GCE endowed it with potential application in discharged wastewater.

        Active vibration control based on modal controller considering structure-actuator interaction

        Active vibration control to suppress structural vibration of the flexible structure is investigated based on a new control strategy considering structure-actuator interaction. The experimental system consists of a clamped-free rectangular plate, a controller based on modal control switching, and a magnetostrictive actuator utilized for suppressing the vibrations induced by external excitation. For the flexible structure, its deformation caused by the external actuator will affect the active control effect. Thus interaction between structure and actuator is considered, and the interaction model based on magnetomechanical coupling is incorporated into the control system. Vibration reduction strategy has been performed resorting to the actuator in optimal position to suppress the specified modes using LQR (linear quadratic regulator) based on modal control switching. The experimental results demonstrate the effectiveness of the proposed methodology. Considering structure-actuator interaction (SAI) is a key procedure in controller design especially for flexible structures.

        miR-143 inhibits proliferation and induces apoptosis of mammary epithelial cells in dairy goat

        MicroRNAs are a class of post-transcriptional regulators of gene expression in multicellular organisms, which play important roles in cell fate, organ morphogenesis and carcinogenesis. In the present study, we demonstrated the critical roles of miR-143 on mammary epithelial cells of dairy goat. The test results of cell count, methylthiazolyldiphenyl-tetrazolium bromide, Hoechst33342/PI (propidium iodide) and flow cytometry showed miR-143-induced G0/G1 phase arrest, inhibited proliferation and promoted apoptosis of mammary epithelial cells; the qRT-PCR test of marker genes related to cell proliferation and apoptosis, BAX and BCL-2, supported the same conclusions. Our study presents evidence that miR-143 is an important post-transcription regulator involving in mammary cells survival, and it may have a value function in mammary gland development, lactation or involution.

        Identification and Function Prediction of Novel MicroRNAs in Laoshan Dairy Goats

        MicroRNAs are a class of endogenous small RNAs that play important roles in post-transcriptional gene regulation by directing degradation of mRNAs or facilitating repression of target gene translation. In this study, three small RNA cDNA libraries from the mammary gland tissues of Laoshan dairy goats (Capra hircus) were constructed and sequenced, individually. Through Solexa high-throughput sequencing and bioinformatics analysis, we obtained 50 presumptive novel miRNAs candidates, and 55,448 putative target genes were predicted. GO annotations and KEGG pathway analyses showed the majority of target genes were involved in various biological processes and metabolic pathways. Our results discovered more information about the regulation network between miRNAs and mRNAs and paved a foundation for the molecular genetics of mammary gland development in goats.
