Metastasis associated genomic aberrations in stage II rectal cancer

Genomic aberrations of rectal carcinoma, especially DNA copy number changes associated with metastasis were largely unclear. We aim to identify the metastasis associated biomarkers in stage II rectal cancer. Formalin-fixed, paraffin-embedded primary tumor tissues of stage II rectal carcinoma were analyzed by array-based comparative genomic hybridization, and genomic aberrations were identified by Genomic Workbench and SAM software. Copy number changes and mRNA expressions were validated by Real-time PCR in an independent rectal cancer samples. The results showed that the most frequent gains in stage II rectal cancer were at 1q21.2-q23.1, 3p21.31, 11q12.2-q23.3, 12q24.11-q24.31, 12q13.11-q14.1 and losses in 18q11.2-q23, 17q21.33-q22, 13q31.1-q31.3, 21q21.1-q21.3, 8p23.3-p23.1 and 4q22.1-q23. Twenty-two amplifications and five homozygous deletions were also identified. We further found that S100A1 (1q21.3-q23.1), MCM7 (7q22.1) and JUND (19p13.11) were amplified and overexpressed in stage II rectal cancer. Interestingly, the genomic aberrations affected 14 signaling pathways including VEGF signaling pathway and fatty acid metabolism. Most importantly, loss of 13q31.1-q34 and gain of 1q44 were associated with distant metastasis. Our results indicated that these metastasis associated genomic changes may be useful to reveal the pathogenesis of rectal cancer metastasis and identify candidate biomarkers.

Although realgar bioleaching solution (RBS) has been proved to be a potential candidate for cancer therapy, the mechanisms of RBS anticancer are still far from being completely understood. Dosed with RBS in C. elegans, the multivulva phenotype resulting from oncogenic ras gain-of-function was inhibited in a dose dependent manner. It could be abrogated by concurrent treatment C. elegans with RBS and the radical scavenger DMSO. However, RBS could not induce DAF-16 nuclear translocation in TJ356 or the increase of HSP 16.2 expression in CL2070, which both could be aroused visible GFP fluorescent variation to represent for oxidative stress generation. Treatment C. elegans with superoxide anion generator paraquat, similar results were also obtained. Our results indicated that RBS suppress excessive activated ras by increasing reactive oxygen species (ROS) in C. elegans. Secondly, ROS induced by RBS significantly accumulated on a higher level inC. elegans with amutational ras than that with wild ras, thus leading to oxidative stress on ras gain-of-function background rather than on normal ras context. Our results firstly demonstrated that using C. elegans as amodel organism for evaluating prooxidant drug candidates for cancer therapy.

Background: Colorectal cancer (CRC) is a leading cause of death worldwide. Chronic gut inflammation is recognized as a risk factor for tumor development, including CRC. American ginseng is a very commonly used ginseng species in the West. Methods: A genetically engineered $Apc^{Min/+}$ mouse model was used in this study. We analyzed the saponin composition of American ginseng used in this project, and evaluated its effects on the progression of high-fat-diet-enhanced CRC carcinogenesis. Results: After oral ginseng administration (10-20 mg/kg/d for up to 32 wk), experimental data showed that, compared with the untreated mice, ginseng very significantly reduced tumor initiation and progression in both the small intestine (including the proximal end, middle end, and distal end) and the colon (all p < 0.01). This tumor number reduction was more obvious in those mice treated with a low dose of ginseng. The tumor multiplicity data were supported by body weight changes and gut tissue histology examinations. In addition, quantitative real-time polymerase chain reaction analysis showed that compared with the untreated group, ginseng very significantly reduced the gene expression of inflammatory cytokines, including interleukin-$1{\alpha}$ (IL-$1{\alpha}$), IL-$1{\beta}$, IL-6, tumor necrosis factor-${\alpha}$, granulocyte-colony stimulating factor, and granulocyte-macrophage colony-stimulating factor in both the small intestine and the colon (all p < 0.01). Conclusion: Further studies are needed to link our observed effects to the actions of the gut microbiome in converting the parent ginsenosides to bioactive ginseng metabolites. Our data suggest that American ginseng may have potential value in CRC chemoprevention.
American ginseng attenuates azoxymethane/dextran sodium sulfate-induced colon carcinogenesis in mice

Background: Colorectal cancer is a leading cause of cancer-related death, and inflammatory bowel disease is a risk factor for this malignancy. We previously reported colon cancer chemoprevention potential using American ginseng (AG) in a xenograft mice model. However, the nude mouse model is not a gut-specific colon carcinogenesis animal model. Methods: In this study, an experimental colitis and colitis-associated colorectal carcinogenesis mouse model, chemically induced by azoxymethane/dextran sodium sulfate (DSS) was established and the effects of oral AG were evaluated. The contents of representative ginseng saponins in the extract were determined. Results: AG significantly reduced experimental colitis measured by the disease activity index scores. This suppression of the experimental colitis was not only evident during DSS treatment, but also very obvious after the cessation of DSS, suggesting that the ginseng significantly promoted recovery from the colitis. Consistent with the anti-inflammation data, we showed that ginseng very significantly attenuated azoxymethane/DSS-induced colon carcinogenesis by reducing the colon tumor number and tumor load. The ginseng also effectively suppressed DSS-induced proinflammatory cytokines activation using an enzyme-linked immunosorbent assay array, in which 12 proinflammatory cytokine levels were assessed, and this effect was supported subsequently by real-time polymerase chain reaction data. Conclusion: AG, as a candidate of botanical-based colon cancer chemoprevention, should be further investigated for its potential clinical utility.
Driving shaft fatigue optimization design of W type profile twin-screw pumps

Under changeable pumped medium and working environment, the twin-screw pump is prone to be broken by fatigue failures. A structure optimization design model and method of the driving shaft are presented based on response surface methodology and finite element analysis. In this model, the shaft diameter, chamfering degree and the shaft extension of the power end are selected as optimization variables, the limit values of the variables and maximal normal deformation of the spindle are considered as the constraint conditions, and the minimization of the equivalent alternating stress on the dangerous shaft section is taken as the optimization objective so as to improve the shaft fatigue reliability. The optimization results of a case show that the equivalent alternating stress on the dangerous spindle section reduces by 26.2 %, and the maximal normal deformation decreases by 25.2 % compared with the original design. In addition, the infinite life reliability and fatigue safety factors both meet the design requirements.
A guided bone regeneration membrane composed of hydroxyapatite and polyurethane

A hydroxyapatite (HA,10 wt%) and polyurethane (PU) composite as a guided bone regeneration (GBR)membrane was obtained from the polycondensation or polyaddition of diisocyanates and hydroxyl groups by a solvent evaporation method. The structure and properties of the membrane were investigated by XRD, IR, SEM, water absorption, wettability and a cell culture test in vitro. The results show that hydroxyapatite particles were dispersed uniformly in the polyurethane matrix and interfacial bonding between hydroxyapatite and polyurethane was formed. The hydroxyapatite/polyurethane membrane has good hydrophilicity and the surface pores can promote cell adhesion and growth. MTT (3-{4,5-dimethylthiazol-2yl}-2,5-diphenyl-2H-tetrazolium bromide) assay and light microscopy observation indicate that the hydroxyapatite/polyurethane membrane demonstrates excellent biocompatibility. The hydroxyapatite/polyurethane membrane will hopefully be selected as a guided bone regeneration and tissue engineering.
Overexpression of Cyclooxygenase-1 Correlates with Poor Prognosis in Renal Cell Carcinoma

The aim of this study was to evaluate expression of COX-1 in renal cell carcinoma (RCC) and its prognostic value. mRNA of COX-1 was detected in 42 paired RCC and adjacent normal tissues with quantitative realtime polymerase chain reaction (qRT-PCR). Expression of COX-1 was also evaluated in 196 RCC sections and 91 adjacent normal tissues with immunohistochemistry. Statistical analysis was performed to assess COX-1 expression in RCC and its prognostic significance. The results of qRT-PCR showed mRNA levels of COX-1 in RCC tissues to be significantly higher than that in adjacent normal tissues (p < 0.001). Immunohistochemical assays also revealed COX-1 to be overexpressed in RCC tissues (p < 0.001). Statistical analysis demonstrated high expression of COX-1 was correlated with tumour size (p = 0.002), pathological stage (p = 0.003), TNM stage (p = 0.003, 0.007, 0.027, respectively), and tumour recurrence (p < 0.001). Survival analysis indicated patients with high expression of COX-1 had shorter survival time (p < 0.001), and COX-1 was an independent predictor. This is the first study to reveal overexpression of COX-1 in RRC and point to use as a prognostic marker in affected patients.
Mechanical behavior of stud shear connectors embedded in HFRC

Hybrid-fiber reinforced concrete (HFRC) may provide much higher tensile and flexural strengths, tensile ductility, and flexural toughness than normal concrete (NC). HFRC slab has outstanding advantages for use as a composite bridge potential deck slab owing to higher tensile strength, ductility and crack resistance. However, there is little information on shear connector associated with HFRC slabs. To investigate the mechanical behavior of the stud shear connectors embedded in HFRC slab, 14 push-out tests (five batches) in HFRC and NC were conducted. It was found that the stud shear connector embedded in HFRC had a better ductility, higher stiffness and a slightly larger shear bearing capacity than those in NC. The experimentally obtained ultimate resistances of the stud shear connectors were also compared against the equations provided by GB50017 2003, ACI 318-112011, AISC 2011, AASHTO LRFD 2010, PCI 2004, and EN 1994-1-1 (2004), and an empirical equation to predict the ultimate shear connector resistance considering the effect of the HFRC slabs was proposed and validated by the experimental data. Curve fitting was performed to find fitting parameters for all tested specimens and idealized load-slip models were obtained for the specimens with HFRC slabs.
