Isolation, Identification, and Performance Studies of a Novel Paraffin-degrading Bacterium of Gordonia amicalis LH3

        Dong-Hui Hao,Xin Song,Jian-Qiang Lin,Yu-Jie Su,Yin-Bo Qu,Jian-Qun Lin 한국생물공학회 2008 Biotechnology and Bioprocess Engineering Vol.13 No.1

In this study, we describe the isolation and identification of a novel long-chain n-alkane degrading strain, Gordonia amicalis LH3. Under aerobic conditions, it utilized approximately 18.0% of paraffin (2% w/v) after 10 day of incubation, and the paraffin compositions of C18C24 alkalines were utilized preferentially. Under anaerobic conditions, paraffin utilization was approximately 1/8 that seen under aerobic conditions, and the compositions of C34 and C36 alkalines were utilized preferentially. The effects of salinity, temperature, and biosurfactants on paraffin degradation were also evaluated. The strain was also demonstrated to grow on oil, and decreased oil viscosity by 44.7% and degraded oil by 10.4% under aerobic conditions. Our results indicated that G. amicalis LH3 has potential applications in paraffin control, microbial enhanced oil recovery (MEOR), and the bioremediation of hydrocarbon-polluted environments.

      • Hiwi Knockdown Inhibits the Growth of Lung Cancer in Nude Mice

        Liang, Dong,Dong, Min,Hu, Lin-Jie,Fang, Ze-Hui,Xu, Xia,Shi, En-Hui,Yang, Yi-Ju Asian Pacific Journal of Cancer Prevention 2013 Asian Pacific journal of cancer prevention Vol.14 No.2

Hiwi, a human homologue of the Piwi family, plays an important role in stem cell self-renewal and is overexpressed in various human tumors. This study aimed to determine whether an RNA interference-based strategy to suppress Hiwi expression could inhibit tumor growth in a xenograft mouse model. A rare population of $SSC^{lo}\;Alde^{br}$ cells was isolated and identified as lung cancer stem cells in our previous study. Plasmids containing U6 promoter-driven shRNAs against Hiwi or control plasmids were successfully established. The xenograft tumor model was generated by subcutaneously inoculating with lung cancer stem cell $SSC^{lo}\;Alde^{br}$ cells. After the tumor size reached about 8 mm in diameter, shRNA plasmids were injected into the mice via the tail vein three times a week for two weeks, then xenograft tumor growth was assessed. In nude mice, intravenously delivery of Hiwi shRNA plasmids significantly inhibited tumor growth compared to treatment with control scrambled shRNA plasmids or the vehicle PBS. No mice died during the experiment and no adverse events were observed in mice administered the plasmids. Moreover, delivery of Hiwi shRNA plasmids resulted in a significant suppressed expression of Hiwi and ALDH-1 in xenograft tumor samples, based on immunohistochemical analysis. Thus, shRNA-mediated Hiwi gene silencing in lung cancer stem cells by an effective in vivo gene delivery strategy appeared to be an effective therapeutic approach for lung cancer, and may provide some useful clues for RNAi gene therapy in solid cancers.

Safety evaluation of four entomopathogenic nematode species against two silkworm species

        Hui Dong,Yu BI,Jia-Lin WANG,Yan-Qun Liu,Huan Wang 한국곤충학회 2020 Entomological Research Vol.50 No.3

Chinese oak silkworm(Antheraea pernyi) andmulberry silkworm(Bombyx mori) are economically important insects used for silk production and food resource. Entomopathogenic nematodes (EPNs) from the families of Steinernematidae and Heterorhabditidae are beneficial organisms currently considered in biological control. In this paper, we evaluated survival of two silkworm species exposed to four Steinernema species which are widely applied in pest control. The results showed that among four Steinernema species, S. bicornutum and S. feltiae did not have an effect on the larval survival to the two silkworm species, whereas S. carpocapsae and S. glaseri did have an effect. Each Steinernema species poses no threat to hatchability of eggs, pupation rate, larval durations and cocoon shell ratio.

The Age-Related Orientational Changes of Human Semicircular Canals

        Hui-Ying Lyu,Ke-Guang Chen,Dong-Ming Yin,Juan Hong,Lin Yang,Tian-yu Zhang,Peidong Dai 대한이비인후과학회 2016 Clinical and Experimental Otorhinolaryngology Vol.9 No.2

Objectives. Some changes are found in the labyrinth anatomy during postnatal development. Although the spatial orientation of semicircular canals was thought to be stable after birth, we investigated the age-related orientational changes of human semicircular canals during development. Methods. We retrospectively studied the computed tomography (CT) images of both ears of 76 subjects ranged from 1 to 70 years old. They were divided into 4 groups: group A (1–6 years), group B (7–12 years), group C (13–18 years), and group D (>18 years). The anatomical landmarks of the inner ear structures were determined from CT images. Their coordinates were imported into MATLAB software for calculating the semicircular canals orientation, angles between semicircular canal planes and the jugular bulb (JB) position. Differences between age groups were analyzed using multivariate statistics. Relationships between variables were analyzed using Pearson analysis. Results. The angle between the anterior semicircular canal plane and the coronal plane, and the angle between the horizontal semicircular canal plane and the coronal plane were smaller in group D than those in group A (P<0.05). The JB position, especially the anteroposterior position of right JB, correlated to the semicircular canals orientation (P<0.05). However, no statistically significant differences in the angles between ipsilateral canal planes among different age groups were found. Conclusion. The semicircular canals had tendencies to tilt anteriorly simultaneously as a whole with age. The JB position correlated to the spatial arrangement of semicircular canals, especially the right JB. Our calculation method helps detect developmental and pathological changes in vestibular anatomy.

      • Drop Interaction in a Burning Drops Stream

( Wei Dong Hsieh ),( Chun Kuei Chen ),( Ta Hui Lin ),( Shuhn Shyurng Hou ) 한국액체미립화학회 2010 한국액체미립화학회 학술강연회 논문집 Vol.2010 No.-

In a spray combustion flow field, a burning drop may interact with other drops flowing in parallel streams or along the same flow stream. Many studies on drop interaction in parallel streams were reported. However, upstream/downstream interaction of drops along a burning drops stream has not been well studied literately. To understand such interesting phenomena, we established an experimental system to examine a burning dodecane drops stream with variable drop inter-spaces in a high-temperature oxygen-rich environment. Effects of the drop inter-space (s/di) and the oxygen concentration ΩO2) in the hot environment on the interaction of burning drops were investigated in the study. Drop inter-spaces were chosen to be 2.5, 5, 10, 30 and 100. Oxygen concentrations of 21%, 25% and 30% in the hot environment were considered. The flame propagation and interactions between the burning drops in the stream were performed. Results showed that the diffusion flame enclosing the drops stream is narrowed with the increase of ΩO2 whereas effect of ΩO2 is not obvious when s/di>30. The overall flame length was longer for lower s/di due to flame propagating across the drops. Evaporation rates remains constant when s/di>30 indicating a weak interaction of drops along the same stream.

Prospective comparison of hybrid capture 2 and SPF10-LiPA for carcinogenic human papillomavirus detection and risk prediction of cervical cancer: a population-based cohort study in China

        Li Dong,Rui-Mei Feng,Li Zhang,Xiao-qian Xu,Xue-Lian Zhao,Margaret Zhuoer Wang,You-Lin Qiao,Fang-Hui Zhao 대한부인종양학회 2017 Journal of Gynecologic Oncology Vol.28 No.5

Objective: To investigate the extent of the cross-reactivity of hybrid capture 2 (HC2) assay andevaluate the potential effect of cross-reactivity on the long-term risk for cervical cancer andprecancers. Methods: Based on the Shanxi Province Cervical Cancer Screening Study-I (SPOCCS-I)cohort from 2005 to 2014 in Shanxi, China, SPF10-line probe assay (LiPA) was performedin all 598 HC2 positive and 300 random-selected HC2 negative cervical specimens. Tenyearcumulative incidence rate (CIR) of cervical intraepithelial neoplasia grade 2 or worse(CIN2+) of these two tests was evaluated using Kaplan-Meier methods. Possible humanpapillomavirus (HPV) types to be cross-reacted by HC2 were also analyzed. Results: The overall agreement between HC2 and SPF10-LiPA for detecting carcinogenic HPVwas 73.27%. The highest 10-year cumulative risk of CIN2+ was observed in both HC2 positiveand LiPA-carcinogenic HPV positive women (25.70%; 95% confidence interval [CI]=23.55%–27.91%), followed by HC2 positive but LiPA-non-carcinogenic HPV positive women (9.97%;95% CI=8.57%–11.50%), HC2 negative but LiPA-carcinogenic HPV positive (2.56%; 95%CI=2.44%–2.70%) and HC2 positive but LiPA-HPV negative (1.85%; 95% CI=1.78%–1.92%)women. The proportion of cross-reactivity of HC2 with untargeted carcinogenic types was8.9%, most of which were attributable to HPV26, 73, 82, 69, 71, 53, 11, 43, and 54. Conclusion: The noticeable high risk of CIN2+ in women infected with cross-reacted noncarcinogenicHPV and low risk in those with miss-to-detective carcinogenic HPV supportedan overall good clinical performance of HC2 for a general cervical cancer screening.

      • Experimental Simulations on Combustion Chamber Deposits of Spray Flames

( Ta Hui Lin ),( Wei Dong Hsieh ) 한국액체미립화학회 2010 한국액체미립화학회 학술강연회 논문집 Vol.2010 No.-

Combustion chamber deposits are usually observed in various internal combustion devices. They result in negative effects on the operation and performance of these devices. Study of deposits formed in the internal combustion devices is not easy and usually time-consuming. Therefore, we proposed an experimental method to simulate the deposit formation on the combustion chamber wall by considering a spray flame impinging onto a stagnation plate. Using this method, deposits can be readily generated on the stagnation plate, and it is easy to extract them for further chemical or physical analysis. In this paper, we first focused on the gasoline deposits, and later on the diesel deposits, generated on the water-cooled stagnation plate. The comparisons of gasoline deposits from the stagnation spray flames and from actual CFR engine tests exhibited good qualitative correlations. Concerning the diesel deposits, they had great difference with the gasoline deposits in the appearance. However, the effect of the cooling water (the wall temperature) and deposit growth history for the diesel deposits were basically similar to those of the gasoline deposits. This method may be used as a preliminary test on understanding deposit formation or for fuel formula developing. However, further studies are needed to justify its applicability to real engine CCD simulation.
