Refolding and Purification of Recombinant Human $Interferon-\gamma$ Expressed as Inclusion Bodies in Escherichia coli Using Size Exclusion Chromatography

Guan Yi-Xin,Pan Hai-Xue,Gao Yong-Gui,Yao Shan-Jing,Cho Man-Gi The Korean Society for Biotechnology and Bioengine 2005 Biotechnology and Bioprocess Engineering Vol.10 No.2

A size exclusion chromatography (SEC) process, in the presence of denaturant in the refolding buffer was developed to refold recombinant human $interferon-\gamma$ ($rhIFN-\gamma$) at a high concentration. The $rhlFN-\gamma$ was overexpressed in E. coli resulting in the formation of inactive inclusion bodies (IBs). The IBs were first solubilized in 8 M urea as the denaturant, and then the refolding process performed by decreasing the urea concentration on the SEC column to suppress protein aggregation. The effects of the urea concentration, protein loading mode and column height during the refolding step were investigated. The combination of the buffer-exchange effect of SEC and a moderate urea concentration in the refolding buffer resulted in an efficient route for producing correctly folded $rhIFN-\gamma$, with protein recovery of $67.1\%$ and specific activity up to $1.2\times10^7\;IU/mg$.

Development of a Serum-Free Culture Method for Endothelial Cells of the Stria Vascularis and Their Pro-inflammatory Secretome Changes Induced by Oxidative Stress

        Ying Yi,Shu-Bin Fang,Guan-Xia Xiong,Xian-Ren Wang,Hui-Ting Chen,Wan-Yi Huang,Li-Xuan Feng 대한이비인후과학회 2023 Clinical and Experimental Otorhinolaryngology Vol.16 No.1

Objectives. Reactive oxygen species in the stria vascularis (SV) of the cochlea may be involved in the pathogenesis of sen-sorineural hearing loss. However, the effects of oxidative stress on SV endothelial cells (SV-ECs) remain largely un-known, and no feasible in vitro cell culture model exists for the functional study of SV-ECs. Methods. We isolated primary SV-ECs from the SV of neonatal mice. The apoptosis-reducing effects of fibronectin in SV-ECs cultured with serum-free medium were determined using β-galactosidase staining and flow cytometry. SV-ECsincubated in serum-free medium were treated with various H2O2 concentrations to evaluate the effects of H2O2 ontheir viability. The secretome of SV-ECs treated with or without H2O2 (100 μM or 500 μM) was analyzed using high-resolution mass spectrometry. The function of the SV-EC secretome was evaluated by a macrophage assay. Results. We successfully isolated and characterized the SV-ECs. Treatment with H2O2 at concentrations up to 500 μM for2 hours and further incubation with serum-free medium in plates precoated with fibronectin showed no significanteffect on apoptosis. Compared to the control SV-ECs, the amount of differential proteins in the secretome of SV-ECsstimulated with 500 μM H2O2 was much higher than in those treated with 100 μM H2O2. Kyoto Encyclopedia ofGenes and Genomes and Gene Ontology analyses suggested that the proteins differentially expressed in SV-ECstreated with 500 μM H2O2 were involved in the regulation of multiple signaling pathways and cellular processes. Thesecretome of H2O2-stimulated SV-ECs exhibited significant pro-inflammatory effects on macrophages. Conclusion. We successfully established an in vitro serum-free culture method, identified the differential proteins releasedby oxidative stress-induced ECs and their functions, and revealed the pro-inflammatory effects of the secretome ofH2O2-stimulated SV-ECs. Therefore, SV-ECs might elicit immunoregulatory effects on bystander cells in the microen-vironment of oxidative stress-induced cochlea, especially cochlear macrophages.

Bimodal Phenomenon of the Stress–Strain Curve During Hot Compression of LA43M Mg–Li Alloy

        Yi Li,Yanjin Guan,Hu Chen,Jiqiang Zhai,Jun Lin,Liang Chen 대한금속·재료학회 2021 METALS AND MATERIALS International Vol.27 No.10

In this study, the samples of LA43M Mg–Li alloy were compressed to the true strains of 0.12, 0.16, 0.36 and 0.60 under300 °C and the strain rate of 0.1 s−1. Under this condition, the stress–strain curves present a special bimodal phenomenon atthe early deformation stage, which is caused by twinning and dynamic recrystallization. In the process of hot compression, extension twins generated first. Twinning accommodated strain and the glide of twinning dislocations dissipated local strainenergy, resulting in the first local stress drop. Then extension twins coarsened and devoured all the matrix, leading to thechange of grain orientation. Subsequently, dislocations accumulated at the grain boundaries and resulted in the increase instress. With further strain, dynamic recrystallization occurred, discontinuous dynamic recrystallization is the main dynamicrecrystallization mechanism. The generation of dynamic recrystallization resulted in the strain softening and leaded tothe second local stress drop. Twinning retards the occurrence dynamic recrystallization and has a crucial influence on themicrostructure development during the hot compression of LA43M.

Applying an Artificial Neural Network to the Control System for Electrochemical Gear-Tooth Profile Modifications

        Yi Jianjun,Guan Yifeng,Ji Baiyang,Yu Bin,Dong Jinxiang 한국정밀공학회 2007 International Journal of Precision Engineering and Vol.8 No.4

Gears, crucial components in modern precision machinery for power transmission mechanisms, are required to have low contacting noise with high torque transmission, which makes the use of gear-tooth profile modifications and gear-tooth surface crowning extremely efficient and valuable. Due to the shortcomings of current techniques, such as manual rectification, mechanical modification, and numerically controlled rectification, we propose a novel electrochemical gear-tooth profile modification method based on an artificial neural network control technique. The fundamentals of electrochemical tooth-profile modifications based on real-time control and a mathematical model of the process are discussed in detail. Due to the complex and uncertain relationships among the machining parameters of electrochemical tooth-profile modification processes. we used an artificial neural network to determine the required processing electric current as the tooth-profile modification requirements were supplied. The system was implemented and a practical example was used to demonstrate that this technology is feasible and has potential applications in the production of precision machinery.

The GntR-Type Regulators GtrA and GtrB Affect Cell Growth and Nodulation of Sinorhizobium meliloti

        Yi Wang,Ai-Min Chen,Ai-Yuan Yu,Li Luo,Guan-Qiao Yu,Jia-Bi Zhu,Yan-Zhang Wang 한국미생물학회 2008 The journal of microbiology Vol.46 No.2

GntR-type transcriptional regulators are involved in the regulation of various biological processes in bacteria, but little is known about their functions in Sinorhizobium meliloti. Here, we identified two GntR-type transcriptional regulator genes, gtrA and gtrB, from S. meliloti strain 1021. Both the gtrA1 mutant and the gtrB1 mutant had lower growth rates and maximal cell yields on rich and minimal media, as well as lower cell motility on swimming plates, than did the wild-type strain. Both mutants were also symbiotically deficient. Alfalfa plants inoculated with wild-type strain 1021 formed pink elongated nodules on primary roots. In contrast, the plants inoculated with the gtrA1 and gtrB1 mutants formed relatively smaller, round, light pink nodules mainly on lateral roots. During the first 3~4 weeks post-inoculation, the plants inoculated with the gtrA1 and gtrB1 mutants were apparently stunted, with lower levels of nitrogenase activity, but there was a remarkable increase in the number of nodules compared to those inoculated with the wild-type strain. Moreover, the gtrA1 and gtrB1 mutants not only showed delayed nodulation, but also showed markedly reduced nodulation competition. These results demonstrated that both GtrA and GtrB affect cell growth and effective symbiosis of S. meliloti. Our work provides new insight into the functions of GntR-like transcriptional regulators.

      • Life Science : Evaluation of physicochemical properties, skin permeation and accumulation profiles of salicylic acid amide prodrugs as sunscreen agent

( Yi Dong Yan ),( Jun Ho Sung ),( Dong Won Lee ),( Jung Su Kim ),( Eun Mi Jeon ),( Dae Duk Kim ),( Dong Wuk Kim ),( Jong Oh Kim ),( Ming Guan Piao ),( Dong Xun Li ),( Chul Soon Yong ),( Han Gon Choi ) 영남대학교 약품개발연구소 2012 영남대학교 약품개발연구소 연구업적집 Vol.22 No.0

Various amide prodrugs of salicylic acid were synthesised, and their physicochemical propertiesincluding lipophilicity, chemical stability and enzymatic hydrolysis were investigated. In vivo skinpermeation and accumulation profiles were also evaluated using a combination of common permeationenhancing techniques such as the use of a supersaturated solution of permeants in an enhancer vehicle, a lipophilic receptor solution, removal of the stratum corneum and delipidisation of skin. Their capacity factor values were proportional to the degree of carbon-carbon saturation in the side chain. All these amides were highly stable in acetonitrile and glycerine. Amide prodrugs were converted tosalicylic acid both in hairless mouse liver and skin homogenates. N-dodecyl salicylamide (C12SM) showed the lowest permeation of salicylic acid in skin compared to the other prodrugs, probably due to its low aqueous solubility. It had a high affinity for the stratum corneum and its accumulation was restricted to only the uppermost layer of skin. Thus, this amide prodrug could be a safer topicalsunscreen agent with minimum potential for systemic absorption.ⓒ2011 Elsevier B.V.All rights reserved.

      • The Brainwave Observation of Synchronization Spectrum Effect under Stimulation of Homemade Binaural Beats Piano Music

        ( Ssu-yi Lu ),( Xiang-guan Deng ),( Sheng-shu Huang ),( Chun-yu Tsai ),( Chyung Ay ) 한국농업기계학회 2018 한국농업기계학회 학술발표논문집 Vol.23 No.1

With the pressure of society and the large population of insomnia, the sleep problem is becoming more and more serious as the population ages. This study hopes to replace medicines with music and enable patients to relax and enter sleep more quickly. According to clinical experiments, hemi-sync can relax the body and mind and improve the difficulty of sleep. In this experiment, 3.4Hz binaural beats with three different styles of our own sleeping piano music were tested on college students. Emotiv 14-channel wireless EEG device was used to measure and record, analyze the experimental data and present as the spectrogram. For each music beats style, 30 students were selected to perform listening experiments. Calculated the success rate of hemi-sync generated by EEG device, and compared the degree of influence of the three music pieces on the listeners and the differences of frequency band in EEG. The experiment results showed that the hemi-sync appear, with a success rate of more than 40%. Different styles of binaural beats piano music and hemi-sync provide a variety of styles of self-created beats piano music for the public to choose, to shorten the time required to get into sleep and solve the problem of insomnia. In addition, we must observe the feasibility of hemi-sync to the subjects. Using the machine learning algorithm, it was found that in the F7 of the brain area, the accuracy of using the RF algorithm can reach 70.1%, while using XGB it reaches 70.4%, which means that the detection of correlation of hemi-sync and F7 in EEG can improve the verification of the influence of binaural beats on EEG. It shows a clear picture in the future that the beats music can influence the brain, so that people with insomnia can be treated with the most suitable music.

      • Exogenous Morphine Inhibits Human Gastric Cancer MGC-803 Cell Growth by Cell Cycle Arrest and Apoptosis Induction

        Qin, Yi,Chen, Jing,Li, Li,Liao, Chun-Jie,Liang, Yu-Bing,Guan, En-Jian,Xie, Yu-Bo Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.4

Morphine is not only an analgesic treating pain for patients with cancer but also a potential anticancer drug inhibiting tumor growth and proliferation. To gain better insight into the involvement of morphine in the biological characteristics of gastric cancer, we investigated effects on progression of gastric carcinoma cells and the expression of some apoptosis-related genes including caspase-9, caspase-3, survivin and NF-${\kappa}B$ using the MGC-803 human gastric cancer cell line. The viability of cells was assessed by MTT assay, proliferation by colony formation assay, cell cycle progression and apoptosis by flow cytometry and ultrastructural alteration by transmission electron microscopy. The influences of morphine on caspase-9, caspase-3, survivin and NF-${\kappa}B$ were evaluated by semi-quantitative RT-PCR and Western blot. Our data showed that morphine could significantly inhibit cell growth and proliferation and cause cell cycle arrest in the G2/M phase. MGC-803 cells which were incubated with morphine also had a higher apoptotic rate than control cells. Morphine also led to morphological changes of gastric cancer cells. The mechanism of morphine inhibiting gastric cancer progression in vitro might be associated with activation of caspase-9 and caspase-3 and inhibition of survivin and NF-${\kappa}B$.
