Myogenic enhancer transcription factor 2c (MEF2c), one of the members of the MEF2 family of transcription factors, plays an important role in mammalian muscle development. However, the role of MEF2c in avian muscle development still remains unclear. To understand the function of MEF2c in avian muscle development, we first cloned the duck MEF2c coding domain sequence (CDS) and analyzed MEF2c expression in duck muscle tissues of embryos from 10 days of incubation to 1 week after birth using real-time PCR technology. The results showed that the duck MEF2c CDS consists of 1,398nucleotides that encode 465 amino acids. The MEF2c duck protein contains a MADS domain, a MEF2 domain and a HJURP_C domain with high homology to related proteins in other organisms. Different expression levels of MEF2c were found in skeletal, smooth and cardiac muscle. Therefore, these results indicated that duck MEF2c has two conserved domains (a MADS and a MEF2 domain), is an indispensable regulator of muscle development, and plays an important role in the development of duck muscle.

Zinc nger proteins (ZFPs) play important roles in plant responses to biotic and abiotic stresses. Through microarray analysis, an Oryza sativa L. multi-stress-responsive gene, OsMSR15, was identied and subsequently cloned from rice Pei’ai 64S (Oryza sativa L.). Expression of OsMSR15 was strongly up-regulated by cold, drought and heat stresses in different tissues at different developmental stages of rice. OsMSR15 contains two C2H2-type zinc nger motifs, a nuclear localization signal (B box), a Leu-rich domain (L-box) and a conserved EAR-motif close to its C-terminus. The OsMSR15-GFP fusion protein was localized to the nucleus. Yeast-one hybrid assay showed that OsMSR15 possesses transcriptional activation ability. Expression of OsMSR15 in Arabidopsis conferred drought tolerance, and transgenic plants showed hypersensitivity to exogenous ABA during the seed germination and post-germination stages. Transgenic plants also showed higher levels of free proline, less electrolyte leakage and increased expressions of a number of stress-responsive genes, including LEA3, RD29A, DREB1A and P5CS1 under drought stress. The obtained results indicate that OsMSR15 is an important regulator involved in plant response to drought stress.
Correlation between fat accumulation and fecal microbiota in crossbred pigs

Backfat thickness (BF) is an important indicator of fat deposition capacity and lean meat rate in pigs and is very important in porcine genetics and breeding. Intestinal microbiota plays a key role in nutrient digestion and utilization with a profound impact on fat deposition of livestock animals. To investigate the relationship between the pig gut microbiome and BF, 20 low-BF (L-BF) and 20 high-BF (H-BF) pigs were selected as two groups from Yunong Black pigs in the present study. Fecal samples from pigs were analyzed for microbial diversity, composition, and predicted functionality using 16S rRNA gene sequencing. The results showed that there were significant differences in microbial β diversity between the two groups. LEfSe analysis revealed a number of bacterial features being differentially enriched in either L-BF or H-BF pigs. Spearman correlation analysis identified the abundance of Oscillospira, Peptococcus, and Bulleidia were significantly positive correlations with BF (P < 0.05), while Sutterella and Bifidobacterium were significantly negatively correlated with BF (P < 0.05). Importantly, the bacteria significantly positively correlated with BF mainly belong to Clostridium, which can ferment host-indigestible plant polysaccharides into shortchain fatty acid (SCFA) and promote fat synthesis and deposition. Predictive functional analysis indicated that the pathway abundance of cell motility and glycan biosynthesis were significantly widespread in the microbiota of the H-BF group. The results of this study will be useful for the development of microbial biomarkers for predicting and improving porcine BF, as well as for the investigation of targets for dietary strategies.

Background: Acupuncture has been widely utilized for irritable bowel syndrome (IBS). However, heterogeneity is large among therapeutic strategies and protocols. The aim of this study was to propose some down-to-earth recommendations and establish an optimized protocol for acupuncture practice in IBS. Methods: A panel of 74 traditional Chinese medicine (TCM) acupuncturists participated in clinical issue investigation. Subsequently, systematic reviews concerning acupuncture for IBS were screened within 3 databases. An initial consensus questionnaire was formed from the results of clinical issue investigation and literature review. Ultimately, a Delphi vote was carried out to determine these issues. 30 authoritative experts with extensive experience were requested to respond with agreement, neutrality, or disagreement for the items. Consensus achievement on a given item was defined as greater than 80% agreement. Results: Following a 2-round Delphi survey, there were 19 items reaching consensus; of which 5 items (26.32%) achieved thorough consensus, and significant agreement was reached for the other 14 items. These items can be classified into the 3 major domains: 1) clinical outcomes that acupuncture can bring for favorable intervention population (5 items), 2) suitable therapeutic principles and parameters of acupuncture (13 items), 3) possible adverse events in the treatment (1 item). Conclusion: Without any ready-made guidelines and lacking of homogeneity in the published literatures, such expert consensus could be valuable for TCM acupuncturists in daily practice and patients with IBS to obtain appropriate and standardized acupuncture treatment. In addition, it also points out the clinical focus which need to be further explored in future trials.

Liver cancer is one of leading digestive malignancies with high morbidity and mortality. There is an urgent need for the development of novel therapies for this deadly disease. It has been proven that asparagus polysaccharide, one of the most active derivates from the traditional medicine asparagus, possesses notable antitumor properties. However, little is known about the efficacy of asparagus polysaccharide as an adjuvant for liver cancer chemotherapy. Herein, we reported that asparagus polysaccharide and its embolic agent form, asparagus gum, significantly inhibited liver tumor growth with transcatheter arterial chemoembolization (TACE) therapy in an orthotopic hepatocellular carcinoma (HCC) tumor model, while significantly inhibiting angiogenesis and promoting tumor cell apoptosis. Moreover, asparagine gelatinous possessed immunomodulatory functions and showed little toxicity to the host. These results highlight the chemotherapeutic potential of asparagus polysaccharide and warrant a future focus on development as novel chemotherapeutic agent for liver cancer TACE therapy.

Rapid recommendation is a novel methodological framework for developing clinical practice guidelines and this framework shares the basic features of classical guidelines but differs from classical clinical practice guidelines in its ‘rapid’ development process (typically within 90 days) with an aim of translat-ing practice-changing studies to recommendations. A recent global innovation of guideline development methodology is the proposal of a rapid recommendation framework for Traditional Chinese Medicine (TCM), which has the potential to add value to the translation of evidence to practice for TCM inter- ventions. Up to now, more than 180 rapid recommendations have been published, but none of them is pertaining to TCM interventions. Due to the nature of multi-dimensional evidence sources for TCM inter- ventions, including classical randomized controlled trials and real world evidence, a more sophisticated methodological approach to synthesize and evaluate the totality of evidence about effects of TCM in- terventions is required. Therefore, appropriate modification to the rapid recommendation framework is necessary. In the efforts to respond to these needs, we have proposed a specific approach to developing rapid recommendations for TCM interventions the Multi-dimensional Evidence Synthesis, Evaluation and Recommendations for TCM interventions (MESERT)
