        Effects of zinc bearing palygorskite supplementation on the growth performance, hepatic mineral content, and antioxidant status of broilers at early age

        Objective: This study was conducted to investigate effects of zinc (Zn) bearing palygorskite (ZnPal) supplementation on growth performance, hepatic mineral content, and antioxidant status of broilers at early age. Methods: A total of 240 1-day-old Arbor Acres broiler chicks were allocated into 5 treatments with 6 replicates of 8 chicks each. Birds in 5 treatments were fed a basal diet supplemented with 0 (Control group; Analyzed Zn content: 81 mg/kg), 20, 40, 60, and 80 mg/kg Zn as ZnPal for 21 days, respectively. Blood, liver and intestinal mucosa were collected at 21 days of age. Results: Treatments did not affect growth performance of broilers during the 21-day study (p> 0.05). The contents of hepatic Zn and magnesium (Mg) were linearly increased (p<0.001) by ZnPal supplementation. ZnPal inclusion linearly (p = 0.007) reduced malondialdehyde (MDA) concentration in serum. The activity of total superoxide dismutase (T-SOD) in liver increased linearly (p = 0.001) with concentration of ZnPal in diet. ZnPal inclusion linearly (p = 0.036) and quadratically (p = 0.005) increased T-SOD activity, and linearly (p = 0.012) increased copper/zinc superoxide dismutase (Cu/Zn SOD) activity in jejunal mucosa. The maximum responses of hepatic and jejunal antioxidant enzymes activities (T-SOD and Cu/Zn SOD) were found when supplementing the basal diet with 60 mg/kg Zn as ZnPal. Furthermore, ZnPal supplementation quadratically (p = 0.001) increased Cu/Zn SOD activity in ileal mucosa, and its maximum activity was observed in the diet supplemented with 20 mg/kg Zn as ZnPal. Conclusion: ZnPal supplementation did not alter growth performance of broilers. Dietary ZnPal inclusion could increase concentrations of hepatic trace minerals (Zn and Mg) and inhibit lipid peroxidation by reducing serum MDA accumulation, with the optimal dosage of Zn from ZnPal being 80 mg/kg diet (analyzed Zn content in the diet: 165 mg/kg), and 60 mg/kg Zn as ZnPal (analyzed Zn content in the diet: 148 mg/kg) was the optimum dosage for broilers to achieve maximum antioxidant enzyme activities.

        Hepatocyte growth factor (HGF) promotes cardiac stem cell differentiation after myocardial infarction by increasing mTOR activation in p27kip haploinsufficient mice

        Myocardial infarction (MI) is a major cause of death worldwide. The treatment of MI has improved during the last decade; however, the loss of myocytes remains a problem because the cells cannot be renewed. Cardiac stem cells were recently discovered and were thought to represent a promising treatment for MI. However, the efficiency of cardiac stem cell differentiation into myocyte is not sufficient. Hepatocyte growth factor (HGF) is related to the differentiation and proliferation of cardiac stem cells. p27kip1 is a potent cell cycle inhibitor in most organs, especially the heart. In this study, we investigated the relationship among p27, HGF and cardiac stem cells in post-MI cardiac repair. We found that p27 haploinsufficient mice exhibited preserved cardiac function and improved cardiomyocyte renewal compared with wild-type mice. In addition, p27 haploinsufficiency may not increase cardiac stem cell proliferation but could improve their differentiation by increasing mammalian target of rapamycin expression.

        Determination of meaty peptide in enzymatic hydrolyzate of beef protein by HPLC-MS

        The purpose of this study is to detect beefy meaty peptide (BMP) in beef hydrolyzate. The synthesized BMP is used as a standard sample in the study. High performance liquid chromatography (HPLC)/ion trap electrospray ionization mass spectrometry (ESI-MS) with UV detection was applied in qualitative analysis of the peptides. Six beef protein enzymatic hydrolyzate samples were separated on a Surveyor HPLC system through a SUPELCO Discovery® C18 analytical column (5 μm, 15 cm×2.1 mm i.d.). The column was eluted at a flow rate of 0.2 mL/min in a linear gradient elution mode of acetronitrile-water solution with 0.1% trifluoroacetic acid. The concentration of acetronitrile was increased from 5% to 50% in 40 minutes. A Finnigan LCQ Advantage MAX instrument was used as detector to analyze with ESI-MS and ESI-MS/MS in positive mode. Among the six samples of beef protein enzymatic hydrolysate, the BMP is detected and confirmed in sample No.4 with a higher intensity of characteristic peak and is further investigated by ESI-MS/MS. As a result, BMP exists in sample No.4. The study proves that HPLC-ESI-MS/MS is a simple, rapid, sensitive method to analyze target peptides from complex polypeptides

        Nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor of three biflavonoids, isolated from roots of Stellera chamaejasme

        Aphelenchoides besseyi and Ditylenchus destructor can cause serious problems for a number of important agricultural crops and vegetables. In this study, the ethanol extract of Stellera chamaejasme L. roots showed strong nematicidal activity against Aphelenchoides besseyi and Ditylenchus destructor. By using a bioactivity-driven fractionation, three biflavonoids were isolated from the extract and their structures were identified by mass and nuclear magnetic resonance spectral data. Nematicidal activity bioassays revealed that isoneochamaejasmin A had the strongest nematicidal activity against A. besseyi and D. destructor with LC 50 values of 2.32 and 0.18 mM at 72 h, respectively. Chamaejasmenin B displayed weaker nematicidal activity against A. besseyi with an LC 50 value of 3.94 mM at 72 h. Neochamaejasmin B induced the lowest mortality against D. destructor with an LC 50 values of 15.6 mM at 72 h. These results suggested that the kind and position of substitutions and the relative configuration of 2-H/3-H and 2”-H/3”-H could be considered as important factors responsible for the nematicidal activity of these purified C-3/C-3″ biflavonoids.

        ZIF-8 Modified Nanofiber Composite Window Screen for Efficient Indoor PM2.5 and Formaldehyde Removal

        Because people spend a lot of time indoors every day, the presence of particulate matter with a diameter less than2.5 μm (PM2.5) and volatile organic molecular substances in indoor air adversely affect public health. These toxic compoundscan be removed from the air using high-efficiency air-filter materials with adsorption capacity. Metal-organic frameworks(MOFs) are among the most suitable materials for air purification as they are synthetic porous materials with excellentadsorption ability. In this study, the zeolitic imidazolate framework-8 (ZIF-8) is uniformly grown on the surface ofpolyacrylonitrile (PAN) nanofibers using an in situ growth method to prepare ZIF-8@PAN nanofiber composite membranes. The nanofiber composite membrane effectively removes PM2.5 and formaldehyde from the air. After loading the optimalamount of ZIF-8, the filtration efficiency of the nanofiber membrane for salt aerosol with a diameter of 0.3 μm increases from90.3 % to 96.9 %, and the removal rate of formaldehyde becomes 98 % within 20 min in the enclosed space of the laboratory. Moreover, after five repeated tests, the formaldehyde adsorption rate remains above 80 %. In addition, ZIF-8@PANnanofiber membranes are used to fabricate anti-haze window screens, which have great potential to improve indoor airquality and mitigate related health risks.

        An annotated database of Arabidopsis mutants of acyl lipid metabolism

        <P><B><B><I>Key message</I></B></B></P><P><B>We have constructed and annotated a web-based database of over 280 Arabidopsis genes that have characterized mutants associated with Arabidopsis acyl lipid metabolism.</B></P><P><B>Abstract</B></P><P>Mutants have played a fundamental role in gene discovery and in understanding the function of genes involved in plant acyl lipid metabolism. The first mutant in Arabidopsis lipid metabolism (<I>fad4</I>) was described in 1985. Since that time, characterization of mutants in more than 280 genes associated with acyl lipid metabolism has been reported. This review provides a brief background and history on identification of mutants in acyl lipid metabolism, an analysis of the distribution of mutants in different areas of acyl lipid metabolism and presents an annotated database (ARALIPmutantDB) of these mutants. The database provides information on the phenotypes of mutants, pathways and enzymes/proteins associated with the mutants, and allows rapid access via hyperlinks to summaries of information about each mutant and to literature that provides information on the lipid composition of the mutants. In addition, the database of mutants is integrated within the ARALIP plant acyl lipid metabolism website (http://aralip.plantbiology.msu.edu) so that information on mutants is displayed on and can be accessed from metabolic pathway maps. Mutants for at least 30 % of the genes in the database have multiple names, which have been compiled here to reduce ambiguities in searches for information.         Anti-androgen-independent Prostate Cancer Effects of Ginsenoside Metabolites In Vitro: Mechanism and Possible Structure-Activity Relationship Investigation

        Treatment of androgen-independent prostate cancer (AIPC) remains unsatisfactory. In our present experiment, natural occurring ginsenosides (NOGs) and intestinal bacterial metabolites (IBMs) were employed to investigate their anti-AIPC cell growth activity using PC-3 cells. Our results showed that the IBMs exerted more portent anti-AIPC activity than NOGs, by decreasing survival rate, inhibiting proliferation, inducing apoptosis, and leading to cell cycle arrest in AIPC PC-3 cells. The increase of LogP and decrease of C-6 steric hindrance, which were caused by deglycosylation by intestinal bacteria, may be the reason for the higher anti-AIPC activity of IBMs.

        Pin-Slot Clearance Joints in Multibody Systems

        The pin-slot joint is one of the most commonly used form of kinematic pairs, which with clearance will have a significant effect on the motion and dynamic response of multibody systems. In this study, the multibody system with a pin-slot clearance joint is modeled and the kinematic and dynamic effects are studied. Firstly, the generalized coordinate systems were established and the motion constraint equations of the pin-slot clearance joint were derived. Then, the theories of kinematics and dynamics were used to deduce equations of motion and dynamics. Finally, using slider-crank mechanism as the application example, the simulation study of the multibody system with a pin-slot clearance joint was carried out. The results show that there is an impact phenomenon in the moving process of the end slider, and with the increase of the slot angle, the impact phenomenon is earlier and the impact strength is weaker, however, with the increase of the slot value, the time when the impact phenomenon appearing doesn't change while the impact strength increasing. Furthermore, the maximum displacement of the end slider reduces with the increase of the slot value while the maximum value of driving torque doesn't change and the fluctuation of driving torque increased.
