      • 거북밀깍지벌레(Cerostegia japonicus)의 發生消長과 그 防除에 관한 硏究

        거북밀깍지벌레의 效果的인 防除法을 규명하기 위하여 釜山地域에서의 發生消長을 검토하고, 4종류의 寄主別로 기계유유제, 有機燐劑 3종 및 Carbamate劑 3종을 공시하여 殺蟲率을 Bliss의 Probit법으로 분석하고 農藥의 농도에 따른 蟲體의 呼吸代謝를 검토 하였다. 1. 釜山地方에서의 거북밀깍지벌레의 孵化器는 6月15日∼7月12日이였으며 孵化最盛期는 7月 初旬이였다. 2. 防除適期는 8月10日경이었다. 3. 8月3日에 처리한 寄主別의 본 害蟲의 LD?이 가장 낮은 것은 자목련에서는 DEP, 감나무에서는 BUX,사철나무와 치자나무에서는 BUX와 DEP로서 전체적으로 볼 때 BUX 및 DEP가 가장 效果的이고 그 다음이 Diazinon이였다. 4. 8月18日 처리에서 LD?이 가장 낮은 것은 자목련과 감나무에서는 Diazinon이, 사철나무와 치자나무에서는 BUX와 DEP로서 전체적으로 보면 Diazinon, BUX와 DEP등이 感受性이 높았다. 5. 8月3日의 처리에서 LD?이 가장 낮은 농약은 BUX로서 寄主別 농도 범위는 0.011∼0.043%였다. 6. 8月18日의 처리에서 LD?이 가장 낮은 농약은 BUX와 Diazinon으로서 寄主別 농도 범위는 각각 0.011∼0.1076%, 0.028∼0.074%였다. 7. 本 害蟲의 正常 酸素消費量은 9.25∼10.02㎕O₂/㎎ body weight/hour였으며 寄主에 따른 차이를 인정할 수 없었다. 8. Diazinon과 BUX에서는 농도가 높아질수록 산소소비량은 정상호흡량 보다 훨씬 증가하였으나 기계유유제에서는 감소되었다. In order to investigate the effective control of Florida was scale (Cerostegia japonicus), the seasonal occurrences of 1st in star in Busan area was examined, the insecticides tested with the different four hosts were carbamates(NAC,BUX BPMC), organicphosphates(DEP,MEP,Diazinon)and Machine-oil emulson, and the mortality was analyzed by the probit analysis. Also the effects of insecticide's concentration on respiratory metabolism of Florida wax scale was examined. The results obtained were as follows 1. The periods of the larva occurrence was from June 15 to July 12, and the major peak of its occurrence was found to be in the early July. 2. The adequate period for the effective control of Florida wax scale was about August 10. 3. The lowest insecticides of LD 50 values to Florida wax scale with each hosts in August 10 was DEP in the people magnolia, BUX in the persimmon tree, BUX and DEP in the spindle tree and the gardenia. So generally the most effective insecticides for controlling it were BUX and DEP, Diazinon was the next. 4. In the second experiments treated in August 18, the lowest insecticide of LD50 values to it was Diazinon in the purple magnolia and the persimmon tree, BUX and DEP in the spindle tree and the gardenia. So the susceptibility of it to the insecticides was highest in Diazinon, BUX and DEP. 5. The lowest insecticide of LD90 values to it was BUX and the concentration range by the hosts was from. 0.011% to 0.043%. 6. In the second experiment treated in August 18, the lowest insecticides of LD90 values to it were BUX and Diazinon, and the concentration ranges by the hosts were from 0.011 to 0.1076%, 0.028 to 0.074% respectively. 7. The normal oxygen consumption of Florida wax scale was from 9.25 to 10.02㎕O₂/㎎ body weight/hour and wasn't recognized the significance by the hosts. 8. The oxygen consumption of it in Diazinon and BUX was higher with increasing of insecticides concentration than the normal respiratory, but it was decreased in machine oil emulsion.

      • 감나무(부유)의 관수기간이 수체생육 및 과실품질에 미치는 영향

        These study was carried out to obtain the basic data which are necessary for producing good quality of fruits required for strengthening the competitiveness of sweet persimmon in the markets. In 1997, there were 4 irrigation periods: April to July, April to August, April to September and April to October, and in 1998 and 1999, 2 irrigation periods: April to September and from April to October, and persimmon trees were irrigated in amount of 15mm when rainfall was less than 30mm in two weeks during these irrigation periods. The rainfall in 1997 during September and October, which were the main fruit expansion phase, was shorter than average year's rainfall, but no water shortage in 1998 and 1999. During August, September and October, the monthly soil moisture in non-irrigation field was deficit in 1997, as compared to irrigation field, but no differences as affected by irrigation treatments in 1998 and 1999. In 1977 in which rainfall was comparatively shorter than average year's rainfall during fruit growing season, the fruit weight were increased to 19%, 14% in irrigation periods from April to September and from April to October, respectively, as compared to control, but there were no differences in peel color and soluble solids as affected by the irrigation treatments.

        Streptozotocin(STZ) may produce a permanent form of insulin-dependent diabetes (IDD) in experimental animals. This STZ-induced diabetes has become an useful model to study the pathogenesis of IDD in man primarily because it approximate clinical findings and also because it lends itself easily to controlled manipulation. The diabetogenic activity of STZ, however, is known to be modified by several agents such as nicotinamide, diazoxide, adrenergic blockers and pertussis vaccine (PV). The present study was undertaken to observe the protective effect of PV or boiled pertussis vaccine(bPV; incubating the PV at 80℃ for 30 minutes) against the development of IDD induced by STZ in young rats, and also the effect of PV on the activities of glycolytic & gluconeogeneic enzymes in both liver and muscle of rat. PV at a dose of 1.2×10^(10) microorganism was administrated intrapntoneally on 3days before & 7days after a single injection of STZ (GO㎎/㎏ body wt). The blood glucose and insulin levels were measured at 1st wk, 2nd wk, 4th wk, 6th wk and 8th wk after STZ injection, and the enzyme activities were exemined at 8th wk. The following results were obtained. 1. No significant difference was noted in blood glucose levels between STZ group and STZ+PV or STZ+bPV groups (P>0.05). 2. Blood insulin levels in STZ group decreased significantly from 2nd wk(P<0.05). Compared with STZ group, blood insulin levels in STZ+PV & STZ+bPV groups were higher, but not a significant value except at 2nd wk in STZ+PV grouP (P<0.05). 3. Compared with STZ group, hepatic glycolytic enzyme activities were significantly increased in STZ+PV or STZ+bPV groups, and muscular glycolytic enzyme activities were significantly increased in STZ+PV or STZ+bPV groups. 4. Compared with STZ group, hepatic gluconeogeneic enzyme activities were significantly increased in STZ+PV or STZ+bPV groups. These results suggest that the PV has somewhat protective effects on blood insulin levels and tissue glycolytic enzyme activities in rats, but it could not reduced blood glucose levels significantly, probably because of excessive increase of hepatic gluconeogeneic enzymes.

        The purpose of this study was verify effects of participation in squash for adult women's mental health. The populatidn was the women belongihg to the K city as 1998. The participants in the questionnaire were 350, but the data for actual analysis induded 300. The statistics analysis method adatede t- test, one- way ANOBA. The concusions of thiy study was as follows: For the frequendy of participation, the participants who have enjoyed 2-3times of 4-5times in a Week possesed higher levels of mental health to compare the samples who had a chance for month, once a week, or less than 3 times participation.

        <P>p53 is a widely known tumor-suppressor gene product that plays a key role in apoptotic cell death induced by DNA-damaging chemotherapeutic agents. Human glioma cells with functional p53 are known to be more resistant to 관-radiation. The aim of this study was to investigate whether the mutant glioblastoma cells (U87MG) transfected with human papilloma virus-type 16 E7 (HPV16 E7) genes were capable of increasing sensitivity towards irradiation and hypoxia-induced cell death. The pLXSN retroviral vector expressed HPV-16E7 genes and was infected into the p53 mutated U87MG cell line. A specific amplification band of HPV16 E7 genes was detected in E7 genes and transfected in the U87MG cell line using a reverse transcriptase polymerase chain reaction. The experimental groups included the mutant glioblastoma cell line (U87MG), empty vector (pLXSN) transfected to mutant glioblastoma cell line (U87MG-LXSN), and retrovirus carrying HPV16 E7 genes transfected to the mutant glioblastoma cell line (U87MG-E7). Hypoxic conditions were optimized using LDH assay and the subjects were exposed to hypoxia (16 and 20 h) and irradiation (9 h). Hoechst-propidium iodide (PI) staining results showed that hypoxia and irradiation increased the number of dead cells in the U87MG-E7 cells compared to U87MG and U87MG-LXSN cells. Results of the FACS analysis showed a similar pattern and recorded 80.44 and 58.94% of apoptotic cells in U87MG-E7 and U87MG cells, respectively. Cell cycle analysis by FACS revealed that, following irradiation and hypoxia, cells showed G2-M arrest. Additionally, the Western blot analysis results showed altered expression of E2F-1, Rb and p53 in the irradiation- and hypoxia-induced U87MG-E7 cells compared to U87MG and U87MG LXSN cells. In conclusion, the over-expression of HPV16 E7 genes in U87MG cell lines increasd cell apoptosis and E2F1 expression compared to the HPV non-infected U87MG cells following irradiation and hypoxia. These results indicate that tumor-specific therapies that increase sensitivity towards radiation and hypoxic conditions may be beneficial for suppression of cancers.</P>

        p21-Activated Kinase 4 (PAK4) as a Predictive Marker of Gemcitabine Sensitivity in Pancreatic Cancer Cell Lines

        <P><B>Purpose</B></P><P>p21-activated kinases (PAKs) are involved in cytoskeletal reorganization, gene transcription, cell proliferation and survival, and oncogenic transformation. Therefore, we hypothesized that PAK expression levels could predict the sensitivity of pancreatic cancer cells to gemcitabine treatment, and PAKs could be therapeutic targets.</P><P><B>Materials and Methods</B></P><P>Cell viability inhibition by gemcitabine was evaluated in human pancreatic cancer cell lines (Capan-1, Capan-2, MIA PaCa-2, PANC-1, Aspc-1, SNU-213, and SNU-410). Protein expression and mRNA of molecules was detected by immunoblot analysis and reverse transcription polymerase chain reaction. To define the function of PAK4, PAK4 was controlled using PAK4 siRNA.</P><P><B>Results</B></P><P>Capan-2, PANC-1, and SNU-410 cells were resistant to gemcitabine treatment. Immunoblot analysis of signaling molecules reported to indicate gemcitabine sensitivity showed higher expression of PAK4 and lower expression of human equilibrative nucleoside transporter 1 (hENT1), a well-known predictive marker for gemcitabine activity, in the resistant cell lines. Knockdown of PAK4 using siRNA induced the upregulation of hENT1. In resistant cell lines (Capan-2, PANC-1, and SNU-410), knockdown of PAK4 by siRNA resulted in restoration of sensitivity to gemcitabine.</P><P><B>Conclusion</B></P><P>PAK4 could be a predictive marker of gemcitabine sensitivity and a potential therapeutic target to increase gemcitabine sensitivity in pancreatic cancer.</P>

        Thyroid storm (thyrotoxicosis) is rarely encountered during pregnancy or the puerperium, even in untreated women and characterized by hypertension, hyperthermia, and multiple systems involvement. Much more common is heart failure, apparently caused by the long-term myocardial effects of thyroid hormone and intensified by other pregnancy complications that include severe preeclampsia, infection, anemia, or combinations of these. Prompt diagnosis and treatment of thyroid storm are essential in reducing morbidity and mortality of a patient from this disorder. We experienced a case of triiodothyroxine induced thyroid storm (T3 thyrotoxicosis) developed after Cesarean section. This case highlights both physicians should be aware of the symptoms and risk factors and reversing of heart failure successfully by reducing cardiac workload is important through immediate medical treatment-adequate hydration, thermoregulation, and control of hypertension, infection and anemia et al.

        Evaluation of Pear Pollen Germination through Colorimetric and Cytometric Analyses

        This study shows that the germination rate differs based on the method by which pollen is applied to the medium. After 3 h of incubation, the germination rate in the liquid medium (52.65%) was 2.46 times higher than that in the solidified medium (21.37%). This clearly demonstrates that there is a limitation to determining the germination rate in vitro using only the pollen identified from microscopic images, as opposed to all the pollen used in the assay. Colorimetric and cytometric analysis was performed to reduce drawbacks with respect to reliability, subjectivity of counts, and analysis time in the conventional method and improve the accuracy of measurement. The Congo Red (CR) absorbance of newly elongated pollen tubes was gradually increased at pollen concentrations of 1.25 to 5.0 mg·mL-1, and its trend line was expressed by the equation y = 0.127x + 0.0255 with R2 = 0.9938. Although no formula was established for calculating the actual germination rate, the slope of the equation obtained can be interpreted as reflecting the actual germination rate. Differences in pollen distribution between the positive control (PC) and negative control (NC) groups in the Q1-LR quadrant were observed, and the high-density region in the PC group on the dot plot formed a shifted transverse distribution to the right compared to that in the NC group. Although our observations need to be confirmed using larger numbers of pollen samples, we found that the percentage changes in events in the PC groups were expressed as a linear equation (y = 21.302x + 68.972) with a high R-squared value (0.9009). Taken together, these results demonstrate that monitoring the CR absorbance of newly elongated pollen tubes and the changes in pollen distribution along the FSC and SSC parameters will help verify pollen performance.
