PREPARATION OF CINOBUFAGIN-LOADED BOVINE SERUM ALBUMIN NANOPARTICLES FOR HEPATOCARCINOMA THERAPY

Cinobufagin-loaded bovine serum albumin nanoparticles were prepared for treating hepatocellular carcinoma. In this report, cinobufagin-bovine serum albumin-nanoparticles (Cino-BSA-NP) were prepared by an aqueous desolvation process. The physicochemical properties, toxicity, and cancer-related applications of Cino-BSA-NP were investigated. Cino-BSA-NP had a uniform spherical morphology with a particle size in the range of 50–240 nm and an average size of 86.3 nm. The zeta potential of the nanoparticles was -49 mV. The overall embedding ratio was 79.5% and the drug loading was 24.1%. Cino-BSA-NP gave cinobufagin release of up to 53.5% within 3 h, followed by slower controlled release. Cino-BSA-NP inhibited growth of hepatocarcinoma cells in vitro to a similar extent as free cinobufagin, but with a much higher median lethal dose (LD50). Hepatic histomorphological changes indicated that hepatic damage was much less severe with Cino-BSA-NP than with free cinobufagin (2.19 mg/kg). The survival time of nude mice with orthotopic transplantation tumors treated with Cino-BSA-NP was prolonged significantly. The results confirm that Cino-BSA-NP renders cinobufagin completely dispersible in aqueous media, meeting the key requirements for intravenous injection, and show controlled release, thus significantly improving cinobufagin's antitumor activity while reducing its side effects.
A Real-Time DSP-Based Imbalance Analysis System for Rotating Machine with Vibration Signal

This paper describes a new digital signal processor (DSP) imbalance measurement system dedicated to real-time vibration analysis on rotating machine. To accomplish real-time analysis, the vibration signals are on-line acquired and processed to analyze the mass imbalance and phase position. This is achieved through the use of FFT and Lissajous diagram. The method followed to analyze the mass imbalance with the chosen hardware and software solutions are described in detail in this paper. Several experimental tests demonstrate the efficiency and accuracy in imbalance analysis performance of the DSP system.

Objective: To study synergistic effects of nedaplatin and cisplatin on three human carcinoma cell lines (esophageal carcinoma cell line Eca-109, ovarian carcinoma Skov-3 and cervical carcinoma Hela). Methods: Inhibition effects were evaluated by MTT assay and cell apoptosis was detected by flow cytometry. In addition, changes of Ki-67, Bax and Bcl-2 at mRNA and protein levels were quantified by RT-PCR and Western blotting. Results: Growth inhibition in each cell lines was dose-dependent after exposure to nedaplatin or cisplatin alone. The interaction of the two drugs was synergistic at higher concentrations according to the median-effect principle. The inhibition rates with nedaplatin, cisplatin and combined treatment were $41.9{\pm}4.1%$, $47.4{\pm}2.9%$, $52.5{\pm}0.9%$(Eca-109), $39.0{\pm}1.26%$, $45.0{\pm}1.45%$, $56.2{\pm}1.44%$ (Skov-3) and $44.8{\pm}2.11%$, $46.9{\pm}0.99%$, $56.6{\pm}1.83%$ (Hela) respectively, with increase in apoptosis. Compared with the nedaplatin or cisplatin alone treatment group, the combinative treatment group's Ki-67 and bcl-2 mRNA (protein) expression was decreased while that of Bax mRNA (protein) was increased. Conclusion: Compared to the effects of nedaplatin or cisplatin alone at high concentrations, combination of nedaplatin and cisplatin at low concentrations proved to be much more effective for inhibition of proliferation and the induction of apoptosis in the Eca-109, Skov-3 and Hela cell lines.

Structure and property of a vertical cutting Ca0.4Sr0.6Bi4Ti4O15 ferroelectric ceramic

Ca0.4Sr0.6Bi4Ti4O15 ceramics with a-axis preferred orientation were prepared by a solid-state reaction method. The cylindrical Ca0.4Sr0.6Bi4Ti4O15 ceramics were cut into two types of plates: one was along the cylinder axis (A), and the other was perpendicular to the direction of the cylinder axis. The effects of sintering temperature on the structure and electrical properties of the Ca0.4Sr0.6Bi4Ti4O15 ceramics were investigated. When the sintering temperature was 1180 oC, sample A gave the highest a-axis (200) orientation with a value of about 27.1%. At this sintering temperature, sample A showed better dielectric properties and ferroelectric properties, the relative dielectric constant (εr), dielectric loss (tan δ), remnant polarization (Pr), and coercive field (Ec) were 217.6, 0.0027, 14.2 μC/cm2 and 49.6 kV/cm, respectively.
Dynamic analysis of Ca^(2+) level during bovine oocytes maturation and early embryonic development

Mammalian oocyte maturation and early embryo development processes are Ca^(2+)-dependent. In this study, we used confocal microscopy to investigate the distribution pattern of Ca^(2+) and its dynamic changes in the processes of bovine oocytes maturation, in vitro fertilization (IVF), parthenogenetic activation (PA) and somatic cell nuclear transfer (SCNT) embryo development. During the germinal vesicle (GV) and GV breakdown stage, Ca^(2+) was distributed in the cortical ooplasm and throughout the oocytes from the MI to MII stage. In IVF embryos, Ca^(2+)was distributed in the cortical ooplasm before the formation of the pronucleus. In 4-8 cell embryos and morulas, Ca^(2+) was present throughout the blastomere. In PA embryos, Ca^(2+) was distributed throughout the blastomere at 48 h, similar to in the 4-cell and 8-cell phase and the morula. At 6 h after activation, there was almost no distribution of Ca^(2+) in the SCNT embryos. However, Ca^(2+) was distributed in the donor nucleus at 10 h and it was distributed throughout the blastomere in the 2-8 cell embryos. In this study, Ca^(2+) showed significant fluctuations with regularity of IVF and SCNT groups, but PA did not. Systematic investigation of the Ca^(2+) location and distribution changes during oocyte maturation and early embryo development processes should facilitate a better understanding of the mechanisms involved in oocyte maturation, reconstructed embryo activation and development, ultimately improving the reconstructed embryo development rate.
A Practical Method of Balancing a Rigid Rotor

Diagnosis and repair tasks of an unbalanced rigid rotor reduce the chances of unexpected failure and the consequent losses in production, time, and money. This paper presents investigation of a balancing system for equilibration of rigid rotor unbalance. A practical vibration signal based method is developed for unbalance diagnosis using wavelet technology and a Lissajous diagram. This paper shows that a mass unbalance can be efficiently estimated through an appropriate narrow-band filter used to extract the required spectra component. The wavelet technology is used to design specified narrow filter bank. A modified Lissajous diagram is also introduced with statistical analysis to compute the phase position. Several experimental tests demonstrate the effectiveness in balancing the mass unbalance of a rigid rotor.
Preparation of Ca0.4Sr0.6Bi4Ti4O15 templated thin films on NaCl (100) substrates

Free-standing Ca0.4Sr0.6Bi4Ti4O15 oriented ferroelectric thin films were deposited on water soluble NaCl (100) substrates using a sol-gel method at annealing temperatures ranging from 650 ο C to 725 ο C. When annealed at 700 ο C, the Ca0.4Sr0.6Bi4Ti4O15 thin film with spherical shaped grains had the smallest strain: 0.2557%. The average size of the spherical shaped grains and surface roughness were close to 100 nm and 1.75 nm respectively.
