PREPARATION OF CINOBUFAGIN-LOADED BOVINE SERUM ALBUMIN NANOPARTICLES FOR HEPATOCARCINOMA THERAPY

Cinobufagin-loaded bovine serum albumin nanoparticles were prepared for treating hepatocellular carcinoma. In this report, cinobufagin-bovine serum albumin-nanoparticles (Cino-BSA-NP) were prepared by an aqueous desolvation process. The physicochemical properties, toxicity, and cancer-related applications of Cino-BSA-NP were investigated. Cino-BSA-NP had a uniform spherical morphology with a particle size in the range of 50–240 nm and an average size of 86.3 nm. The zeta potential of the nanoparticles was -49 mV. The overall embedding ratio was 79.5% and the drug loading was 24.1%. Cino-BSA-NP gave cinobufagin release of up to 53.5% within 3 h, followed by slower controlled release. Cino-BSA-NP inhibited growth of hepatocarcinoma cells in vitro to a similar extent as free cinobufagin, but with a much higher median lethal dose (LD50). Hepatic histomorphological changes indicated that hepatic damage was much less severe with Cino-BSA-NP than with free cinobufagin (2.19 mg/kg). The survival time of nude mice with orthotopic transplantation tumors treated with Cino-BSA-NP was prolonged significantly. The results confirm that Cino-BSA-NP renders cinobufagin completely dispersible in aqueous media, meeting the key requirements for intravenous injection, and show controlled release, thus significantly improving cinobufagin's antitumor activity while reducing its side effects.

Aims: To study effects of down-regulation of pituitary tumor-transforming gene (PTTG) on proliferation and metastasis ability of the SCL-1 cutaneous squamous cell carcinoma (CSCC) cell line and explore related mechanisms. Methods: SCL-1 cells were divided into 3 groups (untreated, siRNA control and PTTG siRNA). Cell proliferation assays were performed using a CCK-8 kit and proliferation and metastasis ability were analyzed using Boyden chambers. In addition, expression of MMP-2 and MMP-9 was detected by r-time qPCR and Western blotting. Results: Down-regulation of PTTG could markedly inhibit cell proliferation in SCL-1 cells, compared to untreated and control siRNA groups (P < 0.05). Real-time qPCR demonstrated that expression levels of PTTG, MMP-2 and MMP-9 in the PTTG siRNA group were 0.8%, 23.2% and 21.3% of untreated levels. Western blotting revealed that expression of PTTG, MMP-2 and MMP-9 proteins in the PTTG siRNA group was obviously down-regulated. The numbers of migrating cells ($51.38{\pm}4.71$) in the PTTG siRNA group was obviously lower than that in untreated group ($131.33{\pm}6.12$) and the control siRNA group ($127.72{\pm}5.20$) (P < 0.05), suggesting that decrease of proliferation and metastasis ability mediated by PTTG knock-down may be closely correlated with down-regulation of MMP-2 and MMP-9 expression. Conclusion: Inhibition of PTTG expression may be a new target for therapy of CSCC.

Language is an important means of human perception and the accumulation of knowledge. Language and cognition are closely related. It is not established by the people through long social practice. Because different people have different growth environment, social background and cultural background, these different factors will affect people``s cognitive activities and cognitive ability. According to the theory of cognitive linguistics, human’s own physical experience is the basis of human cognition, the starting point of cognition, and also a tool of cognition. The experiences gained from bodies are spread to the cognitive world by way of metaphors and other cognitive schemas. That’s how concept system formed. Human cognitive activities are rooted in the physical experiences of human beings. In other words, people``s basic experience plays avery important role in the process of human cognition. The “嘴/口” related words are very important words in the human body vocabulary. People need to survive by the usage of “嘴/口” to complete the most basic “eating” and “speaking“ behavior, so the “嘴/口” related words in Chineseand Korean for semantic analysis have high research value and significance. Based on the results of previous studies, the dictionary meaning of words in Chinese and Korean “嘴/口” as well as the related theories of cognitive linguistics and contrastive linguistics, this article aims at analyzing the similarities and differences of the structure and function of the “嘴/口” related words in Chinese and Korean in the semantic expansion . Hoping this article can provide a reference for the Chinese and foreign language vocabulary teaching.

Meiotic maturation in many species is initiated by the activation of maturation-promoting factor (MPF) with concomitant inactivation of counteracting phosphatases, most notably protein phosphatase 2A (PP2A). Recently, Greatwall (Gwl) has been identified as a cell cycle regulator that inhibits PP2A activity. In this study, we demonstrate that Gwl is required for meiotic maturation in porcine oocytes. Gwl is expressed from germinal vesicle (GV) to metaphase II (MII) stages of porcine oocytes and dramatically decreases with the progression of the meiotic cell cycle. Gwl is initially localized in the nucleus of GV oocytes and is associated with spindle fibers following GV breakdown (GVBD). Depletion of Gwl inhibits or delays meiotic maturation secondary to defects in chromosome congression and spindle formation. Conversely, overexpression of Gwl overcame meiotic arrest and initiated progression to the MII stage. However, these oocytes had severe spindle defects. Furthermore, MII oocytes depleted of Gwl progressed to pronuclear formation. Taken together, our data demonstrate that Gwl is required not only for meiotic maturation but also for maintenance of MII arrest in porcine oocytes.

按照认知语言学的理论,人类自身的身体经验是人类认知世界的基础,是认知的出发点,也是认知的工具。脚,是指人和某些动物身体最下部接触地面的部分,是人体重要的负重器官和运动器官。汉韩“足”类词在中韩两国语言中均属核心词汇。 本文主要研究由汉韩“足”类词语素构成的单纯词和合成词,将其语义取象特征分为“结构义”和“功能义”两种类型。“结构义”指“形貌、位置、构成”的综合,“功能义”指人体部位自身的使用功能,人体词的语义功能、词性功能等语法功能。通过对比分析得出它们之间的共性与差异,以期为对外汉语教学、对外韩国语教学、汉韩人体词汇学、汉韩双语翻译学领域提供有效、可信的语义例证及理论性研究依据。 According to the theory of cognitive linguistics, the human body experience is the basis of human cognition of the world, the starting point of cognition and the tool of cognition. Feet refers to the lower part of the human body and some animals that touch the ground, and are important weight-bearing organs and movement organs of the human body. Chinese and Korean foot words are the core vocabulary in both Chinese and Korean languages. This article mainly studies simple words and compound words composed of Chinese and Korean “foot” morphemes, and divides their semantic image features into two types: “structural meaning” and “functional meaning”. “Structural meaning” refers to the synthesis of “shape, location, and composition”, and “functional meaning” refers to the grammatical functions of the body parts themselves, the semantic functions of human words, and part of speech functions. Through comparative analysis, the similarities and differences between them can be found, in order to provide effective and credible semantic examples and theoretical research basis for teaching Chinese as a foreign language, teaching Korean as a foreign language, Chinese-Korean human lexics, and Chinese-Korean bilingual translation.

The mammalian target of rapamycin (mTOR) signaling pathway is upregulated in the pathogenesis of many cancers, including colorectal cancer (CRC). DEPTOR is an mTOR inhibitor whose expression is negatively regulated by mTOR. However, the role of DEPTOR in the development of CRC is not known. The aim of this study was to investigate the expression of DEPTOR and mTORC1 activity (P-S6) in a subset of CRC patients and determine their relation to tumor differentiation, invasion, nodal metastasis and disease-free survival. Here, Immunohistochemical expression of P-S6 (S235/236) and DEPTOR were evaluated in 1.5 mm tumor cores from 90 CRC patients and in 90 samples of adjacent normal mucosa by tissue microarray. The expression of P-S6 (S235/236) was upregulated in CRC, with the positive rate of P-S6 (S235/236) in CRC (63.3%) significantly higher than that in control tissues (36.7%, 30%) (p<0.05). P-S6 (S235/236) also correlated with high tumor histologic grade (p=0.002), and positive nodal metastasis (p=0.002). In contrast, the expression level of DEPTOR was correlated with low tumor histological grade (p=0.006), and negative nodal metastasis (p=0.001). Interestingly, P-S6 (S235/236) expression showed a significant negative association with the expression of DEPTOR in CRC (p=0.011, R= -0.279). However, upregulation of P-S6 (S235/236) (p=0.693) and downregulation of DEPTOR (p=0.331) in CRC were not significantly associated with overall survival. Thus, we conclude that expression of DEPTOR negatively correlates with mTORC1 activity and tumor progression in CRC. DEPTOR is a potential marker for prognostic evaluation and a target for the treatment of CRC.
