Approximately, 7-10 million people in the world suffer from Parkinson``s disease (PO), Recently, increasing evi-dence has suggested the protective effect of estrogens against nigrostriatal dopaminergic damage in PD. In this study, we investigated whether estrogen affects l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine (MPTP)-induced behavioral impairment in estrogen receptor alpha (ERa)-deficient mice. MPTP (15 mg/kg, four times with l.5-h interval)-induced dopaminergic neurodegeneration was evaluated in ERα wild-type (WT) and knockout (KO) mice. Larger dopamine depletion, behavioral impairments (Rotarod test, Pole test, and Gait test), activation of mi-croglia and astrocytes, and neurointlammation after MPTP injection were observed in ERα KO mice compared to those in WT mice.Immunostaining for tyrosine hydroxylase (TH) after MPTP injection showed fewerTH-positive neurons in ERα KO mice than WT mice. Levels of dopamine and 3,4-<lihydroxyphenylacetic acid (OOPAC, metab-olite of dopamine) were also lowered in ERα KO mice after MPTP injection. Interestingly, a higher immunoreac-tivity for monoamine oxidase (MAO) B was found in the substantia nigra and striatum of ERα KO mice after MPTP injection. We also found an increased activation of p38 kinase (which positively regulates MAO B expression) in ERα KO mice. In vitro estrogen treatment inhibited neuroinflammation in l-methyl-4-phenyl pyridium (MPP +)-treated cultured astrocyte cells; however, these inhibitory effects were removed by p38 inhibitor. These results indicate that ERα might be important for dopaminergic neuronal survival through inhibition of p38 pathway.

과학영재교육원 학생들이 성공적으로 과학 탐구를 수행하기 위해서는 문제 인식, 가설 설정, 탐구 설계, 결론 도출 등의 과학 통합 탐구 능력이 요구된다. 이 연구에서는 김유향(2013)이 개발한 ‘과학 탐구 사고력 측정을 위한 서술형 평가도구’를 사용하여 과학영재교육원 학생들의 과학 통합 탐구 능력을 평가 요소별, 성별, 이수 과정별, 전공 분야별로 비교하였다. 이 평가도구는 탐구 사고력의 주요 요소들인 탐구 문제 도출, 변인 추출, 가설 설정, 실험 설계, 자료 변환, 자료 해석, 결론 도출, 일반화, 평가에 관한 문항이 각 2개씩 총 18문항으로 구성되어 있다. 이 평가도구를 2013년 현재 C 대학교 부설 과학영재교육원에 다니고 있는 학생들 중 102명 학생들(초등과학반 15명, 중등과학심화Ⅰ반 58명, 중등과학심화Ⅱ반 29명)을 대상으로 시행하였다. 그 결과, 과학 통합 탐구 능력의 요소 중 가설 설정, 변인 추출의 점수는 상대적으로 높았고, 자료 변환, 자료 해석, 평가의 점수는 상대적으로 낮았다. ‘자료 변환’에 해당하는 선 그래프 그리기 문항과 ‘평가’에 해당하는 실험 설계의 개선점 찾기 문항의 정답률은 약 40 %로 상대적으로 낮게 나타 났다. 영재교육을 받은 단계별로 비교하면 과학 통합 탐구 능력의 요소들 중 일반화의 경우 유의미한 차이가 있었다. 남학생과 여학생의 점수 차이는 유의미하지 않았다. 이 연구에서 사용한 평가도구가 생물 분야의 소재를 다루고 있어 생물 분야의 영재 학생들과 다른 분야(물리, 화학, 지구과학)의 영재 학생들과 비교한 결과, 유의미한 차이가 없었다. 과학영재교육원 학생들의 과학 통합 탐구 능력 향상을 위하여 자료 변환, 자료 해석, 평가에 관한 교수·학습을 강화하고, 과학 탐구와 관련된 구체적인 교수·학습 자료를 개발할 필요가 있다. The purpose of this study was to investigate science integrated process skill of the students in science education center for the gifted. In order to do this, ``free-response test for the assessment of science process skills`` developed by Yu-Hyang Kim(2013) was administered to 102 students(15 in elementary school science class, 58 in middle school science classⅠ, and 29 in middle school science classⅡ) who attend the program of science education center for the gifted in C university. The assessment tool measured 9 skills ; formulating inquiry questions, recognizing variables, formulating hypotheses, designing experiment, transforming data, interpreting data, drawing conclusions, formulating generalizations, and evaluating the designed experiments. As a result, the students in science education center for the gifted had relatively high scores in the area of ``formulating hypotheses`` and ``recognizing variables``, but they had relatively low scores in the area of ``transforming data``, ``interpreting data``, and ``evaluating the designed experiments``. The 2 items` percentage of correct answers were below 40% ; one is about a drawing a line graph in ``transforming data``, and the other requires finding improvements of the experimental design in ``evaluation``. There was no significant difference between boys`` scores and girls`s one, and between the scores of students in the field of biology and those of students in the other fields(physics, chemistry, and earth science) in science integrated process skills. And there was significant difference according to the periods receiving the gifted education in ``formulating generalizations``. The teaching and learning has to focus on improving science integrated process skills in the program of science education center for the gifted and teaching and learning materials needs to be developed.
Fast temporal detection of intracellular hydrogen peroxide by HyPer

HyPer is the genetically encoded biosensor of intracellular hydrogen peroxide (H2O2), the most stable of the reactive oxygen species (ROS) generated by living cells. HyPer has a high sensitivity and specificity for detecting intracellular H2O2 by confocal laser microscopy. However, it was not known whether high speed ratiometric imaging of H2O2 by HyPer is possible. We thus investigated the sensitivity of HyPer in detecting changes to the intracellular H2O2 levels in HEK293 and PC12 cells using a microfluorometer imaging system. Increase in the HyPer ratio were clearly evident on stimulations of more than 100 μM H2O2 and fast changes in the HyPer ratio were observed on ratiometric fluorescent images after H2O2 treatment. These results suggest that HyPer is a potent biosensor of the fast temporal production of intracellular H2O2.

Detection of Differentially Expressed Genes in Glioblastoma by Suppression Subtractive Hybridization

Objective: A variety of genetic alterations in human glioblastoma comprises signal transduction and cell cycle arrest control of cellular processes. Subtractive hybridization is potentially a faster method for identifying differentially expressed genes associated with a particular disease state. Using the technique of subtraction, we isolated novel genes that are overexpressed in glioblastoma tissue as compared to normal brain tissue. Methods: We evaluated the differential expression of genes in each of hybridizing tester and driver cDNAs to digested 130 clones. After sequencing of 130 clones and homology search, this study performed to determine mRNA expression of the unknown gene, "clone 47", in brain tissue, glioblasoma, and several cancer cell lines by reverse transcription-polymerase chain reaction (RT-PCR). To test the time course for Go-phase arrest, serum stimulation and expression at various times for RT-PCR performed. Results: We identified 23 novel genes by BLAST of the digested 130 clones. The expressions of "clone 47" mRNA of glioblastoma and several cancer lines were significantly higher than normal brain tissues and several normal cell lines. We confirmed the mRNA expression of "clone 47" was up-regulation for $0.5{\sim}1hr$ of WI-38 cell differentiation. Conclusion: The novel gene, "Clone 47" is upregulated in glioblastoma tissue and several cancer cell lines. This gene is time dependent activation during time course of serum stimulation. This result suggests that "clone 47" playa role in brain tumorigenesis and the activation of this "clone 47" may be necessary for the development of cancer.

Biliary atresia is a rare disease, and it is defined as partial or total absence of the biliary tract. If not treated, this disease may cause death within 5 years. Treatment methods for biliary atresia include conservative methods and surgical methods, such as a Kasai operation or/ and liver transplantation. Oral symptoms associated with biliary atresia are green pigmented teeth, enamel hypoplasia, rampant caries, enlargement of the pulp chamber and root canals, and delayed eruption of teeth. Among these symptoms, green pigmentation of teeth is the most significant and unesthetic complication affecting a child`s social psychology. The majority of case reports about green pigmented teeth caused by biliary atresia showed that it affected the deciduous dentition rather than the permanent dentition. This case report describes the dental management of a patient with biliary atresia, who had undergone liver transplantation at age two years, and presented with green pigmentation in both deciduous and permanent dentition.
Optimal hemoglobin level for anemia treatment in a cohort of hemodialysis patients

Background: Anemia is a major risk factor that contributes to mortality in patients with chronic kidney disease. There is controversy over the optimal hemoglobin (Hb) target in these patients. This study investigated the association between Hb level and mortality in a cohort of hemodialysis (HD) patients in Korea. Methods: This study was a multicenter prospective observational study of maintenance HD patients that was performed for 5 years in western Seoul, Korea. Three hundred and sixty-two participants were enrolled. Laboratory values and mortality were accessed every 6 months. Repeated measures of laboratory values in each interval were averaged to obtain one semiannual mean value. The Hb values were divided into six groups: (1) Hbo9 g/dL; (2) 9 g/dLrHbo10 g/dL; (3) 10 g/dLrHbo11 g/dL; (4) 11 g/dLrHbo12 g/dL; (5) 12 g/dLrHbo13 g/dL; and (6) HbZ13 g/dL. We analyzed the odds ratio for all-cause mortality, based on the Hb group, and adjusted for demographics and various laboratory values. Statistics were performed with SAS, version 9.1 software (SAS Institute Inc., Cary, NC, USA). Results: Mortality odds ratios relative to the reference group (10-11 g/dL) in the fully adjusted model were 3.61 foro9 g/dL; 3.17 for 9-10 g/dLn; 4.65 for 11-12 g/dLn; 5.50 for 12-13 g/dLn; and 2.05 forZ13 g/dL (n indicates Po0.05). Conclusion: In this study, a Hb level of 10-11 g/dL was associated with the lowest mortality among the groups with Hb levelo13 g/dL. Larger interventional trials are warranted to determine the optimal Hb target for Korean HD patients. Copyright & 2015. The Korean Society of Nephrology. Published by Elsevier.
