        Silencing circPalm2 inhibits sepsis-induced acute lung injury by sponging miR-376b-3p and targeting MAP3K1

        The apoptosis and inflammation of pulmonary epithelial cells are important pathogenic factors of sepsis-induced acute lung injury (ALI). Upregulation of circPalm2 (circ_0001212) expression levels has been previously detected in the lung tissue of ALI rats. Herein, the biological significance and detailed mechanism of circPalm2 in ALI pathogenesis were investigated. In vivo models of sepsis-induced ALI were established by treating C57BL/6 mice with cecal ligation and puncture (CLP) surgery. Murine pulmonary epithelial cells (MLE-12 cells) were stimulated with lipopolysaccharide (LPS) to establish in vitro septic ALI models. MLE-12 cell viability and apoptosis were evaluated by CCK-8 assay and flow cytometry analysis, respectively. The pathological alterations of the lung tissue were analysed based on hematoxylin-eosin (H&E) staining. Cell apoptosis in the lung tissue samples was examined by TUNEL staining assay. LPS administration suppressed the viability and accelerated the inflammation and apoptotic behaviours of MLE-12 cells. CircPalm2 displayed high expression in LPS-stimulated MLE-12 cells and possessed circular characteristics. The silencing of circPalm2 impeded apoptosis and inflammation in LPS-stimulated MLE-12 cells. Mechanistically, circPalm2 bound with miR-376b-3p, which targeted MAP3K1. In rescue assays, MAP3K1 enhancement reversed the repressive effects of circPalm2 depletion on LPS-triggered inflammatory injury and MLE-12 cell apoptosis. Furthermore, the lung tissue collected from CLP model mice displayed low miR-376b-3p expression and high levels of circPalm2 and MAP3K1. CircPalm2 positively regulated MAP3K1 expression by downregulating miR-376b-3p in murine lung tissues. Importantly, circPalm2 knockdown attenuated CLP-induced inflammation, apoptosis, and pathological alterations in lung tissues collected from mice. Silenced circPalm2 inhibits LPS-induced pulmonary epithelial cell dysfunction and mitigates abnormalities in lung tissues collected from CLP-stimulated mice via the miR-376b-3p/MAP3K1 axis in septic ALI.

        Selection of candidate genes affecting meat quality and preliminary exploration of related molecular mechanisms in the Mashen pig

        Objective: The aim of this study was to select the candidate genes affecting meat quality and preliminarily explore the related molecular mechanisms in the Mashen pig. Methods: The present study explored genetic factors affecting meat quality in the Mashen pig using RNA sequencing (RNA-Seq). We sequenced the transcriptomes of 180-day-old Mashen and Large White pigs using longissimus dorsi to select differentially expressed genes (DEGs). Results: The results indicated that a total of 425 genes were differentially expressed between Mashen and Large White pigs. A gene ontology enrichment analysis revealed that DEGs were mainly enriched for biological processes associated with metabolism and muscle development, while a Kyoto encyclopedia of genes and genomes analysis showed that DEGs mainly participated in signaling pathways associated with amino acid metabolism, fatty acid metabolism, and skeletal muscle differentiation. A MCODE analysis of the protein-protein interaction network indicated that the four identified subsets of genes were mainly associated with translational initiation, skeletal muscle differentiation, amino acid metabolism, and oxidative phosphorylation pathways. Conclusion: Based on the analysis results, we selected glutamic-oxaloacetic transaminase 1, malate dehydrogenase 1, pyruvate dehydrogenase 1, pyruvate dehydrogenase kinase 4, and activator protein-1 as candidate genes affecting meat quality in pigs. A discussion of the related molecular mechanisms is provided to offer a theoretical basis for future studies on the improvement of meat quality in pigs.

        Rectal cancer-derived exosomes activate the nuclear factor kappa B pathway and lung fibroblasts by delivering integrin beta-1

        Numerous studies have revealed the importance of tumor-derived exosomes in rectal cancer (RC). This study aims to explore the influence of tumor-derived exosomal integrin beta-1 (ITGB1) on lung fibroblasts in RC along with underlying mechanisms. Exosome morphology was observed using a transmission electron microscope. Protein levels of CD63, CD9, ITGB1, p-p65 and p65 were detected using Western blot. To determine ITGB1's mRNA expression, quantitative real-time polymerase chain reaction was used. Moreover, levels of interleukin (IL)-8, IL-1β, and IL-6 in cell culture supernatant were measured via commercial ELISA kits. ITGB1 expression was increased in exosomes from RC cells. The ratio of p-p65/p65 as well as levels of interleukins in lung fibroblasts was raised by exosomes derived from RC cells, while was reduced after down-regulation of exosomal ITGB1. The increased ratio of p-p65/p65 as well as levels of pro-inflammatory cytokines caused by exosomes from RC cells was reversed by the addition of nuclear factor kappa B (NF-κB) inhibitor. We concluded that the knockdown of RC cells-derived exosomal ITGB1 repressed activation of lung fibroblasts and the NF-κB pathway in vitro.

        Topology Optimization Based Methods and the Realization Programs for Designing Microstructures of Patched Metamaterials with Prescribed Electromagnetic Properties

        This paper aims to establish a design tool to design the metamaterialmicrostructures with specific electromagnetic properties easily andconveniently, including the design methods of metamaterial microstructuresand the corresponding program codes. For the patch type microstructureand several typical metamaterials (such as the single-negative metamaterialswith specific negative permeability, left-handed metamaterials withspecific material constants at the prescribed frequency; single negativemetamaterials and left-handed metamaterials in prescribed frequencybands), the topology optimization models, solving schedules andcorresponding implementation program codes for microstructure designare presented in detail. Several typical metamaterial microstructures withdifferent design requirements are designed concretely. The resultsillustrated the correctness and validity of the design methods and thecorresponding program codes. The designing method proposed and theprogram codes developed in this paper provide an effective tool for thedesign of metamaterial microstructure with specific function for thedesigners.

        Efficacy of an adhesive nanopesticide on insect pests of rice in field trials

        Nanopesticides with antiwashing capacity on leaves are the most promising new approaches for sustainable pest management and have been fully evaluated in the laboratory. However, few studies have tested these nanopesticides on pests, and their efficacy under field conditions has not been investigated. In this study, an adhesive hollow mesoporous silica hybrid with well-defined spherical shape and good monodispersity was used as a nanocarrier of cyantraniliprole (CNAP) to fabricate an adhesive nanopesticide (CNAP-HMS-PDAAM). The control efficacy of CNAP-HMS-PDAAM was tested under field conditions. The results indicated that the efficacy of four doses of CNAP-HMS-PDAAM (30.0–69.0 g a.i./ha) against Cnaphalocrocis medinalis (Guénee) 3, 7, and 14 days after spraying did not significantly differ from that of Benevia (34.5 g a.i./ha). Twenty-eight days after spraying, the efficacy of all four doses of CNAP-HMS-PDAAM was significantly better than that of Benevia. Additionally, the efficacy of CNAP-HMS-PDAAM at doses of 34.5, 39.0 and 69.0 g a.i./ha against Chilo suppressalis (Walker) were significantly higher than that of Benevia (34.5 g a.i./ha). Thus, CNAP-HMS-PDAAM showed long-term control efficacies against C. medinalis (Guénee) and C. suppressalis (Walker), mainly due to its strong adhesive property on rice leaves and its sustained release properties. In addition, the nanocarriers showed good biocompatibility and had no obvious influence on the growth of rice.

        Influence of temperature on the carbonization context, oxidation properties and mechanism of a porous biomorphic carbon template

        A porous biomorphic carbon template (BCT) was prepared using a carbonized native aspen under an Ar atmosphere. Microstructural properties of the BCT were characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDS) and X-ray diffraction (XRD). Non-isothermal oxidation properties and the mechanism of the BCT formation were studied by thermogravimetric analysis (TGA). Experimental results show that microstructure of the BCT exhibits a honeycomb interconnected porous network and double-peaked distribution of pore diameters. The BCT phase belongs to amorphous carbon, with an increase of the carbonization temperature, the (002) peak of the XRD spectrum becomes stronger, the interplanar spacing decrease, and the structure of BCT slowly evolved towards that of ideal graphite. The non-isothermal oxidation process of BCT is controlled by a chemical reaction and gas diffusion together, and the two stages have different influences on the whole reaction rate with an increase of the conversion (a). The corresponding activation energy (E a) is also calculated.

        A Novel Method to Design Single-Sided Left-Handed Metamaterials with a Specific Frequency Band

        In this paper, a configuration of single-sided left-handed metamaterial (LHM) was constructed by placing one ring and metallic wire rod on the same side of a substrate. Based on this configuration, a novel approach was proposed to design LHM with a specific resonant frequency or two resonant frequencies. A LHM structure with 7.50 GHz resonant frequency was designed by optimizing the sizes of the structure, and the LHM with two resonant frequencies was subsequently constructed by placing two rings and metallic wire rod on the same side of the substrate. The left-handed properties of the proposed structures were verified by both experiments and numerical simulations.

        ESPO Based Course-tracking Control of Ships with Input Delay

        Consider the course-tracking control issue of ships with the dynamic parameter uncertainties, unknown time-varying ocean environment disturbance, and input delay. This paper creates a novel ship course-tracking control solution scheme under a uniformed framework such that the ship course tracks the desired course. Therein, an extended state predictor observer (ESPO) is constructed to estimate the course and the yaw rate of ships as well as the total disturbance caused by ship dynamic parameter uncertainties and unknown time-varying environment disturbance at the future instants, and whereby eventually contributes to compensating both the input delay and the total disturbance. Theoretical analyses indicate that our developed ship course-tracking control law with ESPO achieves the uniform stability of ship course-tracking closed-loop control system. Simulation results and simulation comparisons on a frigate demonstrate the effectiveness and superiority of our developed control law for ships with input delay.

        The role of botulinum toxin type A related axon transport in neuropathic pain induced by chronic constriction injury

        Background: The mechanism of peripheral axon transport in neuropathic pain is still unclear. Chemokine ligand 13 (CXCL13) and its receptor (C-X-C chemokine receptor type 5, CXCR5) as well as GABA transporter 1 (GAT-1) play an important role in the development of pain. The aim of this study was to explore the axonal transport of CXCL13/CXCR5 and GAT-1 with the aid of the analgesic effect of botulinum toxin type A (BTX-A) in rats. Methods: Chronic constriction injury (CCI) rat models were established. BTX-A was administered to rats through subcutaneous injection in the hind paw. The pain behaviors in CCI rats were measured by paw withdrawal threshold and paw withdrawal latencies. The levels of CXCL13/CXCR5 and GAT-1 were measured by western blots. Results: The subcutaneous injection of BTX-A relieved the mechanical allodynia and heat hyperalgesia induced by CCI surgery and reversed the overexpression of CXCL13/CXCR5 and GAT-1 in the spinal cord, dorsal root ganglia (DRG), sciatic nerve, and plantar skin in CCI rats. After 10 mmol/L colchicine blocked the axon transport of sciatic nerve, the inhibitory effect of BTX-A disappeared, and the levels of CXCL13/CXCR5 and GAT-1 in the spinal cord and DRG were reduced in CCI rats. Conclusions: BTX-A regulated the levels of CXCL13/CXCR5 and GAT-1 in the spine and DRG through axonal transport. Chemokines (such as CXCL13) may be transported from the injury site to the spine or DRG through axonal transport. Axon molecular transport may be a target to enhance pain management in neuropathic pain.

        The Prognostic Value of CD8+ and CD45RO+ T Cells Infiltration and Beclin1 Expression Levels for Early Postoperative Cholangitis of Biliary Atresia Patients after Kasai Operation

        Background: Postoperative cholangitis is a common but severe complication after Kasai portoenterostomy for biliary atresia (BA). This study aimed to identify its prognostic factors Methods: Two sets of liver paraffin-embedded tissue samples were collected from BA patients who received Kasai portoenterostomy (n = 25 and n = 31, respectively). Patients were divided into non-cholangitis and cholangitis groups. The infiltration of CD4+ , CD8+ , CD45RO+ , CD68+ cells and expression of Beclin1 were quantitatively evaluated in immunohistochemical analysis. Results: Cholangitis group had a significantly lower CD8+ T cell infiltration but a higher CD45RO+ cell infiltration, and a lower Beclin1 level than non-cholangitis group (all P < 0.01). Multivariate logistic regression analysis indicated that infiltration of CD8+ cells (odds ratio [OR], 0.112; 95% confidence interval [CI], 0.022–0.577) and CD45RO+ cells (OR, 3.88; 95% CI, 1.37–11.03), and Beclin1 level (OR, 0.088; 95% CI, 0.018–0.452) were independent influence factors for early postoperative cholangitis. Receiver operating characteristic (ROC) analysis showed that area under ROC curve (AUROC) values for CD8+ cells, CD45RO+ cells and Beclin1 were 0.857, 0.738 and 0.900, respectively. Conclusion: Our findings demonstrated the CD8+ cells, CD45RO+ cells and Beclin1 level possessed the prognostic value for early postoperative cholangitis following Kasai operation, which may be helpful to develop new prevention and treatment strategies for postoperative cholangitis.
