      • A cleaved amplified polymorphic sequence(CAPS) derived from a conserved missense mutation of the CmACS-7 for selecting monoecious melon plants (Cucumis melo L.)

        Most of the melon(Cucumis melo L.) breeding lines in Korea show andromonoecious (male-perfect flowers) sex expression, which requires laborious hand emasculation to produce the F1 seeds. There is a high demand for developing monoecious (male-female flowers) elite germplasm. The present study was carried out to develop molecular markers for selecting monoecious plants based on the CmACS-7 gene [a locus with 1-aminocyclopropane-1-carboxylic acid synthase(ACS) activity] responsible for ethylene synthesis and sex determination in melon. The full length sequences of the CmACS-7 were cloned from a monoecious inbred ‘Mo23’ and an andromonoecious inbred ‘Am24’. Sequence alignment revealed a major SNP(C170T) in exon1 and 18bp indel in intron4 of the CmACS-7, and a CAPS (SNP-C170T) and SCAR (ID4-18) were developed from the SNP and indel, respectively. A total of 453 F2 plants derived from ‘Mo23’ x ‘Am24’ were determined for their sex expression and genotyped using the SCAR marker. A Mendelian ratio of 3(monoecy): 1(andromonoecy) was observed from the F2 population, and sex type of 449 plants (except for four plants that showed incomplete monoecy) cosegregated with the SCAR marker, demonstrating that CmACS-7 is a single dominant gene conferring monoecy of ‘Mo23’. Allele variation of the CmACS-7 was evaluated by genotyping 114 melon accessions with diverse geographical origins using the CAPS and SCAR. C170T-SNP in exon1 of the CmACS-7 was highly conserved in melon germplasm and perfectly matched with the phenotype, whereas the 18bp-indel mutation in intron4 existed in various forms. The results demonstrated that CAPS marker SNP-C170T can be useful for marker-assisted selection(MAS) of monoecious melon plants

      • Molecular Tagging of the Gene Controlling Fruit Peel Color in Tomato (Solanu lycopersicum L.)

        Fresh market tomato cultivars are divided largely based on fruit color appearance (red or pink), which is attributed by the trait of peel. It had been reported that mutation of the Slmyb12 gene suppresses synthesis of yellow-colored flavonoid (naringenin chalcone) in peel and causes pinkish tomato fruit. Whereas wild-type tomato plants synthesizing naringenin chalcone produce yellow-colored peel, which resulting in the fruit appearance to be red. The present study was performed to investigate the association between the Slmyb12 and fruit color of domestic tomato inbreed lines. A SCAR marker was developed from an Indel mutation site (72bp insertion in exon3) of the Slmyb12, and tested on 22 and 18 red and pink-fruited inbred lines, respectively. Unexpectedly, the results showed that all inbred lines tested had wild-type Slmyb12. The full length sequences of the Slmyb12 were cloned from two inbred lines (FCR1 and FCP1), but the sequence alignment did not identify any nucleotide variations within this gene. Furthermore, scanning of SNPs between FCR1 and FCP1 using SolCAP Tomato SNP array) found no SNPs for Slmyb12. To delimit the genomic region of the gene conferring fruit color of domestic tomato lines, we are analyzing SNPs in the genes adjacent to the Slmyb12 using an F2 population derived from FCR1 x FCP1. So far, one SNP located at 1,750kbp downstream from the stop codon of Slmyb12 was mapped using 54 F2 plants and 83% of phenotype-marker association was revealed, demonstrating that the fruit color is controlled bySlmyb12 indeed, or other neighboring gene(s) involved in the pathway of naringenin chalcone synthesis. Further study with more SNPs will clear up this question.

        <P>Phosphatidylinositol 4,5-bisphosphate (PtdIns[4,5]P2) is a phosphoinositide involved in a variety of cellular functions, including signal transduction, organelle trafficking, and actin dynamics. Although the role of PtdIns[4,5]P2 in endocytosis is well established, the precise trafficking steps relying on normal PtdIns[4,5]P2 balance in the endosomal pathway have not yet been elucidated. Here we show that decrease in intracellular PtdIns[4,5]P2 levels achieved by the overexpression of the 5-phosphatase domain of synaptojanin 1 or by siRNA knock-down of PIP5Ks expression lead to severe defects in the internalization of transferrin as well as in the recycling of internalized transferrin back to the cell surface in COS-7 cells. These defects suggest that PtdIns[4,5]P2 participates in multiple trafficking and/or sorting events during endocytosis. Coexpression of the PtdIns[4,5]P2 synthesizing enzyme, PIP5KI gamma, was able to rescue these endocytic defects. Furthermore, decreased levels of PtdIns[4,5]P2 caused delays in rapid and slow membrane recycling pathways as well as a severe backup of endocytosed membrane. Taken together, our results demonstrate that PtdIns[4,5]P2 availability regulates multiple steps in the endocytic cycle in non-neuronal cells.</P>

      • Vitexin, orientin and other flavonoids from Spirodela polyrhiza inhibit adipogenesis in 3T3-L1 cells

        <P>To investigate the adipogenesis inhibitory effect on lipid accumulation, 3T3-L1 cells were treated with fractions and isolated flavonoids of Spirodela polyrhiza. An ethanol extract of S. polyrhiza was fractionated into three fractions. The butanol soluble fraction (SPB) exhibited potent antiadipogenesis activity and decreased C/EBP&agr; and PPAR&ggr; protein expression level in 3T3-L1 cells without significant cytotoxicity. The flavonoids were isolated from SPB and their chemical structures were identified as chrysoeriol (1), apigenin (2), luteolin (3), vitexin (4), cosmosin (5), orientin (6) and luteolin-7-O-&bgr;-d-glucoside (7) by spectroscopic analysis. Studies on the adipogenesis and intracellular triglyceride accumulation inhibitory effect showed that compounds 4 and 6 had the highest activity and decreased C/EBP&agr; and PPAR&ggr; protein expression level in 3T3-L1 cells. These results suggest that the flavonoids isolated from SPB, especially compounds 4 and 6, contribute to the inhibitory activity of S. polyrhiza in 3T3-L1 cells.         유럽시장에서의 한-독 통역 및 교육과정에 대한 시사점

        This paper explores the relations among German-Korean conference interpreters, the European market, and M.A. level training programs in Korea. After categorizing active German-Korean interpreters working in Europe in two different groups and evaluating market demand, the paper introduces data from the 2012 aiic survey such as average working days, market share, special market segments, and distribution of interpreting modes. Data interpretation will be accompanied by interviews with 6 interpreters and insights based on the author’s personal experience as German-Korean conference interpreter in Europe. Finally, the paper reviews prior researches on trends in B.A.-level German Studies in Korea and makes suggestions for interpretation and translation-related courses at the Graduate School of Interpretation and Translation(GSIT) at the Hankuk University of Foreign Studies. It is suggested that GSIT should actively recruit students who are native German speakers, increase specialized courses, and provide English as a potential C-language.

        사용자의 인터렉션 향상을 위한 EPUB eBook 변환 기법

        기존 책 문서인 PDF문서를 전자책에서 사용 가능하도록 전자책의 표준 형태로 변환하는 과정이 필요하다. PDF문서를 전자책의 표준 형태인 EPUB으로 변환할 때, 인쇄 색상 표현방법인 CMYK를 디지털 색상 RGB 형태로 변환하는 과정에서 형태의 차이로 인해 색감이 제대로 변환되지 못하는 문제점이 있다. 그리고 EPUB은 동영상, 자바스크립트 등을 이용해서 시각적인 효과를 줄 수 있으며, 사용자와의 상호작용이 가능하다. 본 연구는 PDF를 EPUB으로 변환 시 발생할 수 있는 문제점에 대해서 연구를 하였다. (1) 첫 번째는 색감을 잃지 않도록 ICC 프로파일을 이용한 변환 연구를 수행하였다. (2) 두 번째는 변환 과정에서 레이아웃 구성에 대한 연구를 수행하였다. (3) 세 번째는 전자책 독자들과 상호작용할 수 있는 시각적인 효과를 제공하기 위하여, 텍스트 중 특정 부분을 인식하여 시각적인 효과를 주는 연구를 수행하였으며 기존 책에서의 퀴즈를 추출하여 변환 시 자바스크립트를 이용한 퀴즈 플랫폼 프로토타입을 구현하였다. 또한 PDF문서를 EPUB에 기반 한 전자책 문서로 변환 후 사용자 평가를 통해 변환유효성 및 편의성을 검증하였다. To access PDF documents on an electronic book, PDF documents need to be converted into EPUB which is a standard format of the electronic book. When converting a PDF document into EPUB format, we need to convert color representations from CMYK into RGB representation. It is possible to give a visual effect and a user interaction using a video and JavaScript supported by EPUB format. The schemes for converting from PDF to EPUB are studied in this paper. (1) The first study is to carry out not to lose the color conversion effect using an ICC profile. (2) The second one is a layout configuration in the conversion process. (3) The third one is to highlight a specific content such as quiz platform to provide interactive visual effect for electronic book readers. Finally, in this paper we will show the usability of EPUB based eBook converting scheme through user study.

        기존 책 문서인 PDF 문서를 전자책에서도 억세스 할 수 있도록 전자책의 표준 형태로 변환하는 과정이 필요하다. PDF 문서를 전자책의 대표적인 표준 형태인 EPUB3.0으로 변환할 때, 인쇄 색상 표현방법인 CMYK를 디지털 색상 RGB 형태로 변환하는데 형태의 차이로 인하여 색감이 제대로 변환되지 못하는 문제점이 있다. 본 연구에서는 변환 시 색감을 잃지 않도록 ICC 프로파일을 이용한 변환 연구를 수행하였다. 또한 전자책 독자들을 위한 상호 작용적인 시각적인 효과를 제공하기 위하여, 많은 부분의 텍스트 중 특정 부분을 인식하여 효과 코드를 넣는 알고리즘을 제안하였다.

        항공용 제트 펌프는 구동 유체를 공급하는 승압펌프와 엔진의 운용 조건에 따른 요구사항을 만족하도록 설계되어야 한다.[1] ANSYS WORKBENCH[2]를 이용한 성능 해석 및 실제 장착되는 형상을 모사한 성능시험을 통해 제트 펌프가 요구사항을 만족하는지 확인 하였고, 이를 기반으로 민수(RTCA/DO-160G) 및 군수(MIL-STD-810G) 환경시험을 통해 항공용 제트 펌프의 신뢰성 평가를 하고자 하였다. Aircraft Jet Pump have to be designed satisfying with requirement by boost pump supplying motive fuel and operating condition of Engine. Through the Fluid Analysis and Fluid Test, We confirmed performance requirements. In this study, we checked the Reliability assessment of Jet pump as Environmental test applying RTCA/DO-160G method and MIL-STD-810G.

        주관적 만족도를 이용한 가구소비 규모탄력성 추정: 한국노동패널 만족도자료를 이용하여

        본 연구는 최근 선진국에서 최저생계비 등 산정에 많이 활용하는 주관적 동등화지수 (subjective equivalence index) 개념을 소개하고 2001-2012년 한국노동패널(KLIPS)의 만족도 설문 응답자료를 이용하여 우리나라의 주관적 동등화지수를 얻기 위해 필수적인 가구소비 규모탄력성 추정을 목적으로 하였다. 국내 기존문헌에서 다루어온 소비자의 지출행위로부터 도출되는 동등화지수와 달리, 주관적 동등화지수는 가계의 주관적 만족도 보고자료로부터 직접 추정되기 때문에 직관적으로 이해하기 쉬운 장점이 있다. 추정 결과 주관적 동등화지수 규모탄력성 추정치가 0.61로 확인되었으며, 이는 김진·송헌재 (2010)의 0.68에 비해 하향조정된 것이다. 또한 추가적인 결과로, 광역시 지역보다 광역도 지역에서 규모탄력성 추정치가 더 높고, 여성 가구주 가구의 추정치가 남성 가구주 가구보다 높은 것으로 나타났다. 가구주 연령별로는 30대, 40대, 50대로 가면서 추정치가 낮아진 반면, 60대의 경우 추정치가 1보다 커서 규모의 불경제가 나타났다. 규모탄력성 추정치는 주관적 동등화지수를 통해 최저생계비 시산, 동등화가계소득의 계측, 불평 등도지수의 계측, 그리고 빈곤선 측정 등에 다양하게 활용될 수 있을 것이다. This study introduces the concept of subjective equivalence index and estimates the scale elasticity by using the satisfaction survey data in the KLIPS (Korean Labor and Income Panel Study). Based on the work of Schwarze (2003), we estimate the scale elasticity for obtaining the subjective equivalence index and compare the subjective equivalence index approach with the consumption expenditure equivalence index approaches. The subjective equivalence index approach has an advantage in obtaining directly and intuitively from the satisfaction degree of the involved consumers. From the data of 2001-2012 in the KLIPS, we obtain the scale elasticity of 0.61 which is less than 0.68 in Kim and Song (2010) from the data of 1998-2007 in the KLIPS. Additionally, we obtain that the scale elasticity of the families in metropolitan cities is less than that in provinces. Additionally, the scale elasticity of the families with female household heads is greater than that with male household heads. As the age of household heads increases, does the scale elasticity decrease. However, the families with household heads over 60 year old have the scale elasticity with greater than 1, showing the diseconomy of scale in family consumption. These findings suggest that policy makers should be keen to consider the family composition and/or the family location in addition to the family size in calculating the minimum cost of living and in designing the other welfare policies. More detailed analyses and profound interpretations are needed for more concrete calculations on the minimum cost of living, as usual.
