Rutin Suppresses Neoplastic Cell Transformation by Inhibiting ERK and JNK Signaling Pathways

Rutin은 메밀에 함유되어 있는 것으로 잘 알려져 있는 flavonoid 물질로서, 최근 연구들에서 rutin의 항염증 및 암예방 활성이 보고되어져 왔다. 그러나, rutin의 암예방 활성과 관련된 분자생물학적 기전에 대한 연구는 아직까지 미비한 실정이다. 따라서, 본 연구에서는 발암 과정 중 하나인 세포의 악성 변형을 EGF로 유도하여 rutin이 이를 억제하는지 여부를 확인하는 실험을 진행하였으며, 그 분자생물학적 기전을 규명하고자 하였다. Soft agar assay 실험 결과, rutin은 EGF로 유도된 세포의 악성 변형을 $25{\mu}M$, $50{\mu}M$, $100{\mu}M$에서 농도별로 감소시켰다. 또한 EGF로 유도된 MEK/ERK 및 MKK4/JNK 신호전달체계의 인산화를 저해하였다. 그러나 이와는 대조적으로 rutin은 EGF로 유도된 MKK3/6/p38 신호전달체계 인산화는 감소시키지 못하는 것으로 확인되었다. 이상의 연구결과들은 rutin이 암화 과정 중 발생되는 세포의 악성변형 과정을 촉진시킨다고 잘 알려져 있는 MEK/ERK 및 MKK4/JNK 신호전달체계의 활성화를 억제함으로써 암예방 활성을 나타낸다는 것을 제시하고 있으며, 이는 메밀의 생리활성 성분인 rutin의 암예방 생리 활성 소재로서의 이용 가능성을 보여주는 중요한 연구 결과라 할 수 있겠다. 또한 위 연구결과는 MEK/ERK 및 MKK4/JNK 신호전달 체계를 표적으로 하는 생리활성 소재 탐색에도 활용 가능할 것으로 생각되어진다. Rutin is a well-known flavonoid found in buckwheat. Recent studies have demonstrated that the biological actions of rutin include anti-inflammatory and anti-cancer effects, but the underlying molecular mechanisms of these actions are not yet fully understood. Neoplastic cell transformation is considered a major event that contributes to carcinogenesis, and the present study aimed to determine whether rutin would exert anti-tumor effects via the results suggest that rutin exerted a potent inhibitory influence on the molecular activity of the MEK/ERK and MKK4/JNK pathways and strongly attenuated EGF-induced neoplastic cell transformation. These findings provide insight into the biological actions of rutin and the molecular basis for the development of new chemoprotective agents.

가치기반 아동금융교육 프로그램 개발 및 효과평가: 초등학교 1~3학년 아동의 개인재무관리 영역별 가치를 중심으로

In this study, a value-based children's financial education program was developed focusing on the values of each financial management area needed for children. According to 'quasi-experimental design - non-equivalent control group design', an experimental group and a control group were formed for children in the lower grades of elementary school, and a value-based child financial education program was executed, and then the program was applied to children's personal financial management. The effect on the value propensity for each area was evaluated. As a result of the quantitative evaluation, it was confirmed that the 8 values of the children in the experimental group were significantly improved compared to the children of the control group, so this program was effective in enhancing the values of each of the 8 areas of personal financial management needed for children. In addition, the children in the experimental group showed significant improvement compared to the children in the control group in the cognitive, justice, and psychodynamic levels of values. It has also been found to be effective in enhancing as a result of the qualitative evaluation, various positive changes were found in the cognitive, affective, and psycho-dynamic dimensions about the values of each financial management area for children. From a positive perspective, it was found that it plays a useful role in cultivating the values of each area of positive personal financial management.

(E,E)-p-Phenylene diacrylic acid derivatives were prepared in moderate to good yields by the palladium catalyzed vinylation of 4-bromoiodobenzene or 4-diiodobenzene with 2 equiv of acrylic acid derivatives in the presence of triethylamine. 4-Diiodobenzene was more reactive than 4-bromoiodobenzene in the above reactions and the reactions were proceeded stereospecifically. (E,E)-p-Phenylene diacrylic acid derivatives and several other 1,4-diolefinic aromatic compounds were also synthesized by utilizing the selective vinylation of 4-bromoiodobenzene. 4-bromoiodobenzene 또는 4-diiodobenzene과 2 당량의 아크릴산 유도체들간의 트리에틸아민 존재하의 팔라듐 촉매를 이용한 비닐화 반응에 의해 (E,E)-p-Phenylene diacrylic acid유도체들을 비교적 좋은 수득율로 쉽게 얻을 수 있었다. 이들 반응에서 4-diiodobenzene이 4-bromoiodobenzene보다 더 좋은 반응성을 나타냈으며 이들 반응들은 입체특이성있게 진행되었다. 또한, 사용된 촉매에 다른 4-bromoiodobenzene의 선택적인 비닐화 반응을 이용하여 (E,E)-p-Phenylene diacrylic acid유도체들과 이외에 몇가지 1,4-diolefinic aromatic compounds를 편리하게 합성할 수 있었다.

3,4-Dihydroxytoluene의 Raf-1 신호전달체계 억제를 통한 암예방 효능

이전 연구들에서 rutin과 quercetin을 포함한 여러 flavonoids의 암예방 활성이 보고되었으나, rutin의 경우 섭취 시 체내에서 HVA, HPAA, DHT라는 대사체로 변형되어 흡수된다. 그러나, 이들 대사체와 관련한 암예방 효능 및 그 분자생물학적 작용기작에 대한 연구 결과는 보고된 바가 없어, 본 연구에서 이를 규명하였다. DHT는 EGF로 유도된 세포 변형을 억제하였으며, AP-1 전사인자의 활성 또한 억제하였다. DHT는 Raf-1 효소 활성을 효과적으로 저해하므로서 MEK 및 ERK의 인산화를 억제하였으며, Raf-1과 ATP는 비경쟁적으로 직접 결합하여 Raf-1 효소 활성을 저해한다는 사실을 밝혀내었다. 이와는 대조적으로, rutin은 EGF로 유도된 세포 변형, AP-1 활성, ERK 신호전달체계, Raf-1 효소 활성을 억제하지 못하였다. 이상의 연구결과는 DHT의 암예방 활성이 발암과정과 밀접한 연관이 있는 Raf-1 효소 활성을 억제하여 세포 변형을 억제하는 것과 관련되어 있다는 것을 제시한다. In summary, the rutin metabolite DHT significantly reduced EGF-induced neoplastic transformation in JB6 P+ cells. This inhibition was mediated mainly by blocking the Raf/MEK/ERK signaling pathway and subsequent suppression of AP-1 activities. DHT attenuated Raf-1 kinase activity by directly binding to Raf-1 in vitro and ex vivo. Taken together, these results suggest that Raf-1 may be a critical molecular target to suppress DHT-induced neoplastic transformation, which is mainly attributable to the chemopreventive potential of several foods containing rutin.
