      • Murine chronic lymph node window for longitudinal intravital lymph node imaging

        <P>Chronic imaging windows in mice have been developed to allow intravital microscopy of many different organs and have proven to be of paramount importance in advancing our knowledge of normal and disease processes. A model system that allows long-term intravital imaging of lymph nodes would facilitate the study of cell behavior in lymph nodes during the generation of immune responses in a variety of disease settings and during the formation of metastatic lesions in cancer-bearing mice. We describe a chronic lymph node window (CLNW) surgical preparation that allows intravital imaging of the inguinal lymph node in mice. The CLNCLNCLNW is custom-made from titanium and incorporates a standard coverslip. It allows stable longitudinal imaging without the need for serial surgeries while preserving lymph node blood and lymph flow. We also describe how to build and use an imaging stage specifically designed for the CLNW to prevent (large) rotational changes as well as respiratory movement during imaging. The entire procedure takes approximately half an hour per mouse, and subsequently allows for longitudinal intravital imaging of the murine lymph node and surrounding structures for up to 14 d. Small-animal surgery experience is required to successfully carry out the protocol.</P>

        Using toponym co-occurrences to measure relationships between places: review, application and evaluation

        While there is consensus that network embeddedness of cities is of great importance for their development, the precise effect is difficult to assess because of a lack of consistent information on relations between cities. This paper presents, applies and evaluates a rather novel method to establish the strength of relationships between places, a method we refer to as ‘the toponym co-occurrence method’. This approach builds the urban system on the basis of co-occurrences of place names in a text corpus. We innovate by exploiting a so far unparalleled amount of data, namely the billions of web pages contained in the commoncrawl web archive, and by applying the method also to small places that tend to be ignored by other methods. The entire settlement system of the Netherlands is consequently explored. In addition, we innovatively apply machine learning techniques to classify these relations. Much attention is paid to solving biases deriving from place name disambiguation. Gravity modelling is employed to assess the resulting spatial organization of the Netherlands. It turns out that the gravity model fits very well with the pattern of relationships between places as found in digital space, which contributes to our assessment that the toponym co-occurrence method is a solid proxy for relationships in real space. Using the method, it is established that the relationships in the Randstad region, by many considered a coherent metropolitan entity, are actually somewhat less strong than expected. In contrast, historically important, but nowadays small cities in the periphery tend to have maintained their prominent position in the pattern of relationships. Suburban, relatively new places in the shadow of a larger city tend to be weakly related to other places. Several suggestions to further improve the method, in particular the classification of relationships, are discussed.

        Interactions of Cationic Drugs and Cardiac Glycosides at the Hepatic Uptake Level: Studies in the Rat in Vivo, Isolated Perfused Rat Liver, Isolated Rat Hepatocytes and Oocytes Expressing oatp2

        (Received May 9, 2002) This paper deals with a crucial mechanism for interaction of basic drugs and cardiac glycosides at the hepatic uptake level. Available literature data is provided and new material is presented to picture the differential transport inhibition of bulky (type2) cationic drugs by a number of cardiac glycosides in rat liver. It is shown that the so called organic anion transporting peptide 2 (oatp2) is the likely interaction site: differential inhibition patterns as observed in oocytes expressing oatp2, could be clearly identified also in isolated rat hepatocytes, isolated perfused rat liver and the rat in vivo. The anticipation of transport interactions at the hepatic clearance level should be based on data on the relative affinities of interacting substrates for the transport systems involved along with knowledge on the pharmacokinetics of these agents as well as the chosen dose regimen in the studied species. This review highlights the importance of multispecific tranporter systems such as OATP, accommodating a broad spectrum of organic compounds of various charge, implying potential transport interactions that can affect body distribution and organ clearance.

        Validation of a method for the determination of thiocyanate in human plasma by UV/VIS spectrophotometry and application to a Phase I clinical trial of GDC-0425

        The development and validation of a method for the determination of concentrations of thiocyanatein human plasma are described here. A modified colorimetric method of Bowler was used withthe following alteration in Monica Manual, Part III. In order to obtain the same sensitivity in lowamounts of clinical samples, quartz SUPRASIL® micro cuvettes have been used. The quantitationrange was between 25–500 μM. Accuracy and precision of the quality control samples, linearity ofthe calibration curve, dilution, spike recovery and stability under various conditions were evaluatedin the validation of the method and all demonstrated acceptable results. All validation results metgood laboratory practice acceptance and FDA requirements to be acceptable for application in clinicaltrials. The validated method has been used for a Phase I clinical study in cancer patients orallyadministered with either 60 mg or 80 mg of GDC-0425 containing a cyanide (CN-) group. The thiocyanatelevels from patients before and after drug administration showed no clinically significantdifferences

      • Nanofire and scale effects of heat

        Combustion is a chemical reaction that emits heat and light. Nanofire is a kind of flameless combustion that occurs on the micro–nano scale. Pt/Al2O3 film with a thickness of 20 nm can be prepared as a catalyst by micro–nano processing. When the methanol-air mixture gas passes through the surface of the catalyst, a chemical reaction begins and a significant temperature rise occurs in the catalyst region. Compared to macroscopic combustion, Nanofire has many special properties, such as large temperature gradients, uniform temperature distribution, and fast temperature response. The large temperature gradient is the most important property of Nanofire, which can reach 1330 K/mm. Combined with thermoelectric materials, it can realize the efficient conversion of chemical energy to electric energy. Nanoscale thickness offers the possibility of establishing thermal gradient. On the other hand, large thermal gradient has an effect on the transport properties of phonons and electrons in film materials. From these we can get the scale effects of heat. This article will provide an overview of the preparation, properties and applications of Nanofire, and then a comprehensive introduction to the thermal scale and thermal scale effects.

        Aspergillus cibarius sp. nov., from Traditional Meju in Korea

        Aspergillus cibarius sp. nov. isolated from meju, a brick of dried fermented soybeans in Korea, is described. The species was also found from black bean, bread and salami in the Netherlands. It is characterized by abundant yellow to reddish brown ascomata and small lenticular ascospores (4.5–5.5 μm)with a wide furrow, low equatorial crests and tuberculate or reticulate convex surface. The species was resolved as phylogenetically distinct from the other reported Aspergillus species with an Eurotium teleomorph based on multilocus sequence typing using partial fragments of the β-tubulin,calmodulin, ITS and RNA polymerase II genes.

        THE BENEFITS OF CATTLE IN MIXED FARM SYSTEMS IN PABNA, BANGLADESH

        The internal and external outputs of the agricultural sub-systems of mixed livestock-crop farms in Pabna Bangladesh were analysed, to clarify the multiple functions of cattle in farm systems. Production systems with grazing areas were relatively more productive than those with no grazing. There was a tremendous variation in farm income, largely because of land area. An increase of one ha in land area was estimated to boost farm income by about 50%. Number of cattle, cattle off-take percentage and milk off-take per average cow related positively to farm income. As land area decreased the relative importance of cattle production increased. Crops were dominant in producing food for home consumption. Cattle contributed only 5-6 per cent to home consumption. Cattle supplied a significant cash income: 45 per cent in the villages with grazing areas and 57 per cent in the other villages. In future, the cash output from cattle will decline and emphasis will shift to the role of cattle in supporting crop production. Any research or development strategy for livestock needs to focus first on the importance of the complex relation between livestock and crops.

        미세입자분산 고분자 현탁액의 3차원 직접수치해석

        We present a new finite-element scheme for direct numerical simulation of particle suspensions in simple shear flow of a viscoelastic fluid in 3D. The sliding tri-periodic representative cell concept has been combined with DEVSS/DG finite element scheme by introducing constraint equations along the domain boundary. Rigid body motion of the freely suspended particle is described by the rigid-shell description and implemented by Lagrangian multipliers on particle boundaries. We present the bulk rheology of suspensions through the numerical examples of single-, two- and many-particle problems, which represent a large number of such systems in simple shear flow. We report the steady bulk viscosity and the first normal stress coefficient, which show shear-thickening behavior for both properties.
