<P><B>Purpose</B></P><P>To compare the physical characteristics of detrusor overactivity (DO) induced by intravesical infusion of saline in awake, sham rats and rats with chronic spinal cord injury (SCI), by simultaneous registrations of intravesical and intraabdominal pressures.</P><P><B>Methods</B></P><P>Male Sprague-Dawley rats, normal or with a spinal vascular clip at the level of Th9, were investigated cystometrically 1 and 4 weeks after SCI. Intravesical pressure (IVP) and intraabdominal pressure (IAP) were recorded simultaneously to evaluate true DO. During the filling phase, the event of IVP rises, defined as increments that exceeded 2 cmH<SUB>2</SUB>O from baseline, were determined as DO according to the absence of simultaneous changes in IAP.</P><P><B>Results</B></P><P>All SCI rats exhibited DO during the filling phase, which was not shown in sham rats. The frequency and pressure of DO had a tendency to decrease with time. The DO frequency of SCI rats after 4 weeks (0.9±0.2 min<SUP>-1</SUP>) was decreased compared with that after 1 week (2.1±0.4 min<SUP>-1</SUP>; P<0.05). The DO pressure of SCI rats after 4 weeks (8.4±1.9 cmH<SUB>2</SUB>O) was decreased compared with that after 1 week (11.6±2.9 cmH<SUB>2</SUB>O; P>0.05).</P><P><B>Conclusions</B></P><P>Cystometric studies in awake male SCI rats showed some significant changes in bladder function after SCI. All SCI rats exhibited DO during the filling phase, and showed different physical characteristics of DO over the course of time. The neurological basis of these time-related changes remains poorly understood, but may provide important prognostic information about long-term urological management in SCI patients.</P>

The objective of present paper is to investigate the engineering properties of concrete using recycling water incorporating saccharic based stabilizing agent in order to improve qualities of concrete in the region of normal strength and high strength. According to test results, fluidity and air content do not exhibit significant difference with an increase in stabilizing agent contents. Bleeding was not observed at 30% of W Ie, regardless of stabilizing agent contents, nonetheless, concrete with stabilizing agent of 0.15% was observed to occur the least bleeding at 40 and 50% of W IC. For setting time, although presence of recycled water accelerates setting time, setting time shows to be retarded gradually with an increase in stabilizing agent content. For compressive strength, use of stabilizing agent enhanced compressive strength of concrete compared with that of concrete without stabilizing agent. In the side of strength enhancement, adequate dosage of stabilizing agent is found to be 0.15%. As dosage of stabilizing agent increased above 0.15%, drying shrinkage exhibited a tendency to decrease.
Targeting treatment of bladder cancer using PTK7 aptamer-gemcitabine conjugate

Gemcitabine (GEM) is one of the first-line chemotherapies for bladder cancer (BC), but the GEMs cannot recognize cancer cells and have a low long-term response rate and high recurrence rate with side effects during the treatment of BC. Targeted transport of GEMs to mediate cytotoxicity to tumor and avoid the systemic side effects remains a challenge in the treatment of BC.Based on a firstly confirmed biomarker in BC-protein tyrosine kinase 7 (PTK7), which is overexpressed on the cell membrane surface in BC cells, a novel targeting system protein tyrosine kinase 7 aptamer-Gemcitabine conjugate (PTK7-GEMs) was designed and synthesized using a specific PTK7 aptamer and GEM through auto-synthesis method to deliver GEM against BC. In addition, the antitumor effects and safety evaluation of PTK7-GEMs was assessed with a series of in vitro and in vivo assays.PTK7-GEMs can specifically bind and enter to BC cells dependent on the expression levels of PTK7 and via the macropinocytosis pathway, which induced cytotoxicity after GEM cleavage from PTK7-GEMs respond to the intracellular phosphatase. Moreover, PTK7-GEMs showed stronger anti-tumor efficacy and excellent biosafety in three types of tumor xenograft mice models.These results demonstrated that PTK7-GEMs is a successful targeted aptamer-drug conjugates strategy (APDCs) to treat BC, which will provide new directions for the precision treatment of BC in the field of biomarker-oriented tumor targeted therapy.

<P><B>Purpose</B></P><P>We investigated bladder function, with a special focus on nonvoiding contractions (NVCs), in awake rats with chronic chemical cystitis and bladder outlet obstruction (BOO) by use of simultaneous registrations of intravesical and intraabdominal pressures. In addition, we tested the effects of tolterodine on the NVCs in these models.</P><P><B>Methods</B></P><P>A total of 20 female Sprague-Dawley rats were used in this study. In eight rats, chemical cystitis was induced by intravesical instillation of HCl. Twelve rats were subjected to sham instillations or partial BOO. Four weeks after intravesical instillation or 2 weeks after partial BOO, cystometrograms were obtained by use of simultaneous recording of intravesical and intraabdominal pressure in all unanesthetized, unrestrained rats in metabolic cages.</P><P><B>Results</B></P><P>A total of 17 rats survived. In the rats with acute injury by HCl, 50% showed detrusor overactivity (DO), which was not seen in the sham group. The cystitis group had lower DO pressure without a difference in DO frequency compared with the BOO group. After the administration of tolterodine, the cystitis group showed no difference in DO frequency or pressure, whereas the BOO group showed decreased values for both parameters.</P><P><B>Conclusions</B></P><P>Our study showed that toleterodine produced no effect on DO during the filling phase in rats with chronic chemical cystitisbut decreased the frequency and pressure of DO in rats with BOO. Clinically, studies are needed to improve the treatment effect of anticholinergic drugs ininterstitial cystitis patients with overactive bladder.</P>

Purpose: The aim of this study was to apply a new surgical procedure that allows for the successful monitoring of intraurethral pressure (IUP) changes in the cystometry of awake Sprague-Dawley rats. Methods: Twenty-six female Sprague-Dawley rats were grouped according to the catheterization method (bladder only; bladder and urethra; or bladder, urethra, and abdomen). Using an arbitrarily determined initial point of the first phase among four rat micturition phases on the simultaneous curves as a reference point, we compared the time differences to the points on an intravesical pressure (IVP) and those on IUP or a detrusor pressure (DP) curve from intra-abdominal pressure (IAP). Results: In awake rat, the start of urethral flow on IUP curve corresponded to the initial point of the second phase, which is same to the results on the anesthetized rat. However, certain results, such as micturition pressure (MP) and intraluminal pressure high-frequency oscillations (IPHFOs), differed between awake and anesthetized rats. Most MP values were checked after the end of urethral flow on the IUP curve, which is due to the peculiar methodology such as transvesical catheterization. Urethral flow was not completely interrupted during the IPHFOs, which suggests the presence of urethral wall tension against the flow during voiding. After removal of the superimposed effects of IAP from IVP, the DP curve clearly showed a peculiar shape, highlighting the possibility of using IAP in place of IUP to detect the flow starting point on the IVP curve. Conclusions: Awake rat cystometry results have been interpreted based on those in anesthetized rats. However, our awake cystometry data were substantially different in terms of voiding time compared to those of anesthetized rats. This discovery warrants careful interpretation of the voiding parameters in awake rat cystometry.

<P><B>Abstract</B></P> <P>Owing to its excellent mechanical properties, processability, and biodegradability, poly(lactic acid) (PLA) has been widely investigated in the past few decades as a biomaterial. However, the poor heat resistance of PLA severely limits its applicability. In this review, several heat resistance modification methods, such as nucleating agent addition, fiber reinforcement, compounding, blending, stereoisomer complexation, and chemical modification, have been reviewed and their related mechanisms have been discussed in brief.</P>
Fracture toughness and surface morphology of polysulfone-modified epoxy resin

The thermal stability, fracture toughness, flexural strength, and surface morphology of a polysulfone (PSF)-modified epoxy resin were investigated using several techniques. The thermal stability of the PSF-modified epoxy resin was similar to that of the neat epoxy resin. The flexural strength of the epoxy resin decreased with the addition of the PSF. The fracture toughness of the PSF-modified epoxy resin significantly improved compared to that of the neat epoxy resin. The SEM micrograph of the modified epoxy resin showed a relatively rough surface with shear deformation and tortuous cracks, thereby inducing higher fracture toughness in the PSF-modified epoxy system
Identification of Prostate Cancer LncRNAs by RNA-Seq

Purpose: To identify prostate cancer lncRNAs using a pipeline proposed in this study, which is applicable for the identification of lncRNAs that are differentially expressed in prostate cancer tissues but have a negligible potential to encode proteins. Materials and Methods: We used two publicly available RNA-Seq datasets from normal prostate tissue and prostate cancer. Putative lncRNAs were predicted using the biological technology, then specific lncRNAs of prostate cancer were found by differential expression analysis and co-expression network was constructed by the weighted gene co-expression network analysis. Results: A total of 1,080 lncRNA transcripts were obtained in the RNA-Seq datasets. Three genes (PCA3, C20orf166-AS1 and RP11-267A15.1) showed a significant differential expression in the prostate cancer tissues, and were thus identified as prostate cancer specific lncRNAs. Brown and black modules had significant negative and positive correlations with prostate cancer, respectively. Conclusions: The pipeline proposed in this study is useful for the prediction of prostate cancer specific lncRNAs. Three genes (PCA3, C20orf166-AS1, and RP11-267A15.1) were identified to have a significant differential expression in prostate cancer tissues. However, there have been no published studies to demonstrate the specificity of RP11-267A15.1 in prostate cancer tissues. Thus, the results of this study can provide a new theoretic insight into the identification of prostate cancer specific genes.
