        Combined Administration of the Mixture of Honokiol and Magnolol and Ginger Oil Evokes Antidepressant-like Synergism in Rats

        Magnolia bark combined with ginger rhizome is a common drug pair in traditional Chinese prescriptions for the treatment of depression. In the present study, we examined antidepressant-like effects of the mixture of honokiol and magnolol (HMM) from magnolia bark and essential oil from ginger rhizome (OGR) alone and in combination in chronic unpredictable mild stress (CUMS) of rats. Behavioral (sucrose intake, immobility time of forced swimming test) and biochemical parameters [serotonin (5-HT) in prefrontal cortex, hippocampus, and striatum, gastric mucosa cholecystokinin (CCK) and serum gastrin (GAS) levels] were simultaneously examined in the CUMS rats. 20 mg/kg HMM alone, but not OGR, significantly increased sucrose intake and reduced immobility time in the CUMS rats. Moreover, 20 mg/kg HMM and 14 mg/kg OGR in combination exhibited significant synergistic effects on sucrose intake increase and immobility time reduction in the CUMS rats. HMM elevated 5-HT levels in various brain regions, and OGR reduced gastric mucosa CCK and serum GAS levels in the CUMS rats. These results suggested that the synergistic antidepressant-like effects of compatibility of HMM with OGR might be mediated simultaneously by regulation of the serotonergic and gastroenteric system functions. These findings also provided a pharmacological basis for the clinical application of this drug pair of magnolia bark and ginger rhizome in traditional Chinese medicine.

        Net energy content of rice bran, corn germ meal, corn gluten feed, peanut meal, and sunflower meal in growing pigs

        Objective: The objective of this experiment was to determine the net energy (NE) content of full-fat rice bran (FFRB), corn germ meal (CGM), corn gluten feed (CGF), solvent-extracted peanut meal (PNM), and dehulled sunflower meal (SFM) fed to growing pigs using indirect calorimetry or published prediction equations. Methods: Twelve growing barrows with an average initial body weight (BW) of $32.4{\pm}3.3kg$ were allotted to a replicated $3{\times}6$ Youden square design with 3 successive periods and 6 diets. During each period, pigs were individually housed in metabolism crates for 16 d, which included 7 days for adaptation. On d 8, the pigs were transferred to the respiration chambers and fed one of the 6 diets at 2.0 MJ metabolizable energy (ME)/$kg\;BW^{0.6}/d$. Total feces and urine were collected and daily heat production was measured from d 9 to d 13. On d 14 and d15, pigs were fed at their maintenance energy requirement level. On the last day pigs were fasted and fasting heat production was measured. Results: The NE of FFRB, CGM, CGF, PNM, and SFM measured by indirect calorimetry method was 12.33, 8.75, 7.51, 10.79, and 6.49 MJ/kg dry matter (DM), respectively. The NE/ME ratios ranged from 67.2% (SFM) to 78.5% (CGF). The NE values for the 5 ingredients calculated according to the prediction equations were 12.22, 8.55, 6.79, 10.51, and 6.17 MJ/kg DM, respectively. Conclusion: The NE values were the highest for FFRB and PNM and the lowest in the corn co-products and SFM. The average NE of the 5 ingredients measured by indirect calorimetry method in the current study was greater than values predicted from NE prediction equations (0.32 MJ/kg DM).

      • Expression of MiR200a, miR93, Metastasis-related Gene RECK and MMP2/MMP9 in Human Cervical Carcinoma - Relationship with Prognosis

        Aim and Background: Cervical cancer remains the third most common cancer in women globally after breast and colorectal cancer. Well-characterized biomarkers are necessary for early diagnosis and to predict metastatic progression and effective therapy. MiRNAs can regulate gene expression, cell growth, differentiation and apoptosis by targeting mRNAs for translational repression or degradation in tumor cells. The present study was conducted to assess expression of miR93, miR200a, RECK, MMP2, MMP9 in invasive cervical carcinoma, and analyze their clinical significance. Method: A total of 116 patients with invasive cervical carcinoma and 100 patients undergoing hysterectomy for benign lesions were retrospectively examined. Quantitative real-time PCR was performed to determine expression of miR93 and miR200a while RECK, MMP2, MMP9 and MVD were assessed by immunohistochemical staining. Results: Cervical carcinoma patients demonstrated up-regulation of miR-93, miR-200a, MMP2 and MMP9, with down-regulation of RECK as compared to benign lesion tissues. RECK was significantly inversely related to invasion and lymphatic metastasis. The 5-year survival rate for patients with strong RECK expression was significantly higher than that with weakly expressing tumors. Conclusion: MiR-93 and miR-200a are associated with metastasis and invasion of cervical carcinoma. Thus together with RECK they are potential prognostic markers for cervical carcinoma. RECK cooperating with MMP2, MMP9 expression is a significant prognostic factor correlated with long-term survival for patients with invasive cervical carcinoma.

        Genes Regulating the ABORTED MICROSPORES (AMS)-Mediated Male Sterility Networks in Melon (Cucumis melo L.)

        The male sterile plants have higher heterosis in the production of hybrid seeds. The ABORTED MICROSPORES (AMS) gene has been demonstrated to be a candidate gene for ms-5. However, the genetic mechanism underlying AMS-mediated male sterility (MS) regulatory networks in melon (Cucumis melo L.) is still not clearly understood. In the present study, we used transcriptome sequencing analysis, yeast hybridization technology, quantitative real-time polymerase chain reaction (qRT-PCR), and bioinformatics analyzed to systematically investigate the AMS-mediated MS regulatory networks in melon. A set of 15 proteins interacting with AMS, including the C. melo L. Zinc Ribbon protein 1 (CmZR1) gene, was identified using the yeast one-hybrid (Y1H) system and further confirmed using the yeast two-hybrid (Y2H) assay. The interaction of the CmZR1 protein with the C. melo L. Pectin Methylesterase Inhibitor 1 (CmPMEI1) protein was identified and further verified by the glutathione S-transferase (GST) pull-down technique. Bioinformatics analyzed the physical and chemical properties, gene structure, and kinship of the melon PMEI family. We proposed a partial regulatory network for melon MS in which the interaction of CmPMEI1 protein with CmZR1 protein regulates the expression of the AMS gene for pollen abortion. These findings provide important information for increasing the understanding of the molecular mechanism of the MS regulatory network in melon.

        Chinese Pediatric Reference Intervals for Serum Cortisol on IMMULITE 2000

        Clinical interpretation of the test results for cortisol based on continuous reference intervals with appropriate partitions improves pediatric diagnosis; however, these values are available only for Caucasians. To develop the pediatric reference intervals for Chinese population, we examined the serum cortisol levels in 1,143 healthy Chinese children aged 4–18 years (566 boys and 577 girls), using an IMMULITE 2000 Immunoassay System (Siemens Healthcare GmbH). Phlebotomy was performed at 7–9 a.m. for 284 boys and 287 girls and at 1–3 p.m. for the others. They were divided into four age groups according to the Clinical and Laboratory Standards Institute guideline EP28-A3c, with the last group further stratified according to sampling time. Separate reference intervals of 49.6–323.7, 70.9–395.3, and 90.1–448.7 nmol/L were established for children aged 4–8, 9–12, and 13–15 years, respectively. Further, reference intervals of 118.2–464.7 and 71.4–446.7 nmol/L were established for morning and afternoon cortisol levels, respectively, in children aged 16–18 years. Further studies are necessary to transfer and validate these reference intervals in other analytical systems and pediatric populations, and to allow for broader applications.

      • Expression of Endogenous Hypoxia Markers in Vulvar Squamous Cell Carcinoma

        Objective: To investigate the expression of endogenous hypoxia-related markers identified as being involved in vulvar squamous cell carcinoma (VSCC). Methods: We performed immunohistochemical staining of hypoxia-inducible factor-$1{\alpha}$ (HIF-$1{\alpha}$), glucose transporter-1 (GLUT-1), carbonic anhydrase 9 (CA-9) and vascular endothelial growth factor (VEGF), on tissue sections of 25 VSCC patients, 10 vulvar intraepithelial neoplasia (VIN) patients and 12 healthy controls. Results: HIF-$1{\alpha}$ expression was found in all sections, with no significant difference between controls, VIN and VSCC sections (all P<0.05). Glut-1 expression was found in 25% of control, 90% of VIN and 100% of VSCC sections. A significant difference between control and VIN or VSCC was observed (all P<0.05), while no difference was found between VIN and VSCC sections (P>0.05). CA-9 expression was negative in control sections, but it was found in 30% of VIN sections and 52% of VSCC sections with strong staining. Similarly, CA-9 expression also showed obvious differences between controls and VIN or VSCC sections (all P<0.05). However, there was no significant difference between VIN and VSCC (P>0.05). There were only 25% of control sections with weak VEGF expression, while strong staining was found in about 60% of VIN sections and 25% of VSCC sections (all P<0.05). In addition, a difference was also found between VIN and VSCC sections (P<0.05). Conclusion: Expression of endogenous hypoxia markers (HIF-$1{\alpha}$, GLUT-1, CA-9 and VEGF) might be involved in the malignant progression of VSCC.

      • Prognostic Role of Hypoxic Inducible Factor Expression in Non-small Cell Lung Cancer: A Meta-analysis

        Introduction: Reported prognostic roles of hypoxic inducible factor (HIF) expression in non-small cell lung cancer (NSCLC) have varied. This meta-analysis aimed to examine the relationship between HIF expression and clinical outcome in NSCLC patients. Methods: PubMed were used to identify relevant literature with the last report up to December $20^{th}$, 2012. After careful review, survival data were collected from eligible studies. We completed the meta-analysis using Stata statistical software (Version 11) and combined hazard ratio (HR) for overall survival (OS). Subgroup specificity, heterogeneity and publication bias were also assessed. All of the results were verified by two persons to ensure accuracy. Results: Eight studies were finally stepped into this meta-analysis in which seven had available data for HIF-$1{\alpha}$ and three for HIF-$2{\alpha}$. Combined HRs suggested that higher expression of $HIF1{\alpha}$ had a negative impact on NSCLC patient survival (HR=1.50; 95%CI=1.07-2.10; p=0.019). The expression of HIF-$2{\alpha}$ was also relative to a poorer survival (HR=2.02; 95%CI=1.47-2.77; p=0.000). No bias existed in either of the two groups. Conclusion: This study suggests that elevations of HIF-$1{\alpha}$ and HIF-$2{\alpha}$ expression are both associated with poor outcome for patients with NSCLC. The data support further and high quality investigation of HIF expression for predicting poor outcome in patients with NSCLC.

        Indirect Determination of Cetirizine Hydrochloride by ICP-AES

        Cetirizine hydrochloride reacted with $BiI_4^-$ in an acidic aqueous solution to form precipitate. After centrifugation, the atomic emission intensity of $Bi^{3+}$ contained in the supernatant solution was measured at the characteristic wavelength of 206.170 nm. The difference between the spectral signal intensity of the blank solution and that of the supernatant, ${\Delta}I$, was linearly related to the concentration of cetirizine hydrochloride. As a result, a new inductively coupled plasmaatomic emission spectrometric (ICP-AES) method was developed for the analysis of cetirizine hydrochloride. The linear range was from 27.7 to 184.8 $mg{\cdot}L^{-1}$, with a correlation coefficient (r) of 0.9961 and a detection limit of 9.6 $mg{\cdot}L^{-1}$. This method is simple and accurate, Without using toxic organic solvents, and is feasible for the quality control of cetirizine hydrochloride tablets and capsules.

        Spontaneous Sm<sup>3+</sup>→Sm<sup>2+</sup> reduction ability of MAl<sub>2</sub>Si<sub>2</sub>O<sub>8</sub> (M = Sr, Ba): Sm

        <P><B>Abstract</B></P> <P>In air atmosphere, the spontaneous reduction abilities of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> in Sm ions doped monoclinic SrAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (SASO) and BaAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M-BASO) as well as hexagonal BaAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (H-BASO) are investigated. Sm ions doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) samples were synthesized using Pechini-type sol-gel technology and sintered in air atmosphere and reducing condition, respectively. By comparing the photoluminescence properties of Sm doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) calcined in air with that in reducing condition, the spontaneous reduction of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> can be detected only in the host lattice H-BASO. The essential that makes the reduction of Sm<SUP>3+</SUP> to Sm<SUP>2+</SUP> was investigated by calculating their chemical bonds parameter quantitatively. The important key environmental factor of Ba<SUP>2+</SUP> in H-BASO is the largest among the three samples MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB>. It indicates that the hexagonal H-BASO sample is the easiest one to realize the spontaneous reduction of Sm<SUP>3+</SUP>→Sm<SUP>2+</SUP> among Sm doped MAl<SUB>2</SUB>Si<SUB>2</SUB>O<SUB>8</SUB> (M = Sr, Ba) samples. The needed energy transferring one electron of the ligand to the central ions Sm<SUP>3+</SUP> in H-BASO:Sm is the lowest. The theoretical analysis agrees well with the experimental result.
