        Genome-Wide Association Analysis to Search for New Loci Associated with Lifelong Premature Ejaculation Risk in Chinese Male Han Population

        Purpose: Genetic factors play an indispensable role in the pathogenesis of lifelong premature ejaculation (LPE). The suscep-tibility genes/SNPs that have been discovered are very limited and can only explain part of the genetic effects of LPE. There-fore, discovering more genetic polymorphisms associated with the occurrence and development of LPE will help reveal the pathogenesis of LPE. Materials Materials and and Methods:Methods: We conducted a genome-wide association study of LPE in 486 Chinese male Han people (cases and controls). We used Gene Titan multi-channel instrument and Axiom Analysis Suite 6.0 software for genotyping. Imputation was performed by IMPUTE2 software and the 1000 Genomes Project (Phase3) was used as reference for haplotype. Finally, logistic regression analysis was performed on all loci that passed the quality control. The odds ratio and 95% confidence in-terval were calculated to determine the association between each SNPs and Chinese male Han population LPE risk. Results:Results: The results showed that a total of 33 genetic variants in 13 genes (LACTBL1 , SSBP3 , ACOT11 , LINC02486 , TMEM154, LINC01098, NONE, HCG27, HLA-C, TNFSF8, TNC, FAM53B, SULF2) have a suggestively significant genome-wide association with LPE risk (p<5×10-6). Conclusions:Conclusions: This study is the first to conduct a GWAS on LPE in Chinese male Han population 33 genetic polymorphisms have a suggestive genome-wide association with LPE risk. This study have provided data supplement for the genetic loci of LPE risk, and laid a scientific foundation for the pathogenesis and the targeted therapy of LPE.

        Correlation between the Position of the Pituitary Stalk as Determined by Diffusion Tensor Imaging and Its Location as Determined at the Time of Surgical Resection of Pituitary Adenomas

        Objective : An important factor during pituitary adenoma surgery is to preserve pituitary stalk (PS) as this plays a role in reduction of the risk of postoperative diabetes insipidus. The hypothalamic-hypophyseal tract (HHT) projects through the PS to the posterior pituitary gland. To reconstruct white matter fiber pathways, methods like diffusion tensor imaging (DTI) tractography have been widely used. In this report we attempted to predict the position of PS using DTI tractography and to assess its intraoperative correlation during surgery of pituitary adenomas. Methods : DTI tractography was used to tract the HHT in nine patients before craniotomy for pituitary adenomas. The DTI location of the HHT was compared with the PS position identified at the time of surgery. DTI fiber tracking was carried out in nine patients prior to the planned craniotomy for pituitary adenomas. In one patient, the PS could not be identified during the surgery. In the other eight patients, a comparison was made between the location of the HHT identified by DTI and the position of the PS visualized at the time of surgery. Results : The position of the HHT identified by DTI showed consistency with the intraoperative position of the PS in seven patients (88.9% concordance). Conclusion : This study shows that DTI can identify the position of the HHT and thus the position of the PS with a high degree of reliability.

        Whole-genome resequencing reveals domestication and signatures of selection in Ujimqin, Sunit, and Wu Ranke Mongolian sheep breeds

        Objective: The current study aimed to perform whole-genome resequencing of Chinese indigenous Mongolian sheep breeds including Ujimqin, Sunit, and Wu Ranke sheep breeds (UJMQ, SNT, WRK) and deeply analyze genetic variation, population structure, domestication, and selection for domestication traits among these Mongolian sheep breeds.Methods: Blood samples were collected from a total of 60 individuals comprising 20 WRK, 20 UJMQ, and 20 SNT. For genome sequencing, about 1.5 μg of genomic DNA was used for library construction with an insert size of about 350 bp. Pair-end sequencing were performed on Illumina NovaSeq platform, with the read length of 150 bp at each end. We then investigated the domestication and signatures of selection in these sheep breeds.Results: According to the population and demographic analyses, WRK and SNT populations were very similar, which were different from UJMQ populations. Genome wide association study identified 468 and 779 significant loci from SNT vs UJMQ, and UJMQ vs WRK, respectively. However, only 3 loci were identified from SNT vs WRK. Genomic comparison and selective sweep analysis among these sheep breeds suggested that genes associated with regulation of secretion, metabolic pathways including estrogen metabolism and amino acid metabolism, and neuron development have undergone strong selection during domestication.Conclusion: Our findings will facilitate the understanding of Chinese indigenous Mongolian sheep breeds domestication and selection for complex traits and provide a valuable genomic resource for future studies of sheep and other domestic animal breeding.       • Trusted Quantum Botnet Research based on Dual-Chord Protocol

        The controller in the traditional chord-based botnets needs to maintain the periodic communication among all the adjacent peerbots so that every peerbot could efficiently master the new routing information. Once some peerbot is captured by the anti-virus software and is treated as a honeypot, the security personnel can capture more peerbots by tracking the deployed honeypot when it updates the routing table so that the robustness of botnet would be affected. To solve the above-mentioned problem, the trusted chord-based quantum botnet is proposed that utilizes the quantum secure channel to achieve the updating of routing table, and introduces the anti-tracking network for capturing the deployed honeypots. The anlysis result showed that the robustness and stability of trusted chord-based quantum botnet could efficiently be improved.

        Single nucleotide polymorphism-based analysis of the genetic structure of the Min pig conserved population

        Objective: The study aims to uncover the genetic diversity and unique genetic structure of the Min pig conserved population, divide the nucleus conservation population, and construct the molecular pedigree. Methods: We used KPS Porcine Breeding Chip v1 50K for SNP detection of 94 samples (31♂, 63♀) in the Min pig conserved population from Lanxi breeding Farm. Results: The polymorphic marker ratio (PN), the observed heterozygosity (Ho), and the expected heterozygosity (He) were 0.663, 0.335, and 0.330, respectively. The pedigreebased inbreeding coefficients (FPED) was significantly different from those estimated from runs of homozygosity (FROH) and single nucleotide polymorphism (FSNP) based on genome. The Pearson correlation coefficient between FROH and FSNP was significant (p<0.05). The effective population content (Ne) showed a continuously decreasing trend. The rate of decline was the slowest from 200 to 50 generations ago (r = 0.95), then accelerated slightly from 50 to 5 generations ago (1.40<r<1.50) and increased significantly in the last 5 generations (r = 2.6). According to the composition of Chinese lineage, we separated the nucleus conservation population (81 individuals) and the candidate conservation population (13 individuals) of Min pig, then the nucleus conservation population of Min pig was divided into 9 families by genomic information matrix. Conclusion: Our study indicated that the genetic diversity of the Min pig conserved population was inadequate. Due to the introgression of European commercial pig breeds and the unscientific breeding process, it is necessary to construct the molecular pedigree of the nucleus conservation population for the Min pig.         Study of the Enhancement of Magnetic Properties of NdFeB Materials Fabricated by Modified HDDR Process

        The HDDR (Hydrogenation-Disproportionation-Desorption-Recombination) process is a special method to produce anisotropic NdFeB powders for bonded magnet. The effect of the modified HDDR process on magnetic properties of Nd₂Fe₁₄B-based magnet with several composition Nd_(11.2)Fe_(66.5-x)Co_(15.4)B_(6.8)Zr_(0.1)Ga_x (x = 0~1.0) and that of microelement Ga, disproportional temperature and annealing temperature on jHc, grain size were investigated in order to produce anisotropic powder with high magnetic properties. It was found that modified HDDR process is very effective to enhance magnetic properties and to fine grain size. The addition of Ga could change disproportionation character remarkably of the alloy and could improve magnetic properties of magnet powder. Increasing annealing temperature induces significant grain growth. And grain size produced by modified HDDR process is significantly smaller than those produced by conventional HDDR process.

      • Fusion of Infrared and Visible Image Based on Compressed Sensing and Nonsubsampled Shearlet Transform

        In order to solve storage and computation cost problems for the traditional whole sampling image fusion algorithms, a new method of infrared and visible light image fusion is put forward based on compressed sensing (CS) theory. Nonsubsampled shearlet transform (NSST) is introduced as the sparse transform. Compressed sensing is applied to fuse the high frequency subbands decomposed by NSST. The high frequency coefficients are compressed for measured values which are fused by the rules of spatial frequency weighting. Regional energy together with regional standard deviation guides the fusion of the low frequency subband. Finally, the fused image is gained through inverse NSST. The experimental results show that high-quality fused images can be obtained with only one layer NSST. The fused image quality is better than the several traditional fusion algorithms based on compressed sensing.

        Open source board based acoustofluidic transwells for reversible disruption of the blood–brain barrier for therapeutic delivery

        Background Blood–brain barrier (BBB) is a crucial but dynamic structure that functions as a gatekeeper for the central nervous system (CNS). Managing sufficient substances across the BBB is a major challenge, especially in the development of therapeutics for CNS disorders. Methods To achieve an efficient, fast and safe strategy for BBB opening, an acoustofluidic transwell (AFT) was developed for reversible disruption of the BBB. The proposed AFT was consisted of a transwell insert where the BBB model was established, and a surface acoustic wave (SAW) transducer realized using open-source electronics based on printed circuit board techniques. Results In the AFT device, the SAW produced acousto-mechanical stimulations to the BBB model resulting in decreased transendothelial electrical resistance in a dose dependent manner, indicating the disruption of the BBB. Moreover, SAW stimulation enhanced transendothelial permeability to sodium fluorescein and FITC-dextran with various molecular weight in the AFT device. Further study indicated BBB opening was mainly attributed to the apparent stretching of intercellular spaces. An in vivo study using a zebrafish model demonstrated SAW exposure promoted penetration of sodium fluorescein to the CNS. Conclusions In summary, AFT effectively disrupts the BBB under the SAW stimulation, which is promising as a new drug delivery methodology for neurodegenerative diseases.

        An integrated strategy for extraction and pre-concentration of four astragalosides from Radix Astragali by a formulated surfactant aqueous system

        Integrated and environmentally friendly methods for extraction and enrichment of natural products havegained increasing interests. The proposed preparation process provides the possibility that extractionand pre-concentration for astragalosides from Radix Astragali roots can be conducted with the surfactantaqueous system in one single procedure. The formulated surfactant system composing of 2% TX-114 assurfactant and 0.03% (w/v) of Gemini 16-5-16 as co-surfactant showed higher extraction efficiency withultrasonic-assisted method and four astragalosides could be enriched by the cloud point phenomenon ofsurfactant. This work represents a valuable alternative for the eco-friendly extraction and enrichment ofsecondary metabolites from plants.
