        Ordered Reverse k Nearest Neighbor Search via On-demand Broadcast

        The Reverse k Nearest Neighbor (RkNN) query is valuable for finding objects influenced by a specific object and is widely used in both scientific and commercial systems. However, the influence level of each object is unknown, information that is critical for some applications (e.g. target marketing). In this paper, we propose a new query type, Ordered Reverse k Nearest Neighbor (ORkNN), and make efforts to adapt it in an on-demand scenario. An Order-k Voronoi diagram based approach is used to answer ORkNN queries. In particular, for different values of k, we pre-construct only one Voronoi diagram. Algorithms on both the server and the clients are presented. We also present experimental results that suggest our proposed algorithms may have practical applications.

        DISTURBANCE ATTENUATION FOR A CLASS OF DISCRETE-TIME SWITCHED SYSTEMS WITH EXPONENTIAL UNCERTAINTY

        The disturbance attenuation problem for a class of discretetime switched linear systems with exponential uncertainties via switched state feedback and switched dynamic output feedback is investigated, respectively. By using Taylor series approximation and convex polytope technique, exponentially uncertain discrete-time switched linear system is transformed into an equivalent switched polytopic model with additive norm bounded uncertainty. For such equivalent switched model, one designs its switching strategy and associated state feedback controllers and dynamic output feedback controllers so that whole switched model is asymptotical stabilization with H-in nity disturbance attenuation base on switched Lyapunov function and LMI approach. Finally, two numerical examples are presented to illustrate our results.

        CircTRRAP (hsa_circ_0081234) participates in prostate cancer progression and glycolysis by HOXA1 via functioning as a miR‑515‑5p sponge

        Dysregulated circular RNAs (circRNAs) are implicated in prostate cancer (PCa) progression. Hsa_circ_0081234 (circTRRAP) has been revealed as a facilitator in PCa, but the mechanisms associated with circTRRAP in PCa progression are largely unclear. The present study was to explore the regulatory mechanism of circTRRAP-mediated PCa progression. A total of 50 PCa tissues and normal tissues were collected. RNA levels of circTRRAP, microRNA (miR)-515-5p and homeobox A1 (HOXA1) were detected by quantitative real-time polymerase chain reaction (qRT-PCR) or western blot. Cell viability, proliferation, migration, and invasion were estimated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide, 5-ethynyl-2ʹ-deoxyuridine (EdU) and transwell assays, respectively. Cell glycolysis was assessed by measuring glucose uptake and lactate production. The target interaction between circTRRAP or HOXA1 and miR-515-5p was investigated by the dual-luciferase reporter assay. We observed the overt upregulaiton of circTRRAP in PCa samples and cells. Silencing of circTRRAP lowered tumor growth in vivo and restrained PCa cell viability, proliferation, migration, invasion, and glycolysis in vitro. miR-515-5p was negatively regulated by circTRRAP and its deficiency reversed the inhibiting effects of circTRRAP knockdown on PCa cell malignancy and glycolysis. HOXA1 was confirmed as a miR-515-5p target and miR-515-5p overexpression lessened PCa cell malignancy and glycolysis by decreasing HOXA1 expression. Importantly, circTRRAP mediated HOXA1 expression by functioning as a miR-515-5p sponge. In conclusion, circTRRAP took part in PCa progression and glycolysis through mediating the miR-515-5p/HOXA1 axis, suggesting that circTRRAP can serve as a potential therapeutic target for PCa patients.

        Effect of electrochemical treatment on consolidation of soft clay

        In this study, a method of electrochemical consolidation is applied. This method utilizes electro-osmosis, which is an effective ground improvement technique for soft clays, and soil treatment using lime, which is the oldest traditional soil stabilizer. The mechanism of lime treatment for soil involves cation exchange, which leads to the flocculation and agglomeration. Five representative laboratory tests-an electro-osmotic test and four electrochemical tests with various proportions of lime-were performed on dredged marine clay. The objectives of this study are to investigate the effect of electrochemical treatment and to determine the optimum dose for optimal consolidation performance of dredged marine clay. The results show that a better consolidation effect was achieved in terms of current, temperature, and vane shear strength by using electrochemical treatment. The best results were observed for the electrochemical test using 4% lime content.

        Time Series Data Cleaning Method Based on Optimized ELM Prediction Constraints

        Affected by external factors, errors in time series data collected by sensors are common. Using the traditionalmethod of constraining the speed change rate to clean the errors can get good performance. However, they areonly limited to the data of stable changing speed because of fixed constraint rules. Actually, data with unevenchanging speed is common in practice. To solve this problem, an online cleaning algorithm for time series databased on dynamic speed change rate constraints is proposed in this paper. Since time series data usually changesperiodically, we use the extreme learning machine to learn the law of speed changes from past data and predictthe speed ranges that change over time to detect the data. In order to realize online data repair, a dual-windowmechanism is proposed to transform the global optimal into the local optimal, and the traditional minimumchange principle and median theorem are applied in the selection of the repair strategy. Aiming at the problemthat the repair method based on the minimum change principle cannot correct consecutive abnormal points,through quantitative analysis, it is believed that the repair strategy should be the boundary of the repaircandidate set. The experimental results obtained on the dataset show that the method proposed in this paper canget a better repair effect.

      • Discovery of the leinamycin family of natural products by mining actinobacterial genomes

        <P>Nature's ability to generate diverse natural products from simple building blocks has inspired combinatorial biosynthesis. The knowledge-based approach to combinatorial biosynthesis has allowed the production of designer analogs by rational metabolic pathway engineering. While successful, structural alterations are limited, with designer analogs often produced in compromised titers. The discovery-based approach to combinatorial biosynthesis complements the knowledge-based approach by exploring the vast combinatorial biosynthesis repertoire found in Nature. Here we showcase the discovery-based approach to combinatorial biosynthesis by targeting the domain of unknown function and cysteine lyase domain (DUF-SH) didomain, specific for sulfur incorporation from the leinamycin (LNM) biosynthetic machinery, to discover the LNM family of natural products. By mining bacterial genomes from public databases and the actinomycetes strain collection at The Scripps Research Institute, we discovered 49 potential producers that could be grouped into 18 distinct clades based on phylogenetic analysis of the DUF-SH didomains. Further analysis of the representative genomes from each of the clades identified 28 lnm-type gene clusters. Structural diversities encoded by the LNM-type biosynthetic machineries were predicted based on bioinformatics and confirmed by in vitro characterization of selected adenylation proteins and isolation and structural elucidation of the guangnanmycins and weishanmycins. These findings demonstrate the power of the discovery-based approach to combinatorial biosynthesis for natural product discovery and structural diversity and highlight Nature's rich biosynthetic repertoire. Comparative analysis of the LNM-type biosynthetic machineries provides outstanding opportunities to dissect Nature's biosynthetic strategies and apply these findings to combinatorial biosynthesis for natural product discovery and structural diversity.</P>

        The protective effect of procyanidinson tert-butyl hydroperoxide induced human hepatocyte HL7702 cell injury

        The present study aims to investigate the mechanism of the protective action of Procyanidins (PC) against tertiary butyl hydroperoxide (TBHP)-induced cytotoxicity and cell death in human hepatocyte-derived cell line HL7702 cells. Incubation of HL7702 cells with PC prevented TBHP-induced loss in cell viability and reduction in GSH/GSSG ratio and activities of antioxidant enzymes have also been found to be prevented by PC.Western blot was used to evaluate the levels of cytochrome c, Bcl-2, Bax, Sirt3 and IDH2. The results showed that PC effectively protect TBHP-treated HL7702 cells from death, and increase superoxide dismutase activity and significantly decrease malondialdehyde production. PC increased the ratio of GSH/GSSG and NADPH/NADP+, and reduced the release of cytochrome c from mitochondria and decreased the Bax expression, and also increased the expression of Bcl-2, IDH2 and Sirt3. PC prevented TBHP-induced oxidative stress injury in HL7702 cells and functioning via Sirt3-dependent mitochondria pathways.

      • Wind tunnel investigation on wind characteristics of flat and mountainous terrain

        Wind tunnel test is often adopted to assess the site-specific wind characteristics for the design of bridges as suggested by current design standards. To investigate the wind characteristics of flat and mountainous terrain, two topographic models are tested in a boundary layer wind tunnel. The wind characteristics, including the vertical and horizontal mean wind speed distributions, the turbulence intensity, and the wind power spectra, are presented. They are investigated intensively in present study with the discussions on the effect of wind direction and the effect of topography. It is indicated that for flat terrain, the wind direction has negligible effect on the wind characteristics, however, the assumption of a homogenous wind field for the mountainous terrain is not applicable. Further, the non-homogeneous wind field can be defined based on a proposed approach if the wind tunnel test or on-site measurement is performed. The calculated turbulence intensities and wind power spectra by using the measured wind speeds are also given. It is shown that for the mountainous terrain, engineers should take into account the variability of the wind characteristics for design considerations

        Identification of genes involved in inbreeding depression of reproduction in Langshan chickens

        Objective: Inbreeding depression of reproduction is a major concern in the conservation of native chicken genetic resources. Here, based on the successful development of strongly inbred (Sinb) and weakly inbred (Winb) Langshan chickens, we aimed to evaluate inbreeding effects on reproductive traits and identify candidate genes involved in inbreeding depression of reproduction in Langshan chickens. Methods: A two-sample t-test was performed to estimate the differences in phenotypic values of reproductive traits between Sinb and Winb chicken groups. Three healthy chickens with reproductive trait values around the group mean values were selected from each of the groups. Differences in ovarian and hypothalamus transcriptomes between the two groups of chickens were analyzed by RNA sequencing (RNA-Seq). Results: The Sinb chicken group showed an obvious inbreeding depression in reproduction, especially for traits of age at the first egg and egg number at 300 days (p<0.01). Furthermore, 68 and 618 differentially expressed genes (DEGs) were obtained in the hypothalamus and ovary between the two chicken groups, respectively. In the hypothalamus, DEGs were mainly enriched in the pathways related to vitamin metabolism, signal transduction and development of the reproductive system, such as the riboflavin metabolism, Wnt signaling pathway, extracellular matrix-receptor interaction and focal adhesion pathways, including stimulated by retinoic acid 6, serpin family F member 1, secreted frizzled related protein 2, Wnt family member 6, and frizzled class receptor 4 genes. In the ovary, DEGs were significantly enriched in pathways associated with basic metabolism, including amino acid metabolism, oxidative phosphorylation, and glycosaminoglycan degradation. A series of key DEGs involved in folate biosynthesis (gamma-glutamyl hydrolase, guanosine triphosphate cyclohydrolase 1), oocyte meiosis and ovarian function (cytoplasmic polyadenylation element binding protein 1, structural maintenance of chromosomes 1B, and speedy/RINGO cell cycle regulator family member A), spermatogenesis and male fertility (prostaglandin D2 synthase 21 kDa), Mov10 RISC complex RNA helicase like 1, and deuterosome assembly protein 1) were identified, and these may play important roles in inbreeding depression in reproduction. Conclusion: The results improve our understanding of the regulatory mechanisms underlying inbreeding depression in chicken reproduction and provide a theoretical basis for the conservation of species resources.
