      • W(110) 표면에 CO의 흡착

        The interaction of CO with W(110) surface was investigated through LEED, TDS, and photoelectron spectroscopy using synchrotron radiation under UHV condition. After CO saturation at RT, two desorption states, called α and β , were observed at about 400 and 1150 K in thermal desorption spectra, respectively. The kinetics of 3-CO followed the first order kinetics, indicating the existence of molecular CO on W(110) surface. This is contrary to the previous results. The O 1s BE(binding energy) of CO adsorbed on W(110) surface at room temperature was 529.9 eV. On the other hand, the O 1s BE of β-CO after heating to 900 K was different from that of oxygen adsorbed W(110) surface, suggesting a different adsorption state. According to the UP valence band spectra, we observed two peaks at near -10.7 eV (4σ) and -7.0 eV (5σ+1π), indicating the molecular adsorption of CO at room temperature. Furthermore we could see the 4σ peak at the various photon energy and elevated temperatures. Comparing the energy separation, Δ(4σ-1π) , between 4σ and 1π UP peaks of chemisorbed CO, we found that an increased separation reflects an decreased C-O bond strength. Therefore on the basis of TDS and photoelectron spectroscopy, we could suggest that β state of CO on W(110) may not be dissociated and has an adsorption geometry of lying-down mode.

      • FBW7 Upregulation Enhances Cisplatin Cytotoxicity in Non-small Cell Lung Cancer Cells

        Introduction: Lung cancer is extremely harmful to human health and has one of the highest worldwide incidences of all malignant tumors. Approximately 80% of lung cancers are classified as non-small cell lung cancers (NSCLCs). Cisplatin-based multidrug chemotherapy regimen is standard for such lesions, but drug resistance is an increasing problem. F-box/WD repeat-containing protein 7 (FBW7) is a member of the F-box protein family that regulates cell cycle progression, and cell growth and differentiation. FBW7 also functions as a tumor suppressor. Methods: We used cell viability assays, Western blotting, and immunofluorescence combined with siRNA interference or plasmid transfection to investigate the underlying mechanism of cisplatin resistance in NSCLC cells. Results: We found that FBW7 upregulation significantly increased cisplatin chemosensitivity and that cells expressing low levels of FBW7, such as NCI-H1299 cells, have a mesenchymal phenotype. Furthermore, siRNA-mediated silencing or plasmid-mediated upregulation of FBW7 resulted in altered epithelial-mesenchymal transition (EMT) patterns in NSCLC cells. These data support a role for FBW7 in regulating the EMT in NSCLC cells. Conclusion: FBW7 is a potential drug target for combating drug resistance and regulating the EMT in NSCLC cells.

      • Genetic Variants at 6p21.1 and 7p15.3 Identified by GWASs of Multiple Cancers and Ovarian Cancer Risk: a Case-control Study in Han Chinese Women

        A recent study summarized several published genome-wide association studies (GWASs) of cancer and reported two pleiotropic loci at 6p21.1 and 7p15.3 contributing to multiple cancers including lung cancer, noncardia gastric cancer (NCGC), and esophageal squamous-cell carcinoma (ESCC) in Han Chinese. However, it is not known whether such genetic variants have similar effects on the risk of gynecologic cancers, such as ovarian cancer. Hence, we explored associations between genetic variants in 6p21.1 and 7p15.3 and ovarian cancer risk in Han Chinese women. We performed an independent case-control study by genotyping the two loci (rs2494938 A > G at 6p21.1 and rs2285947 A > G at 7p15.3) in a total of 377 ovarian cancer cases and 1,034 cancer-free controls using TaqMan allelic discrimination assay. We found that rs2285947 at 7p15.3 was significantly associated with risk of ovarian cancer with per allele odds ratio (OR) of 1.33 [95% confidence interval (CI): 1.08-1.64, P=0.008]. However, no significant association was observed between rs2494938 and ovarian cancer risk. Our results showed that rs2285947 at 7p15.3 may also contribute to the development of ovarian cancer in Han Chinese women, further suggesting pleiotropy of 7p15.3 in multiple cancers.

        BMB Reports : Characterization of a novel posttranslational modification in polypyrimidine tract-binding proteins by SUMO1

        Polypyrimidine tract-binding protein 1 (PTBP1) and its brainspecific homologue, PTBP2, are associated with pre-mRNAs and influence pre-mRNA processing, as well as mRNA metabolism and transport. They play important roles in neural differentiation and glioma development. In our study, we detected the expression of the two proteins in glioma cells and predicted that they may be sumoylated using SUMOplot analyses. We confirmed that PTBP1 and PTBP2 can be modified by SUMO1 with co-immunoprecipitation experiments using 293ET cells transiently co-expressing SUMO1 and either PTBP1 or PTBP2. We also found that SUMO1 modification of PTBP2 was enhanced by Ubc9 (E2). The mutation of the sumoylation site (Lys137) of PTBP2 markedly inhibited its modification by SUMO1. Interestingly, in T98G glioma cells, the level of sumoylated PTBP2 was reduced compared to that of normal brain cells. Overall, this study shows that PTBP2 is posttranslationally modified by SUMO1.         Clinicopathologic and Prognostic Significance of the Zinc Finger of the Cerebellum Family in Invasive Breast Cancer

        Purpose: Five members of the zinc finger of the cerebellum (ZIC) family—ZIC1, ZIC2, ZIC3, ZIC4, and ZIC5—have been shown to be involved in various carcinomas. Here, we aimed to explore the clinicopathologic and prognostic roles of ZIC family members in invasive breast cancer patients using immunohistochemical analysis, western blotting analysis, and real-time quantitative polymerase chain reaction (RT-qPCR). Methods: A total of 241 female invasive breast cancer patients who underwent radical mastectomy between 2009 and 2011 were enrolled. ZIC proteins in 241 pairs of breast tumors and corresponding normal tissues were investigated using immunohistochemistry and the clinicopathologic roles of proteins were analyzed using Pearson’s chi-square test. Kaplan-Meier curves and Cox regression analysis were also used to analyze the prognostic value of the ZIC proteins. In addition, 12 pairs of fresh-frozen breast tumors and matched normal tissues were used in the western blotting analysis and RT-qPCR. Results: Only ZIC1 expression in normal tissues was obviously higher than that in tumors (p<0.001). On multivariate analysis, ZIC1 expression (in overall survival analysis: hazard ratio [HR], 0.405, 95% confidence interval [CI], 0.233–0.702, p=0.001; in disease-free survival analysis: HR, 0.395, 95% CI, 0.234–0.669, p=0.001) was identified as a prognostic indicator of invasive breast cancer. Conclusion: ZIC1, but not the other proteins, was obviously decreased in breast tumors and associated with clinicopathologic factors. Thus, ZIC1 might be a novel indicator to predict the overall and disease-free survival of invasive breast cancer patients.

        Recent advances in the food application of electrospun nanofibers

        Electrospinning (e-spinning) has excellent advantages in producing functional and versatile nanofibersand electrospun (e-spun) nanofibers have a variety of potential applications, including in food industry. This paper reviews the recent progress of e-spinning and food-related e-spun nanofibers, covering ediblenanofibers (natural materials and food ingredients), active food packaging (antibacterial packaging,antioxidant packaging, high temperature and humidity resistant packaging), food filtration membranes,and food sensors. Additionally, we discussed the new trend of combining nanomaterials with food andthe challenges of applying e-spinning in food science. The results indicated that e-spinning is a promisingand efficient alternative for the production of daily and functional food.

        Pentacyclic Triterpenoids from <i>Astilbe rivularis</i> that Enhance Glucose Uptake via the Activation of Akt and Erk1/2 in C2C12 Myotubes

        <P>Glucose uptake into insulin-sensitive tissues is important for the regulation of blood glucose. This study has investigated whether the pentacyclic triterpenoids substituted with a carboxylic acid at the C-27 position isolated from <I>Astilbe rivularis</I> can enhance glucose uptake and subsequently to also examine their underlying molecular mechanisms. The structure of the new pentacyclic triterpenoid <B>1</B> was assigned by spectroscopic data interpretation. To evaluate the activity of compounds <B>1</B> and <B>2</B>, glucose uptake and glucose transporter 4 (GLUT4) translocation were measured in C2C12 myotubes. The C-27-carboxylated triterpenoids <B>1</B> and <B>2</B> significantly increased basal and insulin-stimulated glucose uptake and GLUT4 translocation to plasma membrane. Both compounds stimulated the phosphorylation of insulin receptor substrate-1 (IRS-1), protein kinase B (Akt), and extracellular signal-regulated kinase 1/2 (Erk1/2). Pretreatment with the Akt inhibitor triciribine or the Erk1/2 inhibitor U0126 decreased the ability of both compounds to enhance basal- and insulin-stimulated glucose uptake and stimulate GLUT4 translocation. These results indicate that compounds <B>1</B> and <B>2</B> activated both the IRS-1/Akt and Erk1/2 pathways and subsequently stimulated GLUT4 translocation, leading to enhanced glucose uptake.         Study on Risk Analysis and Emergency Response of Problematic Vaccine Events in China

        In recent years, frequent vaccine safety events in China have become a serious public health problem. Especially in the new media era, vaccine safety events have received close attention from the public as well as the government. If they are not handled properly, many potential risk factors will be brought to social stability, the credibility and authority of the government will be affected. And many negative impacts will be brought to China national immunization program. In this paper, we summarized the inadequacies of vaccine event research, and analyzed the risk factors of vaccine in R&D, transportation, storage, vaccination and other parts. Based on the double perspectives of supervision and disposal, we constructed a risk analysis framework for vaccine safety events. Taking the 2016 Shandong Vaccine Event as an example, we analyzed the mechanism of the occurrence, development, evolution and termination of vaccine evens, and summarized the possible social risks caused by vaccine events. Taking the 2018 Changchun Changsheng Vaccine Event as an example, we analyzed the emergency response network attributes of vaccine event based on the complex network theory, including structural characteristics, organizational functions and overall performance. In view of the above research conclusions, we put forward countermeasures for vaccine events that are suitable for China’s national conditions, with a view to improving the risk control and emergency response capabilities of public health emergencies.
