        Gene Expression of Heart and Adipocyte Fatty Acid-binding Protein in Chickens by FQ-RT-PCR

        This study was to detect the expression of heart fatty acid-binding protein (H-FABP) and adipocyte fatty acid-binding protein (A-FABP) gene mRNA in different tissues of Rugao and Luyuan chickens at 56 d and 120 d by real-time fluorescence quantitative reverse transcription polymerase-chain reaction (FQ-RT-PCR). The primers were designed according to the sequences of HFABP, A-FABP and GAPDH genes in Gallus gallus, which were used as target genes and internal reference gene, respectively. The levels of H-FABP and A-FABP gene expression were detected by SYBR Green I FQ-RT-PCR. The relative H-FABP and A-FABP gene mRNA expression level was calculated with 2-$^{{\Delta}Ct}$. Melting curve analysis showed a single peak of three genes. Intramuscular fat (IMF) content in breast muscle and leg muscle of the two chicken breeds at 120 d was higher than at 56 d. IMF content in breast muscle and leg muscle at 56 d and 120 d in Luyuan was significantly higher than in Rugao, however, abdominal fat of Luyuan was significantly lower than that of Rugao. The relative H-FABP gene mRNA expression level in cardiac muscle was the highest in both chicken breeds. The relative H-FABP and A-FABP gene expression of different tissues in Luyuan was higher than in Rugao. H-FABP gene mRNA expression had a negative effect on IMF of leg and breast muscles, and was significantly negatively correlated with IMF content. The relative A-FABP gene mRNA level in abdominal fat was higher than in liver. The A-FABP gene mRNA was not expressed in leg, breast and cardiac muscles. A-FABP gene mRNA expression level was significantly positively correlated with abdominal fat and had a significant effect on abdominal fat but not IMF content.

        Gasification of rice husk in a cyclone gasifier

        The experimental results of air gasification of rice husk in the cyclone gasifier were presented at the fuel rate of 20.1 kg/h. With the equivalence ratios varied in the range of 0.21-0.32, the heating value of the producer gas decreases from 6.98 MJ/N㎥ to 3.11 MJ/N㎥ and the cold gas efficiency decreases from 64% to 31%. However, the tar content in the prouder gas decreases with the increase of the equivalence ratio. The rice husk and ash were examined under a scanning electron microscope (SEM) and energy dispersive X-ray (EDX) elemental analysis. The outer surface of the fuel particle which is of scale structure does not change basically during the gasification. The pyrolyzed gas is mainly released from the inner surface of the fuel particle.

        PIV measurement and numerical investigation on flow characteristics of simulated fast reactor fuel subassembly

        The flow characteristics of reactor fuel assembly always intrigue the designers and the experimentalists among the myriad phenomena that occur simultaneously in a nuclear core. In this work, the visual experimental method has been developed on the basis of refraction index matching (RIM) and particle image velocimetry (PIV) techniques to investigate the detailed flow characteristics in China fast reactor fuel subassembly. A 7-rod bundle of simulated fuel subassembly was fabricated for fine examination of flow characteristics in different subchannels. The experiments were performed at condition of Re=6500 (axial bulk velocity 1.6 m/s) and the fluid medium was maintained at 30℃ and 1.0 bar during operation. As for results, axial and lateral flow features were observed. It is shown that the spiral wire has an inhibitory effect on axial flow and significant intensity of lateral flow mixing effect is induced by the wire. The root mean square (RMS) of lateral velocity fluctuation was acquired after data processing, which indicates the strong turbulence characteristics in different flow subchannels.

        Identification of genes involved in inbreeding depression of reproduction in Langshan chickens

        Objective: Inbreeding depression of reproduction is a major concern in the conservation of native chicken genetic resources. Here, based on the successful development of strongly inbred (Sinb) and weakly inbred (Winb) Langshan chickens, we aimed to evaluate inbreeding effects on reproductive traits and identify candidate genes involved in inbreeding depression of reproduction in Langshan chickens. Methods: A two-sample t-test was performed to estimate the differences in phenotypic values of reproductive traits between Sinb and Winb chicken groups. Three healthy chickens with reproductive trait values around the group mean values were selected from each of the groups. Differences in ovarian and hypothalamus transcriptomes between the two groups of chickens were analyzed by RNA sequencing (RNA-Seq). Results: The Sinb chicken group showed an obvious inbreeding depression in reproduction, especially for traits of age at the first egg and egg number at 300 days (p<0.01). Furthermore, 68 and 618 differentially expressed genes (DEGs) were obtained in the hypothalamus and ovary between the two chicken groups, respectively. In the hypothalamus, DEGs were mainly enriched in the pathways related to vitamin metabolism, signal transduction and development of the reproductive system, such as the riboflavin metabolism, Wnt signaling pathway, extracellular matrix-receptor interaction and focal adhesion pathways, including stimulated by retinoic acid 6, serpin family F member 1, secreted frizzled related protein 2, Wnt family member 6, and frizzled class receptor 4 genes. In the ovary, DEGs were significantly enriched in pathways associated with basic metabolism, including amino acid metabolism, oxidative phosphorylation, and glycosaminoglycan degradation. A series of key DEGs involved in folate biosynthesis (gamma-glutamyl hydrolase, guanosine triphosphate cyclohydrolase 1), oocyte meiosis and ovarian function (cytoplasmic polyadenylation element binding protein 1, structural maintenance of chromosomes 1B, and speedy/RINGO cell cycle regulator family member A), spermatogenesis and male fertility (prostaglandin D2 synthase 21 kDa), Mov10 RISC complex RNA helicase like 1, and deuterosome assembly protein 1) were identified, and these may play important roles in inbreeding depression in reproduction. Conclusion: The results improve our understanding of the regulatory mechanisms underlying inbreeding depression in chicken reproduction and provide a theoretical basis for the conservation of species resources.
