        Associations of serotonergic genes with poststroke emotional incontinence

        <P><B>Objectives</B></P><P>Poststroke emotional incontinence (PSEI) has been associated with serotonergic dysfunction. Polymorphisms of serotonin transporter (5‐HTT) and serotonin 2a receptor (5‐HTR2a) genes may regulate serotonergic signaling at brain synapses, and this study was to investigate associations with PSEI in an East Asian population.</P><P><B>Methods</B></P><P>In 276 stroke cases, PSEI was diagnosed by Kim's criteria. Covariates included age, gender, education, history of depression or stroke, current depression, and stroke severity and location. Genotypes were ascertained for 5‐HTT gene‐linked promoter region (5‐HTTLPR), serotonin transporter intron 2 variable number tandem repeat, 5‐HTR2a 1438A/G, and 5‐HTR2a 102 T/C. Associations with PSEI were estimated by using logistic regression models, and gene–gene interactions were investigated by using the generalized multifactor dimensionality reduction method.</P><P><B>Results</B></P><P>PSEI was present in 37 (13.4%) patients. The 5‐HTT gene‐linked promoter region <I>s</I>/<I>s</I> genotype was independently associated with PSEI. No associations with STin2 VNTR and 5‐HTR2a genes were found, and no significant gene–gene interactions were identified.</P><P><B>Conclusions</B></P><P>Stroke patients with 5‐HTTLPR <I>s</I> allele had higher susceptibility to PSEI, which underlines the potential role of serotonergic pathways in its etiology.         예비유아교사 임용직전 교육프로그램 개발 및 교사역량에 미치는 효과

        본 연구는 예비유아교사가 정교사로 임용되기 직전 교사로서의 역량을 강화하지 위한 교육프로그램을 개발하고 이 프로그램이 예비유아교사의 교사역량에 미치는 효과를 살펴보고자 하는 데 그 목적이 있다 이러한 목적을 달성하기 위하여 설정한 연구문제는 다음과 같다. 첫째, 예비유아교사 임용직전 교육프로그램 구성은 어떠한가? 둘째, 예비유아교사 임용직전 교육프로그램이 교육신념, 전문적 자질, 교사효능감에 미치는 효과는 어떠한가? 교사역량을 갖춘 유야교사가 되기 위한 예비유아교사 임용직전교육프로그램의 개발은 문헌고찰, 예비유아교사와 현직유아교사의 인식 조사, 전문가 의견 조사를 거쳐 프로그램의 목표 및 교육내용, 교수-학습 방법을 구성하였다. 예비유아교사 대상 임용직전 교육프로그램의 효과를 살펴보기 위하여 대구시내에 소재한 B 전문대학 유아교육과 3학년 재학생 30명을 실험집단, 30명을 비교집단으로 구분하였다. 실험집단을 대상으로 주 3회, 총 12회기에 걸쳐 실험을 진행하였다. 수집된 자료는 SPSS 18.0 프로그램을 이용하여 실험집단과 비교집단의 사전검사, 사후검사, 추후검사의 평균과 표준편차를 산출하고 시기별 점수 차이가 있는지 알아보기 위하여 집단과 시기를 독립변인으로 한 반복측정 분산분석(repeated measures ANOVA)을 실시하였다. 본 연구의 결과는 다음과 같다. 첫째, 예비유아교사의 교사역량을 향상시키기 위한 임용직전 교육프로그램은 교육신념, 전문적 자질, 개인적 자질, 교사효능감 증진을 위한 12회기 프로그램으로 개발되었다. 교수-학습 내용 및 방법은 교사역할에 대한 자신감 및 구체적 기술을 다양한 활동과 반성적 과정을 통해 학습할 수 있도록 구성하였다. 둘째, 예비유아교사 임용직전 교육프로그램은 교육신념, 전문적 자질, 개인적 자질, 교사효능감 증진에 긍정적인 효과가 있다. 본 연구를 통하여 예비유아교사에게 필요한 현장적응 역량 강화교육의 중요성이 강조된다. The purpose of this study was to develop an educational program for pre-service early childhood teachers and to investigate the program`s effects on the teachers` teaching competence. The research questions were as follows. First, what is the purpose, contents, teaching-learning methods, and the assessment of the educational program for pre-service early childhood teachers? Second, what are the effects of the program on teaching beliefs, professional disposition, and teaching efficacy of the pre-service early childhood teachers? The educational program for pre-service early childhood teachers was constructed through literature reviews, surveys on the perceptions of in-service and pre-service teachers, and interviews of early childhood professionals to develop the purpose, the contents, the teaching-learning methods, and the assessment of the educational program. The effects of the programs were investigated by carrying out an experLn1ental study. A total of 30 pre-service early childhood teachers were assigned to the experimental group and 30 pre-service early childhood teachers were assigned to the comparative group. The experimental group participated in 3 sessions per week for a total of 12 sessions. The teaching beliefs was examined by Moon`s (1993) scale, the personal and the professional disposition by Ki`s (2008) scale, and the leaching efficacy by Kim`s (2000) scale. The collected data were analyzed using SPSS 18.0. A pre-test, a post-test, and an afterward-test were carried out to compare the means of the experimental and the comparative groups using a repeated measures ANDV A. The results of this study were as follows. First, the teaching competence of pre-service early childhood teachers were composed of the teaching beliefs, the personal and the professional disposition, and the teaching efficacy. The educational program for pre-service early childhood teachers was developed for 12 sessions to promote the teaching competence through reflective and hands-on activities. Second, the experimental group`s teaching beliefs, personal and professional disposition and disposition and teaching efficacy improved by participating in the educational program developed by this study. The results of this study emphasize the importance of educating the early childhood pre-service teachers to promote their practical knowledge applicable to their teaching profession.

      • Secretome analysis of differentially induced proteins in rice suspension-cultured cells triggered by rice blast fungus and elicitor

        <P>Secreted proteins were investigated in rice suspension-cultured cells treated with rice blast fungus Magnaporthe grisea and its elicitor using biochemical and 2-DE coupled with MS analyses followed by their in planta mRNA expression analysis. M. grisea and elicitor successfully interacted with suspension-cultured cells and prepared secreted proteins from these cultures were essentially intracellular proteins free. Comparative 2-D gel analyses identified 21 differential protein spots due to M. grisea and/or elicitor over control. MALDI-TOF-MS and μLC-ESI-MS/MS analyses of these protein spots revealed that most of assigned proteins were involved in defense such as nine chitinases, two germin A/oxalate oxidases, five domain unknown function 26 (DUF 26) secretory proteins, and β-expansin. One chitin binding chitinase protein was isolated using chitin binding beads and strong enzymatic activity was identified in an in-gel assay. Interestingly, their protein abundance correlated well at transcript levels in elicitor-treated cultures as judged by semi-quantitative RT-PCR. Each identified differentially expressed protein group was compared at transcript levels in rice leaves inoculated with incompatible (KJ401) and compatible (KJ301) races of M. grisea. Time-course profiling revealed their inductions were stronger and earlier in incompatible than compatible interactions. Identified secreted proteins and their expression correlation at transcript level in suspension-cultured cells and also in planta suggest that suspension-cultured cells can be useful to investigate the secretome of rice blast–pathogen interactions.</P>

        Therapeutic Effect of Total Ginseng Saponin on Skin Wound Healing

        In this study, we investigated the effects of total ginseng saponin (TGS) on the cutaneous wound healing process using histological analysis. A total of 24 ICR mice, 5-weeks-old, were used for all in vivo experiments. Mice were divided into control and TGS-treated groups and four equidistant 1-cm full-thickness dorsal incisional wounds were created. The wounds were extracted at days 1, 3, 5, and 7 post-injury for histomorphometrical analysis including wound area and contracture measurements, keratinocyte migration rate, and calculation of infiltrating inflammatory cells. The results showed that the wound area was smaller and keratinocyte migration rate was higher in the TGS-treated group than that of the control group from days 3 to 7. Inflammatory cells in the TGS-treated group at days 1 and 3 were reduced compared to the control group. Wound contraction in the TGS-treated group was greater than in the control group on days 3 to 5, and collagen deposition in the TGS-treated group was higher than in the control group during wound healing. The results indicate a beneficial effect of TGS when used to treat skin wounds.

        <P>We studied the role of the additives trehalose and poly(vinyl pyrrolidone) in the physical and pharmacokinetic properties of peptide drug incorporated hyaluronic acid microneedles. Poly(vinyl pyrrolidone) increases the mechanical strength of microneedles and ameliorates drug bioavailability <I>in vivo</I>, suggesting that poly(vinyl pyrrolidone) can be a promising additive in the fabrication of peptide drug-encapsulated fully dissolving microneedles.</P>

      • Fabrication of pre-alloyed Al–Li powders with high Li content via thermal dehydrogenation of LiH and rapid solidification process

        <P><B>Abstract</B></P> <P>Pre-alloyed Al–Li powders having high Li content were fabricated using thermal dehydrogenation of LiH. This, together with a rapid solidification process (RSP), led to strong suppression of the vaporization of Li. The uniform distribution of the Li present in the Al matrix was proved by confirming the random arrangement of δ'-AlLi precipitates seen in the microscopic images in the qualitative analysis. Spectroscopic analysis provided quantitative results for the concentration of Li. The dehydrogenation from LiH to Li was determined by H/O/N measurement, density reduction, and thermal analysis. Herein, we present that Al–Li powders with pre-determined 3.45wt.% Li, can be prepared via thermal dehydrogenation with 5.00wt.% LiH (4.37wt.% Li), and that the resulting material could be used in high-performance powder metallurgical (P/M) applications for aircraft and aerospace structures.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Al–Li alloy having high Li content is prepared via thermal decomposition of LiH. </LI> <LI> Fabrication of pre-alloyed Al–Li powders is accomplished using RSP. </LI> <LI> Arrangement of δ'-AlLi precipitates in Al–Li alloy is confirmed by SEM and TEM. </LI> <LI> Al–Li powders are characterized by microscopic, spectroscopic, and physical analyses. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

      • Triblock copolymer matrix-based capillary electrophoretic microdevice for high-resolution multiplex pathogen detection

        <P>Rapid and simple analysis for the multiple target pathogens is critical for patient management. CE-SSCP analysis on a microchip provides high speed, high sensitivity, and a portable genetic analysis platform in molecular diagnostic fields. The capability of separating ssDNA molecules in a capillary electrophoretic microchannel with high resolution is a critical issue to perform the precise interpretation in the electropherogram. In this study, we explored the potential of poly(ethyleneoxide)-poly(propyleneoxide)-poly(ethyleneoxide) (PEO-PPO-PEO) triblock copolymer as a sieving matrix for CE-SSCP analysis on a microdevice. To demonstrate the superior resolving power of PEO-PPO-PEO copolymers, 255-bp PCR amplicons obtained from 16S ribosomal RNA genes of four bacterial species, namely Proteus mirabilis, Haemophilus ducreyi, Pseudomonas aeruginosa, and Neisseria meningitidis, were analyzed in the PEO-PPO-PEO matrix in comparison with 5% linear polyacrylamide and commercial GeneScan™ gel. Due to enhanced dynamic coating and sieving ability, PEO-PPO-PEO copolymer displayed fourfold enhancement of resolving power in the CE-SSCP to separate same-sized DNA molecules. Fivefold input of genomic DNA of P. aeruginosa and/or N. meningitidis produced proportionally increased corresponding amplicon peaks, enabling correct quantitative analysis in the pathogen detection. Besides the high-resolution sieving capability, a facile loading and replenishment of gel in the microchannel due to thermally reversible gelation property makes PEO-PPO-PEO triblock copolymer an excellent matrix in the CE-SSCP analysis on the microdevice.</P>
