A loader mechanism mounted on tractor was modeled for kinematic analysis, and a computer simulation program was developed by using Cartesian coordinate approach, in order to remove the burden of routine derivation and computation from the engineer. The results of kinematic analysis was compared with those of a commercial CAD program. The results of this study was summarized as follows. 1. The results of the simulation showed good agreement with results of the commercial CAD program. This program could be used for predicting the kinematic properties of each links on the loader mechanism. 2. The velocity of bucket which affect the performance of loader increased very quickly just before the finishing point of boom operation, and acceleration also increased sharply at this point. Thus it is concluded that when bucket is rolling, maximum reaction force arises. 3. Velocity and acceleration of each links on the loader mechanism changed very quickly when the bucket cylinder started operation.

Sinus pericranii is a rare vascular anomaly characterized by abnormal venous communication between the inner and outer regions of the cranial cavity. Here, we report a case of sinus pericranii and venous malformations in the right periorbital region of a 2-year-old girl. Radiologic findings showed venous malformations in the right parietal region communicating with the superior sagittal sinus in the intracranial region. There were notable improvements following surgical resection for the abnormal venous lesions and several sclerotherapies. Presence of a bluish and pulsating mass on the scalp, which showed bruit on auscultation, may indicate sinus pericranii, which should be included in the differential diagnosis.

Background: The number of confirmed cases of individuals with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection increased rapidly due to the Omicron variant. Correctional facilities are vulnerable to infectious diseases, and they introduced rapid antigen tests (RATs) to allow for early detection and rapid response. We aimed to evaluate the diagnostic performance and usefulness of SARS-CoV-2 RATs in newly incarcerated people. Materials and Methods: We conducted a cross-sectional study at correctional facilities in Korea from 9 March to 22 May 2022. The study population was newly incarcerated people who were divided into two groups. In one group, 799 paired SARS-CoV-2 RATs and real-time polymerase chain reaction (RT-PCR) were conducted simultaneously in 522 individuals in March 2022. In the other group, 4,034 paired RATs and RT-PCR consecutively in 4,034 participants; only individuals with negative RATs results underwent RT-PCR from April to May 2022. We analyzed data using descriptive statistics and a logistic regression model. Results: Among the 799 specimens in March, RT-PCR was positive in 72 (9.0%), and among the 4,034 specimens in April - May 2022, RT-PCR was positive in 40 (1.0%). Overall, the RATs had a sensitivity of 58.3%, specificity of 100.0%, positive predictive value (PPV) of 100.0%, and negative predictive value (NPV) of 96.0%. Asymptomatic individuals constituted 98.2% of the study group, and symptomatic individuals 1.8%. In asymptomatic cases, the sensitivity of RATs was 52.5%, specificity was 100.0%, PPV was 100.0%, and NPV was 96.3%. In symptomatic cases, the sensitivity of RATs was 84.6%, specificity was 100.0%, PPV was 100.0%, and NPV was 33.3%. Sensitivity (P = 0.034) and NPV (P = 0.004) differed significantly according to the presence and absence of symptoms, and the F1 score was the highest at 0.9 in symptomatic individuals in March. There was a positive linear trend in the proportion of false-negative RATs in newly incarcerated people following the weekly incidence of SARS-CoV-2 (P = 0.033). The best-associated predictors of RATs for SARS-CoV-2 infection involved symptoms, timing of sample collection, and repeat testing. Conclusion: Sensitivity and NPV significantly depend on whether symptoms are present, and the percentage of false negatives is correlated with the incidence. Thus, using RATs should be adjusted according to the presence or absence of symptoms and the incidence of SARS-CoV-2 infection in the community. RATs could be a useful screening tool as an effective first-line countermeasure because they can rapidly identify infectious patients and minimize SARS-CoV-2 transmission in correctional facilities.

<P><B>Background</B></P><P>Curcumin, a yellow ingredient of turmeric (<I>Curcuma longa</I> Linn, Zingiberaceae), has long been used in traditional folk medicine in the management of inflammatory disorders. Although curcumin has been reported to inhibit experimentally-induced colitis and carcinogenesis, the underlying molecular mechanisms remain largely unresolved.</P><P><B>Methods</B></P><P>Murine colitis was induced by dextran sulfate sodium (DSS) which mimics inflammatory bowel disease. Curcumin or tetrahydrocurcumin was given orally (0.1 or 0.25 mmol/kg body weight daily) for 7 days before and together with DSS administration (3% in tap water). Collected colon tissue was used for histologic and biochemical analyses.</P><P><B>Results</B></P><P>Administration of curcumin significantly attenuated the severity of DSS-induced colitis and the activation of NF-κB and STAT3 as well as expression of COX-2 and inducible nitric oxide synthase. In contrast to curcumin, its non-electrophilic analogue, tetrahydrocurcumin has much weaker inhibitory effects.</P><P><B>Conclusions</B></P><P>Intragastric administration of curcumin inhibited the experimentally induced murine colitis, which was associated with inhibition of pro-inflammatory signaling mediated by NF-κB and STAT3.</P>
Improving Culture Performance and Antibody Titer in CHO Cell Fed-batch Culture by Controlling pH

Mammalian cell culture is most common industrial process for the production of recombinant proteins. However, higher cell density and titer are still limited due to factors such as excessive metabolites production, nutrients limitation, and high osmolality. Lactate is the major product from glucose metabolism, and lactate accumulation has been demonstrated to inhibit growth rate and negatively affect antibody production in mammalian cells. To partly overcome this factor, we investigated a simple method using pH control in Chinese hamster ovary (CHO) cells. We performed CHO-DG44 fed-batch cultures under pH conditions (6.9 and 7.0) using rocking bioreactor. As a result, cells grown under low pH condition produced lactate approximately 70% lower than cells grown under high pH condition. We also found that peak viable cell density (PVCD) and final antibody titer were increased in fed-batch culture under low pH condition.
