Beneficial Effects of Cynaroside on Cisplatin-Induced Kidney Injury In Vitro and In Vivo

Anti-cancer drugs such as cisplatin and doxorubicin are effectively used more than radiotherapy. Cisplatin is a chemotherapeutic drug, used for treatment of various forms of cancer. However, it has side effects such as ototoxicity and nephrotoxicity. Cisplatin-induced nephrotoxicity increases tubular damage and renal dysfunction. Consequently, we investigated the beneficial effect of cynaroside on cisplatin-induced kidney injury using HK-2 cell (human proximal tubule cell line) and an animal model. Results indicated that 10 μM cynaroside diminished cisplatin-induced apoptosis, mitochondrial dysfunction and caspase-3 activation, cisplatin-induced upregulation of caspase-3/MST-1 pathway decreased by treatment of cynaroside in HK-2 cells. To confirm the effect of cynaroside on cisplatin-induced kidney injury in vivo, we used cisplatin exposure animal model (20 mg/kg, balb/c mice, i.p., once a day for 3 days). Renal dysfunction, tubular damage and neutrophilia induced by cisplatin injection were decreased by cynaroside (10 mg/kg, i.p., once a day for 3 days). Results indicated that cynaroside decreased cisplatin-induced kidney injury in vitro and in vivo, and it could be used for improving cisplatin-induced side effects. However, further experiments are required regarding toxicity by high dose cynaroside and caspase-3/MST-1-linked signal transduction in the animal model.

BACKGROUND: Chemical fungicides not only may pollute the ecosystem but also can be environmentally hazardous, as the chemicals accumulate in soil. Biological control is a frequently-used environment-friendly alternative to chemical pesticides in phytopathogen management. However, the use of microbial products as fungicides has limitations. This study isolated and characterized a three-antifungal-enzyme (chitinase, cellulase, and ${\beta}$-1,3-glucanase)-producing bacterium, and examined the conditions required to optimize the production of the antifungal enzymes. METHOD AND RESULTS: The antifungal enzymes chitinase, cellulase, and ${\beta}$-1,3-glucanase were produced by bacteria isolated from an sawmill in Korea. Based on the 16S ribosomal DNA sequence analysis, the bacterial strain AM50 was identical to Streptomyces sp. And their antifungal activity was optimized when Streptomyces sp. AM50 was grown aerobically in a medium composed of 0.4% chitin, 0.4% starch, 0.2% ammonium sulfate, 0.11% $Na_2HPO_4$, 0.07% $KH_2PO_4$, 0.0001% $MgSO_4$, and 0.0001% $MnSO_4$ at $30^{\circ}C$. A culture broth of Streptomyces sp. AM50 showed antifungal activity towards the hyphae of plant pathogenic fungi, including hyphae swelling and lysis in P. capsici, factors that may contribute to its suppression of plant pathogenic fungi. CONCLUSION(S): This study demonstrated the multiantifungal enzyme production by Streptomyces sp. AM50 for the biological control of major plant pathogens. Further studies will investigate the synergistic effect, to the growth regulations by biogenic amines and antifungal enzyme gene promoter.

<P>An acinar morphogenesis inhibitor named fusarisetin A (<B>1</B>) that possesses both an unprecedented carbon skeleton and a new pentacyclic ring system has been identified from an in-house fractionated fungal library using a three-dimensional matrigel-induced acinar morphogenesis assay system. Dual deep neural network-based classifiers to detect experimental seizures

Manually reviewing electroencephalograms (EEGs) is labor-intensive and demands automated seizure detection systems. To construct an efficient and robust event detector for experimental seizures from continuous EEG monitoring, we combined spectral analysis and deep neural networks. A deep neural network was trained to discriminate periodograms of 5-sec EEG segments from annotated convulsive seizures and the pre- and post-EEG segments. To use the entire EEG for training, a second network was trained with non-seizure EEGs that were misclassified as seizures by the first network. By sequentially applying the dual deep neural networks and simple pre- and post-processing, our autodetector identified all seizure events in 4,272 h of test EEG traces, with only 6 false positive events, corresponding to 100% sensitivity and 98% positive predictive value. Moreover, with pre-processing to reduce the computational burden, scanning and classifying 8,977 h of training and test EEG datasets took only 2.28 h with a personal computer. These results demonstrate that combining a basic feature extractor with dual deep neural networks and rule-based pre- and post-processing can detect convulsive seizures with great accuracy and low computational burden, highlighting the feasibility of our automated seizure detection algorithm.

While use of mass rearing systems improved poultry production, chances of exposing to contagious diseases have been increased, making flocks more vulnerable to diseases. Diseases of interest which affects egg production adversely include Low pathogenic avian influenza (LPAI), Infectious bronchitis (IB), Avian meta-pneumoviral infection (aMPV) and Egg drop syndrome'76 (EDS'76). This report collected and analyzed 5,385 serum samples, which were collected from 1,330 different chicken flock, provided by Chungbuk National University, Avian Disease Laboratory at 2009. Serums were analyzed based on rearing stages; 0~1.3weeks (wks) (maternal antibody period), >1.3~3 wks (starting period), >3~10 wks (growing period), >10~22 wks (developing period), >22~40 wks (peak laying period), >40~60 wks (late laying period) and over 60 wks (post-molting period). Results showed the 99.7% of the tested flocks were immunized against ND and73.8%, 97.1%, 78,2% and 78% of the flocks were immunized against other 4 agents (LPAI, IB, EDS'76, aMPV). Maternal antibody was transferred to enough quantity for NDV. Generally, antibody titers which were developed at 22 weeks were stabilized permanently for life. In case of IB and aMPV, infection titer emerged as early as 10 weeks and the titer was increased from 99.4% to 100% for life. EDS76 showed increase in titers, reflecting decreased frequency of vaccination programs. Overall, this study displayed general trends of major viral disease in layers, but considering the trend of development of preventive measures and evolution of pathogens, conducting serological surveys on a regular basis is important.
