      • Expression of High Mobility Group Box - B1 (HMGB-1) and Matrix Metalloproteinase-9 (MMP-9) in Non-small Cell Lung Cancer (NSCLC)

        Objective: This study evaluated the expression level of high mobility group box-B1 (HMGB-1) and matrix metalloproteinase-9 (MMP-9) in non-small cell lung cancer (NSCLC) inmorder to reveal any relation with development and prognosis. Methods: NSCLC and normal tissues were selected from 30 patients at age of 30-73, and used for RT-PCR and Western blot analyses of HMGB-1. A total of 100 paraffin embedded NSCLC tissues were also isolated from patients through surgical resection, and used for detection of HMGB-1 by immunohistochemistry. In addition, 50 samples were also applied for MMP-9 detection, and 30 normal tissues were considered as controls. Correlation analysis of HMGB-1 and MMP-9 was carried out by Pearsons correlation coefficient. Results: The average expression level of HMGB-1 in NSCLC patients was significantly higher than in normal lung tissues. In addition, patients in III-IV period exhibit significantly higher positive rate of HMGB-1 when compared with I-II period cases. Furthermore, a positive correlation with HMGB-1 was found in the expression of MPP-9. Conclusion: HMGB-1 was highly expressed in NSCLC, which may become a prognostic and predictive marker for NSCLC. Besides, MPP-9 was positively correlated with HMGB-1.

        An Effective Process for Removing Organic Compounds from Oily Sludge

        Wet air oxidation (WAO) of oily sludge was carried out using $Fe^{3+}$ as catalyst, placed in a 0.5 L batch autoclave in the temperature range of $250-330^{\circ}C$. Experiments were conducted to investigate the effects of temperature, the initial COD, reaction time, concentration of catalyst and $O_2$ excess (OE) on the oxidation of the oily sludge. The results showed that in the WAO 88.4% COD was achieved after 9 min reaction at temperature of $330^{\circ}C$, OE of 0.8 and the initial COD of 20000 mg/L. Temperature was found to have a significant impact on the oxidation of oily sludge. Adding a catalyst significantly improved the COD removal. Homogenous catalyst, $Fe^{3+}$, showed effective removal for pollutants. COD removal was 99.7% in the catalytic wet air oxidation (CWAO) over $Fe^{3+}$ catalyst. The results proved that the CWAO was an effective pretreatment method for the oily sludge.

        An effective process for removing organic compounds from oily sludge using soluble metallic salt

        This study evaluates the application of wet oxidation for the treatment of oily sludge by using hydrogen peroxide as oxidant instead of air at high temperature in a batch reactor. The experimental results show that the quantity of oxidant with 250% to theoretical oxygen demand of oily sludge is comparatively effective and economica1. Adding a catalyst, Fe3+, significantly improved the COD removal. Reaction mechanism for WO of oily sludge is analyzed. Oxidation of intermediate for oily sludge is a rate-limiting step for further oxidation of oily sludge.

        LGR4 protects PC12 against OGD/R-induced oxidative stress and apoptosis through activation of AKT/GSK3β

        Background Leucine-rich repeat-containing G protein-coupled receptor 4 (LGR4) suppresses hypoxia/re-oxygenationinduced injury in hepatocytes, and protects against hepatic and myocardial ischemia/reperfusion (I/R)-induced injury. The role of LGR4 on cerebral I/R injury was investigated in this study. Objective Providing potential therapeutic target of cerebral I/R injury. Results Expression of LGR4 was reduced in OGD/R-induced PC12. Over-expression of LGR4 attenuated OGD/R-induced decrease of cell viability and increase of apoptosis. Increased levels of LDH, ROS, MDA, and MPO, as well as decreased SOD, in OGD/R-induced PC12, were restored by LGR4 over-expression. Over-expression of LGR4 counteracted with the suppressive effects of OGD/R on protein expression of AKT and GSK3β phosphorylation in PC12. Conclusion The protective effect of LGR4 against OGD/R-induced oxidative stress and apoptosis in PC12 was mediated by activation of AKT/GSK3β pathway.

        Progress on the Preparation of Ultra-pure Hydrogen Peroxide

        Progress on the preparation of ultra-pure hydrogen peroxide is reviewed. The technologies include distillation and relative technologies, combination methods of ion exchange and adsorption resin, membrane separation and relative technologies, crystallization, flocculation, and adding addition agents as so well. Distillation is reliable and easy to industrialize, while purity of hydrogen peroxide distilled is not high enough. Crystallization, flocculation, and adding addition agents can only be used as accessory technologies. It is safe for membrane separation, but the life of membrane is short. Ion exchange and adsorption resin processes are simple and the product purity is high, but ion exchange resin is easy to be oxidized. It is the tendency for the preparation of ultra-pure hydrogen peroxide that distillation is used firstly and then the combination of membrane separation technologies or ion exchange and adsorption resin processes are followed.

        Knockdown of EMMPRIN (OX47) in MRMT-1 Carcinoma Cells Inhibits Tumor Growth and Decreases Cancer-Induced Bone Destruction and Pain

        Purpose Bone destruction and pain caused by cancer is one of the most devastating complications of cancer patients with bone metastases, and it seriously affects the quality of patients’ life. Extracellular matrix metalloproteinase inducer (EMMPRIN) is a cell adhesion molecule with increased expression in a variety of tumors. This study focused to clarify the specific function of EMMPRIN in bone metastasis of breast cancer.Materials and Methods Adenovirus with shRNA-EMMPRIN was transfected into MRMT-1 rat breast carcinoma cells, and the MRMT-1 cells with different expression levels of EMMPRIN were implanted into the bone marrow cavity of rat tibia. Next, the effect of down-regulation of EMMPRIN was evaluated as follows: bone damage was detected by X-ray radiological and tartrate-resistant acid phosphatase staining; the tumor burden was evaluated by hematoxylin and eosin staining; the test of pain-related behaviors was assessed used the bilateral paw withdrawal mechanical threshold; and the levels of secretory factors in tumor conditioned medium were determined by using enzyme-linked immunosorbent assay.ResultsWe found that down-regulation of EMMPRIN in tumor cells can simultaneously reduce tumor burden, relieve cancer-induced bone destruction and pain. ConclusionMaterials and Methods EMMPRIN is expected to be a therapeutic target for relieving bone metastasis of breast cancer and alleviating cancerinduced bone destruction and pain. The method of targeting EMMPRIN may be a promising strategy for the treatment of cancer in the future.

        Entangling gates for trapped-ion quantum computation and quantum simulation

        The trapped-ion system has been a leading platform for practical quantum computation and quantum simulation since the frst scheme of a quantum gate was proposed by Cirac and Zoller (Phys Rev Lett 74:4091, 1995). Quantum gates with trapped ions have shown the highest fdelity among all physical platforms. Recently, sophisticated schemes of quantum gates such as amplitude, phase, frequency modulation, or multi-frequency application, have been developed to make the gates fast, robust to many types of imperfections, and applicable to multiple qubits. Here, we review the basic principle and recent development of quantum gates with trapped ions.

      • MUTYH Association with Esophageal Adenocarcinoma in a Han Chinese Population

        Adenocarcinoma of esophagus (AE) is a complex disease, affected by a variety of genetic and environmental factors. Much evidence has shown that the MutY glycosylase homologue (MUTYH) plays a key role in the pathogenesis of many cancers. However, there have been no reports on influence on AE in the Han Chinese population. The objective of this study was to investigate this issue. A gene-based association study was conducted using three single nucleotide polymorphisms(SNPs) reported in previous studies. The three SNPs (rs3219463, rs3219472, rs3219489) were genotyped in 207 unrelated AE patients and 249 healthy controls in a case-control study using polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP). The results revealed that the genotype distribution of rs3219472 differed between the case and control groups (OR=1.66,95%CI=1.11-2.48, P=0.012), indicating that an association may exist between MUTYH and AE. These findings support a signifcant role for MUTYH in AE pathogenesis in the Han Chinese population.

        Identification of a novel promoter region responsible for the embryo-specific expression of SERK1 in pineapple

        Somatic embryogenesis (SE) is a key regeneration process in plant. AcSERK1 is a gene specifi cally expressed in the early stage of SE in pineapple ( Ananas comosus ), suggesting that the promoter of SERK1 might contain specifi c cis acting element regulating SE. To identify embryonic cell-specifi c element in the SERK1 promoter, a series of binary plant transformation vectors with GUS (β-glucuronidase) reporter gene were systematically analyzed by transient gene expression system in wild-type and transgenic pineapple embryogenic callus. Histochemical and quantitative GUS assays demonstrated that the activity of the AcSERK1 upstream regulatory sequence lacking − 921 to -911 or -910 to -880 was signifi cantly reduced in the embryonic callus of the pineapple, and these two regions were needed for the embryonic cell-specifi c. Besides, a promoter lacking − 943 to -922 was shown to signifi cantly increase GUS activity in embryogenic callus, suggesting repressive elements exist in this region. Our data of stable transformation assays confi rmed again the 5’ upstream regulatory sequence (-921 to -880) of the AcSERK1 gene is an essential functional region. Our fi ndings lay the basis for better understanding of the molecular mechanisms of AcSERK1 gene in the regulation in early stage of SE.

      • Regulation Roles of MICA and NKG2D in Human Renal Cancer Cells

        Objective: Our aim was to investigation the roles of MHC class I chain-related gene A(MICA) and natural killer cell group 2D(NKG2D) in human renal cancer cells. Materials and Methods: The expression of membrane MICA (mMICA) on renal cells and NKG2D on NK cells were detected by flow cytometry (FCM); the content of sMICA were detected by enzyme linked immunosorbent assay (ELISA) and the distribution of mMICA on renal tumor tissues by immunohistochemistry; the interaction between MICA and NKG2D was observed by antibody closed method. Results: Our results showed that the expression of mMICA in renal cancer tissues was significantly higher than in controls, where the soluble MICA was not expressed. Cytotoxic activity of NK cells was significantly reduced after exposure to NKG2D and MICA antibodies (P<0.05), and serum containing sMICA can obviously lower the function of NKG2D (P<0.05). Conclusions: The interaction of mMICA and NKG2D play important roles in mediation of cytotoxicity of NK cells in RCC. On the other hand, sMICA may mediate tumor immune escape through down- regulated NKG2D expression.
