        AZ91 합금융체내의 Fe 용해도

        Fe solubility in AZ91 alloy melts was measured by a new method designed for removal of Al_(8)(Mn,Fe)_(5) inclusions. In the AZ91 alloy melts, Al_(8)(Mn,Fe)_(5) precipitates were formed due to supersaturated Mn and Fe, but all precipitates were not settled down from the melts and hence part of them was included in the samples for measuring Fe solubility. This means that the solubility measured from these samples is higher than the true solubility. In the present study, it was shown that these Al_(8)(Mn,Fe)_(5) inclusions could be separated from the samples by the new method. As a result, the Fe solubility measured by the present method was lower than the previous solubility data. Amount of the unsettled Al_(8)(Mn,Fe)_(5) inclusions was estimated to be 59-148 ppm. It was also observed that Fe contents in Al_(8)(Mn,Fe)_(5) were varied with the Fe and Mn content in AZ91 alloy melts.

      • SIRT1 is required for oncogenic transformation of neural stem cells and for the survival of “cancer cells with neural stemness” in a p53-dependent manner

        <P><B>Background</B></P><P>Cancer stemness, observed in several types of glioma stem cells (GSCs), has been demonstrated to be an important barrier for efficient cancer therapy. We have previously reported that cancerous neural stem cells (F3.Ras.CNSCs), derived from immortalized human neural stem cells by a single oncogenic stimulation, form glial tumors in vivo.</P><P><B>Method</B></P><P>We searched for a commonly expressed stress modulator in both F3.Ras.CNSCs and GSCs and identified silent mating type information regulation 2, homolog (SIRT1) as a key factor in maintaining cancer stemness.</P><P><B>Result</B></P><P>We demonstrate that the expression of SIRT1, expressed in “cancer cells with neural stemness,” is critical not only for the maintenance of stem cells, but also for oncogenic transformation. Interestingly, SIRT1 is essential for the survival and tumorigenicity of F3.Ras.CNSCs and GSCs but not for the U87 glioma cell line.</P><P><B>Conclusion</B></P><P>These results indicate that expression of SIRT1 in cancer cells with neural stemness plays an important role in suppressing p53-dependent tumor surveillance, the abrogation of which may be responsible not only for inducing oncogenic transformation but also for retaining the neural cancer stemness of the cells, suggesting that SIRT1 may be a putative therapeutic target in GSCs.</P>

        Acute oral toxicity of the ethyl acetate fraction of <i>Orostachys japonicus</i> in mice

        <P><I>Context</I>: <I>Orostachys japonicus</I> (Crassulaceae) is referred to as Wa-song in Korea. It is used as an anti-inflammatory, antifebrile, hemostatic, and anti cancer agent, and as an antidote.</P><P><I>Objective</I>: The purpose of this study was to evaluate the acute toxicity of the ethyl acetate fraction of <I>O. japonicus</I> (OJE) after the oral administration in Balb/c mice of both sexes.</P><P><I>Materials and methods</I>: Mice were oral administered a single doses of 500, 1000, and 2000 mg/kg of body weight and were monitored for 14 d. Biochemical parameters [aspartate amino transferase (AST), alanine amino transferase (ALT), alkaline phosphatase (ALP), total protein (TP), globulin (GB), total cholesterol (TC), triglyceride (TG), blood urea nitrogen (BUN), and creatinine (CR)] and histopathological examination of liver were performed.</P><P><I>Results and conclusion</I>: No animals died and no toxic changes were observed in clinical signs, body weight, and organ weight. The LD<SUB>50</SUB> of orally administered OJE was higher than 2000 mg/kg/d in both sexes. No toxicological findings were found in biochemical parameters. In histophathological examination, neutrophilic infiltration was observed at a dose of 2000 mg/kg group in both sexes. These finding suggest that oral administration of OJE does not produce acute toxicity. Therefore, these results could provide satisfactory preclinical evidence of safety to launch clinical trials on standardized formulation of OJE to be a biohealth product.</P>

        Borderline Personality Pathology in Major Depressive Disorder, Bipolar I and II Disorder, and Its Relationship With Childhood Trauma

        Objective Mood disorder and borderline personality pathology (BPP) are frequently comorbid and relate to childhood trauma. We investigated the relationship between childhood trauma and BPP features in mood disorder patients versus controls.Methods A total of 488 mood disorder patients, particularly major depressive disorder (MDD), bipolar I disorder (BD I), and bipolar II disorder (BD II), and 734 controls were included. We examined between-group BPP-related differences and correlated between BPP and childhood trauma using the Childhood Trauma Questionnaire-Short Form (CTQ) and the Personality Assessment Inventory–Borderline Features Scale.Results BD II patients showed significantly higher BPP. Emotional abuse and neglect were prominently associated with BPP, while affective instability and negative relationships exhibited a stronger association with childhood trauma. We also found a positive relationship between childhood trauma and BPP in MDD, BD I, and BD II patients.Conclusion The findings of the present study imply that BPP features are more likely to be found in patients with BD II than BD I or MDD. Mood disorder patients with severe childhood trauma may have higher BPP features. Thus, further study of the relationship between childhood trauma and BPP features could improve the therapeutic approaches and help understand patients with mood disorders.

        A Repeated-dose Oral Toxicity Study of Orostachys japonicus Extract in Sprague-Dawley Rats

        A 12-week repeated-dose oral toxicity study of water-soluble Orostachys japonicus extract (WOJ) was performed in Sprague-Dawley (SD) rats of both genders. Each group of ten rats was orally administered in doses of either 0 or 250 mg/day over a 12-week period. As a result, no WOJ-related changes were observed in terms of survival rate, clinical signs, body weight, or food intake. In addition, no difference in organ weight between the control and treated groups was detected. Furthermore, serum biochemistry parameters revealed some changes within normal ranges although significant decreases in total-bilirubin in the females. In spite of some alterations in serum biochemistry, the clinical signs, body weight changes from food intake, and autoptical remarks indicated that WOJ was not toxic. This study suggests that repeated treatment of O. japonicus very low toxicity and the NOAEL (no observed adverse effect dose) of WOJ exceeds 250 mg/kg in the SD rats.

        목적 :공공보건의료의 최일선인 보건소는 코로나19 팬더믹 상황에서 선도적인 역할을 하였다. 장기화되는 코로나19로 인한 의료인의 업무 과중, 스트레스에 대한 연구는 국내외에서 활발히 이루어졌으나, 보건소 공무원들에 대한 관심과 연구는 부족하다. 본 연구는 코로나19 관련 업무를 담당한 보건소 공무원의 어려움과 경험에 대해 조사하였다. 방법 : 2020년 10월부터 2021년 4월까지 코로나19 관련 업무를 담당한 보건소 공무원 14명을 대상으로 반 구조화된 질문을 사용하여 대면 인터뷰를 시행하였다. 합의된 질적 연구 방식을 통해 인터뷰 내용을 분석하였다. 결과 : 보건소 공무원들은 업무량 증가, 인력 부족, 불균형적 업무 분장 등의 어려움을 겪었다. 휴일 없이 지속되는 업무에 수면부족, 피로 등 신체 건강 관련 문제가 나타났다. 또한 코로나19의 장기화로 인해 무력감, 사회생활 위축 등에서 오는 정신적 스트레스를 가지고 있었다. 보건소 공무원들은 업무 보람, 직원 간 유대 형성 및 인적 인프라 증가의 긍정적 영향을 언급하였다. 개선점으로는 보건소가 감염병에 대한 전문적 역량을 갖추고, 보건소 공무원에게 적절한 보상과 인정이 필요하다고 하였다. 결론 :코로나19 관련 업무를 담당한 보건소 공무원들은 상당한 스트레스를 받고 있었다. 소진 고위험군 직원들을 조기에 발견하여 개입하고, 회복탄력성과 관련된 훈련 프로그램을 정기적으로 시행해야 한다. 또한 방역의 최일선에서 근무하는 공무원들에 대한 사회적 지지와 인정이 필요하다. Objectives: Public health centers have played a critical role during the COVID-19 pandemic. The workload and stress experienced by medical personnel because of the prolonged COVID-19 have been actively studied; however, interest in and research on public health center officials is lacking. This study investigated the challenges and experiences of public health center officials in charge of COVID-19-related tasks. Methods: Face-to-face interviews were conducted between October 2020 and April 2021 using semi-structured questions with 14 public health center officials in charge of COVID-19-related tasks. A standardized qualitative research methodology was used to analyze the data from the interviews. Results: Public health center officials faced several challenges, such as increased workload, shortage of manpower, and unbalanced division of duties. Physical health issues, such as sleep deprivation and fatigue, manifested when work continued without breaks. Additionally, the prolonged COVID-19 pandemic resulted in mental stress brought on by feelings of helplessness and atrophy in social life. Public health officials spoke about the positive impact of rewarding work, creating bonds among coworkers, and increasing human infrastructure. Conclusion: Health center officials in charge of COVID-19-related work were under considerable stress. Early intervention for employees identified as at high risk of burnout is crucial, along with regular implementation of training programs for resilience. Furthermore, frontline public servants at the forefront of quarantine need social support and recognition.

        Determination of sugar phosphates by high-performance anion-exchange chromatography coupled with pulsed amperometric detection

        <P><B>Abstract</B></P><P>We have developed an improved analytical method for the determination of sugar phosphates using sodium carbonate (Na<SUB>2</SUB>CO<SUB>3</SUB>) for high-performance anion-exchange chromatography–pulsed amperometric detection. The target analytes were separated completely within 10min using eluent containing 20mM NaOH and 35mM Na<SUB>2</SUB>CO<SUB>3</SUB>. The limit of detection (S/N=3) and quantitation (S/N=10) for analytes were 10–30ng/mL and 35–100ng/mL, respectively. Linear dynamic range was 1–30μg/mL (<I>r</I><SUP>2</SUP>≥0.9998). The RSDs for intra- and inter-day assays were found to be of satisfactory results (0.23–3.09%), and the recoveries from blood spots were 97.62–99.69%.</P>

        A Study on Occupational Stress and Coping, Turnover, Knowledge and Practice of Infection Control in Dental Hygienists of COVID-19

        Background: The importance of infection with COVID-19 is being emphasized in dentistry with high risks such as aerosols. The purpose of this study is to investigate the knowledge and practice of infection control, stress and coping, and turnover of dental hygienists. Methods: Questionnaire was conducted knowledge and practice of infection control, occupational stress and coping, turnover. Survey data was investigated about 149 dental hygienists from February to March 2021 Data were analyzed t-test, ANOVA, Pearson’s correlation using statistical programs of PASW Statistics ver. 21.0. Results: Regarding occupational stress, relationship conflict was higher in the group with less than 2 years of experience (p<0.05). Job anxiety, organizational system, inadequate compensation, and workplace culture were highly surveyed in the 3 to 5 year of experience. The group with more than 6 years of experience had the highest perception of lack of job autonomy (p<0.05). The group with higher knowledge of infection control had lower mean inappropriate rewards and stress (p<0.05). The group with high infection control performance had a lower average in items such as job instability, organizational system, inadequate compensation, workplace culture, and stress. And problem-focused coping ability was found to be high (p<0.05). Infection control knowledge and performance were positively correlated (r=0.251, p<0.01), infection control practice and stress were negatively correlated (r=-0.264, p<0.01), and stress and emotional coping were positively correlated (r=0.367, p<0.01). Stress was positively correlated with turnover rate (r=0.549, p<0.01). Conclusion: Infection control training was required to reduce occupational stress. Occupational stress was highly correlated with turnover, a holistic and systemic organizational operation and improvement of the quality of medical care were required to reduce stress.
